Excess phosphorus (P) loading is a key driver of eutrophication and harmful algal blooms (HABs) in aquatic systems worldwide. This relationship is of increasing concern in coastal Louisiana, where climate variability, altered hydrology, and land-use change converge, resulting in increased hydrologic and nutrient loading into coastal systems. This dissertation examines controls on P cycling in the Lake Pontchartrain Basin through a multi-scale framework linking estuarine processes, watershed loading, climate and weather patterns, and sediment dynamics. Man-made levees along the lower Mississippi River prevent delivery of sediment from building and maintaining Louisiana’s coastal wetlands. The Mid-Barataria sediment diversions is designed to reintroduce Mississippi River water, sediment, and nutrients into the sediment-starved Barataria Basin. Phosphorus (P) is an important macronutrient for regulating primary production in coastal marine ecosystems. Wetlands can serve as a sink or source for phosphorus to the overlying water column through various retention and release processes, dependent on concentration. Surface ocean phosphate is commonly below the standard analytical detection limits, leading to an incomplete picture of the global variation and biogeochemical role of phosphate. A global compilation of phosphate measured using high-sensitivity methods revealed several previously unrecognized low-phosphate areas and clear regional differences. Both observational climatologies and Earth system models (ESMs) systematically overestimated surface phosphate. Furthermore, ESMs misrepresented the relationships between phosphate, phytoplankton biomass, and primary productivity. Atmospheric iron input and nitrogen fixation are known important controls on surface phosphate, but model simulations showed that differences in the iron-to-macronutrient ratio in the vertical nutrient supply and surface lateral transport …

In desert soils, phosphorus (P) is an important limiting nutrient and its cycling characteristics are less understood compared to nitrogen and carbon. Phosphorus cycling is controlled by both geochemical and biological factors. Traditionally, P availability has been characterized via sequential extraction analyses such as Hedley fractionation, but newly developed extraction methods allow for the examination of more biologically relevant P fractions, providing insight on mechanisms of P acquisition by plants and microbes. Farms are non-point sources for nutrient contaminants that drain into watersheds and contribute to eutrophication and other environmental problems. Eastern Kentucky University’s Meadowbrook Farm raises both crops and livestock, causing dissolved phosphorus in the form of orthophosphate (PO43-) from fertilizer and animal manure to enter surface and subsurface waters, eventually flowing into Muddy Creek, a tributary of the Kentucky River.

We sampled surface water, springs, and water from French drains that emanate from the farm, and also sampled Muddy Creek waters from May through August 2016. Typically, 1 to 2 days after sampling, we colorimetrically measured dissolved …

A widespread stressor, anthropogenic nitrogen (N) and phosphorus (P) pollution can increase resource nutrient content and alter animal community composition in freshwater ecosystems. In this dissertation, I used ecological stoichiometry theory to examine effects of diet nutrient content and leaf litter type on growth, regulation, and wastes of aquatic invertebrate detritivores. I tested effects of leaf litter diet carbon:phosphorus (C:P) on growth and stoichiometric regulation of the detritivorous caddisfly Pycnopsyche lepida and used results to determine a threshold elemental ratio of oak litter C:P=1620 that confers peak growth of this species. Seasonal and spatial variability in environmental factors may affect dissolved organic matter composition and nutrient transformation and retention in streams. The objective of this research was to quantify and describe seasonality, quantity, and quality of nutrient processing and export of ammonium (NH4), soluble reactive phosphate (SRP), and dissolved organic carbon (DOC) into Lake Superior through intensive study in a small 1st order watershed coupled with snapshot measurements across 12 tributaries that varied in size, location, and wetland coverage. Nitrogen (N2) fixation can give certain species of cyanobacteria a competitive advantage in lake and reservoir phytoplankton. These species of cyanobacteria, along with others that cannot fix N2, can form toxic compounds that impair water quality when present in high concentrations. N2 fixation rates may be substantial in small (< 1.0 km2), temperate reservoirs since these systems experience thermal stratification and often nitrogen (N) limitation throughout a substantial proportion of the year. However, the effects of N2 fixation on N cycling, alleviation of short-term N limitation, and water quality are not well-understood. A mesocosm experiment and ecosystem-scale observational study were conducted to 1) determine the efficiency of N2 fixation under varying N relative to phosphorus (P) supply, 2) examine the effects of N2 fixation on autotrophic biomass accumulation and microcystin production, and 3) measure N2 fixation rates and importance to autotrophic N demand and zooplankton N assimilation. Results of the mesocosm experiment indicated that N2 fixation was increased at low N:P supply under high P. However, N2 fixation was inefficient at alleviating N limitation when fixed N was the primary source of N. Additionally, microcystin production occurred only at high N:P supply when N2 fixation was low, indicating that reducing external N inputs may have a positive effect on water quality. Results of whole-reservoir determination of N2 fixation using seston δ15N natural abundances indicated that N2 fixation rates throughout the warm season were substantial and influenced by water temperature. Annual N2 fixation rates ranged from 2.2 - 6.6 g N m-2 yr-1, and contributed up to 19% of the annual autotrophic N demand. Zooplankton were assimilating fixed N in most of the study reservoirs, representing a possible mechanism of ecosystem fixed N retention. Collectively, these results suggest that N2 fixation plays a substantial role in N cycling in small, temperate reservoirs, but likely cannot alleviate short-term N limitation.

Arsenic and phosphorus are biochemically very similar, and hence arsenate (As5+) is toxic by interfering with the energy metabolism, in particular during P limitation. However, many phytoplankton detoxify As by reducing arsenate to arsenite (As3+), and/or methylating it to mono and dimethyl As. Such As detoxification becomes operative in oligotrophic waters when phosphate concentrations are below those for As; therefore, we evaluated the potential use of these detoxification products as indicators of P-limitation by measuring As speciation during the US GEOTRACES North Atlantic transect. Dissolved inorganic phosphorus (DIP) concentrations in surface water of vast areas of the ocean are extremely low (<10 nM) and phosphorus (P) availability could limit primary productivity in these regions. We explore the use of oxygen isotopic signature of dissolved phosphate (δ18OPO4) to investigate biogeochemical cycling of P in the Sargasso Sea, Atlantic Ocean. Additional techniques for studying P dynamics including 33P-based DIP turnover time estimates and percent of cells expressing alkaline phosphatase (AP) activity as measured by enzyme-labeling fluorescence are also used. In surface waters, δ18OPO4 values were lower than equilibrium by 3–6%, indicative of dissolved organic phosphorous (DOP) remineralization by extracellular enzymes. Phosphorus is a key element in important biochemical compounds, such as RNA and phospholipids, and can become limiting in a variety of marine systems. The uptake of phosphorus into biochemical fractions (protein, low molecular weight (LMW) compounds, lipid, polysaccharide and nucleic acid) in Acartia tonsa fed 33P -labeled Rhodomonas salina was examined. R. salina was cultured on two variations of one media that in one case contained phosphorus in balance and the other out of balance with relation to other standard f/2 components. We examined the physiological responses of steady-state iron (Fe)-replete and Fe-limited cultures of the biogeochemically critical marine unicellular diazotrophic cyanobacterium Crocosphaera at glacial (19 Pa; 190 ppm), current (39 Pa; 380 ppm), and projected year 2100 (76 Pa; 750 ppm) CO2 levels. Rates of N2 and CO2 fixation and growth increased in step with increasing partial pressure of CO2 (pCO2), but only under Fe- replete conditions. The kinetics of sulfate reduction and inorganic nutrient production (ΣCO2, ammonium, and phosphate) were examined in the sediments at five sites in the southern Chesapeake Bay, using long term (> 200 d) sediment decomposition experiments. Average first order rate constants for these processes (at 25oC) decreased from 8.2 to 3.7 yr-1 in the surface sediments (0-2 cm), to 2.1 to 0.2 yr-1 at 12-14 cm. The C/N and C/P ratios of the organic matter undergoing decomposition also increased with depth at these sites. Taken together, these results indicate that the reactivity of the organic matter …
