Targeted Limb Heating Augments The Actions Of Igf1 In The Growth Plate And Increases Bone Elongation In Growing Mice, Holly Lyn Racine
Theses, Dissertations and Capstones
Bone elongation disorders can lead to painful musculoskeletal disabilities in adulthood. Existing treatment options to correct left-right asymmetry in limb length include invasive surgeries and/or drug regimens. These are often only partially effective. Previous studies in weanling mice have shown that a daily application of mild heat (40°C) to limbs on one side of the body could be used to noninvasively enhance bone elongation. However, the impact of heat-treatment on bone at the cellular level remains elusive. The epiphyseal growth plate, the band of cartilage located at each end of long bones, is the main site of longitudinal growth and …

Mental Stress From Animal Experiments: A Survey With Korean Researchers, Minji Kang, Ahram Han, Da-Eun Kim, Troy Seidle, Kyung-Min Lim, Seungjin Bae
Morality and Ethics of Animal Experimentation Collection
Animal experiments have been widely conducted in the life sciences for more than a century, and have long been a subject of ethical and societal controversy due to the deliberate infliction of harm upon sentient animals. However, the harmful use of animals may also negatively impact the mental health of researchers themselves. We sought to evaluate the anxiety level of researchers engaged in animal use to analyse the mental stress from animal testing. The State Anxiety Scale of the State-Trait Anxiety Inventory (STAI) was used to evaluate how researchers feel when they conduct animal, as opposed to non-animal, based experiments …
Detection Of Local Steroidogenic Enzyme Gene Expression In Brown Anoles,

Detection Of Local Steroidogenic Enzyme Gene Expression In Brown Anoles, Ada Spahija
Honors Program Theses
The endocrine system in vertebrates responds to stress by releasing steroid hormones, mainly glucocorticoids (GC), which increase blood glucose levels to supply key organs and muscles with energy needed for survival. Steroid hormones are synthesized via an enzymatic pathway that converts cholesterol into either GCs, androgens, or estrogens in a step-wise manner. The adrenal cortex is known to produce GCs, but evidence suggests that individual organs can also produce steroid hormones de novo in response to stress. This study aims to quantify gene expression of four steroidogenic enzymes, encoded by CYP19A1, CYP17A1, StAR, and HSD17ß3 genes, via qRT-PCR to determine …

Development And Preclinical Evaluation Of Long-Lasting Cocaine Hydrolases For Cocaine Overdose And Cocaine Use Disorder Treatment, Ting Zhang
Theses and Dissertations--Pharmacy
Cocaine is a plant-based illicit drug commonly involved in substance use disorder. Although cocaine overdose and cocaine use disorders cause adverse health consequences to individuals and the economic burden on their family and society, there are no FDA (Food and Drug Administration) approved medications for treatment. Recently, it has been recognized that delivery of cocaine hydrolase (CocH) is a promising therapeutic strategy. Human butyrylcholinesterase (hBChE), the primary enzyme involved in cocaine metabolism in human, have advantages over other candidates for the development of CocH. Previous studies in our laboratory have designed and characterized hBChE mutants that have ~4,000-fold improved catalytic …

Disarming “Nature” As A Weapon: A Queer Ecosemiotic Reimagining Of Futurity And Environmental Ethics Through Memoir, Sam Lauer
Master’s Theses
In this thesis, I posit that the need for an active, conscious, and radical queering of ecocriticism as a literary and cultural theory has arisen in light of the postmodern problematization of “nature” and the “natural,” along with the queerness of society, culture, and science. The way we understand “nature” (in life and in texts), whether of physical environments, inherent selfhood, or normalcy, begs to be appropriately informed by discourses and realities of queerness in order for both social and environmental healing to take place. I have analyzed three works of queer creative nonfiction—memoirs—to illuminate the ways in which the …

Possible Breakdown Of Dopamine Receptor Synergism In A Mouse Model Of Huntington's Disease, Samantha F. Kennedy
LSU New Orleans Theses and Dissertations
The model of basal ganglia function proposed by Albin, Young and Penney (1989) describes two anatomically independent motor pathways, the direct and indirect. However, under normal conditions striatal dopamine (DA) is required for the expression of motor behavior, and DAergic control of the two pathways (via D1 and D2 receptors, respectively) is dependent on co-activation. We tested for a possible breakdown of D1/D2 synergism using transgenic R6/1 mice bearing the human huntingtin allele (Htt). Motor stereotypy, observed prior to the onset of HD-related symptoms, was rated on a 5-point scale following activation of: A) D1 receptors alone, B) D2 receptors …

Effects Of Crude Oil On Tumor Suppressor P53 Polymorphisms In Laboratory-Exposed Atlantic Killifish, Fundulus Heteroclitus, Allison Margaret Nadler
Seton Hall University Dissertations and Theses (ETDs)
Polycyclic aromatic hydrocarbons (PAHs), constituents of crude oil, are implicated as a potent source of adverse toxicological effects on living organisms. To model the effects of PAHs in response to environmental oil spill disasters a species of killifish (Fundulus heteroclitus) was captured and exposed to crude oil in a laboratory-controlled setting. Over a period of 7 days, fish were dosed with crude oil by gavage, culled, and organs were harvested for analysis. Excitation-emission matrix spectroscopy (EEMS) of gall bladder tissue homogenates was used to verify exposure. Effects of PAHs on the p53 gene were evaluated as an indicator …

Small-conductance Ca2+-activated K+ (SK) channels play essential roles in the regulation of cellular excitability and have been implicated in neurological and cardiovascular diseases through both animal model studies and human genetic association studies. Over the past two decades, positive modulators of SK channels such as NS309 and 1-EBIO have been developed. Our previous structural studies have identified the binding pocket of 1-EBIO and NS309 that is located at the interface between the channel and calmodulin. In this study, we took advantage of four compounds with potencies varying over three orders of magnitude, including 1-EBIO, NS309, SKS-11 (6-bromo-5-methyl-1H-indole-2,3-dione-3-oxime) and …

Effects Of Trophic Relationships On Oyster Reef Restoration Success In The Mississippi Sound, Virginia Robin Fleer
Dissertations
Natural and anthropogenic changes resulting from altered hydrology, hurricanes, variable precipitation, and the BP oil spill have all taken their toll on oyster reefs in Mississippi. In response, oyster reef restoration efforts are currently underway within the Northern Gulf of Mexico. In order to understand why these efforts succeed or fail, it is crucial to consider predator-prey relationships within the context of the trophic dynamics of oyster reefs. Thus, for this dissertation study I integrated a multidisciplinary approach to understanding key trophic interactions affecting oyster recruitment, growth and survival, comprising field sampling, manipulative lab experiments, and individual-based modeling. Spat settlement …

Signature Whistle Production During A Bottlenose Dolphin Group Integration, Megan S. Broadway
Dissertations
Bottlenose dolphins are an important species of interest because they possess a variety of abilities that are relatively rare in the animal kingdom, one being complex acoustic communication. Signature whistles - distinctive calls that are unique for each individual – are one of the most studied call types, but we know little about how these calls are used in various contexts, such as during an introduction. Looking at the socio-behavioral context in which signature whistles are used is likely the best way of learning how these whistles are used in a particular context (Caldwell, Caldwell, & Tyack, 1990). For this …

The competing needs of maintaining productivity within abattoirs, and maintaining high standards of animal welfare, provide fertile grounds for applied research in animal behavior. However, there are challenges involved in capturing useful behavioral data from the supply chain (from paddock to processing plant). The challenges identified in this report are based on a review of the scientific literature as well as field study observations. This article describes those challenges as they relate to collecting behavioral data on livestock-herding dogs, humans and livestock as they interact in abattoirs, and provides insights and recommendations for others embarking on animal studies in confined …

The hippocampus is critical for episodic memory, and synaptic changes induced by long-term potentiation (LTP) are thought to underlie memory formation. In rodents, hippocampal LTP may be induced through electrical stimulation of the perforant path. To test whether similar techniques could improve episodic memory in humans, we implemented a microstimulation technique that allowed delivery of low-current electrical stimulation via 100 μm-diameter microelectrodes. As thirteen neurosurgical patients performed a person recognition task, microstimulation was applied in a theta-burst pattern, shown to optimally induce LTP. Microstimulation in the right entorhinal area during learning significantly improved subsequent memory specificity for novel portraits; participants …

Ependymal cilia protrude into the central canal of the brain ventricles and spinal cord to circulate the cerebral spinal fluid (CSF). Ependymal cilia dysfunction can hinder the movement of CSF leading to an abnormal accumulation of CSF within the brain known as hydrocephalus. Although the etiology of hydrocephalus was studied before, the effects of ethanol ingestion on ependymal cilia function have not been investigated in vivo. Here, we report three distinct types of ependymal cilia, type-I, type-II and type-III classified based upon their beating frequency, their beating angle, and their distinct localization within the mouse brain-lateral ventricle. Our studies …

Elucidating The Mechanical, Structural, Functional, And Molecular Mechanisms Involved In Irreversible Vascular Changes In Aortic Coarctation, Brandon Wegter
Master's Theses (2009 -)
Coarctation of the aorta (CoA) is a constriction of the thoracic aorta and is one of the most common congenital cardiovascular defects. Treatment by surgical correction has saved the lives of thousands of children, but many still have a reduced lifespan due to hypertension. Previous results using our novel rabbit model showed that the current treatment guideline of a 20 mmHg blood pressure gradient (BPG) induces irreversible vascular changes, which persisted despite correction. Preliminary data of the downregulation of natriuretic peptide receptor C (NPR-C) in proximal aortic tissue of human patients with CoA serves as the possible underlying mechanism for …

Habitat destruction and anthropogenic activities cause wild birds to migrate towards urban areas in search of food and sometimes nesting in high-roof buildings, where they are caught by local people using different techniques and are further used for hunting or recreational purposes. An injured Golden Eagle (Aquila chrysaetos) was found entangled in bushes near the vicinity of Balkasar Research Complex, Chakwal, Pakistan. After complete physical examination, a wound on the right wing and closed leg fracture was diagnosed. The successful treatment was done by anti-parasitic and anti-bacterial drugs with careful monitoring. Splint was adjusted after aligning the fractured …

In a recent publication in Lab Animal,1 a nonhuman primate (NHP) researcher at the California National Primate Research Center, USA, argued for the continued importance of experiments on macaques for better understanding HIV and AIDS, and for tackling the virus and the disease with therapies and vaccines. Though the stance of the author is far from surprising, given his background and current employer (an organisation which houses around 5000 monkeys for research), the paper, even as comprehensive as it is, is not sufficiently critical of NHP experiments and overlooks a number of their deficiencies and established problems with them. Given …
