Purification And Characterization Of The Prostate Cancer-Associated Antigen: 7e11-C5, Qi Feng

A murine monoclonal antibody (MAb), 7E11-C5, recognizes a prostate organ-specific antigen. Under reducing conditions, the 7E11-C5 antigen exhibited major 100-Kd and minor 70-Kd bands, suggesting that the antigen comprises two peptide chains. The antigen was detected in pooled normal human seminal plasma, xenograft tissue extract of a prostate adenocarcinoma cell line, LNCaP, and tissue extracts of benign prostatic hyperplasia (BPH) and prostate carcinomas, but was not identified in any of the non-prostate tissue extracts tested. Antibodies of the three major prostate-associated antigens (prostatic acid phosphatase (PAP), prostate specific antigen (PSA) and prostate secretory protein (PSP)) were unable to block the …

This study describes morphological changes in the lung at several time intervals following a single injection of 1-Nitronaphthalene (100 mg/kg intraperitoneal) in male Sprague-Dawley rats using transmission and scanning electron microscopy. Dilation of the smooth endoplasmic reticulum and mitochondrial distension in the nonciliated bronchiolar (Clara) cells was apparent as early as 1 hour after injection, while adjacent ciliated cell showed minimal changes. At 6 hours, when respiratory distress was apparent in the animals, Clara and ciliated cells exhibited chromatin clumping, loss of mitochondrial cristae and cytoplasmic vacuolization. Glycerophospholipids of mammalian cells exist as chemically diverse structures with various fatty acids at the sn-1 and sn-2 positions. Arachidonic acid, a polyunsaturated fatty acid, which may be converted to biologically active eicosanoids such as prostaglandins, thromboxanes, and leukotrienes, is found predominantly in the sn-2 position of glycerophospholipids. The purpose of this study was to examine, at the level of the individual molecular species, the incorporation of arachidonate into phospholipids and its release from phospholipids during stimulation. There is an expressed need to develop a greater variety of safe contraceptive methods which would find acceptance worldwide, particularly in developing countries. Immunization against spermatozoa might be such a method as judged by the accumulated evidence from studies on human females and in a number of female animal species.

Two extraction techniques were used for sperm membrane antigen isolation. The first technique involved NP-40 detergent for antigen extraction from human motile sperm and the second technique employed homogenized human testis for antigen extraction. This research project was undertaken to understand the process of desorption/removal of organic compounds from soil matrices under the influence of temperature and gas purging. The project was divided in two parts :

A. Plug deposition experiments: Columns packed with soil matrices were purged with an inert gas under isothermal conditions. Organic compounds were then introduced as a plug on the front of the column by on-line injection. The rate of passage (due to sequential adsorption/desorption) of the plug through the various soil matrices was measured by a gas chromatograph equipped with a flame ionization detector (FID). A method was developed for extraction of plasmid DNA from bacterial cells embedded in agarose blocks. Cell - containing blocks were treated with various lysing reagents and inserted into the wells of an agarose gel. Upon electrophoresis the plasmid DNA migrated out of the embedding block and into the gel leaving intact chromosomal DNA in the well. This method was tested with various organisms and found to be effective for plasmid isolation. In comparison to "traditional" procedures, this new method is less tedious since chemical separation of plasmids is not required prior to electrophoresis. Also, a higher yield of plasmid …

Experiments were conducted to determine whether a continuous process similar to the common activated sludge process could be operated at a relative steady state with phenol and 2-chlorophenol as the sole carbon sources. After a number of unsuccessful trials, process conditions were developed which allowed for operation at a relative steady state with respect to the concentration of microorganisms in the reactor and the concentration of phenol and 2-chlorophenol in the effluent.

Phenol and 2-chlorophenol were removed at better than 99 percent efficiency on a continuous basis. The mixed liquor grown under the conditions of the experiment was highly filamentous …

The sizing chemical ASA is emulsified to make i t compatible with the papermaking process. Unfortunately, emulsified ASA is chemically reactive with water at the elevated temperatures found in papermaking systems. As an alternative, encapsulated ASA was made and investigated for its stability towards water at elevated temperatures.

A handsheet procedure was developed to analyze emulsified and encapsulated ASA. It was concluded that the procedure gave statistically reproducible results. Investigations with the procedure revealed important variables concerning the laboratory and m ill application of ASA. Evidence indicated that retention of ASA was critical to sizing development.

Male Sprague Dawley rats (250g) were exposed to benzyl chloride vapor at a concentration of 220 ppm for 6 hours. During inhalation, the blood benzyl chloride concentration increased to reach a steady state level of 9-10 ug/ml between 4 and 6 hours of exposure. A small concentration of benzyl chloride (0.86 ug/ml) was present in the blood four hours after the termination of exposure. The absorption half life was 0.53 hours (0.39-0.81) and the elimination half life was l.12 hour (0.93-l.40).

DTIC and BCNU have been demonstrated to have enhanced chemotherapeutic effects in mice bearing Bl6 mouse melanoma cells. The order of administration seems to be important in maintaining optimum effects. This was shown is a study by Hill et al. (unpublished).

In the present study, mouse melanoma cells, Bl6CL4, were treated with either DTIC or BCNU. The drug was allowed to remain on the cells for 2, 3, or 4 days. Safety procedures for performing geotechnical testing with hazardous and toxic materials, excluding radioactive wastes, are developed for the Flexible Wall Permeameter and the Atterberg Limits Tests. The tests were evaluated in detail to determine the critical points at which the technician would be exposed to the hazardous and/or toxic permeant. The biological characteristics of four microbial mixtures were examined after successive exposure to phenol and 2-chlorophenol at room temperature. One of the microbial mixtures was an activated sludge from the Livingston, NJ municipal wastewater treatment plant. The other three mixtures were commercial preparations sold by Polybac Corp.(Hydrobac), Sybron Corp. (BI-CHEM DC 1006/1007), and General Environmental Sciences(LLMO).

The characteristics examined were relative counts and species of gram negative/gram positive bacteria, yeast and fungi, protozoa, and higher organisms. Metallothionein (MT) and glutathione (GSH) have been shown in mammalian research to play a role in the sequestration and depuration of mercury. The sulfhydryl groups on both these peptides have a strong affinity for mercury, MT for Hg2+ and GSH for CH3Hg+. In this study, killifish (Fundulus heteroclitus) were force-fed one piece of squid laced with labelled mercuric nitrate to give a final food concentration of 20 ppm 203Hg2+ and 0.03 -0.05 µg g-1 Hg2+ total dose. Volatile organic species (aliphatic hydrocarbons from hexane to dodecane, cyclohexane and aromatic species including benzene, toluene, ethyl benzene, m,p, and o—xylenes, nitrobenzene, styrene and naphthalene) associated with residential kerosene heater emissions were measured at two sites. Acrylamide causes a peripehral neuropathy. The biochemical mechanism by which it does so is the subject of intense investigation. One of the principal hypotheses for this mechanism centers on acrylamide's ability to inhibit glycolysis. This study examines the effects of in vivo exposure to acrylamide on the enzymes of the nerve terminal particle, the synaptosome.

A method for measuring glycolytic enzyme inhibition in the synaptosome was developed. Acrylamide (500 mg/kg) was administered to rats intraperitoneally. Bacterial populations indigenous to surface (1 m) waters of the Middle Atlantic Outer Continental Shelf were sampled at seasonal intervals to determine the abundance and distribution of petroleum-degrading (HC) and "total" heterotrophic (HET) bacteria. Simultaneously, unweathered South Louisiana crude oil (SLCO) was added to aliquots of surface water samples to evaluate rates and patterns of degradation of hydrocarbon. Degradation was studied using selected nutrient and temperature regimes in both closed flask and prototype "open" or continuous dilution systems. This study was performed to determine the spectral signatures of deer and their natural background elements for censusing purposes. Consideration was given to atmospheric transmittance, acceptable flying weather, and terrain. Possible spectral bands between 0.3 and 14.0 μm were obtained (over a pathlength of 1500 feet at an altitude of 5000 feet) based upon atmospheric transmittance using the LOWTRAN 3B computer program. They are: 0.30 - 1.33, 1.49 - 1.79, 2.00 - 2.50, 3.00 - 3.16, 3.38 - 4.10, 4.59 - 5.05, and 8.00 - 13.33 μm, for transmittance greater than 75%. Determination Of Lead Content In Apical Tissue Following Root Canal Obturation With Sargenti's Formula, G. Merle Hickok

Four Stump-tail monkeys were used to help determine if the lead in Sargenti's formula diffuses out into the apical tissue while confined entirely within the root canal. In each animal two teeth were filled with Sargenti's formula 1-2 millimeters short of the radiographic apex, and two teeth were filled with gutta percha and Proco-Sol sealer for controls, making a total sample size of sixteen. The time ranged from 48 hours to 120 days. Lead analysis using the spectrophotometric method showed a higher lead content in the teeth filled with Sargenti's formula than with the gutta percha controls.

This makes irrigation of vegetables necessary to their production in South Dakota since summer rainfall is normally both inadequate and undependable. A numerical method and model were developed to simulate soil moisture during steady-state and transient flow toward a tile drain. Intermittent application of water by drip irrigation and rainfall were simulated by use of finite difference approximation equations. No known analytical solution for these flow situations exists because of the complex mathematics describing unsaturated flow in the soil. The computer model is based on the electrical resistance network analog. The explicit finite difference equations were solved by the successive over-relaxation method for steady-state flow and by iteration for transient soil moisture flow. In-stream concrete structures were studied through model tests and river tests. The model studies indicated that four designs provided good habitat in the model stream. These structures were the inverted weir, the "V" structure, the slab with legs and the cylinder. Through the river studies it was determined that these structures did not influence enough of the total river area to be effective in providing good fish habitat. Also an appreciable amount of yearly maintenance would be required to free the structures from silting in, debris, and vandalism. The slab with legs was the only promising structure.

The specific-locus mutation frequency resulting from 300 R of acute X irradiation has been determined for the germ cells present in male mice at 0, 2, 4, 6, 8, 10, 14, 21, 28, and 35 days of age and also for female mice at 0 days of age, Sample size was much larger for the males irradiated on day 0 than for other age groups but in all groups it was large enough to insure that an extremely high rate would be noticed.
