The rise in gun violence in the United States has increased the demand on forensic agencies to process evidence promptly and accurately. The exchange of imperceptible traces among objects, suspects, and victims can provide critical insights into the interactions and sequence of events during a crime. Gunshot residue (GSR) is a type of trace evidence that holds significant investigative value in shooting reconstructions. GSR can help identify potential shooters, intermediate targets, and bullet trajectories; however, its analysis is often expensive and time-consuming. Forensic firearm analysis involves the comparison of test fires from a suspect firearm to an unknown sample, typically collected from a crime scene. Test fires and the unknown sample are examined for agreement in class characteristics and sufficient agreement in individual characteristics using a comparison microscope. The forensic analysis of firearms has been challenged both in court and by governmental organizations for its subjective nature and the inability to accurately report error rates.

This study investigated the repeatability and reproducibility of minutiae mark-up. The use of fingerprints as a mode of identification has been around for thousands of years with contributions from figures like Sir William Hershel, Thomas Taylor, Henry Faulds, Sir Francis Galton and many more. Latent fingerprint impressions can be deposited by the perpetrator of a crime on a variety of surfaces at a crime scene. Shoeprints deposited during the commission of a crime vary in quality as a function of numerous factors, such as substrates, media, and the specific physical activities carried out by perpetrators. This variability in impression quality impacts the amount of information available from questioned crime scene impressions, and therefore an examiner's confidence in providing opinions of source attribution. In response, this research attempted to characterize footwear impression quality based on image sharpness, contrast, entropy, homogeneity, noise, quantity of information, and complexity, and to integrate these hand-engineered feature descriptions with subjective opinions of impression quality.

Latent prints have been processed as a source of touch DNA evidence in the hopes of attaining partial or even full DNA profiles. Forensic footwear impression analysis is a valuable tool in criminal investigations. Extracting useful features from images of footwear impressions is a critical step in this process. However, the quality of these images can vary widely, making feature extraction challenging. In order to give a quality assessment rating to a footwear impression image, the image should first be analyzed to extract features from the impression. To determine whether a questioned fire is the result of arson or not, forensic chemists will often attempt to extract and identify any ignitable liquid residues present in the fire debris. Forensic chemists use gas chromatography-mass spectrometry (GC-MS) to identify ignitable liquids based on similarities between chromatograms of fire debris extracts and those of reference ignitable liquids that have been evaporated (weathered) to different extents. Stimulant drugs comprise one of the top drug categories abused in the United States. Due to its accessibility, low price, and manufacturing simplicity, methamphetamine is frequently placed within the top 10 seized drugs in the country. As of March 2023, methamphetamine is the most seized controlled substance in the United States, with 34,291 kg. In 2022, the United States seized over 79,000 kg of methamphetamine. The recovery of known-source shoes for the purpose of comparison to crime scene impressions often occurs with a temporal lag. During this passage of time, the outsole can be altered due to continued wear. These changes may impact forensically relevant characteristics of use known as randomly acquired characteristics (RACs). Forensic genetic genealogy has grown in both popularity and controversy in recent years. The genetic information comes from large numbers of single nucleotide polymorphism (SNP) markers. Polymorphism variations can be traced through familial lineage, determining how closely individuals are related to each other based on the similarities and differences in SNPs within their DNA. Efficient methods for DNA analysis are desperately needed in laboratories due to the influx in DNA analysis requests. The current DNA processing methods are costly, time consuming, and involve multiple tube changes, increasing the risk of contamination. However, the use of direct PCR can simplify the DNA analysis process by eliminating the extraction, purification and quantitation steps. Presently, the FBI Quality Assurance Standard 9.4 requires all evidence samples to be quantitated. Mitochondrial DNA (mtDNA) analysis has several forensic applications such as criminal investigations, identification of human remains, and missing person investigations. It is also the only type of DNA that is available from certain sample types such as hair shafts. The presence of mtDNA heteroplasmy within and between tissue types can lead to mtDNA sequence differences when comparing samples originating from the same individual. Studies on mtDNA heteroplasmy are increasingly being carried out for their implications in forensic interpretation of mtDNA sequences. With the current state of the field of forensic biology, collecting DNA from evidence samples may pose some difficulty. Traditional methods for collection of DNA from biological fluids and touch samples include cuttings, scrapings, and the double swab method. While all of these methods have benefits, they also have their drawbacks. In order to more efficiently collect DNA from an evidence sample, novel approaches need to be explored. Counterfeit pharmaceuticals are an actively developing health and economic threat worldwide. Particularly prevalent are counterfeit pharmaceuticals distributed in emerging nations and through internet pharmacies or e-pharmacies. In fire debris analysis, ignitable liquid residues are commonly identified using gas chromatography-mass spectrometry (GC-MS). The detection and identification of an ignitable liquid can help arson investigators determine whether a fire was intentional or accidental. To assist with the identification of ignitable liquid residues, chromatograms of questioned samples are typically compared to those of known ignitable liquids that have been weathered (evaporated) to different extents. Pressure sensitive tapes are a common evidence type in a variety of forensic cases, including violent crimes, drug trafficking, and terrorism. Forensic laboratories are often requested to analyze tape samples for composition and potential source identification. Tape samples manufactured from different sources are often distinguishable when using a combination of sensitive analytical techniques. Nonetheless, the interpretation of the data remains challenging due to a lack of standard criteria and guidelines from which examiners can support their conclusions. In this work we demonstrate the power of compound-specific isotope analysis (CSIA) to analyze proteinaceous biological materials in three distinct forensic applications, including: 1) linking necrophagous blow flies in different life stages to their primary carrion diet; 2) identifying the harvesting area of oysters for food authentication purposes; and 3) the ability to predict biometric traits about humans from their hair.
