Recently, structural variation in the genome has been implicated in many complex diseases. Using genomewide single nucleotide polymorphism (SNP) arrays, researchers are able to investigate the impact not only of SNP variation, but also of copy-number variants (CNVs) on the phenotype. The most common analytic approach involves estimating, at the level of the individual genome, the underlying number of copies present at each location. Once this is completed, tests are performed to determine the association between copy number state and phenotype. Several issues need to be better understood before breast tissue engineering becomes a clinically viable option. One of the most important aspects is the interaction between cells and the microtopography of the implant surface. The aim of this study was to evaluate the efficacy of D1 cells, multipotent mouse bone marrow stromal precursors, in differentiating to adipocytes and to characterize their metabolic activity (lactic acid released and glucose consumed), leptin secretion and lipid production when cultured on patterned poly(L-lactide) (PLLA) films. This dissertation introduces a new metric in the area of High Performance Computing (HPC) application reliability and performance modeling. Derived via the time-dependent implementation of an existing inequality measure, the Failure index (FI) generates a coefficient representing the level of volatility for the failures incurred by an application running on a given HPC system in a given time interval. This coefficient presents a normalized cross-system representation of the failure volatility of applications running on failure-rich HPC platforms. The dynamic treatment effect literature considers multiple treatments administered over time, with some treatments affected by interim outcomes. But the literature overlooks the possibility of individuals acting in anticipation of future treatments. This lack of anticipation aspect may not matter in the drug–response relationships which motivated the literature. But human beings (or animals with some intelligence) do not just respond to current and past treatments, but also ‘reflect and anticipate’ future treatments. New methodology has been proposed in recent years for evaluating the improvement in prediction performance gained by adding a new predictor, Y, to a risk model containing a set of baseline predictors, X, for a binary outcome D. We prove theoretically that null hypotheses concerning no improvement in performance are equivalent to the simple null hypothesis that the coefficient for Y is zero in the risk model, P(D = 1|X, Y ). Despite gains in prenatal care (PNC) usage and birth outcomes for minority women during the past few decades, observed disparities between non-Hispanic Whites, Blacks, and Hispanics persist. Using the National Center for Health Statistics’ (NCHS) natality files from 1981 through 1998, Alexander, Kogan, & Nabukera (2002) examined live births of U.S. residents by trimester in which PNC was initiated and the appropriateness of that care based on the Adequacy of Prenatal Care Utilization Index (APNCU) (Kotelchuck, 1994). They found racial disparities between White and Black women in both the trimester of PNC initiation and the number of PNC visits made. …

The Dempster-Shafer Theory, a generalization of the Bayesian theory, is based on the idea of belief and as such can handle ignorance. When all of the required information is available, many data fusion methods provide a solid approach. Yet, most do not have a good way of dealing with ignorance. Recent research efforts strive to address the growing need for dismount surveillance, dismount tracking and characterization. Current work in this area utilizes hyperspectral and multispectral imaging systems to exploit spectral properties in order to detect areas of exposed skin and clothing characteristics. Because of the large bandwidth and high resolution, hyperspectral imaging systems pose great ability to characterize and detect dismounts. A cross country flight, in two and three dimensional space, is considered, as well as a vertical drop in three dimensional space. American Indian and Alaska Native (AI/AN) women currently have some of the highest mortality rates from breast cancer for any racial/ethnic group in the United States and some of the lowest screening rates. However, current data are not available for regional differences in screening, which can result in dramatically different stage at diagnosis and mortality. We conducted surveys with 120 focus group participants in a needs assessment of mammography among AI/AN in the greater Kansas City metropolitan area and parts of Northeast Kansas. Objective: To examine black-white differences in hepatitis C treatment within the Veterans Administration (VA) and determine whether racial variation in specialty consultation explains differences in hepatitis C treatment between blacks and whites.

Methods: We performed a retrospective cohort study of 1040 veterans meeting VA eligibility criteria for hepatitis C treatment. We used multiple imputation to handle missing race data. Specialty consultation was determined from the VA outpatient medical dataset and hepatitis C treatment was determined from the VA decision support system. Objectives. This study investigated the associations between poverty rates, Appalachian mountaintop coal mining, and age-adjusted total mortality rates to determine if persons exposed to this form of mining experience greater poverty and higher death rates compared to other types of mining or other areas of Appalachia.

Methods. Mortality rates, poverty rates, Appalachian designation and mining activity were examined for counties in Kentucky, Tennessee, Virginia and West Virginia (N=403). Linear least squares models tested for annual group differences from 2000-2007 in total and child poverty, and total mortality, based on mining type and Appalachian location. Background

Preterm birth is the principal factor contributing to adverse outcomes in multiple pregnancies. Randomized controlled trials of progestogens to prevent preterm birth in twin pregnancies have shown no clear benefits. However, individual studies have not had sufficient power to evaluate potential benefits in women at particular high risk of early delivery (for example, women with a previous preterm birth or short cervix) or to determine adverse effects for rare outcomes such as intrauterine death.

Methods/design

We propose an individual participant data meta-analysis of high quality randomized, double-blind, placebo-controlled trials of progestogen treatment in women with a twin pregnancy. The …

The development of high throughput screening (HTS) assays in the field of nanotoxicology provide new opportunities for the hazard assessment and ranking of engineered nanomaterials (ENM). It is often necessary to rank lists of materials based on multiple risk assessment parameters, often aggregated across several measures of toxicity and possibly spanning an array of experimental platforms. Bayesian models coupled with the optimization of loss functions have been shown to provide an effective framework for conducting inference on ranks. We describe a method to estimate pollution profiles and contribution levels for distinct prominent pollution sources in a region based on daily pollutant concentration measurements from multiple measurement stations over a period of time. In an extension of existing work, we will estimate common source profiles but distinct contribution levels based on measurements from each station. In addition, we will explore the possibility of extending existing work to allow adjustments for synoptic regimes—large scale weather patterns which may effect the amount of pollution measured from individual sources as well as for particular pollutants. Maximal oxygen consumption (VO2max) is considered by many to be the best overall measure of an individual's cardiovascular health. Collecting the measurement, however, requires subjecting an individual to prolonged periods of intense exercise until their maximal level, the point at which their body uses no additional oxygen from the air despite increased exercise intensity, is reached. Collecting VO2max data also requires expensive equipment and great subject discomfort to get accurate results. Because of this inherent difficulty, it is often avoided despite its usefulness. Nonparametric inference methods on the mean difference between two correlated Functional processes are proposed. We compare methods that: 1) incorporate different levels of smoothing of the mean and covariance; 2) preserve the sampling design; and 3) use parametric and nonparametric estimation of the mean functions. We apply our method to estimating the mean difference between average normalized δ-power of sleep electroencephalograms for 51 subjects with severe sleep apnea and 51 matched controls in the first 4 hours after sleep onset. In this paper, we study the impact of unmeasured confounding on inference about a two-way interaction in a mean regression model with identity, log or logit link function. Necessary and sufficient conditions are established for a two-way interaction to be nonparametrically identified from the observed data, despite unmeasured confounding for the factors defining the interaction. A lung cancer data application illustrates the results.

The phenomenon of accelerated tumor growth following surgery has been observed repeatedly and merits further study. Inflammatory breast carcinoma (IBC) is widely recognized as an extremely aggressive malignancy characterized by micrometastasis at the time of diagnosis, with one interesting subgroup defined as secondary IBC where pathologically identifiable IBC appears after surgical treatment of a primary non-inflammatory breast cancer. One possible mechanism can be related to the stimulation of dormant micrometastasis through local angiogenesis occurring as part of posttraumatic healing. Left restriction semigroups have appeared at the convergence of several flows of research, including the theories of abstract semigroups, of partial mappings, of closure operations and even in logic. For instance, they model unary semigroups of partial mappings on a set, where the unary operation takes a map to the identity map on its domain. This perspective leads naturally to dual and two-sided versions of the restriction property. Missing data in longitudinal studies can create enormous challenges in data analysis when coupled with the positive-definiteness constraint on a covariance matrix. For complete balanced data, the Cholesky decomposition of a covariance matrix makes it possible to remove the positive-definiteness constraint and use a generalized linear model setup to jointly model the mean and covariance using covariates (Pourahmadi, 2000). However, this approach may not be directly applicable when the longitudinal data are unbalanced, as coherent regression models for the dependence across all times and subjects may not exist. In many mass tort cases, separately trying all individual claims is impractical, and thus a number of trial courts and commentators have explored the use of statistical sampling as a way of efficiently processing claims. Most discussions on the topic, however, implicitly assume that sampling is a “second best” solution: individual trials are preferred for accuracy, and sampling only justified under extraordinary circumstances.
