Understanding interactions between nanoparticles and biological systems is fundamental for the development of emerging nano-biotechnology applications. In this thesis, I present an investigation of zinc oxide (ZnO) nanoparticles interactions with biomolecules in two separate studies. The first section of my thesis covers tracking and detection of ZnO nanoparticles using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). ZinkicideTM is a bactericidal ZnO nanoparticle which has been developed for agriculture. The characterization of Zinkicide in biological media and in solution has been difficult due to its high dispersibility and ultra-small size. Laser ablation in liquid has proven to be a universal and green method to synthesize nanocrystals and fabricate functional nanostructures. This study demonstrates the superiority of femtosecond laser-mediated plasma in enhancing photoredox of metal cations for controllable fabrication of plasmonic nanostructures in liquid. Through employing upstream high energetic plasma during laser-induced microexplosions, single/three-electron photoreduction of metallic cations can readily occur without chemical reductants or capping agents. Experimental evidences demonstrate that this process exhibits higher photon utilization efficiency in yield of colloidal metal nanoparticles than direct irradiation of metallic precursors. This project presents a corrosion-resistant and high capacity implantable TiN nanotube electrode to work as a neural probe. Smaller electrode size allows for reduced tissue damage while the nanopore morphology of the TiN gives a larger surface area and stability to the electrode. The electrode offers improved biocompatibility, reduced tissue damage, reduced cost, and increased usability over time.

Solar-grade single or multiple crystalline wafers are needed in large quantities in the solar cell industry, and are generally formed by a top-down process from crystal ingots, which causes a significant waste of materials and energy during slicing, polishing, and other processing. Here, a bottom-up technique that allows the growth of wafer-size hybrid perovskite multiple crystals directly from aqueous solution is reported. Single-crystalline hybrid perovskite wafers with centimeter size are grown at the top surface of a perovskite precursor solution. It remains unclear how the visual system is able to extract affective content from complex scenes even with extremely brief (< 100 millisecond) exposures. One possibility, suggested by findings in machine vision, is that low-level features such as unlocalized, two-dimensional (2-D) Fourier spectra can be diagnostic of scene content. To determine whether Fourier image amplitude carries any information about the affective quality of scenes, we first validated the existence of image category differences through a support vector machine (SVM) model that was able to discriminate our intact aversive and neutral images with ~ 70% accuracy using amplitude-only features as inputs. This model allowed us to confirm that scenes belonging to different affective categories could be mathematically distinguished on the basis of amplitude spectra alone. The next question is whether these same features are also exploited by the human visual system. Subsequently, we tested observers’ rapid classification of affective and neutral naturalistic scenes, presented briefly (~33.3 ms) and backward masked with synthetic textures. We tested categorization accuracy across three distinct experimental conditions, using: (i) original images, (ii) images having their amplitude spectra swapped within a single affective image category (e.g., an aversive image whose amplitude spectrum has been swapped with another aversive image) or (iii) images having their amplitude spectra swapped between affective categories (e.g., an aversive image containing the amplitude spectrum of a neutral image). Despite its discriminative potential, the human visual system does not seem to use Fourier amplitude differences as the chief strategy for affectively categorizing scenes at a glance. The contribution of image amplitude to affective categorization is largely dependent on interactions with the phase spectrum, although it is impossible to completely rule out a residual role for unlocalized 2-D amplitude measures.

Solute segregation at dislocations can impede the motion of dislocations, strengthening materials. Here, we study the formation and role of solute segregation at dislocations in CrFeCoNiMo high-entropy alloys (HEAs) by high-angle annular dark-field scanning transmission electron microscopy imaging and mechanical testing both deformed and annealed samples. Mo atoms exhibit pronounced segregation into the planar-extended core of dislocations, i.e. stacking faults, causing the increase in the yield strength while the loss of the ductility. This work suggests that mechanical properties of HEAs can be tailored by alloying additional elements that are in favor of segregation into dislocations.

A probe for traversing a tubular passage includes an introducer to be supported at an entrance to the tubular passage. Diseases of pregnancy are the leading cause of maternal and neonatal morbidity and mortality affecting more than 20% (26 million) of all pregnancies annually. Those diseases include preeclampsia, preterm labor, intrauterine growth restriction and gestational diabetes, many of which are caused by compromised functions of the placenta. Placenta is a specialized organ that is only present during pregnancy where it creates a maternal-fetal interface that is responsible for many functions that contribute to the development of the fetus. Hydrophobic thiol coated gold nanoparticles have recently been investigated for their ability to self-assemble into robust, ultra-thin, porous membranes at a liquid-vapor interface. Due to the well-ordered, hexagonal close-packed nanoparticle arrays formed during the self-assembly process, these 2-dimensional sheets have very well-defined pore structures and have been shown to span gaps of several microns under ideal conditions. Radiosensitization is a novel targeted therapy strategy where chemical compounds are being explored to enhance the sensitivity of the tissue to the effects of ionizing radiation. Among the different radiosensitizers alternatives, nanomaterials have shown promising results by enhancing tumor injury through the production of free radicals and reactive oxygen species (ROS). We studied the possibility of achieving an order of magnitude reduction in the energy dissipation needed to write bits in perpendicular magnetic tunnel junctions (p-MTJs) by simulating the magnetization dynamics under a combination of resonant surface acoustic waves (r-SAW) and spin-transfer-torque (STT). The magnetization dynamics were simulated using the Landau-Lifshitz-Gilbert equation under macrospin assumption with the inclusion of thermal noise. We studied such r-SAW assisted STT switching of nanomagnets for both in-plane elliptical and circular perpendicular magnetic anisotropy (PMA) nanomagnets and show that while thermal noise affects switching probability in in-plane nanomagnets, the PMA nanomagnets are relatively robust to the …

A direct numerical simulation of single plume flow in thermal convection with polymers was carried out in a domain with 1:3 as the width to height ratio. The heat transport ability is weakened by adding polymers within the here-investigated governing parameter range. However, it is promoted when the maximum polymer extension L is increased. The distribution of vertical velocity and temperature indicates that the plume in the polymer solution case is speeded up and widens bigger as compared to that in the Newtonian fluid case. The nitrogen-vacancy (NV) center is a point defect in diamond with unique properties for use in ultra-sensitive, high-resolution magnetometry. One of the most interesting and challenging applications is nanoscale magnetic resonance imaging (nano-MRI). While many review papers have covered other NV centers in diamond applications, there is no survey targeting the specific development of nano-MRI devices based on NV centers in diamond. Aluminum (Al) alloys have been widely used in the transportation industry. However, most highstrength Al alloys to date have limited mechanical strength, on the order of a few hundred MPa, which is much lower than the flow stress of high-strength steels. In this study, we show the fabrication of nanocrystalline Al alloys with high-density growth twins enabled by a few atomic percent of Co solute. In situ uniaxial compression tests show that the flow stress of Al–Co solid solution alloys exceeds 1.5 GPa, while good work hardening capability is maintained. Silver nanoparticles (AgNP) are widely used in a variety of consumable products as antibacterial to prevent or treat infection. Unfortunately, evidence exits that AgNP induces inflammation which can worsen with repeated human exposure. However, there is little or no research on how to mitigate these adverse effects due to AgNP induced-toxicity. Here, we investigated if surface modification of AgNP by liposomal encapsulation suppresses AgNP-mediated inflammatory responses in THP1 monocytes and THP1 differentiated macrophages (TDM). Here we carried out a comparative study on two cost and time effective methods of encapsulating silver nanoparticles (AgNP) in dipalmitoyl-phosphatidyl choline (DPPC)/cholesterol-based liposome to enhance nanoparticle cytotoxicity, and evaluated the effect of this on a blood cell line (THP1 monocytes) often involved in uptake of nanoparticles during human exposure. DLS and Zeta potential analyses over a 6-months period showed the extruded Lipo-AgNP (Ex-Lipo-AgNP) exhibited more stable characteristics when compared with the probe-sonicated Lipo-AgNP (PB-Lipo-AgNP). SEM microscopy indicated agglomeration of the PB-Lipo-AgNP which was not observed in Ex-Lipo-AgNP. Ultrasound is a prominent nondestructive testing modality for the detection, localization, and sizing of defects in engineering materials. Often, inspectors analyze ultrasonic waveforms to determine if echoes, which stem from the scattering of ultrasound from a defect, exceed a threshold value. In turn, the initial selection of the threshold value is critical. In this letter, a time-dependent threshold or upper bound for the signal envelope is developed based on the statistics governing the scattering of ultrasound from microstructure. Graft repair of aortic coarctation is commonly used to mimic the physiological aortic arch shape and function. Various graft materials and shapes have been adopted for the surgery. The goal of this work is to quantitatively assess the impact of graft materials and shapes in the hemodynamics and wall mechanics of the restored aortic arch and its correlation with clinical outcomes. Mg/Nb nanolaminates synthesized through vapor deposition techniques exhibit high flow strength without conventional twinning in Mg. In this work, we investigated the influence of laminated microstructures on deformation mechanisms of Mg nanolayers. Using molecular dynamics simulations, we explored that (0001)-oriented Mg layers transform or re-orient to {10¯10}-oriented Mg layers through nucleation and growth of {10¯12} twins by atomic shuffling, instead of conventional {10¯12} twinning shear. Such a reorientation accommodates in-plane compressive strain and out-of-plane tensile strain when Mg/Nb laminates are subjected to compression parallel to the Mg/Nb interfaces. The container wall also includes an interior surface configured to be in fluid communication with the working fluid. We propose a new structure for piezoelectric gyroscopes. It is made from multilayered thin films of AlN or ZnO with alternating c-axes along the film thickness. It is shown theoretically that when such a film is electrically driven into higher-order overtone thickness-shear vibration in one of the two in-plane directions of the film and is rotating about the film normal, the Coriolis force due to the rotation causes a higher-order overtone thicknessshear vibration in a perpendicular direction with an electrical output that can be used to measure the angular rate of the rotation. Cold working individual layers during additive manufacturing (AM) by mechanical surface treatments, such as peening, effectively “prints” an aggregate surface integrity that is referred to as a glocal (i.e., local with global implications) integrity. Printing a complex, pre-designed glocal integrity throughout the build volume is a feasible approach to improve functional performance while mitigating distortion. However, coupling peening with AM introduces new manufacturing challenges, namely thermal cancellation, whereby heat relaxes favorable residual stresses and work hardening when printing on a peened layer. Thus, this work investigates glocal integrity formation from cyclically coupling LENS® with laser peening on 420 stainless steel.

Nanotwinned metals exhibit outstanding radiation tolerance as twin boundaries effectively engage, transport and eliminate radiation-induced defects. However, radiation-induced detwinning may reduce the radiation tolerance associated with twin boundaries, especially at elevated temperatures. Here we show, via in-situ Kr ion irradiation inside a transmission electron microscope, that 3 at. % Fe in epitaxial nanotwinned Cu (Cu97Fe3) significantly improves the thermal and radiation stability of nanotwins during radiation up to 5 displacements-per-atom at 200 °C. Deformation twins are three-dimensional domains, traditionally viewed as ellipsoids because of their two-dimensional lenticular sections. In this work, we performed statistical analysis of twin shapes viewing along three orthogonal directions: the ‘dark side’ (DS) view along the twin shear direction (η1), the twinning plane normal (TPN) view (k1) and the ‘bright side’ (BS) view along the direction λ(=k1 × η1). Our electron back-scatter diffraction results show that twins in the DS and BS views normally exhibit a lenticular shape, whereas they show an irregular shape in the TPN view. We developed an Adaptive Reference-Digital Image Correlation (AR-DIC) method that enables unbiased and accurate mechanics measurements of moving biological tissue samples. We applied the AR-DIC analysis to a spontaneously beating cardiomyocyte (CM) tissue, and could provide correct quantifications of tissue displacement and strain for the beating CMs utilizing physiologically-relevant, sarcomere displacement length-based contraction criteria. The data were further synthesized into novel spatiotemporal parameters of CM contraction to account for the CM beating homogeneity, synchronicity, and propagation as holistic measures of functional myocardial tissue development.
