C1q TNF Related Protein 3 (CTRP3), is a cytokine that is primarily secreted from adipose tissue, which classifies it as an adipokine. Our previous research has shown that CTRP3 prevents alcoholic fatty liver disease (ALD) in male mice. However, even when accounting for confounding factors such as absolute and relative alcohol intake, females are more sensitive to the effects of consumption compared to male mice. Therefore, the goal of this project was to determine whether CTRP3 prevented ALD in female mice. Membrane fusion is key to organism homeostasis and occurs when a transport vesicle fuses with a target compartment, merging the two membranes into one while releasing the contents of the transport vesicle into the target compartment. This process is controlled by SNARE proteins. Syntaxin-17 protein plays a crucial role in the fusion of the autophagosome membrane with the lysosome membrane, allowing for degradation of misfolded intracellular components and pathogens. Forage-based ruminant diets supplemented with condensed tannins or polyphenolic-containing legumes may alter nitrogen (N) metabolism in the animal and reduce gas emissions from soil receiving excreta. The objective of the study was to determine if soil receiving urine or manure from sheep fed forage diets supplemented with condensed tannin or polyphenolic-containing legumes would decrease N gas emissions. Acute myeloid leukemia (AML) is an often devastating hematologic malignancy with 5-year overall survival lingering near 20%. Acquiring a deeper understanding of molecular underpinnings of leukemogenesis will provide a basis for developing more effective therapeutic strategies for patients with AML.
Here, we identified overexpression of hnRNP K as a recurrent abnormality in a subset (~20%) of AML patients. High levels of this RNA-binding protein associated with inferior clinical outcomes in de novo AML. The modern broiler is growing at a rapid rate generating tremendous amounts of heat. A sensitive Net Energy (NE) system is needed to measure body heat production (HP) generated primarily by daily maintenance and synthesis and degradation of myofibrillar and sarcoplasmic protein. The first two chapters present evaluation of the classic way to calculate NE versus a new methodology; the Arkansas NE (Ark NE) system, with birds from two genetic lines fed diets with different AA content or different ME content in two different environmental temperatures.

The hypothalamo-pituitary-adrenal (HPA) axis is the regulatory system for the neuroendocrine stress response within vertebrates. Within the HPA axis corticotropin releasing hormone (CRH) is a major regulator and driving hormone. A structure named the nucleus of the hippocampal commissure (NHpC) has been found to contain CRH neurons and also these neurons respond to early food deprivation stress significantly prior to the paraventricular nucleus (PVN), the major driving nucleus of the classic neuroendocrine HPA axis. Necrotic enteritis (NE) has re-emerged following restriction of antimicrobial usage and costs $6 billion every year worldwide. The primary objective of the studies was to evaluate prevention and treatment of NE using a microbiota metabolic product, secondary bile acid deoxycholic acid (DCA), in drinking water. Woody Breast (WB) myopathy causes significant economic losses to the global poultry industry and the lack of objective, non-invasive and rapid methods for its detection as well as profitable alternatives for its utilization are contributing factors. Today, the most widely used class of anthelmintic used to treat parasitism, is the macrocyclic lactone. Many studies have conflicting results on the efficacy of macrocyclic lactones (ML) efficacy against internal parasitism. Reproductive performance is crucial for sustained financial success in the beef cattle industry. This dissertation includes a population study that quantified the incidence of Neospora caninum infections in the central region of the United States and tested its relationship with reproductive performance in beef cattle. Trial one of that study concluded that 6.9% of open, replacement heifers (n = 1306) tested seropositive. The second trial in that project found that 9.6% of the breeding age females (n = 500) tested were seropositive for Neospora caninum; and that state in which the cattle lived and age impacted (P < 0.05) infection rate. Breed composition, number of farm dogs on the ranch, and use of total mixed rations were not associated (P > 0.1) with seropositive …

Cattle grazing wheat pasture have the potential to gain BW exceptionally well, but excessive nitrogen intake results in increased excretion and increased greenhouse gas (GHG) emissions. Supplemental concentrates with the addition of an ionophore given to ruminants grazing wheat is a potential practice for producers to increase nitrogen efficiency while decreasing GHG emissions. Therefore, the objective of this experiment was to quantify the effects of energy (2.95 kg/d) and lasalocid (200 mg/hd/d) supplementation on nutrient intake, energy metabolism, respiratory gas fluxes, and performance of grazing cattle. The developing brain is susceptible to extensive neurotoxicity following exposure to sedative/anesthetic drugs (SADs). Every year hundreds of thousands of children around the world are exposed to SADs with no viable non-neurotoxic agents approved for clinical use. Allopregnanolone (AlloP) has well-established sedative effects in adults and neonates. AlloP and many SADs produce sedation/anesthesia through allosteric modulation of GABAA receptors, which is one of two principal mechanisms behind SAD-induced neurotoxicity. The ability to sequence genome‐scale data from herbarium specimens would allow for the economical development of data sets with broad taxonomic and geographic sampling that would otherwise not be possible. Here, we evaluate the utility of a basic double‐digest restriction site–associated DNA sequencing (ddRADseq) protocol using DNAs from four genera extracted from both silica‐dried and herbarium tissue.
Methods
DNAs from Draba, Boechera, Solidago, and Ilex were processed with a ddRADseq protocol. The effects of DNA degradation, taxon, and specimen age were assessed.

Anchored hybrid enrichment (AHE) has emerged as a powerful tool for uncovering the evolutionary relationships within many taxonomic groups. AHE probe sets have been developed for a variety of insect groups, though none have yet been shown to be capable of simultaneously resolving deep and very shallow (e.g., intraspecific) divergences. In this study, we present NOC1, a new AHE probe set (730 loci) for Lepidoptera specialized for tiger moths and assess its ability to deliver phylogenetic utility at all taxonomic levels. Mist netting is the most popular field technique used to estimate avian species abundance. It can, however, provide a biased estimate of species abundance as not all birds are equally likely to be captured. This is partially due to net avoidance, the purposeful evasion of mist nets by birds. Most benthic organisms living in the intertidal zone have planktonic larvae that reside temporarily in the water column before settling in their adult habitats. Larvae aggregate in offshore larval pools, and transport horizontally and vertically in the water to remain in the nearshore and during their pelagic life. While some horizontal transport of larvae can be attributed to advection, behavioral responses, like vertical swimming and buoyancy control, allow larvae to position themselves at depths where flow direction can be exploited. Gout is a chronic arthritis caused by the deposition of poorly soluble monosodium urate monohydrate (MSU) crystals in peripheral joints. Resident macrophages initiate inflammation in response to MSU mediated by NF-κB nuclear translocation and NLRP3 inflammasome activation. We investigated the role of CD44, a transmembrane receptor, in mediating MSU phagocytosis by macrophages. We used an antibody that sheds the extracellular domain (ECD) of CD44 to study the role of the receptor and its associated protein phosphatase 2A (PP2A) in macrophage activation. When two species compete for food, one species typically outcompetes the other. Subordinate species can alleviate costs of competition by reducing diet overlap, promoting coexistence. Non-native coyotes (Canis latrans) and historically native gray foxes (Urocyon cineroargenteus) have expanded their range in Maine and may compete with native red foxes (Vulpes vulpes). To investigate competition, I analyzed stable isotopes and stomach contents to determine if coyotes, red foxes, and gray foxes differed in use of anthropogenic foods and trophic position. El Niño Southern Oscillation (ENSO) is commonly known as the atmospheric and oceanographic powerhouse of the Southern Pacific Ocean. From phytoplankton to apex predators, ENSO controls the stability of species’ populations within this biodiverse ocean environment. El Niño’s 9-15 month alteration of the heat storage in the tropical Pacific drastically shifts the temperature, nutrient, and circulation gradient its marine life is accustomed to. Climate change is posing threats to many ecosystems world wide but will have a more substantial impact on the volcanoes in the tropics because they do not have a seasonally altered climate.
