This dissertation examined the potential integration of exercise therapy and psychotherapy as a treatment approach for mood-related difficulties experienced by former elite athletes during the transition out of competitive sport. Retirement from athletics is often associated with significant biopsychosocial challenges, including disruptions in identity, healthy routines, social support, and emotional well-being. Existing literature suggests that exercise may serve as an effective adjunctive intervention for improving mood and overall psychological functioning, though limited research has explored its application among retiring athletes within psychotherapeutic contexts. Acute And Chronic Oral Administration Of Cannabidiol-Dominant Cannabis Products: Sex- And Formulation-Dependent Effects On Anxiety-Like Behavioral Patterns In Rats, Carlos R. Maestas-Olguin

Interest in the anxiolytic-like effects of cannabidiol (CBD), the non-intoxicating phytocannabinoid found within the genus, Cannabis, has grown significantly in recent years. However, preclinical investigations of CBD’s anxiolytic potential often elect to forgo translationally relevant models that maximize external validity in favor of greater internal and mechanistic validity. As a result, the effects of relatively low dose (< 3mg/kg) oral CBD administration on anxiety-like behaviors are not yet fully characterized. The present studies examined the impact of two orally delivered CBD-dominant cannabis products – a “full-spectrum” formulation derived from the whole plant and a CBD isolate derived from the same chemovar – on innate defensive behaviors exhibited by adult male and female rats in various tests of anxiety. Results indicated that acute and chronic low dose oral CBD administration attenuated anxiety-like behavior in rats, regardless of sex or formulation. These findings extend the growing corpus of literature describing anxiolytic-like effects of CBD.

Fetal Alcohol Spectrum Disorders involve morphologic and neurobiological abnormalities in offspring subjected to prenatal alcohol exposure. Although not as well studied as high exposure, moderate prenatal alcohol exposure represents the most common and undetected form in humans. While learning and memory deficits have been well characterized at the behavioral level in humans with Fetal Alcohol Spectrum Disorders and in animal models of moderate prenatal alcohol exposure, few studies have determined the circuit or systems level mechanisms of the behavioral deficits. Single treatments with psychedelics produce rapid, long-lasting antidepressant effects. Psychedelics may also exert anxiolytic effects and be useful in treating more complex psychiatric conditions, such as PTSD. However, less is known about the impact of these agents on PTSD-related symptoms. Thus, we aimed to determine whether a single administration of psilocybin could reverse PTSD-like sequelae induced by stress-enhanced fear learning in rats. Single treatments with psychedelics have demonstrated rapid, long-lasting antidepressant effects. However, psychedelics often produce unfavorable psychoactive effects that could worsen existing psychological distress. Thus, we compared the effects of psilocybin and the related tryptamine, norbaeocystin, on anxiety-like behavior in male and female rats. In Experiment 1, rats were treated with 1 mg/kg psilocybin (PSIL), 1 mg/kg norbaeocystin (NOR), or vehicle 30 min before a 5-min trial on an elevated plus maze (EPM) or in an open field test (OFT). Most individuals consume caffeine on a daily basis. Research has shown that caffeine stimulates the release of glucocorticoids and catecholamines and can enhance cognition. Caffeine also amplifies stress-induced increases in arousal and aggravates PTSD-like symptoms. Thus, we examined the impact of caffeine on memory for a stressful experience and intrusive memories related to that experience. Undergraduate students ingested caffeine or a placebo. Thirty minutes later, they were exposed to the Trier Social Stress Test (TSST; stress condition) or friendly-TSST (f-TSST; control condition), both of which were designed to enable participant memory for the experiences to be quantified. Oral contraceptives (OCs) are often initiated during adolescence and emerging adulthood, a period of continued maturation of the hypothalamic–pituitary–gonadal (HPG) axis. Because hormonal exposure during this window may shape emotional, biological, and reproductive outcomes, it is important to understand the specific formulations young women take and the hormonal profiles these formulations produce. This study characterizes OC formulation features and measured hormone levels in young adult OC users (n = 30).

Ethinyl estradiol (EE) doses ranged from 0.01–0.035 mg (mean = 0.022 mg), with 70% using ≤0.02 mg. Pill regimens included 21/7 packs (83%), 24/4 packs (10%), and 84/7 regimens (3%). …

Gambling advertising has become increasingly prevalent across various media platforms such as TV, social media, real-life advertising, etc. With advertisements designed to influence attitudes, intentions, and behaviors of potential and existing consumers, this review aims to compile existing research to examine the measurable effects of gambling advertising. Studies indicate that young people and problem gamblers are especially vulnerable to these advertisements, which normalize gambling and contribute to increases in gambling behavior. This effect on behavior is dose-dependent; increased exposure to advertising is associated with greater gambling engagement. In a pre-registered online study, we investigated the relationship between ostracism and overclaiming. Ostracism is the act of deliberately or unintentionally excluding and ignoring others. Overclaiming refers to the behavior of exaggerating one’s knowledge or familiarity with specific topics, often as a way of attempting to present oneself as more knowledgeable or competent than one actually is. Drawing from the temporal need-threat model, we hypothesized that ostracized individuals, as a compensatory response, would likely overclaim more compared to included individuals. Opioid use disorder occurs alongside impaired risk-related decision-making, but the underlying neural correlates are unclear. We developed an approach-avoidance conflict task using a modified conditioned place preference procedure to study neural signals of risky opioid seeking in the prefrontal cortex, a region implicated in executive decision-making. Following morphine conditioned place preference, rats underwent a conflict test in which fear-inducing cat odor was introduced in the previously drug-paired side of the apparatus. Fear-related disorders are often treated with exposure therapy, a technique based on the concept of extinction. However, this type of therapy is ineffective for many individuals, so finding a pharmacological adjunct that augments the extinction of fear could lead to better treatment outcomes. In recent preclinical work, investigators have shown that psychedelics can accelerate the extinction of fear, potentially through their impact on neurotrophic signaling. We previously found that psilocybin exerted sex-dependent effects on the extinction of conditioned fear to an isolated cue (i.e., a tone). Hormonal contraceptives contain a synthetic estrogen (e.g., ethinyl estradiol/EE) and/or a synthetic progestin (e.g., levonorgestrel/LEVO) and can exacerbate nicotine intake. However, whether altered intake reflects changes in nicotine reinforcement or enhancement by nicotine of co-occurring reinforcers is unknown. This reinforcer-enhancement effect is evidenced in rats when nicotine self-administration is increased by the presence of a visual stimulus (VS) reinforcer. A few studies suggest that the placebo effect can influence physiological changes in people. In recent years, there has been an increase in public fascination with serial killers, however current criminological explanations continue to focus on social and psychological factors, lacking commentary on potential biological implications. This thesis addresses a gap in the literature by examining how biological processes, specifically epigenetic, neurobiological, and endocrinological mechanisms, interact with environmental stressors to shape serial homicidal behavior. Breast cancer is the most commonly diagnosed malignancy in women, with more than two million new cases diagnosed world-wide each year. Improvements in screening technology and diagnostics have increased survival rates, but cancer-associated sleep disruptions persist for the majority of breast cancer survivors well beyond the treatment period. The propensity to attribute incentive salience to reward-related cues, or sign-tracking (ST) has been linked to addiction in animal models and some initial human data. The current study explores the role of sign-tracking (ST), as a potential risk factor for problematic slot machine gambling, which abundantly features audiovisual reward-related cues, henceforth sensory features. Mild traumatic brain injuries (mTBIs) often lead to acute symptoms like disorientation and discoordination, but the underlying mechanisms are unknown. Although most patients recover quickly from a single mTBI, repeated injuries can be debilitating. After an mTBI, a neurometabolic cascade produces metabolic burden and vulnerability. Spreading depolarizations (SDs) have been observed after mTBIs. Here we investigated SDs’ role in short-term motor behavioral deficits post-mTBI. The primary intention of this study was to further understand the impact of assessing cognitive impairment in psychiatric patients, as a mediating factor on readmission rates. Mild cognitive dysfunction impacts a patient’s functional outcomes (Bowie & Harvey, 2006; Davis et al., 2012; Marcantonio, et al., 2001). Little information exists to guide best practices in the treatment of adults with cognitive impairment who are hospitalized for acute conditions (Davis et al., 2012). Major depression is a devastating disorder that consists of multiple symptoms such as low mood and motivational dysfunction. It has been shown that motivational dysfunction can be studied in animal models by using effort-based choice paradigms, which vary in their response requirements. It has been reported that dopamine depletion in the nucleus accumbens decreases ratio-scheduled lever-pressing in a manner related to the size of the ratio requirement. Previously, we discovered that pregnant mice (dams) that experienced chronic variable stress (CVS) during puberty display a blunted hypothalamic-pituitary-adrenal (HPA) response when exposed to an acute stressor. Evidence suggests that rats given long access (LgA) sessions to self-administer (SA) opioids escalate their intake, while also showing greater withdrawal severity and drug-induced reinstatement compared to rats maintained on short access (ShA) daily SA sessions. Little is known about the neural changes that occur during opioid escalation that may impact withdrawal and relapse. Past work examining opioid SA using ShA sessions in rodents has identified the central amygdala (CeA) as an area of interest that becomes hyperactive in acute withdrawal and may be involved in the incubation of craving that occurs after protracted withdrawal. Adolescent stress exposure increases the likelihood of alcohol misuse and alcohol use disorder (AUD) in adulthood; however, it is not clear how genetic and environmental factors interact to increase risk. This study examined how adolescent social isolation affects adult binge-like ethanol drinking and levels of the stress hormone corticosterone in male and female mice with a genetic predisposition toward high alcohol preference (HAP). Major depressive disorder (MDD) is a leading cause of disability worldwide, with a lifetime prevalence rate of approximately 20%. Inadequate pharmacological treatment methods for MDD are a significant debilitating factor. Patient estimates suggest that the treatment resistance rate for pharmacological interventions is over 30%. Postmortem analyses of human tissue of individuals diagnosed with MDD have shown an increase in Poly (ADP-ribose) polymerase 1 (PARP-1) mRNA gene expression in prefrontal cortical white matter when compared to psychiatrically normal brain tissue.
