Evergreen Tree Species For Urban Use In Eastern Tennessee, Vivian Orr Potter

The objective of this study was to determine which of five tree species was best suited for urban conditions in eastern Tennessee with respect to certain criteria. The species included eastern white pine (Pinus strobus L.), eastern hemlock [Tsuga canadensis (L.) Carr.], Norway spruce [Picea abies (L.) Karst.], southern magnolia (Magnolia grandiflora L.), and eastern red-cedar (Juniperus virginiana L.). The criteria used for comparing the species were resistance to disease, resistance to insect pests, range of soil fertility adaptability, range of soil pH adaptability, and aesthetic appeal.

The purposes of this study were to characterize High and Low performance rated Agents, to determine the relationship between performance rating level, attitude toward the assignment and other attributes of SPIFFY Agents in Tennessee. It was also proposed to identify selected aspects of the SPIFFY programs conducted by High and Low rated Agents.

The population of this study included all 37 SPIFFY Agents in the State of Tennessee. The data were collected by means of a mail questionnaire. A secondary source of data consisted in personnel records.

Partial Purification And Characterization Of Pectin Methyl Esterase In Southern Peas (Vigna Sinesis), Kun Kook Park

The purposes of this study were to partially purify pectin methyl esterase (PME) of Southern peas (Vigna sinensis), to determine optimum pH and NaC1 concentration for maximum enzyme activity, to measure the energy of activation and kinetic reactions, and to ascertain the presence of multiple molecular forms of the enzyme and their isoelectric points.

PME was extracted from an acetone powder preparation and purified by a two-step ammonium sulfate fractionation and dialysis. The activity of the partially purified enzyme was significantly (p < 0.05) influenced by pH, NaC1 concentration, and the interaction between pH and NaC1 concentration. The optimum pH and NaC1 concentration were 7.9 and 0.25 M, respectively. The energies of activation were calculated to be 4,900, 5,950, and 6,900 calories/mole at pH 6.0, 7.0, and 7.0, respectively. PME had a Km of 0.781% pectin N.F. and a Vmax of 24.34 PME units.

The plant growth regulator 2-chloroethylphosphonic acid (ethephon) was applied to summer squash to determine its effects on the flowering pattern. Treatments with all pistillate flowers pruned at anthesis, with and without ethephon applications were also compared to control plants to investigate the factors regulating flowering and fruit set of summer squash.

It was found that plants in the pruned treatments had a greater carbohydrate accumulation than plants in the nonpruned treatments, indicating that carbohydrate accumulation may be inversely associated with fruit development. Ethephon treatments produced more pistillate flowers per plant but also had a large number of aborted flowers. Thus, …

The purposes of this study were: to investigate the influence of dietary calcium, dietary phosphorus, and their ratios as observed in vegetarian and non-vegetarian subjects on their bone densities and to compare bone density, the dietary intakes of calcium and of phosphorus, and the ratio of calcium to phosphorus in vegetarian and non-vegetarian subjects.

After obtaining informed consent, data were collected on 43 vegetarian and 36 non-vegetarian women, 40 to 92 years old, who consumed self-selected diets. The objectives of this study were: (1) to make a theoretical comparison of progress expected by selecting for a single trait with progress expected by selection based on an index including two traits and (2) to compare estimates of repeatability of cow productivity when the effect of sire is removed from the data and when it is not.

Data were collected over a period of seven years from 581 bull calves and 552 heifer calves from the purebred Angus herd at The University of Tennessee Plateau Experiment Station, Crossville.

Ten tissues from each of four species of fish-large­mouth bass (Micropterus salmoides), black crappie (Pomoxis nigromaculatus), walleye (Stizostedion vitreum) and rainbow trout (Salmo gairdneri) from Lake Powell were analyzed and compared for the concentrations of ten heavy metals: Fe, Ca, Mg, Cu, Cr, Cd, Zn, As, Se, and Pb.

Samples were digested with nitric and perchloric acids, and analyses were performed by atomic absorption spectro­photometry. Concentrations were expressed on a dry weight basis for all tissues, and were not corrected for percent recovery.

As expected, high levels of iron were observed in the blood tissues, with low concentrations being observed …

To collect and examine the organisms associated with newly inundated terrestrial vegetation in Lake Powell, small plastic Christmas trees were "planted" along a sloping sandy bottom at depths of 2, 3, 4.5, 6, 8, and 10 meters. For the 17 months from July, 1973, through October, 1974, the trees were harvested and replanted by scuba divers at 4- and 12-week intervals. The macroinvertebrate and diatom components of the periphytic community associated with each tree were collected and evaluated for number of individuals per unit area (density) and species composition. The most desirable way of treating endocrine deficiencies would be by the successful transplantation of functional endocrine tissue into the deficient animal. However, unless the donor and recipient are identical twins or from an inbred strain of animals, an immune response is usually elicited in the recipient by the foreign tissues leading to rejection of the transplant. Thyroid gland cells which have been enzymatically dispersed from thyroid tissue and cultured in vitro are capable of reforming functional thyroid tissue when implanted into animals of the same strain. Beetles of Trypophloeus populi attacked green bark of unhealthy aspen (Populus tremuloides) and hastened the death of the tree. Beetles of Procryphalus mucronatus favored dead bark and were of little significance in the death of the aspen. These two beetle species were distinguished from each other by their primary galleries, eggs, larvae, larval mines, and mating behavior. In T. populi, there were three larval instars and one to one and one-half generations per year; only larvae overwintered, and the eggs of the first generation were present by mid-July. Therapeutic lung lavage is a procedure by which the interior of the lung of a living animal is “washed” with a suitable solution. Broncho­pulmonary lavage, a special type of lung lavage in which the bronchioles and alveoli are washed, is the major concern of this thesis. Broncho­pulmonary lavage was investigated in 1920 as a possible treatment for lungs damaged by irritant gases and in the late 1950‘s as a method of hemodialysis. A survey of the endoparasites of 100 stray cats in central Utah was conducted. Each cat was euthanized, and the entire alimentary tract, gall bladder, bile duct, heart, respiratory and excretory systems were excised and examined for endoparasites. Fecal samples were examined by a sugar flotation technique. Parasites identified and number of cats infected were: Isospora felis 5; I. rivolta, 4; Dipylidium caninum, 1; Hydatigera taeniaformis, 10; Toxocara mystax, 43; and Physaloptera sp., 2. One ectoparasite, Cheyletiella parasitivorax, was identified in the feces of 4 cats. Dipylidium caninum and T. mystax are known to cause accidental infection in humans and …

Phytoplankton samples were collected biweekly from two stations, one in each branch of Lynnhaven Bay, Virginia over the period January 24 to June 9, 1975. Water temperature, salinity, pH, oxygen, ammonia, urea, reactive nitrite, reactive nitrate, reactive phosphorus, productivity, transparency, and weather conditions were recorded concurrently. Data were manipulated employing analysis of variance, simple correlation, correlation matrices, and forward stepwise multiple regression. Total phytoplankton numbers exhibited a unimodal pattern. A pre-bloom pulse was recorded at both stations in late February. The spring bloom lasted from April through May. Phytoplankton growth was primarily a function of nutrient supply and temperature. Sex pheromone activity in the foveal glands of female Dermacentor andersoni (Stiles) and Dermacentor variabilis (Say) is described. Autoradiography revealed the accumulation of 36Cl in the foveal glands of females during their first 120 hours of feeding on a 36Cl-labeled host. Emissions collected from these females contained a 36Cl-labeled product. This product, however, was absent in emissions collected from the same females with their foveae dorsales obstructed.

Accumulation of 36Cl in the foveal gland, in conjunction with the release of a 36Cl-labeled product through the foveae dorsales, suggest localization of sex pheromone activity within these …

The properties of the coenzyme, NAD, binding site of chicken muscle L-∝-glycerophosphate dehydrogenase were studied. Adenosine monophosphate, adenosine diphosphate and adenosine diphosphoribose were sha.vn to inhibit the enzyme competitively with respect to NAD. The presence of adenosine, pyrophosphate and ribose regions in the coenzyme binding site of the enzyme was suggested by the inhibitor constants obtained for these compounds.

Disodium monoalkyl phosphates, n-butyl to n-dodecyl phosphate, inclusive, were also shown to inhibit the L-∝-glycerophosphate dehydrogenase catalized reaction competitively with respect to NAD. A positive chain length effect was observed in the binding of these compounds to the enzyme, suggesting the …

Purification of Yeast NAD+-dependent isocitrate dehydrogenase was partially successful when utilizing an NAD+-ribosyl-hydrazide Sepharose affinity chromatography column. Although no specific elution system was found, a maximum purification of sixty-five fold was achieved with 0.5M KCl elution. A 38-fold purification was obtained when 20mM NAD+ was employed for elution.

Yeast and calf liver NADP+-dependent isocitrate dehydrogenase were also found to bind to NAD+-ribosyl-hydrazide Sepharose as well as the NAD+-dependent enzyme. Again, elution was achieved with either o,5M KCl or 20 mM NADP+.

Concentrations of cAMP increased with time in pea hypocotyls assayed at 24 hour intervals over a period of four days. The highest concentration was 20.1 pmoles/mg protein which occurred in 96 hour hypocotyl tips. Ninety-six hour root bodies (dissected region between tip and embryo) achieved a concentration of 8.6 pmoles/mg. Concentrations of cAMP in embryos remained constant around one pmole/mg for three days and values doubled on the fourth day.

Hypocotyls, after germination for 72 hours, were dissected and incubated for various lengths of time in water and 3xl0-8M IAA. Tips incubated for two hours showed a tenfold …

Several coatings colors were prepared and applied. The coated sheets were tested with great emphasis on keeping the conditions constant throughout the course of the experimental work. The study utilized several types of insolubilizers: urea formaldehyde, melamine formaldehyde, glyoxal and latex. Two types of starches were also used: regular and cationic.

Results showed the importance of drying on wet rub development of both urea and malamine formaldehyde as compared to latex and glyoxal. Results also proved the superiority of using insolubilizer additions with cationic starch rather than regular starch.

Prairie rattlesnakes, of the species Crotalus viridis virldis, were orally, infected with tetrathyridia of Mesocestoides corti to determine the host response to the parasite. Snakes were maintained at constant temperatures of 25, 30, and 35 C for 4--24 weeks. Blood chemistry evaluations of the host were made by standard hematological methods. Eosinophil counts, total proteins, and serum gamma globulins increased while packed-cell volumes and hemoglobins decreased. Autopsy studies revealed the first documented asexual multiplication of Mesocestoides corti in a laboratory infected ectothemic host, with the small intestine being the preferred organ. The preference for a rocky situation was analyzed under laboratory conditions. In three-compartmented choice boxes, Peromyscus californicus and P. eremicus chose a compartment with rocks and a sandy floor over a compartment with just a sandy floor but P. maniculatus did not show a preference. In a similar experiment, in continual total darkness, the three species chose the compartment with the rocks. UM-key did not prefer the compartment with rocks when the rocks were encased in plastic and were under a normal light regime. The effect of temperature on calcification in Bossiena orbigniana orbigniana (Decaisne, 1842) [Manza, 1940; Silva, 1957] (Corallinaceae) was studied using calcium-45 labeling of terminal segments under laboratory conditions. B. orbigniana has optimum calcium uptake for the temperatures 17-21°C within a temperature range of 3-37°C. These results suggest that optimum calcification in B. orbigniana should take place from late spring to early autumn. Q10 values for 3-21°C averaged 3.39 indicating an enzyme-governed reaction. It is proposed that thallus growth rate and calcification are not directly related. It is believed that the optimum temperatures (17-21°C) affect the enzymes towards an increase …

The possible effects of ablastin on Schistosoma mansoni were investigated by (1) establishing a concurrent infection of Trypanosoma musculi and Schistosoma mansoni in mice, and (2) by obtaining Trypanosoma lewisi immune rat serum (ablastin) and injecting it into S. mansoni infected mice. These procedures were followed on days 4, 14 and 28 of S. mansoni infection. For the groups that were given immune rat serum, controls were kept into which normal rat serum was injected.

Ablastin does not have any effect on the development or morphology of the S. mansoni worms as tested in this study. Although ablastin inhibits glycolytic …

The assay of Bursa-derived and Thymus-derived lymphocytes (B and T cells) in man has become increasingly useful during the past few years in normal and in clinical conditions. There has been quite a range reported in the literature in the percent of B and T cells in normal subjects by different investigators.

The purpose of this study was to see if monocyte contamination was one cause of non-specific rosette formation. Hypothalamic and cortical extracts from various cariogenically grouped rats which had been maintained on either a Purina diet or a high sucrose diet (HSD) for three months were infused intravenously into 28 days old rats to determine if there was a titer of parotid hormone releasing factor (PRF) capable of facilitating fluid movement (FM) in the teeth. The hypothalamic and cortical extracts obtained from Purina-fed rats with 0-caries and the hypothalamic extracts from BSD-fed rats conclusively demonstrated FM stimulatory ability.
