How Low Can You Go? Feature Selection For Drug Discovery, Derek Jones, Sally R. Ellingson, W. A. De Jong

The cost of bringing a drug to market depends on how quickly a candidate drug can be “discovered” and evaluated to ensure safety and effectiveness. In this work we develop a method for predicting whether a given drug and protein compound will “bind.” Our aim is to select a set of features to predict drug-protein interactions.

This study focuses on kinases. Kinase inhibitors are the largest class of new cancer therapies. Selective inhibition is difficult due to high sequence similarity, leading to off-target interactions and side-effects. Pictured here human c-SRC.

Health information technology (HIT), which includes electronic health record (EHR) systems and clinical data analytics, has become a major component of all health care delivery and care management. The adoption of HIT by physicians, hospitals, post-acute care organizations, pharmacies and other health care providers has been accepted as a necessary (and recently, a government required) step toward improved quality, care coordination and reduced costs: “Better coordination of care provides a path to improving communication, improving quality of care, and reducing unnecessary emergency room use and hospital readmissions. This dissertation proposes novel texture feature-based computational models for quantitative analysis of abnormal tissues in two neurological disorders: brain tumor and stroke. Applied behavior analysis (ABA) is considered an effective treatment for individuals with autism spectrum disorder (ASD), and many researchers have further investigated factors associated with treatment outcomes. However, few studies have focused on whether treatment intensity and duration have differential influences on separate skills. The aim of the current study was to investigate how treatment intensity and duration impact learning across different treatment domains, including academic, adaptive, cognitive, executive function, language, motor, play, and social. Mental health related disorders are common diseases, especially among the elder. Among the various mental health diseases, one potential threat to ageing-in-place is the risk of depression. In this paper, we propose a simple unobtrusive sensing system using passive infra-red motion sensors to monitor the activities of daily living of elderly, who are living alone. A feature extraction module comprising of three layers-states, events, and activities, and the corresponding algorithms are proposed to extract features. Objective: Quality assurance of large ontological systems such as SNOMED CT is an indispensable part of the terminology management lifecycle. We introduce a hybrid structural-lexical method for scalable and systematic discovery of missing hierarchical relations and concepts in SNOMED CT.

Material and Methods: All non-lattice subgraphs (the structural part) in SNOMED CT are exhaustively extracted using a scalable MapReduce algorithm. Four lexical patterns (the lexical part) are identified among the extracted non-lattice subgraphs. Non-lattice subgraphs exhibiting such lexical patterns are often indicative of missing hierarchical relations or concepts. In this work we have developed a multi-tiered computational platform to study protein-drug interactions. At the beginning of the workflow more efficient and less accurate methods are used to enable large libraries of proteins in many conformations and massive chemical libraries to be screened. At each subsequent step in the workflow a subset of input data is investigated with increased accuracy and more computationally expensive methods. We demonstrate the developed workflow with the investigation of the lymphocyte-specific kinase LCK, which is implicated as a drug target in many cancers and also known to have toxic effects when unintentionally targeted. It is essential to proactively detect mental health problems such as loneliness and depression in the independently-living elderly for timely intervention by caregivers. In this paper, we introduce an unobtrusive sensor-enabled monitoring system that has been deployed to 50 government housing ats with the independent-living elderly for two years. Then, we also present our initial findings from the 6-month sensor data between August 2015 and April 2016 as well as the survey data to measure the subjective well-being indicator. In this thesis, I will expand upon each step in the process of acquiring and analyzing electroencephalogram (EEG) for the classification of benign childhood epilepsy with centrotemporal spikes. Despite huge advancements in the field of health informatics—natural language processing, machine learning, predictive modeling—there are significant barriers to the access of clinical data. These barriers include information blocking, privacy policy concerns, and a lack of stakeholder support. Unlike diseases with simple genetic risk components, there exists no single marker determining as to whether someone will develop AD. This study evaluated the potential impact of an online spiritual care educational program on pediatric nurses’ attitudes toward and knowledge of spiritual care and their competence to provide spiritual care to children with cancer at the end of life. It was hypothesized that the intervention would increase nurses’ positive attitudes toward and knowledge of spiritual care and increase nurses’ level of perceived spiritual care competence. Allosteric interactions in the Hsp70 proteins are linked with their regulatory mechanisms and cellular functions. Despite significant progress in structural and functional characterization of the Hsp70 proteins fundamental questions concerning modularity of the allosteric interaction networks and hierarchy of signaling pathways in the Hsp70 chaperones remained largely unexplored and poorly understood. In this work, we proposed an integrated computational strategy that combined atomistic and coarse-grained simulations with coevolutionary analysis and network modeling of the residue interactions. This list of resources was designed to complement a project funded by the National Network of Libraries of Medicine (NN/LM) New England Region (NER) entitled “Empathy Learned Through an Extended Medical Education Virtual Reality Project." The project used a virtual reality (VR) experience for 1st year medical students developed by Embodied Labs. The interactive “Alfred Lab” immerses users in the story of a 74-year-old patient who has macular degeneration and hearing loss, allowing users to experience these conditions from the patient’s perspective as he interacts with his family and doctor.

The main aim of the article is to build a method to assess risk perception in real time in order to early detect and prevent risk behaviors and possible human errors. To this end, the relation between mental workload and stress as critical factors affecting risk perception has been investigated. In particular the mental-physical activation generated by an increment of the workload has the effect of reducing the resources needed to perceive risk increasing the worker vulnerability. In efforts to increase safety and reduce peak workload situations in the cockpit, a tool with a different interaction style was developed for use in case of a runway change instructed by Air Traffic Control during approach. In an experiment a workload comparison was made between the new tool and the conventional cockpit. Workload was measured by means of a self-rating after each experiment run, as well as eye blink frequency during each run. This study investigates what strategies technology development marketing leaders utilized to commercialize a new product innovation in the healthcare market. Analysis of data generated 3 major themes: strategies implemented during the prelaunch phase, strategies implemented during the pilot customer phase, and strategies implemented in the broader market launch phase of the CoI process. Findings from this study may contribute to maximizing commercial success and the diffusion of new product innovations in healthcare, which might lead to better patient outcomes and enhanced ways of practicing medicine.
