Background

The phase 3 WAYPOINT study (NCT04851964) reported that tezepelumab improved outcomes in patients with chronic rhinosinusitis with nasal polyps (CRSwNP), including nasal polyp size, nasal congestion, and sinonasal symptoms, and reduced the need for surgery and systemic corticosteroids (SCS).

Objective

To evaluate the efficacy and safety of tezepelumab across Japanese Epidemiological Survey of Refractory Eosinophilic Chronic Rhinosinusitis-defined eosinophilic chronic rhinosinusitis (ECRS) subgroups.

Methods

Unplanned 30-day hospital readmission remains a fundamental challenge in US healthcare, associated with increased risk to patient recovery and representing an estimated $52.4 billion in annual expenses (Beauvais et al., 2022). While the rigorously validated LACE index serves as the clinical standard for readmission modeling, its linear structure and four explanatory variables lack the complexity to capture the high-dimensional and interactive nature of patient risk. Through random sampling, sample-based path planners enable autonomous agents to quickly find paths without human intervention. However, due to the paths' randomness, sample-based path planners currently require additional verification, partially nullifying agents' ability to act autonomously. I set out to characterize this uncertainty so humans know what to expect from these path planners and know how to alter the path planner to desired specifications. To ensure the results are theoretical as well as practical, I first create a stochastic model of path length uncertainty using the trade-off between sampling time and optimality. Fine-grained Temporal Action Segmentation (TAS) has become a cornerstone of video understanding, offering dense frame-level predictions essential for clinical assessment, surgical skill evaluation, and human-computer interaction. While TAS methods have delivered strong results on coarse-grained benchmarks, two fundamental challenges persist: (1) global attention mechanisms dilute boundary information critical for subsecond precision, a phenomenon we term the temporal granularity bottleneck, and (2) dense frame-level annotation remains prohibitively expensive, with most datasets requiring exhaustive labeling of lengthy untrimmed videos. This thesis documents the design, deployment, and forward-test evaluation of an evolutionary multi-agent algorithmic trading system on Polymarket, the largest decentralized prediction market. The system pairs a locally-hosted 72-billion-parameter language model with a gradient-boosted statistical filter and an evolutionary selection mechanism that maintains a population of approximately 500 autonomous trading agents. Each agent generates a probability estimate for an event, compares it to the prevailing market price, and trades the resulting disagreement.

Research on Nash equilibrium existence for infinite games has grown into a patchwork of technical preconditions and counterexamples. This paper presents a unified program in equilibrium theory by revising the predominant model of mixed strategies based on countable additivity. A game is specified by a nonempty set of players and, for each player, a nonempty action set and a bounded von Neumann-Morgenstern utility function. Every such game is shown to admit a Nash equilibrium in finitely additive mixed strategies. We compare five numerical approaches for approximating solutions to the Black–Scholes partial differential equation for pricing European call options: FTCS, BTCS, Crank– Nicolson, Monte Carlo simulation, and a physics–informed neural network (PINN). These methods span finite difference techniques, probabilistic simulation, and machine learning. Performance is evaluated based on computational efficiency and accuracy relative to the analytical Black–Scholes solution.

Among the methods, Crank–Nicolson and the PINN demonstrated the strongest overall performance. Crank–Nicolson achieved the highest accuracy but exhibited increased runtime as the number of underlying stock price grid points grew. The Marshall–Olkin family of distributions has gained increasing attention in fields such as reliability engineering, survival analysis, financial risk modeling, and actuarial science because of its flexibility in modeling dependence among events and its wide range of extensions. Despite its growing relevance, a systematic understanding of how research on Marshall–Olkin models has evolved over time is still limited. Deeply virtual exclusive scattering processes (DVES) serve as precise probes of nucleon quark and gluon distributions in coordinate space. These distributions are derived from generalized parton distributions (GPDs) via Fourier transform relative to proton momentum transfer. QCD factorization theorems enable DVES to be parameterized by Compton form factors (CFFs), which are convolutions of GPDs with perturbatively calculable kernels. Accurate extraction of CFFs from DVCS, benefiting from interference with the Bethe–Heitler (BH) process and a simpler final state structure, is essential for inferring GPDs. Translation of Islamic religious texts poses unique challenges requiring both linguistic and theological expertise. This study explores the application of neural machine translation (NMT) models to Arabic-English hadith translation while analyzing semantic similarity patterns across different human translations. Using the complete Sahih Bukhari corpus (7,550 hadiths) as the primary dataset, we adopt a dual approach combining transfer learning and comprehensive neural network analysis to demonstrate the critical impact of corpus size on model performance.

Liver Transplantations are crucial treatment for end-stage liver disease. However, a persistent deficit of donor organs necessitates maximizing the utility of each available graft to minimize failure rates. We evaluated whether donor–recipient molecular immunogenicity metrics - Electrostatic and Hydrophobic Mismatch Scores (HMS/EMS) and eplet-based counts - improve post–liver-transplant survival prediction. The analytic cohort comprised adult, first time, single-organ deceased-donor transplants drawn from Scientific Registry of Transplant Recipients; follow-up was truncated at five years, and the endpoint was all-cause graft failure (earliest of graft failure or death; otherwise, censored). This thesis presents an extension of the Kalman filter to handle nonlinear and non-Gaussian systems. The standard Kalman filter is optimal under Gaussian assumptions but struggles with more complex noise models. This work introduces a novel loss function based on the Mahalanobis distance, which incorporates the covariance structure of measurement errors, enabling the filter to adapt to non-Gaussian scenarios. The neural network framework is applied to predict the system’s process model, while retaining the classical Kalman measurement update. Segmentation of curvilinear structures such as water contours, cracks in cement, and vascular networks in biomedical imaging, poses unique challenges due to extreme class imbalance, irregular morphology, low contrast against complex backgrounds, and the need to preserve global connectivity while detecting fine-scale details. We propose a Multiscale Variational U-Net (MSVU-Net) architecture designed specifically to address these challenges. This dissertation investigates surrogate modeling for fixed-location environmental forecasting using novel data-combination techniques. The work surveys the landscape of observational measurements and numerically generated data, identifying similar research and gaps in current methodologies. The ratio-coupled training framework is introduced to combine two data sources per predicted feature through a tunable parameter that weights training signal strength. An optimization scheme is developed to simultaneously tune surrogate weights and the coupled signal ratio, allowing relative influence between signals to act as an explicit regularizer. This qualitative study examined representation of historically marginalized students in STEM instructional content at the higher education level and its impact on their learning experiences. Despite growing diversity initiatives in STEM enrollment, curricular materials often fail to reflect the identities of underrepresented students. Using critical theory and interpretivist approaches, this research investigated how representation—or its absence—shapes students' sense of belonging, academic identity formation, and persistence. Through semi-structured interviews with undergraduate students from historically marginalized backgrounds, and purposeful sampling, this study captured the lived experiences of students engaging with STEM instructional materials. High-cardinality categorical variables remain difficult to model in tabular data, where classical encoders encounter sparsity, susceptibility to leakage, and the loss of meaningful relational structure. This dissertation develops a unified framework for learning, evaluating, and synthesizing representations of such variables using both traditional encoders and modern embedding methods, including Word2Vec, FastText, Node2Vec, TF–IDF/SVD, and supervised entity embeddings. The framework is applied across three benchmark datasets (Adult, PetFinder, Breast Cancer) and a hierarchical educational case study (IPEDS/CIP). This thesis develops a discrete stochastic linear systems interpretation of age–stage demographic evolution grounded in Leslie operators and realized in a discrete-event simulation implemented with salabim. The central claim is that one annual cycle of the simulation constitutes a cone-preserving, stochastic affine transformation on a high- dimensional population state vector indexed by age, sex, marital status, household type, employment, and education, and that the composition of yearly operators yields a random matrix product whose top Lyapunov exponent is the stochastic counterpart of the Perron–Frobenius growth rate (Caswell, 2001; Tuljapurkar, 1997)[1, 2]. The rapid evolution of DNA foundation models promises to revolutionize genomics, yet comprehensive evaluations are lacking. Here, we present a comprehensive, unbiased benchmark of five models (DNABERT-2, Nucleotide Transformer V2, HyenaDNA, Caduceus-Ph, and GROVER) across diverse genomic and genetic tasks including sequence classification, gene expression prediction, variant effect quantification, and topologically associating domain (TAD) region recognition, using zero-shot embeddings. Our analysis reveals that mean token embedding consistently and significantly improves sequence classification performance, outperforming other pooling strategies. Potential evapotranspiration (PET) exhibits substantial spatial and temporal variability across large landscapes, necessitating site-specific estimation for accurate environmental and water resource assessments. However, obtaining PET or ET data for specific locations across an entire state remains challenging due to the limited number of weather stations and associated environmental datasets. This study aimed to develop an automated geospatial modeling framework to map PET distribution across South Carolina, USA, using PET estimated by the temperature-based Hargreaves–Samani (H–S) method with daily weather data from 59 NOAA stations. Around 36 million people in the world are blind and an additional 217 million have moderate to severe vision impairment. In higher education, four percent of 54,204 undergraduates who participated in the 2022 American College Health Association survey reported to be blind or have low vision. Those students frequently do not have access to data visualizations we generally teach and use in postsecondary statistics and data science classes. Transfer learning is the predominant method for adapting pre-trained models on another task to new domains while preserving their internal architectures and augmenting them with requisite layers in Deep Neural Network models. Training intricate pre-trained models on a sizable dataset requires significant resources to fine-tune hyperparameters carefully. Most existing initialization methods mainly focus on gradient flow-related problems, such as gradient vanishing or exploding, or other existing approaches that require extra models that do not consider our setting, which is more practical. Cancer is mainly caused by a relatively small portion of somatic genome alterations (SGAs), called cancer drivers. Despite success in identifying a good number of cancer drivers, many more remain to be discovered to explain various cancers. Moreover, limited tools are available to identify potential interactions among cancer drivers for a better understanding of oncogenesis. To tackle these challenges, we have developed a novel approach called individualized Bayesian inference using a decision tree (IBI-DT).
