How Perception Of Decision Environment And Future Information Affects Changes In Delay Discounting Rates: Differences Across U.S. And China, Differences Before And After The U.S. 2018 Midterm Elections, Fran Walsh Oct 2019

How Perception Of Decision Environment And Future Information Affects Changes In Delay Discounting Rates: Differences Across U.S. And China, Differences Before And After The U.S. 2018 Midterm Elections, Fran Walsh

Masters Theses

In this thesis, I will explore the idea that choices between present, smaller value options and future, larger value options depend on how much individuals trust the future to deliver the reward. Due to this aspect of trust, the individual must build their estimate of trust based on information for their present environment and their future expectations. This estimate of future trust can change across different time points in the same environment (i.e., before and after a national election) and between environments in the same time point (i.e., between two countries experiencing different economic rates of change). In this set …

Characterizing The Interaction Between Non-Pathogenic Fusarium Oxysporum And Arabidopsis Thaliana To Determine Beneficial Effects Conferred To The Model Plant Host, Kathryn Isabelle Vescio Oct 2019

Characterizing The Interaction Between Non-Pathogenic Fusarium Oxysporum And Arabidopsis Thaliana To Determine Beneficial Effects Conferred To The Model Plant Host, Kathryn Isabelle Vescio

Masters Theses

Fusarium oxysporum (Fo) is a soil-borne fungal pathogen that causes vascular wilt disease on a broad range of plants, including agricultural crops and the model plant Arabidopsis thaliana. There are non-pathogenic members of the Fo species complex that confer defense benefits against other pathogens to the host plant, however alteration to the host’s physiology through interaction with one of these strains, Fo47, have not been described. In this study, we aimed to establish the Fo47-A. thaliana interaction and determine if Fo47 reduces disease severity of a pathogenic Fo isolate, Fo5176. Additionally, we sought to use bioinformatics to mine …

The Effects Of Acidosis On Calcium Dependent Binding Of A Single Crossbridge, Matthew Unger Oct 2019

The Effects Of Acidosis On Calcium Dependent Binding Of A Single Crossbridge, Matthew Unger

Masters Theses

Intracellular acidosis is a putative agent of skeletal muscle fatigue, in part, because acidosis depresses the calcium (Ca2+) sensitivity and force production of muscle (18, 50). However, the molecular mechanisms behind this depression in Ca2+ sensitivity and force production are unknown. This gap in knowledge poses a significant challenge in generating a complete understanding of the fatigue process. To close this gap, the ability of myosin to bind to a single actin filament was measured under acidic conditions, in a laser trap assay, with and without regulatory proteins. Decreasing pH from 7.4 to 6.5 reduced the frequency …

A Novel Infield Metagenomic Approach To Evaluating Surface Water Quality In Lake Warner, Brooke Stebbins Oct 2019

A Novel Infield Metagenomic Approach To Evaluating Surface Water Quality In Lake Warner, Brooke Stebbins

Masters Theses

In January 2010, a magnitude 7.0 earthquake devastated Haiti, one of the poorest countries in the Western Hemisphere. Haiti’s weak sanitation infrastructure and limitations in the public health system made the country susceptible to the spread of waterborne diseases. Following the earthquake, cholera rapidly spread through Haiti, killing 4,672 people in 5 months and leaving thousands hospitalized (MSNBC, 2010). Cholera is an infectious diarrheal disease caused by the pathogen, Vibrio cholerae, which results in severe dehydration with a high mortality risk. The source of the epidemic was traced to the Artibonite River, the island’s longest and most essential drinking …

Arboriculture Safety Around The World, Jamie Lim Oct 2019

Arboriculture Safety Around The World, Jamie Lim

Masters Theses

Arboricultural work is inherently dangerous, with more serious injuries and fatalities than most other professions. Safety standards exist in some jurisdictions, but it is unclear how many standards exist, how they compare to one another, and whether (and how many) jurisdictions share standards. To establish a baseline understanding of these issues, my objectives were to (i) develop a database of existing standards, (ii) identify the most frequently occurring safety topics and (iii) describe similarities and differences in safety topics among standards from different countries. I worked with a variety of contacts and traditional university library resources to identify, obtain, analyze, …

Application Of Flow Cytometry As Novel Technology In Studying Lipid Oxidation In Oil-In-Water Emulsions, Peilong Li Oct 2019

Application Of Flow Cytometry As Novel Technology In Studying Lipid Oxidation In Oil-In-Water Emulsions, Peilong Li

Masters Theses

The body of literature on the impact of emulsion particle size on oxidation rates is unclear. This could be because emulsions are typically polydisperse and the oxidation rate of individual droplets is impossible to discern. Flow cytometry is a technique for studying individual cells and their subpopulations using fluorescence technologies. It is possible that individual emulsion droplets could also be characterized by flow cytometry as a novel approach for studying lipid oxidation. Typical emulsion droplets are too small to be visualized by flow cytometer, so emulsions were prepared to have droplets > 2 μm; weighting agent and xanthan gum were added …

The Efficacy Of Atp Monitoring Devices At Measuring Organic Matter On Postharvest Surfaces, Kristin Lane Oct 2019

The Efficacy Of Atp Monitoring Devices At Measuring Organic Matter On Postharvest Surfaces, Kristin Lane

Masters Theses

The Food Safety Modernization Act (FSMA), specifically the Produce Safety Rule (PSR), requires growers to clean and sanitize food-contact surfaces to protect against produce contamination. The ATP monitoring device is a potential sanitation tool to monitor the efficacy of an on-farm cleaning and sanitation program that could help growers meet regulatory expectations mandated by PSR. The ATP device uses bioluminescence to detect all ATP (found in bacteria and produce matter cells) from a swabbed surface. Little work has been done to test the efficacy of these tools under postharvest conditions. The present study evaluated ATP measurement for postharvest surface cleanliness …

Patterns Of Morphological Plasticity In Metriaclima Zebra And Danio Rerio Suggest Differently Canalized Phenotypes Due To Form-Function Relationships, Dylan Jockel Oct 2019

Patterns Of Morphological Plasticity In Metriaclima Zebra And Danio Rerio Suggest Differently Canalized Phenotypes Due To Form-Function Relationships, Dylan Jockel

Masters Theses

In order to ascertain the degree of compatibility in developmental restructuring and behavioral plasticity between two fish species frequently made subject of laboratory research (Metriaclima zebra & Danio rerio), alternative trophic niche exposure experiments utilizing novel three-prong feeding treatments were conducted to obtain morphometric data, which demonstrated both species do bear some degree of plasticity. The results are somewhat complicated by differences in locality of detectable restructuring, which may be due to disparity in the form-function relationship for each species’ lineage. Each is notable in the manner of respective species’ jaw protrusion, as it is driven by anterior …

Markov Decision Process Approach To Strategize National Breast Cancer Screening Policy In Data-Limited Settings, Vijeta Deshpande Oct 2019

Markov Decision Process Approach To Strategize National Breast Cancer Screening Policy In Data-Limited Settings, Vijeta Deshpande

Masters Theses

Early diagnosis is a promising strategy to reduce premature mortalities and for optimal use of resources. But the absence of mathematical models specific to the data settings in LMIC’s impedes the construction of economic analysis necessary for decision-makers in the development of cancer control programs. This thesis presents a new methodology for parameterizing the natural history model of breast cancer based on data availabilities in low and middle income countries, and formulation of a control optimization problem to find the optimal screening schedule for mammography screening, solved using dynamic programming. As harms and benefits are known to increase with the …

Sex Specific Electrophysiology Of Aromatase Neurons In The Medial Amygdala, Marcelo Henrique Correia Oct 2019

Sex Specific Electrophysiology Of Aromatase Neurons In The Medial Amygdala, Marcelo Henrique Correia

Masters Theses

The medial amygdala (MeA) is a central node in the interwoven circuits that regulate social behavior based on pheromones. Aromatase-expressing (arom+) neurons in the MeA are key for the establishment and maintenance of sex differences. Here, we characterized the intrinsic electrophysiological properties of arom+ neurons and non-aromatase (arom-) neurons in the MeA of male and female mice. Most electrophysiological properties were similar for arom+ neurons in the MeA between sexes, but the relative refractory period was twice as large in female mice. We also show that the firing pattern and firing frequency is markedly …

Optical Meets Mechanical: Use Of Luminescence Spectroscopy To Assess Ageing In Biodegradable Films, Louis Colaruotolo Oct 2019

Optical Meets Mechanical: Use Of Luminescence Spectroscopy To Assess Ageing In Biodegradable Films, Louis Colaruotolo

Masters Theses

With the growing concern of the accumulation of plastic-based food packaging waste, the search for bio-based biodegradable packages is on the rise. These materials differ from their petro-based counterparts in their degradation rates, which are much higher in the former. Not only do bio-based biodegradable materials degrade faster during post-usage processes but also they age faster during usage and storage, which affects their performance and functionality. The application of noninvasive testing methods with the capability to report on the matrix’s state could assist in the development of a more ubiquitous way to assess ageing in food packaging, particularly in biodegradable …

Development Of Colloids-Based Food Delivery Systems To Enhance The Beneficial Effects Of Nutraceuticals For Human Health, Zipei Zhang Aug 2019

Development Of Colloids-Based Food Delivery Systems To Enhance The Beneficial Effects Of Nutraceuticals For Human Health, Zipei Zhang

Doctoral Dissertations

In current studies, hydrogel particles were formed by polysaccharides (i.e., alginate or pectin) and protein (i.e., casein or gelatin) through complex coacervation. Our results indicated that the encapsulation of protein or polyunsaturated lipid droplets nanoparticles within the hydrogel particles could improve their chemical or physical stability during storage. The lipid droplets encapsulated within the hydrogel particles could be released under simulated oral conditions, which was triggered by a pH or temperature change. The current study further fabricated the hydrogel particles (beads) using alginate or carrageenan based on an injection-gelation method. We found carrageenan beads had a relatively fragile structure that …

Sleep-Dependent Consolidation Of Locomotor Sequence Learning And Locomotor Adaptation, Gabriela Borin Castillo Jul 2019

Sleep-Dependent Consolidation Of Locomotor Sequence Learning And Locomotor Adaptation, Gabriela Borin Castillo

Doctoral Dissertations

Walking is a complex task that requires precise coordination of many muscles and joints. The nervous system must continually learn how to control gait patterns as changes occur to the body (e.g., injury and fatigue) or environment (e.g., slippery floor). Motor learning refers to processes that improve the spatial and/or temporal accuracy of a movement through motor practice. Although additional hours of practice can improve motor skill performance (online learning), time without additional practice (offline learning) can further enhance motor learning. Consolidation refers to the process by which motor (procedural) memory becomes more robust and stable after the end of …

Determining Kinetic Parameters Of Thiamine Degradation In Three Nasa Spaceflight Foods In Thermal Processing And Long-Term Storage And Methods For Analyzing Microstructure And Precipitant Development In Real And Model Wines, Timothy R. Goulette

Doctoral Dissertations

The current NASA spaceflight food program encompasses a wide variety of commercial and custom-made edible products. These foods are designed to ensure each crewmember receives the Recommended Dietary Allowance (RDA) of vitamins and minerals to maintain adequate health during the entire spaceflight mission. With the desire for long-duration and exploration-class missions, such as the impending mission to Mars, the retention of important nutrients such as labile vitamins in spaceflight foods is critically important. For this reason, multiple studies were conducted to determine the vitamin degradation behavior of thiamine (vitamin B1) within three spaceflight foods which demonstrated both significant …

Physical And Biological Drivers Of Juvenile Fish Distributions In Unstructured Shallow Tropical Nearshore Habitats, Christopher Haak Jul 2019

Physical And Biological Drivers Of Juvenile Fish Distributions In Unstructured Shallow Tropical Nearshore Habitats, Christopher Haak

Doctoral Dissertations

The structural complexity of aquatic habitats can influence the ecological processes that occur within them, as fine-scale topographic features act as refugia for small fishes, buffering the effects of environmental stressors. Accordingly, the habitat requirements of juvenile demersal fishes in shallow littoral zones are often defined by their associations with distinct benthic microhabitats, such as densely vegetated substrates. However, an array of ecologically-important juvenile fishes also associate with topographically-homogeneous, sparsely-vegetated substrata. Absent the benefits offered by structural refugia, such fishes may be more affected by environmental variability and may have evolved distinct strategies for coping with stressors. I examined this …

Electrophysiological Correlates Of Natural Language Processing In Children And Adults, Margaret Ugolini Jul 2019

Electrophysiological Correlates Of Natural Language Processing In Children And Adults, Margaret Ugolini

Doctoral Dissertations

To understand the causes of differences in language ability we must measure the specific and separable processes that contribute to natural language comprehension. Specifically, we need measures of the three language subsystems – semantics, syntax, and phonology – as they are used during the comprehension of real speech. Event-Related Potentials (ERPs) are a promising approach to reaching this level of specificity. Previous research has identified distinct ERP effects for each of the subsystems – the N400 to semantic anomalies, the Anterior Negativity and P600 to syntactic anomalies, and the Phonological Mapping Negativity to unexpected speech sounds. However, these studies typically …

The Prevention Of Obesity-Associated Colorectal Cancer Via Dietary Suppression Of Inflammation-Driven Wnt-Signaling, Jinchao Li Jul 2019

The Prevention Of Obesity-Associated Colorectal Cancer Via Dietary Suppression Of Inflammation-Driven Wnt-Signaling, Jinchao Li

Doctoral Dissertations

Colorectal cancer (CRC) is the third leading cause of cancer deaths in the United States. A number of population studies have established that modifiable lifestyle factors such as obesity plays an important role in colorectal carcinogenesis. In the United States, more than one-third of adults are obese and obesity prevalence rates have no sign of decrease. Therefore, the development of effective strategies to prevent obesity-induced CRC is a public health priority. This study aimed to investigate whether genetic or dietary strategies can prevent obesity-induced CRC and determine the potential molecular mechanisms underlying the prevention effects of these strategies. We used …

Walking For Object Transport: An Examination Of The Coordinative Adaptations To Locomotor, Perceptual, And Manual Task Constraints, Avelino Amado Jul 2019

Walking For Object Transport: An Examination Of The Coordinative Adaptations To Locomotor, Perceptual, And Manual Task Constraints, Avelino Amado

Doctoral Dissertations

The goal of this dissertation was to understand how the intrinsic dynamics of gait adapt to support the performance of an ecologically relevant object transport task. A common object transport task is walking with a cup of water. Because the water can move relatively independent of the cup, the cup and water system is classified as a complex object. To model this task participants carried a cup with a wooden lid placed on top. On the lid there was a circular region with the same circumference as the cup and a ball. The object of the task was to keep …

Let-7 Mirnas Program The Fate Of Cd8 T Cells, Alexandria Wells Jul 2019

Let-7 Mirnas Program The Fate Of Cd8 T Cells, Alexandria Wells

Doctoral Dissertations

CD8 T cells are the cytotoxic effectors of the adaptive immune response, clearing virally infected and cancerous cells within the host. CD8 T cells acquire their cytotoxic function by differentiating into cytotoxic T lymphocytes (CTLs). Once the CTLs have cleared the antigen, the majority of responding cells will die during contraction; however, a small population of the antigen-specific responders will remain, differentiating into long-lived memory cells that provide potent protection to the host upon re-encounter with the antigen. These differentiation programs often become disturbed in cases of chronic infection and cancer. Instead, CD8 T cells are subverted into the so-called …

Feasibility, Acceptability, And Preliminary Efficacy Of An Academically-Integrated Physical Activity Program On Classroom Behavior In Preschoolers, Sarah Burkart Jul 2019

Feasibility, Acceptability, And Preliminary Efficacy Of An Academically-Integrated Physical Activity Program On Classroom Behavior In Preschoolers, Sarah Burkart

Doctoral Dissertations

Maladaptive classroom behaviors (i.e., hyperactivity, inattention) are common in preschoolers, yet elevated levels may lead to future academic difficulties or attention-deficit/hyperactivity disorder (ADHD). Physical activity (PA) may be one way to alleviate these maladaptive behaviors within the classroom, yet preschoolers are not meeting PA guidelines. Implementing academically-integrated PA may serve a two-fold benefit of enhancing PA and classroom behavior. However, process evaluation data describing academically-integrated PA interventions are sparse. Therefore, the purpose of this study was to examine the feasibility, acceptability, and preliminary efficacy of a 12-week PA intervention integrated into early learning standards on classroom behavior in preschoolers. Two …

Metabolic Cost And Stability Of Locomotion In People With Lower Limb Amputation, Ryan Wedge Jul 2019

Metabolic Cost And Stability Of Locomotion In People With Lower Limb Amputation, Ryan Wedge

Doctoral Dissertations

It is generally accepted that metabolic energy expenditure and gait stability are key factors that influence the selection of able-bodied locomotor patterns. It is unclear how energy expenditure and gait stability are prioritized during walking in people with lower limb amputation. People with lower limb amputation generally have greater metabolic energy expenditure during walking and increased incidence of falls. People with unilateral lower limb amputation spend more time on the intact limb compared with the prosthetic limb, while able-bodied individuals generally walk with symmetrical timing between limbs. Restoring symmetry is often a goal of rehabilitation and assistive devices, yet the …

The Bioactive Effects Of Chicoric Acid As A Functional Food Ingredient, Ye Peng Jul 2019

The Bioactive Effects Of Chicoric Acid As A Functional Food Ingredient, Ye Peng

Doctoral Dissertations

Chicoric acid, a hydroxycinnamic acid, has been reported to possess a variety of health benefits, including antivirus, anti-oxidation, anti-inflammation, obesity prevention and neuroprotection effects. However, its role on aging and glucose homeostasis are largely unknown. Therefore, the bioactive effects of chicoric acid on aging and glucose homeostasis were determined using Caenorhabditis elegans (C. elegans) and C2C12 myotubes, respectively. Our study showed that chicoric acid (25 and 50 µM) significantly extended the lifespan of C. elegans and increased median survival rates. The declines of pumping rate and locomotive activity, two indicators of aging, were delayed by chicoric acid. Chicoric …

Brachypodium Distachyon Gnrf, Swam1 And Swam4 Are Transcriptional Regulators Of Secondary Cell Wall Biosynthesis, Sandra Romero-Gamboa Jul 2019

Brachypodium Distachyon Gnrf, Swam1 And Swam4 Are Transcriptional Regulators Of Secondary Cell Wall Biosynthesis, Sandra Romero-Gamboa

Doctoral Dissertations

Plant cell walls are complex structures that contain a matrix of cellulose, lignin and hemicellulose. The regulation of the biosynthesis of these components has been well-studied in the eudicot plant Arabidopsis thaliana, and a transcriptional network has been elucidated. Several NAC and MYB family transcription factors are key regulators of secondary cell wall biosynthesis, and their functional characterization provides significant insight into the complex underlying transcriptional network. Genetic and structural evidence suggests that genes controlling this process might be different between eudicots and monocots. Here, the model grass Brachypodium distachyon has been selected to characterize the function of GNRF …

Modeling Fire Observations, Ignition Sources, And Novel Fuels To Understand Human Impacts On Fire Regimes Across The U.S., Emily Fusco Jul 2019

Modeling Fire Observations, Ignition Sources, And Novel Fuels To Understand Human Impacts On Fire Regimes Across The U.S., Emily Fusco

Doctoral Dissertations

Fire is a natural, and necessary, component of many ecosystems. However, people are changing the spatial and temporal distribution of wildfires in the U.S. at great economic and ecological costs. My dissertation addresses the impacts of humans on U.S. fires both through the introduction of ignition sources and flammable grasses. Further, I evaluate fire datasets that are widely used to investigate these phenomena over large spatial and temporal scales. Finally, I create an aboveground carbon map that can be used to estimate the potential carbon loss consequences in western U.S. ecosystems most at risk to fire. My work shows that …

Multiscale Simulations Of Intrinsically Disordered Proteins, Xiaorong Liu Jul 2019

Multiscale Simulations Of Intrinsically Disordered Proteins, Xiaorong Liu

Doctoral Dissertations

Intrinsically disordered proteins (IDPs) lack stable secondary and/or tertiary structures under physiological conditions. The have now been recognized to play important roles in numerous biological processes, particularly cellular signaling and regulation. Mutation of IDPs are frequently associated with human diseases, such as cancers and neuron degenerative diseases. Therefore, it is important to understand the structure, dynamics, and interactions of IDPs, so as to establish the mechanistic basis of how intrinsic disorder mediates versatile functions and how such mechanisms may fail in human diseases. However, the heterogeneous structural ensembles of IDPs are not amenable to high resolution characterization solely through experimental …

Acute Estrogen Synthesis And Action In The Auditory Cortex Of Developing Male Zebra Finches (Taeniopygia Guttata), Daniel Vahaba Jul 2019

Acute Estrogen Synthesis And Action In The Auditory Cortex Of Developing Male Zebra Finches (Taeniopygia Guttata), Daniel Vahaba

Doctoral Dissertations

Birdsong, as with human speech, is learned during an age- and experience-dependent sensitive period early in life. Songbirds must first memorize their parents’ song during a sensory phase, then refine their own burgeoning vocalizations to match the auditory memory of their parents’ song during a sensorimotor phase. While the error-correction aspect of the sensorimotor phase of song learning is comparatively well understood, it is largely unknown how auditory memories are formed and how auditory processing may change across development to facilitate song memorization. The songbird caudomedial nidopallium (NCM) is a brain region that encodes complex communication signals like song and …

Feasibility And Efficacy Of A Recess-Based Combined Fitness Intervention On Cognition And Academic Performance In Elementary School Children, Christine St Laurent Jul 2019

Feasibility And Efficacy Of A Recess-Based Combined Fitness Intervention On Cognition And Academic Performance In Elementary School Children, Christine St Laurent

Doctoral Dissertations

Youth physical activity and fitness have been reported to influence cognition and academic related outcomes. Despite the potential benefits of muscular fitness, few intervention studies have examined the impact of an intervention that has incorporated both cardiorespiratory and muscular fitness training on cognition and academic performance in children. Previous studies have mainly been implemented either during the school day or immediately after-school. Although recess may be an ideal time to promote physical activity because it does not compete with other academic demands, it has been an understudied setting. Therefore, the purpose of this dissertation project was to examine the feasibility, …

Faba bean is a multi-benefit cool-season grain legume that can be integrated into cropping systems of a shorter-growing season regions such as New England. A comprehensive research study was conducted to explore the ecological, nutritional, medicinal, and financial benefits of faba bean as a new multi-purpose crop for Northeast U.S.A. It was revealed that the faba bean genotypes exhibited dramatic variations in thermal units required to reach various phenological stages. Using phenology, morphology and physiological growth pattern of six faba bean varieties showed that larger seeds and later maturity did not necessarily produced higher yield. Aquadulce, the earliest maturity with …

Ni Site Structure And Function In Biological Sensing And Enzyme Activity, Hsin-Ting Huang Jul 2019

Ni Site Structure And Function In Biological Sensing And Enzyme Activity, Hsin-Ting Huang

Doctoral Dissertations

Ni(II) is one of the important cofactors involved in various enzyme functions. For organisms utilizing Ni(II), a regulation system is required to maintain Ni(II) homeostasis and prevent toxicity. The focus of this dissertation is on investigating the relationship between the Ni(II) site structure and the function of proteins, a Ni(II) sensor and a Ni(II) enzyme. RcnR, a Ni(II)/Co(II) sensor in E. coli, controls the expression of the Ni(II)/Co(II) exporter proteins, RcnAB. Due to the lack of structural information, the mechanism of metal induced allosteric regulation and metal selection is not fully elucidated. Results presented here show that binding of …

The Spatial Organization Of Mycobacterial Membrane, Julia Puffal Jul 2019

The Spatial Organization Of Mycobacterial Membrane, Julia Puffal

Doctoral Dissertations

Mycobacteria comprises a large group of organisms including the pathogenic species Mycobacterium tuberculosis, the causative agent of tuberculosis. A fast- growing saprophytic member of this genus, however, Mycobacterium smegmatis, is oftentimes used as a model organism for the pathogenic species. With a unique cell envelope architecture and unconventional polar growth, spatial coordination of cell envelope biosynthesis is vital for proper assembly of this complex structure. Here, we provide a comprehensive overview of known lateral heterogeneities in mycobacterial plasma membrane, with a particular focus on the intracellular membrane domain (IMD), a spatially distinct region of the plasma membrane with diverse functions. …
