Sex Differences In Lateral Hypothalamic Extracellular Glucose Concentrations During Cumulative Dosing Of Methamphetamine In Rats, Joshua A. Jolton

The present study experimentally investigated sex differences in lateral hypothalamic glucose concentrations following cumulative dosing of methamphetamine. Male (n=17) and female (n=11) rats were surgically implanted with enzyme-based glucose biosensors in the lateral hypothalamus. Rats then received a saline injection followed by four methamphetamine doses (0.025 mg/kg, 0.05 mg/kg, 0.1 mg/kg, and 0.2 mg/kg) spaced 55 minutes apart. We found that while there was a general decrease in glucose baseline throughout the study for both sexes, the magnitude of the decrease was significantly greater in females compared to males. This paper clarifies the conceptual space of discussion of legal insanity by considering the virtues of the ‘medical model’ model that has been used in Norway for almost a century. The medical model identifies insanity exclusively with mental disorder, and especially with psychosis, without any requirement that the disorder causally influenced the commission of the crime. We explore the medical model from a transdisciplinary perspective and show how it can be utilised to systematise and reconsider the central philosophical, legal and medical premises involved in the insanity debate. In the target article Crump et al. present 8 criteria to assess whether decapods experience pain. Four of these -- sensory integration, motivational trade-offs, flexible self-protection, and associative learning -- could be used to assess sentience in general. In this commentary I discuss difficulties with using these criteria to provide evidence of sentience in decapods, particularly if this evidence is to change public opinion and policies. The complexity of human reproductive behavior has necessitated its examination through a variety of scientific disciplines, each focusing on specific elements of our biology, behavior, and society. However, this complexity also necessitates that we reintegrate the information learned from each discipline into a single framework, one rooted in the evolutionary principles that have shaped the development of all life on earth. This study utilized a novel design to investigate the sensitivity of D2 dopamine receptors to modulating compounds through multiple exposures over early development of zebrafish larvae. Zebrafish were dosed for 30 minutes from 5-8 days post fertilization (dpf) with 16µ/mol of either a D2 antagonist, haloperidol, or a D2 agonist, quinpirole hydrochloride. Two other groups were then dosed with these compounds from 9-12dpf. The effects of D2 receptor modulation were measured by analyzing motor activity on measures of movement distance, frequency, and velocity. Results indicated that larvae dosed with haloperidol on 5dpf had increased activity after the first dosage, but …

Parkinson’s disease (PD) is estimated to impact nearly 10 million people globally and is estimated to increase in the future. PD is a progressive neurodegenerative disease that worsens through continuous cell death of dopaminergic neurons. This cell death can create motor symptoms such as bradykinesia, tremor, and muscular rigidity. Subthalamic nucleus deep brain stimulation, STN DBS, is a surgical intervention which places stimulating electrodes in the STN greatly improving motor symptoms. The ability to represent approximate quantities appears to be phylogenetically widespread, but the selective pressures and proximate mechanisms favouring this ability remain unknown. We analysed quantity discrimination data from 672 subjects across 33 bird and mammal species, using a novel Bayesian model that combined phylogenetic regression with a model of number psychophysics and random effect components. This allowed us to combine data from 49 studies and calculate the Weber fraction (a measure of quantity representation precision) for each species. We then examined which cognitive, socioecological and biological factors were related to variance in Weber fraction. We found contributions of phylogeny …

Memory dysfunction is a common symptom of aging, neuropsychiatric disorders, and neurodegenerative disorders, yet truly effective treatments for memory loss do not exist. De novo gene transcription is a molecular requirement for long-term memory formation. The transcription of genes related to synaptic plasticity and learning are regulated in part by histone acetylation, an epigenetic mechanism that regulates chromatin accessibility. Pharmacological compounds that maintain histone acetylation, called histone deacetylase inhibitors (HDACi), enhance memory by preventing deacetylation of core histone proteins, which initiates binding of transcriptional machinery to open chromatin. It has been suggested that the human species may be undergoing an evolutionary transition in individuality (ETI). But there is disagreement about how to apply the ETI framework to our species, and whether culture is implicated as either cause or consequence. Long-term gene–culture coevolution (GCC) is also poorly understood. Some have argued that culture steers human evolution, while others proposed that genes hold culture on a leash. The present study investigates the presence of a molecular genetic basis of the quality of aspiration and motivation for behavior described by Ryan and Deci’s self-determination theory and Gray’s biopsychological theory of personality respectively. In this study, investigation was carried out to determine the impact of DRD4 receptor gene polymorphism on intrinsic and extrinsic aspiration as well as approach motivation using the Aspiration Index scales and Behavioral Inhibition and Activation (BIS/BAS) scales as our psychometric instrument of evaluation. Post-traumatic stress disorder (PTSD) is a trauma-related disorder characterized by intense fearful memory formation. Women are twice as likely as men to develop PTSD, indicating there may be sex differences in the underlying neural circuits. Given that serotonin (5-HT) dysfunction is implicated in PTSD, and 5-HT modulates fear learning, we investigated whether there are sex differences in the modulation of fear learning by 5-HT. We administered the selective serotonin reuptake inhibitor (SSRI) citalopram (20mg/kg or 10m/kg, i.p.) once to male and female mice prior to auditory fear conditioning and tested the effects on fear memory the next day. Anxiety disorders are among the most common mental health diagnoses, affecting about a third of the population in their lifetime. However, approximately a third of individuals with anxiety do not respond to current treatment approaches, highlighting the need to identify additional potential therapeutic mechanisms. Safety learning is one such mechanism, but methodological challenges and a dearth of research have prevented the field from advancing the understanding of the role of safety learning in the etiology and remediation of anxiety disorders. Darwin offered an evolutionary perspective on the origins of human morality, suggesting that humans share a biological foundation with nonhuman primates. This paper reviews the current literature on moral and prosocial behaviors of nonhuman primates, specifically examining whether nonhuman primates exhibit behaviors that are typical of empathy and fairness. The literature documents that nonhuman primates exhibit empathetic behaviors regarding emotional contagion and sympathetic concern. There is also evidence that nonhuman primates have a sense of fairness, seen in their reciprocal behaviors and aversion to inequity. Hypofunctionality at the N-Methyl-D-aspartic acid receptor (NMDAR) is a commonly used model of the neurodevelopmental disorder schizophrenia due to the complex circuitry changes that follow NMDAR blockade. While these animal models are very popular for modeling the cognitive deficits seen in schizophrenia, actual treatments for this disorder remain sparse. Childhood maltreatment compromises healthy development, impacts neurobiology, and is associated with lasting alterations to emotional perception, processing, and regulation. Most significantly, childhood maltreatment increases the risk for later development of emotional disorders such as major depressive disorder (MDD). The stress associated with both childhood maltreatment and MDD can lead to lasting alterations to the fronto-limbic circuitry. Using functional and structural magnetic resonance imaging, researchers have observed hippocampal atrophy and amygdala hyperresponsiveness in participants who’ve experienced both childhood maltreatment and MDD. The present studies were aimed to better understand developmental contributions to the risk for disordered drinking, and facilitate the long-term goal of developing effective interventions for individuals at high risk for alcohol use disorders. Experiment 1 assessed the effect of adolescent pre-exposure to ethanol on adult place preference, as well as, sex- and beta-endorphin(bE)-related contributions. Previous studies have suggested that heterosexual women, but not heterosexual men, find same sex beauty rewarding. This finding has been attributed to a “greater bisexual interest among heterosexual women”, but no other explanations have been offered or tested. The current study aimed to explore social comparison tendencies as a potential alternate explanation to this previously observed finding. While the stigma for mental illnesses has greatly declined in the last decade, there is still a disconnect between individuals without neurological illnesses and those with neurological illnesses, especially those that cause individuals to lose contact with reality. The goal of this interdisciplinary paper is to create empathy for these individuals, specifically people with schizophrenia, Alzheimer disease, and post-traumatic amnesia.
