Osteoarthritis (OA) is a form of arthritis that develops in the joint due to overuse and aging causing pain, discomfort, and disability. Total Knee Arthroplasty (TKA) is a surgical procedure performed when OA symptoms are severe with an estimated 600,000 patients in the United States currently receiving TKA. Studies have reported dissatisfaction of the knee for 14-39% of patients. This study collected knee kinematics before and after surgery using stereo radiography for precise measurement of gait and deep knee flexion activities. Synovial inflammation is now recognized as a major contributor to OA progression and pain. Activated synovial macrophages in an OA joint are believed to play a major role in low grade inflammation found in OA. Bisphosphonates such as zoledronic acid (ZA) are known to induce apoptosis specifically in macrophages and several of them are being evaluated as a disease-modifying osteoarthritis drug in clinical trials. Biophysical signals including stiffness and dimensionality influence a myriad of stem cell behaviors including morphology, mechanosensing, and differentiation. 2D stiff environments cause increased cellular spreading and induce osteogenic differentiation whereas 3D soft environments favor rounder cell morphologies attributed to a chondrogenic phenotype. The goal of this study is to create a composite that integrates these divergent biophysical signals within one system. This composite consists of a stiff and porous polycaprolactone (PCL) backbone that provides mechanical stiffness and a 2D environment. Osteoarthritis (OA) is a pathological degenerative condition of the joints that is widely prevalent worldwide, resulting in significant pain, disability, and impaired quality of life. The diverse etiology and pathogenesis of OA can explain the paucity of viable preventive and disease-modifying strategies to counter it. Advances in genome-editing techniques may improve disease-modifying solutions by addressing inherited predisposing risk factors and the activity of inflammatory modulators. Implant geometry is a significant factor in determining knee stability and patient satisfaction following total knee replacement (TKR). Ineffective muscle recruitment, impaired joint functionality and increased implant wear are consequences of an unstable knee replacement. Current knee laxity evaluation techniques are limited in their ability to account for the muscular response to knee instability. Osteoarthritis (OA) consists of a degenerative disease on articular cartilage, which is prone to excessive mechanical loading and frictional resistance that leads to the wear and tear of the tissue. Joint injuries, like anterior cruciate ligament (ACL) tears, can lead to OA due to a change in articular cartilage loading. Gait analysis combined with knee joint finite element modeling (FEM) has been used to predict the articular cartilage loading. Osteoarthritis of the glenoid results in regional bone density variations and bone loss that may compromise early component fixation and support. The two common morphologies, symmetric and asymmetric erosion, were characterized by bone density and morphology, and assessed on the basis of bone removal and bone quality in the context of augmented glenoid components. The bone strain field was also compared when different augmented glenoid components underwent simulated joint loading using finite element analysis.

Post-traumatic osteoarthritis (PTOA) is a prevalent condition following rupture of the anterior cruciate ligament (ACL). While numerous animal models of PTOA exist, most are based on surgical disruption of a stabilizing structure. In the rat, surgical ACL transection is the most commonly employed model, but it may introduce confounding biological factors due to surgery. Purpose: Digital x-ray tomosynthesis (DTS) has the potential to provide 3D information about the knee joint in a load-bearing posture, which may improve diagnosis and monitoring of knee osteoarthritis compared with projection radiography, the current standard of care. Manually quantifying and visualizing the joint space width (JSW) from 3D tomosynthesis datasets may be challenging. This work developed a semiautomated algorithm for quantifying the 3D tibiofemoral JSW from reconstructed DTS images. The algorithm was validated through anthropomorphic phantom experiments and applied to three clinical datasets.
