Tumor xenograft experiments are a popular tool of cancer biology research. In a typical such experiment, one implants a set of animals with an aliquot of the human tumor of interest, applies various treatments of interest, and observes the subsequent response. Efficient analysis of the data from these experiments is therefore of utmost importance. This dissertation proposes three methods for optimizing cancer treatment and data analysis in the tumor xenograft context. When conducting statistical analysis in the Bayesian paradigm, the most critical decision made by the researcher is the identification of a prior distribution for a parameter. Despite the mathematical soundness of the Bayesian approach, a wrongly specified prior can lead to biased and incorrect results. To avoid this, prior distributions should be based on real data, which are easily accessible in the "big data" era. This dissertation explores two applications of Bayesian hierarchical modelling that incorporate information obtained from a meta-analysis.

In system reliability, practitioners may be interested in testing the homogeneity of the component lifetime distributions based on system lifetimes from multiple data sources for various reasons, such as identifying the component supplier that provides the most reliable components.

In the first part of the dissertation, we develop distribution-free hypothesis testing procedures for the homogeneity of the component lifetime distributions based on system lifetime data when the system structures are known. Using the physicochemical properties of wine to predict quality has been done in numerous studies. Given the nature of these properties, the data is inherently skewed. Previous works have focused on handful of sampling techniques to balance the data. This research compares multiple sampling techniques in predicting the target with limited data. For this purpose, an ensemble model is used to evaluate the different techniques. There was no evidence found in this research to conclude that there are specific oversampling methods that improve random forest classifier for a multi-class problem.

The M3-Competition found that simple models outperform more complex ones for time series forecasting. As part of these competitions, several claims were made that statistical models exceeded machine learning (ML) techniques, such as recurrent neural networks (RNN), in prediction performance. These findings may over-generalize the capabilities of statistical models since the analysis measured the total forecasting accuracy across a wide range of industries and fields and with different interval lengths. Influence diagnostics in regression analysis allow analysts to identify observations that have a strong influence on model fitted probabilities and parameter estimates. The most common influence diagnostics, such as Cook’s Distance for linear regression, are based on a deletion approach where the results of a model with and without observations of interest are compared. Here, deletion-based influence diagnostics are proposed for generalized estimating equations (GEE) for correlated, or clustered, nominal multinomial responses. Non-Fungible Tokens (NFTs) enable ownership and transfer of digital assets using blockchain technology. As a relatively new financial asset class, NFTs lack robust oversight and regulations. These conditions create an environment that is susceptible to fraudulent activity and market manipulation schemes. This study examines the buyer-seller network transactional data from some of the most popular NFT marketplaces (e.g., AtomicHub, OpenSea) to identify and predict fraudulent activity. The restricted mean survival time (RMST) is a clinically meaningful summary measure in studies with survival outcomes. Statistical methods have been developed for regression analysis of RMST to investigate impacts of covariates on RMST, which is a useful alternative to the Cox regression analysis. CERN (Conseil européen pour la recherche nucléaire) uses a massive underground particle collider, called the Large Hadron Collider or LHC, to produce particle collisions at extremely high speeds. Alternating recurrent events data arise commonly in health research; examples include hospital admissions and discharges of diabetes patients; exacerbations and remissions of chronic bronchitis; and quitting and restarting smoking. Recent work has involved formulating and estimating joint models for the recurrent event times considering non-negligible event durations. Using U.S. resident survey data from the National Community Survey in combination with public data from the U.S. Census and additional sources, a Voting Regressor Model was developed to establish fair benchmark values for city performance. These benchmarks were adjusted for characteristics the city cannot easily influence that contribute to confidence in local government, such as population size, demographics, and income. Across the United States, record numbers of wildfires are observed costing billions of dollars in property damage, polluting the environment, and putting lives at risk. The ability of emergency management professionals, city planners, and private entities such as insurance companies to determine if an area is at higher risk of a fire breaking out has never been greater. This paper proposes a novel methodology for identifying and characterizing zones with increased risks of forest fires. Methods involving machine learning techniques use the widely available and recorded data, thus making it possible to implement the tool quickly.

Understanding diagnostic tests and examining important features of novel coronavirus (COVID-19) infection are essential steps for controlling the current pandemic of 2020. In this paper, we study the relationship between clinical diagnosis and analytical features of patient blood panels from the US, Mexico, and Brazil. Our analysis confirms that among adults, the risk of severe illness from COVID-19 increases with pre-existing conditions such as diabetes and immunosuppression. These data are collected from the environment and may be missing or measured with error when the observer’s ability to see the animal is obscured. In this dissertation, improved statistical methods for time-series and lifetime data are developed. First, an improved trend test for time series data is presented. Then, robust parametric estimation methods based on system lifetime data with known system signatures are developed.

In the first part of this dissertation, we consider a test for the monotonic trend in time series data proposed by Brillinger (1989). In this paper, we explore a representation methodology for the compression of DNA isolates. Using lossless string compression via tokenization of frequently repeated segments of DNA, we reduce the length of the isolates to be counted as k-mers for classification. With this new representation, we apply a previously established feature sampling method to dramatically reduce the feature space. In understanding the genetic diversity, we also look at conserving biological function across these spaces. Using a random forest model we were able to predict the resistance or susceptibility of bacteria with 85-90\% accuracy, with a 30-50\% reduction in overall isolate length, …

Cell assemblies, defined as groups of neurons forming temporal spike coordination, are thought to be fundamental units supporting major cognitive functions. However, detecting cell assemblies is challenging since they can occur at a range of time scales and with a range of precisions, from synchronous spikes to co-variations in firing rate. In this dissertation, we use a recently published cell assembly detection (CAD) algorithm that is capable of detecting assemblies at a range of time scales and precisions. In this dissertation, we explore sensitivity analyses under three different types of incomplete data problems, including missing outcomes, missing outcomes and missing predictors, potential outcomes in \emph{Rubin causal model (RCM)}. The first sensitivity analysis is conducted for the \emph{missing completely at random (MCAR)} assumption in frequentist inference; the second one is conducted for the \emph{missing at random (MAR)} assumption in likelihood inference; the third one is conducted for one novel assumption, the ``sixth assumption'' proposed for the robustness of instrumental variable estimand in causal inference.
