Genetic Variants In Immune Related Genes As Predictors Of Responsiveness To Bcg Immunotherapy In Metastatic Melanoma Patients., Romela Irene Ramos, Misa A Shaw, Leland Foshag, Stacey L Stern, Negin Rahimzadeh, David Elashoff, Dave Hoon

Articles, Abstracts, and Reports

Adjuvant immunotherapy in melanoma patients improves clinical outcomes. However, success is unpredictable due to inherited heterogeneity of immune responses. Inherent immune genes associated with single nucleotide polymorphisms (SNPs) may influence anti-tumor immune responses. We assessed the predictive ability of 26 immune-gene SNPs genomic panels for a clinical response to adjuvant BCG (Bacillus Calmette-Guérin) immunotherapy, using melanoma patient cohorts derived from three phase III multicenter clinical trials: AJCC (American Joint Committee on Cancer) stage IV patients given adjuvant BCG (pilot cohort; n = 92), AJCC stage III patients given adjuvant BCG (verification cohort; n = 269), and …

Kimmel Cancer Center Faculty Papers

Non-alcoholic fatty liver disease (NAFLD) has become the most common chronic liver disease worldwide. Its incidence has grown alongside the increasing global prevalence of type 2 diabetes, obesity, and metabolic syndrome. The risk of progression to hepatocellular carcinoma for nonalcoholic steatohepatitis patients over 5 years is 8%, and despite targeted and immunotherapy treatment advances, HCC maintains a bleak 5-year survival of 19%. NAFLD’s primary risk factors are components of metabolic syndrome as well as possible sleep disturbances. NAFLD is most common among men 50-60 years of age, though incidence in women catches up after menopause. In the US, Hispanics are …

Sensitivity Analysis Of An Agent-Based Simulation Model Using Reconstructability Analysis, Andey M. Nunes, Martin Zwick, Wayne Wakeland

Systems Science Faculty Publications and Presentations

Reconstructability analysis, a methodology based on information theory and graph theory, was used to perform a sensitivity analysis of an agent-based model. The NetLogo BehaviorSpace tool was employed to do a full 2k factorial parameter sweep on Uri Wilensky’s Wealth Distribution NetLogo model, to which a Gini-coefficient convergence condition was added. The analysis identified the most influential predictors (parameters and their interactions) of the Gini coefficient wealth inequality outcome. Implications of this type of analysis for building and testing agent-based simulation models are discussed.

Climatic Drivers Of Verticillium Dahliae Occurrence In Mediterranean Olive-Growing Areas Of Southern Spain, Juan M. Requena-Mullor, Jose Manuel García-Garrido, Pedro Antonio García, Estefanía Rodríguez

Biology Faculty Publications and Presentations

Verticillium wilt, caused by the soil-borne fungus Verticillium dahliae, is one of the most harmful diseases in Mediterranean olive-growing areas. Although, the effects of both soil temperature and moisture on V. dahliae are well known, there is scant knowledge about what climatic drivers affect the occurrence of the pathogen on a large scale. Here, we investigate what climatic drivers determine V. dahliae occurrence in olive-growing areas in southern Spain. In order to bridge this gap in knowledge, a large-scale field survey was carried out to collect data on the occurrence of V. dahliae in 779 olive groves in Granada …

Malignant Eccrine Adenoma With Sarcomatous (Heterologous) Components: Report Of A Rare Skin Adnexal Neoplasm With Literature Review, Hira Ishtiaq, Muhammad Abdulwaasey, Muhammad Usman Tariq, Saira Fatima

Department of Pathology and Laboratory Medicine

Malignant eccrine spiradenoma (MES) is an exceedingly rare skin adnexal tumor that arises from pre-existing benign eccrine spiradenoma (BES). MES tumors show a wide spectrum of morphological features, posing a diagnostic challenge to the pathologist. Sarcomatous (heterologous) elements are seen in a few of these tumors, further complicating the morphological picture. We herein describe a case of a 66-year-old male who presented with a recently enlarging, ulcerated, nodular skin lesion over the right leg that had been present for the last 25 years. The patient underwent wide local excision of the tumor. Microscopic examination revealed a neoplastic lesion comprising benign …

The pelagic Gulf of Mexico (GoM) is a complex system of dynamic physical oceanography (western boundary current, mesoscale eddies), high biological diversity, and community integration via diel vertical migration and lateral advection. Humans also heavily utilize this system, including its deep-sea components, for resource extraction, shipping, tourism, and other commercial activity. This utilization has had impacts, some with disastrous consequences. The Deepwater Horizon oil spill (DWHOS) occurred at a depth of ∼1500 m (Macondo wellhead), creating a persistent and toxic mixture of hydrocarbons and dispersant in the deep-pelagic (water column below 200 m depth) habitat. In order to assess the …

Faculty Research 2020

Studies of the severely pancytopenic scat mouse model first demonstrated the crucial role of RASA3, a dual RAS and RAP GTPase activating protein (GAP), in hematopoiesis. RASA3 is required for survival in utero; germline deletion is lethal at E12.5-13.5 due to severe hemorrhage. Here, conditional deletion in hematopoietic stem and progenitor cells (HSPCs) using Vav-iCre recapitulates the null phenotype demonstrating that RASA3 is required at the stem and progenitor level to maintain blood vessel development and integrity and effective blood production. In adults, bone marrow blood cell production and spleen stress erythropoiesis are suppressed significantly upon induction of RASA3 deficiency, …

The genetic landscape of diseases associated with changes in bone mineral density (BMD), such as osteoporosis, is only partially understood. Here, we explored data from 3,823 mutant mouse strains for BMD, a measure that is frequently altered in a range of bone pathologies, including osteoporosis. A total of 200 genes were found to significantly affect BMD. This pool of BMD genes comprised 141 genes with previously unknown functions in bone biology and was complementary to pools derived from recent human studies. Nineteen of the 141 genes also caused skeletal abnormalities. Examination of the BMD genes in osteoclasts and osteoblasts underscored …

Molecular Signatures Of Sexual Communication In The Phlebotomine Sand Flies, Paul V. Hickner, Nataliya Timoshevskaya, Ronald J. Nowling, Frédéric Labbé, Andrew D. Nguyen, Mary Ann Mcdowell, Carolina N. Spiegel, Zainulabeuddin Syed Dec 2020

Molecular Signatures Of Sexual Communication In The Phlebotomine Sand Flies, Paul V. Hickner, Nataliya Timoshevskaya, Ronald J. Nowling, Frédéric Labbé, Andrew D. Nguyen, Mary Ann Mcdowell, Carolina N. Spiegel, Zainulabeuddin Syed

Biology Faculty Publications

Phlebotomine sand flies employ an elaborate system of pheromone communication wherein males produce pheromones that attract other males to leks (thus acting as an aggregation pheromone) and females to the lekking males (sex pheromone). In addition, the type of pheromone produced varies among populations. Despite the numerous studies on sand fly chemical communication, little is known of their chemosensory genome. Chemoreceptors interact with chemicals in an organism’s environment to elicit essential behaviors such as the identification of suitable mates and food sources. Thus, they play important roles during adaptation and speciation. Major chemoreceptor gene families, odorant receptors (ORs), gustatory receptors …

Diverse And Complex Muscle Spindle Afferent Firing Properties Emerge From Multiscale Muscle Mechanics, Kyle P. Blum, Kenneth S. Campbell, Brian C. Horslen, Paul Nardelli, Stephen N. Housley, Timothy C. Cope, Lena H. Ting

Physiology Faculty Publications

Despite decades of research, we lack a mechanistic framework capable of predicting how movement-related signals are transformed into the diversity of muscle spindle afferent firing patterns observed experimentally, particularly in naturalistic behaviors. Here, a biophysical model demonstrates that well-known firing characteristics of mammalian muscle spindle Ia afferents – including movement history dependence, and nonlinear scaling with muscle stretch velocity – emerge from first principles of muscle contractile mechanics. Further, mechanical interactions of the muscle spindle with muscle-tendon dynamics reveal how motor commands to the muscle (alpha drive) versus muscle spindle (gamma drive) can cause highly variable and complex activity during …

Key Interplay Between The Co-Opted Sorting Nexin-Bar Proteins And Pi3p Phosphoinositide In The Formation Of The Tombusvirus Replicase, Zhike Feng, Nikolay Kovalev, Peter D. Nagy

Plant Pathology Faculty Publications

Positive-strand RNA viruses replicate in host cells by forming large viral replication organelles, which harbor numerous membrane-bound viral replicase complexes (VRCs). In spite of its essential role in viral replication, the biogenesis of the VRCs is not fully understood. The authors identified critical roles of cellular membrane-shaping proteins and PI(3)P (phosphatidylinositol 3-phosphate) phosphoinositide, a minor lipid with key functions in endosomal vesicle trafficking and autophagosome biogenesis, in VRC formation for tomato bushy stunt virus (TBSV). The authors show that TBSV co-opts the endosomal SNX-BAR (sorting nexin with Bin/Amphiphysin/Rvs- BAR domain) proteins, which bind to PI(3)P and have membrane-reshaping function during …

Early Evidence Of Natal-Habitat Preference: Juvenile Loons Feed On Natal-Like Lakes After Fledging, Brian A. Hoover, Kristin M. Brunk, Gabriella L. Jukkala, Nathan Banfield, Andrew L. Rypel, Walter H. Piper

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

Many species show natal habitat preference induction (NHPI), a behavior in which young adults select habitats similar to those in which they were raised. However, we know little about how NHPI develops in natural systems. Here, we tested for NHPI in juvenile common loons (Gavia immer) that foraged on lakes in the vicinity of their natal lake after fledging. Juveniles visited lakes similar in pH to their natal lakes, and this significant effect persisted after controlling for spatial autocorrelation. On the other hand, juveniles showed no preference for foraging lakes of similar size to their natal one. When …

Evidence For Phytoremediation And Phytoexcretion Of Nto From Industrial Wastewater By Vetiver Grass, Abhishek Roychowdhury, Pallabi Mukherjee, Saumik Panja, Rupali Datta, Christos Christodoulatos, Dibyendu Sarkar

Michigan Tech Publications

The use of insensitive munitions such as 3-nitro-1,2,4-triazol-5-one (NTO) is rapidly increasing and is expected to replace conventional munitions in the near future. Various NTO treatment technologies are being developed for the treatment of wastewater from industrial munition facilities. This is the first study to explore the potential phytoremediation of industrial NTO-wastewater using vetiver grass (Chrysopogon zizanioides L.). Here, we present evidence that vetiver can effectively remove NTO from wastewater, and also translocated NTO from root to shoot. NTO was phytotoxic and resulted in a loss of plant biomass and chlorophyll. The metabolomic analysis showed significant differences between treated and …
