Hormonal contraceptives contain a synthetic estrogen (e.g., ethinyl estradiol/EE) and/or a synthetic progestin (e.g., levonorgestrel/LEVO) and can exacerbate nicotine intake. However, whether altered intake reflects changes in nicotine reinforcement or enhancement by nicotine of co-occurring reinforcers is unknown. This reinforcer-enhancement effect is evidenced in rats when nicotine self-administration is increased by the presence of a visual stimulus (VS) reinforcer. This dissertation examined the effects of EE and LEVO on nicotine reinforcement and reinforcer-enhancement in ovary-intact female Sprague-Dawley rats. The capacity of temporal interference (TI) stimulation to target deep brain regions without affecting nearby surface electrodes remains uncertain. Using artifact-free optical recording, we compare excitation patterns and thresholds in hippocampal neurons stimulated by “pure” and amplitude-modulated sine waves, representing TI waveforms near electrodes and at the target, respectively. We show that pure 2- and 20-kHz sine waves induce repetitive firing at rates that increase up to 60–90 Hz with stronger electric fields. Beyond this limit, action potentials merge into sustained depolarization, resulting in an excitation block. Modulating the sine waves at 20 Hz aligns firing with amplitude “beats” and …

Traumatic brain injury (TBI)-related growth hormone deficiency is often undertreated, despite documented physical, metabolic, and neuropsychiatric effects. Insulin-like growth factor (IGF-1), with neuroreceptors located in brain regions responsible for learning, memory, and mood, regulates cerebral blood flow, neurogenesis, and neuroplasticity. The aim of this study was to determine associations between IGF-1 levels and post-TBI symptom severity, anxiety, and depression. This retrospective observational study at an Academic Brain Injury Center included participants evaluated 3-12 months post-TBI with available IGF-1 values and complete Rivermead Post-Concussion Symptoms Questionnaire (RPQ-13), Generalized Anxiety Disorder-7 (GAD-7), and Patient Health Questionnaire-9 (PHQ-9) responses. This study investigated the lived experiences of individuals living with HIV (PLWH) in the Dallas-Fort Worth (DFW) area of Texas during the COVID-19 pandemic, using an Interpretative Phenomenological Analysis (IPA) framework. The research examined the intersection of health, social dynamics, and psychological resilience among PLWH, who faced compounded vulnerabilities due to systemic inequities and the dual public health crises of HIV and COVID-19. Through semi-structured interviews with ten participants from diverse backgrounds, the study captured the profound impacts of disrupted healthcare systems, heightened stigma, and social isolation on this marginalized population. Early life adversity has been shown to have a variety of negative effects on adult functioning, including parenting quality, with implications for child wellbeing. Adverse early life experiences including poverty have been linked to long-term disruptions in emotional, cognitive, and physiological functioning that compromise the ability to provide sensitive, nurturing care. The present study investigated whether emotional regulation difficulties, physiological regulation, and cognitive function mediate the relationship between early life financial insecurity and parenting quality. A sample of 132 mother-child dyads completed self-report measures, biological samples, and cognitive assessments. A multiple mediation model ran in SPSS, revealed a significant indirect …

This thesis aims to explore how racial health disparities, underrepresentation in gynecology, and socioeconomic barriers contribute to adverse pregnancy outcomes among women of color in New York City. Through an interdisciplinary approach that combines resources from medical anthropology and human biology, this research will examine the biological implications of chronic stress, specifically HPA axis dysregulation, and its role in maternal health outcomes. Childhood poverty has been shown to increase adult risk for obesity above and beyond its direct effects on adult socioeconomic status (SES). One proposed mechanism of these effects is by shifting behavioral patterns of dietary consumption and choice, for example by increasing rapid attention to high-calorie unhealthy foods. Yet, whether such neural mechanisms can explain observed differences in dietary behavior based on childhood SES remains an open question. Substance use disorders impose a staggering toll on the United States (U.S.), straining healthcare systems, destabilizing communities, and profoundly impacting both individuals and families. Approximately one in six Americans aged 12 and older had a past-year substance use disorder in 2022 (Key Substance Use, 2022), and nearly one-third of adults experience a substance use disorder in their lifetime (Wu, 2010), contributing to an economic burden exceeding $400 billion annually (National Drug Threat Assessment, 2011). Perineuronal nets (PNNs) are specialized extracellular matrix structures that surround certain types of neurons in the central nervous system. Research from the past few decades has cemented PNNs as an integral player in the closure and onset of critical periods, memory modulation, and neural plasticity. These effects are thought to occur when PNNs form around a neuron and structurally stabilize its synaptic connections. Vagally mediated heart rate variability (vmHRV) reflects top-down cognitive processes involved in emotion-cognition integration. Using cognitive control can be effortful and increase negative affect. However, this intrinsic affective component of cognitive control has not been well studied, and the role of vmHRV in the association between subjective experience in using cognitive control and behavioral performance remains unknown. Introduction: Neonatal hypoxic-ischemic encephalopathy (HIE) is neurological disease caused by the deprivation of oxygen and blood flow to the brain during the developmentally-critical perinatal period. Therapeutic hypothermia (TH) is the standard of care treatment for HIE, though cognitive deficits can persistent throughout life despite treatment. The nature of these deficits, and the impacts of TH and sex are not well understood, and this presents a key barrier in the development of novel therapeutics.

Objective: The effectiveness of out of hospital lifestyle interventions in healthcare is likely co-dependent on social support from involved one's. We sought to investigate the interrelationships between patient-partner Co-responsibility, Spousal Support, and Self-efficacy and the mechanisms through which they contribute to health behaviors and outcomes relevant to weight loss and maintenance. Methods: Co-responsibility, Spousal Support, self-efficacy, health outcomes and health behaviors like total weight loss, physical activity, OBESI-Q, and health satisfaction were assessed through a questionnaire among 868 individuals attempting to lose weight. Neuroimaging studies have revealed consistent abnormalities in functional connectivity within specific neural networks that may serve as biomarkers for major depressive disorder (MDD). It is important to find inexpensive, non-invasive techniques that target these biomarkers to make diagnosis easier and more objective. EEG microstates are quasi-stable potential topographies that are thought to reflect the quasi-stable network activity of the underlying neural generators. MDD has been shown to alter features of the four canonical EEG microstates (A, B, C, D) with some conflicting results. Poland syndrome is a rare disorder that unilaterally affects the chest, arms, and hands. Despite being a limb disorder, there has not been research done on how Poland syndrome interacts with the sensorimotor cortex. Other limb disorders have documented connections to the sensorimotor cortex, and can be used to infer how Poland syndrome affects the sensorimotor cortex. Limb disorders with extensive research on the sensorimotor cortex are phantom limb syndrome, focal hand dystonia, and congenital one-handedness. Aerobic metabolism is known to generate damaging ROS, particularly hydrogen peroxide. Reactive oxygen species (ROS) are highly reactive molecules containing oxygen that have the potential to cause damage to cells and tissues in the body. ROS are highly reactive atoms or molecules that rapidly interact with other molecules within a cell. Intracellular accumulation can result in oxidative damage, dysfunction, and cell death. Due to the limitations of H2O2 (hydrogen peroxide) detectors, other impacts of ROS exposure may have been missed. Women overrepresent men for sub-optimal sleep, a consequence of hormone fluctuation in the menstrual cycle affecting sleep regulatory pathways. While research has examined the prevalence of sub-optimal sleep through cycle phases, little research has examined how hormonal contraceptives (HC’s) could similarly affect women’s sleep, while also neglecting to utilize subjective sleep measures. In this study, we examine subjective sleep quality among naturally cycling (NC) women, women using different HC types, and between active and inactive phase pill users by subjecting 463 women to a subjective sleep battery. High-stress situations, like police interrogations, can hinder an individual's decision-making and critical-thinking ability (Liston et al., 2009), making them more susceptible to suggestive or leading messages during questioning. While research on suggestibility mainly focuses on children and adolescents, young adults may also be vulnerable to suggestion because of their sensitivity to social feedback (Blakemore & Mills, 2014; Redlich & Goodman, 2003). Mild traumatic brain injuries (mTBIs) often lead to acute symptoms like disorientation and discoordination, but the underlying mechanisms are unknown. Although most patients recover quickly from a single mTBI, repeated injuries can be debilitating. After an mTBI, a neurometabolic cascade produces metabolic burden and vulnerability. Spreading depolarizations (SDs) have been observed after mTBIs. Here we investigated SDs’ role in short-term motor behavioral deficits post-mTBI. We hypothesized that SDs contribute to the deficits, and exacerbated symptoms after repeated mTBIs. Major depressive disorder (MDD) is a mood disorder characterized by persistently depressed mood and/or anhedonia that causes significant functional impairment in daily life. MDD affects nearly 15% of adults in their lifetime, and since 2019, prevalence estimates have increased up to ~30% in young adults (18-25 years old). In addition to its debilitating effects on daily functioning, epidemiological studies link MDD to ~3 times greater risk of cardiovascular disease (CVD), followed by accelerated CVD progression and mortality. Although the mechanisms contributing to MDD-CVD comorbidity are unclear, there is evidence of stress system dysfunction in adults with MDD.

Asthma is a common lung disease that impacts lung functioning through inflammatory based mechanisms. Past research suggests that decreased blood oxygenation due to asthma attacks may impair cognitive capabilities (Irani et al., 2017). Moreover, the observed differences in cognition between those with and without asthma may be associated with disease severity or asthma control respectively in asthma populations. The current study explored differences in cognitive functioning between college students with and without self-reported asthma. Background
Gene-environment interaction (G X E) is likely an important influence shaping individual differences in alcohol misuse (AM), yet it has not been extensively studied in molecular genetic research. In this study, we use a series of genome-wide gene-environment interaction (GWEIS) and in silico annotation methods with the aim of improving gene identification and biological understanding of AM.
Methods
We carried out GWEIS for four AM phenotypes in the large UK Biobank sample (N = 360,314), with trauma exposure and socioeconomic status (SES) as moderators of the genetic effects. Exploratory analyses compared stratified genome-wide association (GWAS) and GWEIS modeling approaches. …

Background
Large-scale cohort and epidemiological studies suggest that PTSD confers risk for dementia in later life but the biological mechanisms underlying this association remain unknown. This study examined this question by assessing the influences of PTSD, APOE ε4 genotypes, DNA methylation, and other variables on the age- and dementia-associated biomarkers Aβ40, Aβ42, GFAP, NfL, and pTau-181 measured in plasma. Our primary hypothesis was that PTSD would be associated with elevated levels of these markers.

The heart–brain interaction is the main mechanism for maintaining normative physiological processes, and its dysregulation underlies the somatic symptoms of various mental disorders. Cortical inhibition, triggered by afferent signals from baroreceptor activation, induces systematic variations in sensorimotor responses within a cardiac cycle, with reaction times (RTs) slower at cardiac systole compared to diastole (known as cardiac cycle time effects). However, recent data suggest that baroreceptor afferents not only inhibit simple responses but also facilitate complex sensorimotor responses during cardiac systole. Background: Depression has been shown to have adverse effects on blood pressure (BP) and is associated with high blood pressure variability (BPV). In turn, high short-term BPV has been related to eventual cardiovascular risk. But it is not clear how early in adulthood the detrimental effects of depression on BPV may be discerned, if being at high risk for depression also compromises BPV, and whether the clinical features of depression moderate its adverse effects. Neurological disabilities cause diverse health and mental challenges, impacting quality of life and imposing financial burdens on both the individuals diagnosed with these conditions and their caregivers. Abnormal brain activity, stemming from malfunctions in the human nervous system, characterizes neurological disorders. Therefore, the early identification of these abnormalities is crucial for devising suitable treatments and interventions aimed at promoting and sustaining quality of life. 4-10% of women who use hormonal oral contraceptives (HOCs) discontinue use after experiencing symptoms of anxiety, irritability, and depression in the first 3 months of administration. Previous studies have acknowledged a relationship between HOCs and emotional dysregulation but have not identified mechanisms by which HOCs have this effect. As a result, effective treatment options for HOC-induced mood disorders are lacking. Current theories implicate combined-HOCs, particularly those containing ethinyl estradiol and levonorgestrel (EE-LNG), in reducing neuroactive steroid levels. Specifically, HOC users show decreases in allopregnanolone, a positive allosteric modulator associated with neuronal inhibition and emotional regulation. The objective of this study …
