Tobacco is one of the most widely used substances across the United States; tobacco consumption is linked with several diseases and is one of the leading causes of preventable death in the country. Nicotine is the active substrate found in tobacco products, acting as a stimulant, which reinforces continued use. The introduction of tobacco-free options such as e-cigarettes and oral pouches has driven an increase in nicotine exposure across the population, especially in young adults. Acute nicotine use induces neuroadaptations in the mesolimbic dopaminergic system, which mediates reinforcement learning and addiction, that potentiates reward-related neurotransmission. Hormonal contraceptives contain a synthetic estrogen (e.g., ethinyl estradiol/EE) and/or a synthetic progestin (e.g., levonorgestrel/LEVO) and can exacerbate nicotine intake. However, whether altered intake reflects changes in nicotine reinforcement or enhancement by nicotine of co-occurring reinforcers is unknown. This reinforcer-enhancement effect is evidenced in rats when nicotine self-administration is increased by the presence of a visual stimulus (VS) reinforcer. This dissertation examined the effects of EE and LEVO on nicotine reinforcement and reinforcer-enhancement in ovary-intact female Sprague-Dawley rats. Nicotine is commonly considered to be the primary addictive constituent in tobacco products. Therefore, research focused on nicotine and its effects is necessary for understanding nicotine dependency and in developing potential cessation treatments. Research has confirmed the relevance of learning processes associated with nicotine and their influence on the development of addiction. Conditioning involving interoceptive stimuli is increasingly cited as playing a role in many diseases, psychopathologies, and drug addiction. Adolescence is a vulnerable developmental period in regards to drug initiation and use. The gateway hypothesis suggests that adolescent cigarette smoking may result in a heightened risk for methamphetamine use. However, little is understood about the role of nicotine on adolescent methamphetamine addiction. The aim of the present study was to determine whether early, late, or continuous adolescent nicotine exposure would alter oral methamphetamine self-administration, extinction, or reinstatement. Addictions, both substance and behavioral, have been conceptualized as having similar biopsychosocial processes with different opportunistic expressions (Shaffer et al., 2004). Biological processes such as the hormonal response to stress as measured by cortisol, dehydroepiandrosterone (DHEA), and the ratio of the DHEA/cortisol may be among the variables underlying the disposition to develop an addictive disorder, regardless of whether it is a substance-based or a so-called behavioral addiction. It is presently unclear whether ovarian hormones, such as estradiol (E2) promote the reinforcing effects of nicotine in females. Thus, we compared extended access to nicotine intravenous self-administration (IVSA) in intact male, intact female, and OVX female rats (Study 1) as well as OVX females that received vehicle or E2 supplementation (Study 2). The E2 supplementation procedure involved a 4-day procedure involving 2 days of vehicle administration and 2 days of E2 administration. Research has revealed that schizophrenics are significantly more likely to smoke cigarettes than the general population, and consume nicotine products at a much more prevalent rate. Further exacerbating this issue, it has been previously demonstrated in clinical populations that the type of antipsychotic treatment administered (typical versus atypical) may result in either an increase or a decrease of already heightened smoking behavior within the schizophrenic population. One of the most common childhood disorders, attention-deficit hyperactivity disorder (ADHD) places individuals at a higher risk for nicotine (NIC) dependence. Approximately 37.2% of individuals with ADHD currently smoke compared to the 18.3% of individuals with no record of mental illness. Methylphenidate (MPH; Trade name Ritalin) is the most commonly prescribed treatment for ADHD. Research regarding the synergistic effects of MPH and NIC, however, is divided. Some research indicates that MPH may enhance susceptibility to NIC effects, whereas other studies report that MPH may inhibit sensitization to NIC. Smoking and tobacco dependence are serious health concerns in the United States and globally. Reward via the pharmacological effects of nicotine are believed to be the principal motivating factor that drive tobacco dependence. Research reveals differences in sensitivity between males and females to the motivational effects of nicotine in tobacco use. Enhancement of reinforcement value of non-nicotine rewards contributes to overall nicotine reward. Similar value-enhancing effects have been observed by the two most commonly prescribed smoking cessation aids, bupropion and varenicline. Nicotine has multiple behavioral effects as a result of its action in the central nervous system. Nicotine strengthens the behaviors that lead to nicotine administration (primary reinforcement), and this effect of nicotine depends on mesotelencephalic systems of the brain that are critical to goal directed behavior, reward, and reinforcement. Nicotine also serves as a ‘reinforcement enhancer’ – drug administration enhances behaviors that lead to other drug and nondrug reinforcers. Nicotine has both unconditioned and conditioned stimulus properties. Conditioned stimulus properties of nicotine may contribute to the tenacity of nicotine addiction. The purpose of this experiment was to use neurohistochemical analysis of rapidly developing c-Fos protein to elucidate neurobiological loci involved in the processing of nicotine as an interoceptive conditioned stimulus (CS). Objective: Iptakalim is a putative ATP-sensitive potassium (KATP) channel opener. It is also a novel nicotinic acetylcholine receptor (nAChR) blocker and can antagonize nicotine-induced increase in dopamine release in the nucleus accumbens. Our recent work also shows that iptakalim exhibits a clozapine-like atypical antipsychotic profile, indicating that iptakalim may possess a dual action against nicotine addiction and schizophrenia.
Methods: The present study examined the potential therapeutic effects of iptakalim on nicotine use in schizophrenia.
