In forensic science, DNA extraction can be a tedious and resource-intensive process. Extraction with Proteinase K is an industry standard but has its drawbacks, such as requiring multiple ionic detergents and washing steps. MicroGEM has developed a new enzyme called forensicGEM that is temperature-dependent and compatible with mesophilic enzymes, offering complete DNA extraction in about 20 minutes in a single tube, limiting contamination, loss of sample, and working time. ForensicGEM can extract DNA from highly degraded samples, potentially leading to more complete STR profiles. The United States has been experiencing a nationwide public health crisis since introducing prescription opioids in the 1990s. A central problem with opioids is their highly addictive nature causing a dose-dependent respiratory depression leading to overdoses and deaths. Drug overdose deaths have risen in the past two decades, with opioids being the dominant driver of this epidemic. Given the circumstances, accurate and efficient confirmatory analysis of opioids in urine is essential to monitoring patients’ compliance and preventing drug diversion in clinical and forensic settings. Luminol, the most common presumptive test for blood at a crime scene, has multiple issues, such as false positive results with chemical agents, no luminescence due to “active oxygen” cleaning agents on bloodstains, and inability to penetrate textile materials. This study aspires to find a new screening approach to trace DNA recovery techniques to yield a higher quantity of trace DNA from larger items of evidence. It takes the path of visualizing trace DNA on items of evidence with potential DNA so analysts can swab a more localized area rather than attempting to recover trace DNA through the general swabbing technique currently used for trace DNA recovery. The first and second parts consisted of observing trace DNA interaction with Diamond Dye on porous and non-porous surfaces.

Forensic science and entomology programs may benefit from using deceased pigs as teaching resources due to anatomical similarities to humans. The use of such animals provides opportunities for students to learn the effects of geographical location and climate on insects; understand insect succession; practice insect collection and identification; and learn post-mortem interval procedures, taphonomic principles of decomposition, and crime scene investigation and management techniques. Forensic entomology and taphonomic studies fit well with educational interests in experiential learning. Submerged items are often thought to lack evidentiary value. However, previous studies have shown the ability to recover DNA from submerged porous items for upwards of six weeks. The crevices or interweaving fibers in porous items are thought to protect DNA from being washed away. It is hypothesized that, because non-porous surfaces do not have the same traits that might aid in DNA retention, then DNA quantities and the number of donor alleles recovered would decrease over longer submersion periods. This qualitative data leads the reader to one ultimate question: what does the rise in terrorism in Africa mean for the United States (U.S.) and U.S. policymakers, National Security academia, students in National Security, if anything at all? An ELISA method was developed and validated to detect ketamine in human hair samples. Ketamine is an anesthetic drug that causes memory loss, dissociative sensations, and hallucinations. Due to these adverse effects, ketamine is a common drug used in drug facilitated sexual assaults (DFSAs). It is very important to be able to detect the substances used in DFSAs over a longer period of time due to the delayed reporting of these crimes. Touch paper evidence could be the source of probative human DNA but recovery is challenging and forensic laboratories instead prioritize processing by the Latent Print and Questioned Document disciplines. Biological evidence is often found at crime scenes and can comprise of DNA from the victim(s) and perpetrator(s) to a crime as well as from individuals with no direct relationship to the incident. This can complicate analysis as DNA mixtures are one of the more difficult sources of biological evidence to interpret. Probabilistic genotyping (PG) has greatly aided in mixture analysis. However, even with PG, standard bulk mixture approaches do not always result in probative results as allele overlap, artifacts, or low-level minor contributors inevitably cause genotype information loss. Efficient methods for DNA analysis are desperately needed in laboratories due to the influx in DNA analysis requests. The current DNA processing methods are costly, time consuming, and involve multiple tube changes, increasing the risk of contamination. However, the use of direct PCR can simplify the DNA analysis process by eliminating the extraction, purification and quantitation steps. Presently, the FBI Quality Assurance Standard 9.4 requires all evidence samples to be quantitated. Mitochondrial DNA (mtDNA) analysis has several forensic applications such as criminal investigations, identification of human remains, and missing person investigations. It is also the only type of DNA that is available from certain sample types such as hair shafts. The presence of mtDNA heteroplasmy within and between tissue types can lead to mtDNA sequence differences when comparing samples originating from the same individual. The shedding propensity of a person can assist data interpretation in casework when assessing the possibility of passive transfer for DNA analysis. Past studies on shedding propensity evaluated palmar skin (washed and unwashed) deposits. This study compared different skin locations with respect to shedding propensity, and explored the potential of tape-lifts as a skin surface collection method. Eight different skin types and samples were collected with adhesive tape disks from 28 participants over three non-consecutive days; the washed and unwashed fingers from both hands, toe, and arm, neck below ear, and nape. Contact traces are an important part of DNA casework, but the probative value of any identified associations depends on the possibility of passive transfer. There is known individual variation in DNA left behind during contact, this DNA shedding propensity has an effect on whose DNA is detected. This study evaluated this variability using a cell staining approach. Volunteers were asked to deposit a fingerprint on a clean glass slide, then wash their hands and deposit a second fingerprint after a 30-minute wait without touching anything. Cocaine overdose deaths are on the rise in the United States. This increase may be due to the co-consumption of cocaine with other drugs. When cocaine is ingested in conjunction with alcohol, a specific metabolite called cocaethylene forms. Information regarding cocaethylene, specifically its interpretive value in post-mortem cases, has not been widely studied. Data from the Office of the Chief Medical Examiner in New York City revealed that there were 195 cases in 2017 positive for cocaethylene. These cases were most likely to involve black (42.1%) males (79.5%) in Manhattan (28.2%) aged 50 - 59 (34.4%). Most of the cases …

Postmortem survival of DNA in human skeletal remains occurs due to the compact microstructure of the skeleton and its ability to provide a strong, protective physical barrier to environmental insults. On a molecular level, DNA preservation in bones/teeth involves electrostatic interactions between the negatively-charged DNA backbone and positively charged calcium residues in hydroxyapatite, the latter of which is one of the main components of bone microstructure. Despite these protections, over time endogenous DNA becomes damaged, limiting our ability to detect it and affecting its utility in making a positive identification. Locard’s Exchange Principle states that “every contact leaves a trace.” The same principle applies when a perpetrator of a homicide decides to bury a victim in a clandestine grave. If a perpetrator originally buried a murder victim in haste to prevent discovery and then decided to later move the victim’s body to a remote location for disposal, decomposition fluids from the victim’s body (which contain cellular material and therefore DNA) would remain in the surrounding grave soil at the original burial site. Trace DNA evidence may be discovered at a crime scene after having been deposited by a person of interest via active or passive transfer. Based on previous studies, passive transfer of one’s DNA is influenced by their shedding propensity, or probability of depositing a detectable amount of DNA through touch. Determining the shedding propensity of a person of interest can aid in trace DNA interpretation in forensic casework. The purpose of this experiment was to test blood detectors against household cleaners. Similar experiments have been run before, but not on the selected surfaces. It is important to understand what one is looking for at a crime scene, and what a cleaned surface may look like. Four surfaces were chosen for this experiment: ceramic tile as a control, concrete, laminated wood flooring, and drywall. For each surface, 0.5mL of pig’s blood was allowed to dry before being cleaned off with either Clorox bleach, Dawn soap and hot water, a Mr. Clean Magic Eraser, or Mrs. Meyer’s Multi-Purpose Concentrate. Forensic entomology is considered a relatively new field of entomology yet insects have been around for millions of years. Insects have been associated with dead animals and humans for centuries. The remains of insects have been recorded from Ancient Egypt to the present time. Effective DNA extraction methods are important for forensic applications. The main goal of this experiment was to determine if a newly developed trypsin based protein/DNA co-extraction method applied to contact traces would yield comparable results to a commercial Proteinase K method (QIAamp DNA Investigator Kit) and a Chelex extraction method with Tween 20 used in forensic laboratories. This was tested on 20 sets of sebaceous fingerprints on glass slides.

A previously developed, non-destructive, homemade vacuum method for collecting biological material from handwritten documents had promising DNA results for white copy paper, preserved indented writing, and latent prints. Prior to casework implementation, additional validation experiments are warranted and here the method was tested for different paper substrates. This work describes testing of notebook paper, bank deposit slips, magazine pages, and manila envelopes. The quantity of recovered DNA varied from donor to donor, but the mean quantities showed a trend that can be explained by the different sizes and surface properties of the tested paper types. Household/commercial bleach (6% NaOCl, sodium hypochlorite) degrades DNA through oxidative damage, production of chlorinated base products, and cleavage of DNA strands (breaking it into smaller and smaller fragments). The presence of these lesions significantly impacts the ability to generate a full genetic profile from an evidentiary sample. In fact, knowledge of the damaging effect of bleach on DNA is the basis for its use in forensic laboratories to clean workbenches and prevent cross-contamination of samples between cases.
