Instrumental disparities and soil moisture are two of the key limitations in implementing spectroscopic techniques in the field. This study sought to address these challenges through two objectives. The first objective was to assess Visible-near infrared (VisNIR) and mid-infrared (MIR) spectroscopic approaches and explore the feasibility of transferring calibration models between laboratory and portable spectrometers. The second objective addressed the challenge of soil moisture and its impact on spectra. The portable spectrometers demonstrated comparable performance to their laboratory-based counterparts in both regions. This project had two primary goals: (1) to explore opportunities to further a deep-ocean vehicle’s reach using alternative pressure spheres, and (2) to implement an existing deep-ocean vehicle (lander) in active scientific research.

I gained a greater understanding of the limitations and design choices made for existing pressure spheres using Finite Element Analysis (FEA). My simplified FEA model predicted sphere failure for the existing 30% Fiber Glass 70% Nylon injection molded spheres at an external pressure of 3,954psi or 2,690m ocean-depth (only a 7.38% error compared to the tested minimum failure depth), so I determined it a valid model. Eimeria species oocysts cause coccidiosis, one of the costliest diseases in the poultry industry. Coccidiosis infections cause significant economic losses due to reduced performance in chickens and turkeys. The objective of this thesis was to determine fecal bacterial recovery from chlorhexidine (CHX) salt suspensions containing Eimeria maxima M6 oocysts and evaluate the effect of CHX salts on oocysts during long-term cold storage. For bacterial recovery experiments, CHX digluconate and CHX gluconate were tested at different concentrations on poultry fecal slurries during oocyst shedding. Bats are an ecologically important taxon that can host zoonotic pathogens. Globally, many bat species are synanthropic and live closely with humans, often roosting in man-made structures. The spatial overlap between humans and bats creates opportunities for human-bat contact, which can lead to human exposure to bat-borne pathogens and conflicts that cause bat mortality. Social isolation is detrimental to health. Isolation and loneliness are linked to all-cause mortality and accelerated disease progression in patients with cancer, dementia, and vascular disease. Efficient replacement of dead cells in epithelial tissue is crucial for maintaining barrier function and tissue homeostasis. Apoptotic cells can signal to neighboring cells to stimulate proliferation and compensate for cell loss and maintain overall cell numbers in normal physiology and cancer. While dying cells can transmit instructive cues to neighboring cells, the molecular mechanisms that induce cell division are not well understood. The regulation of reproductive physiology in animals is mediated by the Hypothalamus-Pituitary-Gonad (HPG) axis. In seasonal breeders, a conserved reciprocal switching of deiodinase enzymes alters thyroid hormone signaling, which controls HPG activity. This study tested the hypothesis that neural regulation of the HPG axis as described in seasonal systems is conserved and repurposed to respond to other cues in aseasonal species. Using water deprivation and nest restriction in the opportunistic Zebra Finch, we were able to create a breeding group and non-breeding group, confirmed by egg laying and changes in ovarian anatomy. Shallow, river-influenced coastal sediments are important for global carbon storage and nutrient cycling and provide a habitat for diverse communities of invertebrates (infauna). Elevated bed shear stress from extreme storms can resuspend, transport, and deposit sediments, disrupting the cohesive structure of muds, and sorting and depositing sand eroded from beaches. These physical disruptions can also resuspend or smother infauna, decreasing abundances and changing community structure. Infaunal activities such as burrowing, tube construction, and feeding can impact sediment structure and stability. The availability of P in feedstuffs and the effect of feedstuffs, feed additives, and anti-nutritional factors on secretory IgA (sIgA) production in chickens were evaluated. A rapid method was developed using the precision-fed rooster assay to evaluate phytic acid disappearance, P digestibility, and phytase efficacy. Further, exogenous phytase was found to be effective at degrading myo-inositol 1,2,3,4,5,6-hexakis, however, the enzyme was less efficient at P release from lower inositol phosphates. The availability of P in feedstuffs was found to be affected by dietary Ca content, where increasing Ca linearly reduced (P < 0.05) both phytic acid hydrolysis and P digestibility. For example, the standardized ileal digestibility of P was reduced from 73 to 47% in soybean meal when the dietary Ca content was increased from 0.30 to 0.75%. The digestibility of P in distillers dried grains with solubles (DDGS) and corn fermented protein (CFP), however, was high across all Ca levels (71 to 80%). Relative P bioavailability based on tibia bone ash was also found to be high for DDGS and CFP, being 81 and 78%, respectively. Upon further evaluation, it was observed that utilizing low dietary Ca levels (0.30%) when P digestibility in individual plant-based feedstuffs was determined overestimated the P digestibility in practical grower diets by up to 16 percentage units. Utilization of higher Ca levels (0.75%) when P digestibility in individual plant-based feedstuffs was determined, however, resulted in calcualted P digestibility values that were within 1 to 3 percentage units of the measured digestibility in practical grower diets. Aside from P, feedstuffs were also evaluated to determine their effect on sIgA production in the intestine. Roosters excreted approximately 6 to 30 mg of sIgA / 24 h. Further, the proportional contribution of sIgA to total endogenous amino acid (AA) losses ranged from 1 to 13%, with the greatest contribution being observed for Trp. The addition of casein to a protein-free diet increased (P < 0.05) sIgA production in both roosters and broiler chickens. Genetic analysis in model systems using bioinformatic approaches provides a rich context for a concrete and conceptual understanding of gene structure and function. With the intent to engage students in research and explore disease biology utilizing the nematode Caenorhabditis elegans model, we developed a semester-long course-based undergraduate research experience (CURE) in a hybrid (online/in-person) learning environment—the gene-editing and evolutionary nematode exploration CURE (GENE-CURE). Primary cilia from the brain microvascular endothelial cells (ECs) are specialized cell-surface organelles involved in mediating sensory perception, cell signaling, and vascular stability. Immunofluorescence (IF) analysis of human primary brain microvascular ECs reveals two cilia per cell. To confirm the in vitro observation of the two-cilia phenotype in human primary brain ECs, ECs isolated from mouse brain were cultured and stained for cilium. Indeed, brain ECs from a ciliopathic mouse (polycystic kidney disease or Pkd2−/−) also possess more than one cilium. Primary cilium emerges from the mother centriole. Purpose: The metabolic alterations due to chronic hyperglycemia are well-known to cause diabetes-associated complications. Short-term hyperglycemia has also been shown to cause many acute changes, including hemodynamic alterations and osmotic, oxidative, and inflammatory stress. The present study was designed to investigate whether diabetes-associated hyperglycemia can cause rapid-onset detrimental effects on the tear film, goblet cells, and glycocalyx and can lead to activation of an inflammatory cascade or cellular stress response in the cornea.

Liposomes were synthesized using the thin film method. A lyophilized power of egg-derived phosphatidylcholine, stearylamine, and cholesterol were added to ethanol and dried under argon to form a lipid cake. The lipid cake was rehydrated with dPBS and sonicated at 60°C forming a heterogenous batch of liposomes. Our results revealed the average size of the liposomes, determined by Dynamic Light Scattering, was approximately 223.1nm, while demonstrating a weakly positive zeta-potential of 1.9± 8.07mv. Chronic hypoxia may have a huge impact on the cardiovascular and renal systems. Advancements in microscopy, metabolomics, and bioinformatics provide opportunities to identify new biomarkers. In this study, we aimed at elucidating the metabolic alterations in kidney tissues induced by chronic hypoxia using untargeted metabolomic analyses. Reverse phase ultrahigh performance liquid chromatography-mass spectroscopy/mass spectroscopy (RP–UPLC–MS/MS) and hydrophilic interaction liquid chromatography (HILIC)–UPLC–MS/MS methods with positive and negative ion mode electrospray ionization were used for metabolic profiling. In this paper, we propose a new form of multidisciplinary academic collaboration that goes beyond the traditional modes of knowledge exchange. We argue that most research collaboration today is based on interactions between closely related disciplines, in which researchers share data, methods, and insights within a common framework or problem. However, such collaboration may not foster the development of the communication and management skills essential to a multi-disciplinary research career. Therefore, we suggest establishing a network of researchers from divergent, yet complementary, disciplines who are interested in improving these skills through regular interactions and feedback.
