Inflammatory cytokines such as interleukin-6 (IL-6) and interleukin-12 (IL-12) are central regulators of immune signaling and key biomarkers of disease, yet existing assays for their detection remain slow, invasive, and lack multiplexing ability. This dissertation advances the development of single-walled carbon nanotube (SWCNT) optical nanosensors capable of real-time, multiplexed, and molecularly specific cytokine detection through innovative use of single-stranded DNA (ssDNA) interfaces.

First, an IL-6-specific DNA aptamer was employed as both a dispersing agent and recognition probe for SWCNT fluorescence sensing. Phosphorescent materials are essential in bioimaging, anti-counterfeiting, and other applications due to their prolonged light emission. However, current phosphorescent materials face challenges such as complex synthesis, high costs, and instability. Carbon dots (CDs), with size less than 10 nm, are a promising alternative due to their biocompatibility, tunable optical properties, and simple synthesis. This thesis explores the synthesis of CDs using ethylenediamine (EDA) and phosphoric acid via a microwave-assisted method, followed by silica encapsulation using a modified Stöber process to form CDs@SiO2, aiming to enhance their stability and optical properties. Illicit and hazardous substances pose a serious effect on the human body which can create longer-term issues on the mental and physical health which potentially lead to fatalities. Current methods for detecting illicit substances include analytical techniques like mass spectrometry (MS) and gas/liquid chromatography (GC/LC) which are known for accuracy and sensitivity. However, these instruments are bulky, costly, time-consuming, and require highly trained personnel. Metal-organic supercontainers (MOSCs), a breakthrough development in our laboratory since 2012, have emerged as promising enzyme mimics, showcasing their prowess in encapsulating guest molecules and catalyzing confined chemical reactions. The intrigue surrounding MOSCs stems from their extraordinary traits, encompassing multiple binding domains, high stability, large surface area, tunable pore size, and structural modularity. Primarily comprising sulfonylcalix[4]arenes, divalent metal ions, and carboxylate linkers, MOSCs' quintessential architecture comprises both endo- and exo- nano-cavities, amenable to guest interaction, with geometry and functionality manipulation achievable through precise precursor selection. Fluorinated functional groups have attracted great interest in various aspects of chemistry applications including pharmaceuticals, organic electronics, and polymers. The aim of this study is to explore the impact of fluorinated functional groups on 1) optimization of crystal structures and electronic properties of organic semiconductor molecules, 2) electrochemical polymerization of fluorinated aromatics, 3) photochemical reactivity of carbonyl compounds, and 4) generating new nucleophilic fluorination reagents. Our initial work demonstrates that long fluoroalkyl side chains on planar aromatic systems give rise to fluorine-fluorine (F---F) attractive interactions, assisting formation of lamellar packing motif in the solid state. In the past, we have demonstrated the photophysical properties of several anthraquinone derivatives that act as a luminescent host for various environmental and biological cations. Here we extend our expertise in photochemistry to the class of chemicals containing the pyridoxal moiety. Several ligands were synthesized using eco-friendly, simple, one-step reactions via Schiff base and condensation reactions. The optical properties of these ligands were examined with a series of cations. Orotone photographs are positive photographic images on glass with a gold-colored metallic coating or backing. Although the materials and process used in the production of orotones have been previously documented, there is limited published scientific research on the subject. This study focused on expanding those published works by analyzing a greater number of photographs, including eleven orotones, four hand-colored orotones, two silvertones, and one uncoated orotone. The photographs, dating from the early to mid-20th century, were from the University of Washington (UW) Libraries' collection, except for one orotone that was from the Portland Art Museum.

De novo protein design offers many interesting prospects both as a means to better understand natural protein dynamics and as a potential resource in biomedical and industrial applications. In this work I describe the modification of a simple, well-characterized heme-binding protein by altering side chain residue identities on the hydrophilic surface of the protein to produce variants with a range of net external charges. These charge modifications had a significant impact on nearly every measurable character of the protein. Sol-gel silica is a porous glasslike material that can be prepared at room temperature. In this work, sol-gel method was employed to develop sol-gel sensors for pH measurement and sol-gel silica sorbents for removal of heavy metals.

Sol-gel pH sensors were prepared by entrapping pH indicators in porous solgel silica films. The feasibility of developing optical pH sensors with linear response over a wide pH range by using mixtures of multiple indicators was demonstrated. The Diels-Alder dimerization of trans-1-substituted butadienes is expected to yield a cyclohexene adduct with the substituents in the 3 and 4 positions cis to one another. This prediction is based on past observations of other Diels-Alder additions. The cis-isomer is the only one consistent with a two-stage mechanism proposed by Woodward and Katz.

In the case of trans-phenylbutadiene, the expected adduct is cis-3-phenyl 1-4-(trans-styry1) cyclohexene. Alder, Haydn and Vogt, however, reported that the corresponding trans-isomer is the dimerization product. There is reason to believe that the observation of the trans-isomer may have been the result of product isomerization during purification. …

Using Fe57Mössbauer spectroscopy, two alternative sites occupied by cobalt (10-4 at. % ) impurity atoms in aluminum have been isolated. The substitutional site A is the stable position after annealing the sample above 840 K, followed by a rapid quench. The impurity atoms in site A are characterized by a single line Mössbauer spectrum (indicative of a cubic environment), a room-temperature f = 0.502 (r. m. s. displ. 0.071 A) and an I. S. (reI. to Fe) = -0.421 mm/sec. For anneals below 770 K the cobalt atoms migrate to site B, which is characterized by a …

The purpose of this research project was to study the αᵧ phase in Alnico 6. Other phases were studies for comparison. Also the effect of an applied magnetic field on the formation of the αᵧ phase was investigated.

Three single crystals of Alnico 6 were heat treated for one hour--one at 1250°C, one at 1000°C, and one at 800°C--and water-quenched. The microstructure of each sample was observed with the optical microscope, and X-ray diffraction patterns were taken to determine the crystal structure of the phases present.
