Here, we present a protocol to isolate progenitor cells from mouse epididymal visceral adipose tissue and construct bulk RNA and assay for transposase-accessible chromatin with sequencing (ATAC-seq) libraries. We describe steps for adipose tissue collection, cell isolation, and cell staining and sorting. We then detail procedures for both ATAC-seq and RNA sequencing library construction. This protocol can also be applied to other tissues and cell types directly or with minor modifications.

The growth of scientific papers in the past decades calls for effective claim extraction tools to automatically and accurately locate key claims from unstructured text. Such claims will benefit content-wise aggregated exploration of scientific knowledge beyond the metadata level. One challenge of building such a model is how to effectively use limited labeled training data. In this paper, we compared transfer learning and contrastive learning frameworks in terms of performance, time and training data size. We found contrastive learning has better performance at a lower cost of data across all models. Our contrastive-learning-based model ClaimDistiller has the highest performance, boosting …

Popular approaches to building data from unstructured text come with limitations, such as scalability, interpretability, replicability, and real-world applicability. These can be overcome with Context Rule Assisted Machine Learning (CRAML), a method and no-code suite of software tools that builds structured, labeled datasets which are accurate and reproducible. CRAML enables domain experts to access uncommon constructs within a document corpus in a low-resource, transparent, and flexible manner. CRAML produces document-level datasets for quantitative research and makes qualitative classification schemes scalable over large volumes of text. The advent of the Internet was heralded as a revolutionary development in the democratization of information. It has emerged, however, that online discourse on social media tends to narrow the information landscape of its users. This dynamic is driven by the propensity of the network structure of social media to tend toward homophily; users strongly prefer to interact with content and other users that are similar to them. This study aims to analyze the needs of researchers in a regional comprehensive university for research data management services; discuss the options for developing a research data management program at the university; and then propose a phased three-year implementation plan for the university libraries. The method was to design a survey to collect information from researchers and assess and evaluate their needs for research data management services. This chapter provides an orientation to the history of technology, work, and the theology of work and then explores three visions of the future of work—a literary dystopia, a philosophical utopia, and a theological apocalypse—as resources for understanding the significance of work and imagining its future. In the first vision, found in Kurt Vonnegut’s speculative novel Player Piano, automation leads to the end of meaningful work and nearly renders humans obsolete. I propose a method for tracking and assessing scientific progress using a prediction consensus algorithm designed for the purpose. The protocol obviates the need for centralized referees to generate scientific questions, gather predictions, and assess the accuracy or success of those predictions. It relies instead on crowd wisdom and a system of checks and balances for all tasks. It is intended to take the form of a web-based, searchable database. This study focused on the development of a mathematical model required for estimating the number of adopters of ICT devices among libraries located in Nigeria. Data for this study was collected from 121 respondents selected based on a research survey approach using simple random sampling. 9 ICT devices were identified, namely: PCs, printers/fax machines, search engines, e-library systems, bulk SMS services, library management systems, bar/QR code readers, projectors and video conferencing. Purpose – In this paper, a call to the library and information science community to support documentation and conservation of cultural and biocultural heritage has been presented.

Design/methodology/approach – Based in existing Literature, this proposal is generative and descriptive— rather than prescriptive—regarding precisely how libraries should collaborate to employ technical and ethical best practices to provide access to vital data, research and cultural narratives relating to climate.

This Grounded theory study explores US adults' modern information search process as they pursue information through digital search user interfaces and tools. To study the current search process, a systematic grounded theory methodology and two data collection methods, a think-aloud protocol and semi-structured interviews, are used to develop the theory. The emerging theory addressed two tightly connected research questions that asked, “What is the process by which humans search and discover information?” and “What is the process by which search and discovery interfaces and tools support the modern search process?”
