Cartridges and spent cartridge casings can be probative pieces of evidence. Unfortunately due to a combination of factors such as exposure to high temperatures and initially low amounts of biological material on the surface, DNA testing so far has been mostly unsuccessful for these items. Typing other marker systems, such as protein polymorphisms, on the same biological evidence would add power of discrimination. To explore this option we developed a DNA-protein trypsin-based co-extraction method that was optimized for unfired and fired cartridges. DNA testing on touched objects is a valuable tool in forensic investigations, but DNA is usually present in low amounts, causing poor STR typing results. For touch DNA evidence, there is a clear need for additional individualization, especially for highly probative samples. This could be achieved by testing genetically variable proteins. The goal of this project was to develop a DNA/protein co-extraction method to facilitate DNA and protein testing on the same evidence item. The colonization of decomposing remains by necrophagous insects such as blow flies is of forensic importance because the progression through the various stages of insect development can be correlated to time of death. The ability to infer this information hinges on accurate determination of the fly species that are associated with the entomological evidence collected. This evidence can include eggs, larvae, pupae, and puparial casings. Determination of the egg’s identity is particularly challenging because the eggs of multiple species are morphologically very similar. Tape-lifting and swabbing are two methods commonly used for collecting biological samples in the United Kingdom and United States to investigate vehicle crimes. Determining the optimal collection method may lead to an increase in generating DNA profiles and crime-solving. The objective of this study is to evaluate the efficiency of adhesive tape and the double-swab collection methods for investigating vehicle crimes with possible touch DNA samples. Two experiments were conducted to evaluate the use of tape-lifts and swabs on spiked common vehicle fabric materials. The backlog of untested sexual assault kits is a national problem. Numerous federal funding opportunities offer the forensic science and law enforcement communities valuable resources needed to test the kits, but issues still remain. The majority of resources are focused on the collection and testing of sexual assault kits, but the tracking of the kits has not been a primary focus. This research highlights improvements that can be made to better understand the current backlog and improve the future processing and tracking of kits. Hyperspectral remote sensing is an innovative technology with applications in many sciences and is a non-destructive method that may offer more precise aging within development stages. Hyperspectral reflectance measurements from the anterior, midsection, and posterior of Lucilia sericata (Meigen) larvae and pupae were conducted daily from samples of the developing insects beginning at second instar. Only midsection measurements were conducted on second instar larvae due to their size, to ensure that the measurement was not of reflective surroundings. The use of genetic evidence in criminal cases is well established and has improved the public opinion and credibility of forensic science. However, several shortcomings associated with current genetic profiling techniques exist. Scientific research aimed at increasing the overall knowledge and understanding of biological factors will lead to the development of methods capable of improving the discriminating power of DNA evidence, overcoming limitations associated with DNA evidence, or complementing current methods of DNA profiling. The Golden Eagle (Aquila chrysaetos) and Bald Eagle (Haliaeetus leucocephalus) are the largest avian predators in North America, and are thus species of great ecological importance and cultural significance. There is a long history of human use of eagle body parts, and this use continues today: Bald and Golden eagles are among the North American birds most affected by the illegal wildlife trade. Detached eagle talons are often recovered in both law enforcement and archaeological contexts, but data to allow morphological identification of these talons have been lacking. BACKGROUND: Cells release extracellular membrane vesicles including microvesicles known as exosomes. Exosomes contain microRNAs (miRNAs) however the full range within colorectal cancer cell secreted exosomes is unknown. OBJECTIVE: To identify the full range of exosome encapsulated miRNAs secreted from 2 colorectal cancer cell lines and to investigate engineering of exosomes over-expressing miRNAs. METHODS: Exosomes were isolated from HCT-116 and HT-29 cell lines. RNA was extracted from exosomes and microRNA array performed. Cells were engineered to express miR-379 (HCT-116-379) or a non-targeting control (HCT-116-NTC) and functional effects were determined. In Forensic Science casework, identifying the source of a tissue can assist in crime scene reconstruction. Currently, presumptive testing methods to identify tissue type are utilized in crime scene laboratories, but there is a need for a more reliable confirmatory test for tissue type identification. INTRODUCTION AND SUMMARY OF ARGUMENT Professor Scherr agrees with petitioner that review is warranted because the Maryland Court of Appeals decision is erroneous. The Fourth Amendment does not sanction police harvesting of DNA without probable cause and a warrant and without the subject’s knowledge or consent, to be used however the authorities deem appropriate and without restriction. The Maryland Court of Appeals’ decision is contrary to the Supreme Court’s jurisprudence as articulated in the Riley v. California – Maryland v. King – United States v. Jones trilogy. Students who participate in active and experiential learning activities develop a better understanding of basic scientific principles and are also more likely to be retained in a STEM discipline. Developing such activities, however, can be difficult. In the present study, we sought to develop classroom activities to engage university students and increase understanding of the theory and practice of forensic science. One common accessory amongst the target audience was the ever-present media, both social and popular (entertainment). In order to facilitate optimal plant DNA quantitation and identification, an assay has been developed that uses generic plant PCR primers that amplify a region in the chloroplast genome of plant samples. The assay uses the SYBR green detection dye to detect the PCR product with a universal PCR primer set to the large subunit of ribulose bisphosphate carboxylase, rbcL, but can be used with any of the universal barcode primers for land plants (rbcL, matK, trnH, psbA). MicroRNAs (miRNAs) have been found to be stable in the circulation of cancer patients raising their potential as circulating biomarkers of disease. The specific source and role, however, of miRNAs in the circulation is unknown and requires elucidation to determine their true potential. In this study, along with primary tissue explants and primary stromal cells, three breast cancer cell lines were employed, including T47D, MDA-MB-231 and SK-BR-3. Tissue explants were harvested in theatre, with informed patient consent, and included tumour, tumour associated normal, and diseased lymph node samples. Effect Of Fluctuating Temperatures On The Development Of A Forensically Important Blow Fly, Protophormia Terraenovae (Diptera: Calliphoridae), Jodie-A. Warren, Gail S. Anderson

Experiments were conducted to compare the immature development of Protophormia terraenovae (Robineau-Desvoidy) at fluctuating temperatures of 4-28 and 9-23°C to their mean constant temperature, 16°C. Overall development was fastest at the greater fluctuation and slowest at the constant temperature but showed similar percentages of development time in each stage. The rate summation effect is suspected to have caused this difference in development rate because fluctuations above the mean increase the rate relatively more than temperatures below the mean can lower the rate.

Forensic entomology is a science used to estimate a post-mortem interval (PMI). Larvae develop at predictable rates and the time interval for this development can be used to estimate the PMI. Environmental temperatures are not constant and fluctuate with the photoperiod. In early and late insect seasons, temperatures can drop to below threshold temperatures where development essentially ceases. Threshold temperatures differ for different species and should be determined by raising insects at the extremely low temperatures. The lower threshold temperature for Protophormia terraenovae for egg hatch is 10.3°C ; for first instar larvae to molt to second instar larvae it …
