Biological dinitrogen (N₂) fixation, the energetically expensive conversion of N₂ gas to ammonia, plays an essential role in balancing the nitrogen budget in the ocean. Accumulating studies show detectable N₂ fixation rates below the euphotic zone in various marine systems, revealing new insights of marine N₂ fixation. However, the reported rates are highly variable and frequently fall close to detection limits, raising the question of the ubiquity and significance of N₂ fixation in the global dark ocean. Using highly sensitive isotopic labeling incubation including a set of control incubations, we confirm the occurrence of mesopelagic N₂ fixation in the South …

The OOI Biogeochemical Sensor Data Best Practices and User Guide is intended to provide current and prospective users of data generated by biogeochemical sensors deployed on the Ocean Observatories Initiative (OOI) arrays with the information and guidance needed for them to ensure that the data is science-ready. This guide is aimed at researchers with an interest or some experience in ocean biogeochemical processes. We expect that users of this guide will have some background in oceanography, however we do not assume any prior experience working with biogeochemical sensors or their data. Agricultural activities often contaminate watersheds with excess nutrients leading to poor water quality and eutrophication. Eastern Kentucky University (EKU) Meadowbrook Farm raises crops and livestock, which contribute dissolved nutrients to the neighboring Muddy Creek watershed. Consequently, the Farm is developing methods to sequester phosphorous and limit nutrient contamination.
Before phosphorous sequestration methods can be tested, Farm surface water and groundwater geochemistry must be better understood to determine hydrological pathways for nutrients. We use naturally-occurring dissolved cations, pH, oxidation-reduction potential (ORP), specific conductivity (SC), dissolved oxygen (DO%), total hardness, and alkalinity as chemical tracers to parse the contribution of dissolved ions …

We assayed dissolved nutrient levels in surface and subsurface waters of Eastern Kentucky University’s Meadowbrook Farm in order to assess levels of dissolved nutrients leaving its farmland and draining into the Muddy Creek watershed. The Farm raises both crops and livestock so that nutrient sources include fertilizer and manure. We sampled springs, runoff, and subsurface pipe drainage as well as Muddy Creek on six days from May to August 2016 under a variety of weather conditions. Riparian vegetation along streams has many positive effects on water quality and macroinvertebrate communities, especially in agricultural areas. Some of these effects include erosion prevention, pollutant removal, and lower summer water temperatures. There has been much research done examining the link between these riparian areas and streams; however, riparian vegetation is understudied in karst areas. Watershed biogeochemistry throughout Midwestern agroecosystems have been altered through hydrologic manipulation and over-application of nitrogenous fertilizers. As a result, nitrate (NO3-N) export from subsurface drainage has negative impacts on local and downstream ecosystem health. Wetland installation has proven to be a viable option for targeted management where a large proportion of NO3-N is removed through the bacterially mediated process denitrification. For denitrification to maintain high rates under prolonged NO3-N saturation, a stable supply of labile dissolved organic carbon (DOC) is required. Under iron (Fe3+)-reducing conditions where aqueous Fe2+ and unreduced solid Fe3+-oxides commonly coexist, soil Fe2+ oxidation has been shown to be coupled with nitrate (NO3-) reduction. One possible secondary reaction is the involvement of NO3- and nitrite (NO2-) with Fe-oxide minerals found in many natural environments. Using an instrumented water quality network in Morro Bay Estuary, California from 2007 to 2010 (15 min sampling frequency), this study addressed the two objectives of constructing a nitrate budget and assessing the influence of sampling frequency on water quality parameters. These two objectives led to the submission of an original report of research (Appendix A) and a note (Appendix B) to peer-reviewed journals.

15N-nitrate (NO3 -) pool dilution experiments show that ammonia (NH3) is oxidized to nitrate in geothermal springs up to at least 85C; however, nitrite (NO2 -)- oxidizing microorganisms are only known to grow up to 66°C. We hypothesize that thermophilic microorganisms oxidize nitrite to nitrate at high temperatures. Alternatively, it is possible that nitrite is oxidized abiotically. We propose to test these hypotheses by setting up microbial enrichments designed to grow thermophilic nitrite oxidizing bacteria by varying incubation temperature (50, 65, 80°C), oxygen concentration (20% and 5%), and cultivation media.
