Pyridazines are nitrogen-containing compounds that have been found to show a number of pharmaceutical applications. They exhibit antimicrobial, antifungal, and antibacterial properties to name a few. In particular: 3,4,6-triphenylpyridazine derivatives are known to possess anticancer properties. The purpose of this project is to synthesize tri-substituted pyridazine derivatives using microwave-assisted reactions, and to check its biological applications. Microwave synthesis will help shorten reaction times and produce high, efficient yields. Various catalysts, bases, reaction times, and temperatures will be explored to study the scope of the reactions.

Novel eco-friendly tetramethylguanidinium propanesulfonic acid trifluoromethylacetate ([TMGHPS][TFA]) ionic liquid was developed as catalyst and medium for the Fischer indole synthesis of a wide variety of hydrazines and ketones. The indole products were isolated in high yields and with minimal amounts of organic solvent. This reaction showed that [TMGHPS][TFA] can be regenerated and reused with reproducible yields without eroding the integrity of the ionic liquid.

This work is focused on the design, synthesis and characterization of block copolymers (BCPs) containing both conjugated and polyelectrolyte blocks. This type of rod-coil BCP is considered to have microphase separation which is different compared to coil-coil BCPs. The conjugated block used for studies is regioregular poly(3-alkylthiophene) (P3AT), which can be synthesized by Grignard metathesis (GRIM) polymerization. The polyelectrolyte block used is polystyrene sulfonate with well-defined structure, which can be synthesized by atom transfer radical polymerization (ATRP). Thirty-one complexes of bis-(benzimidazole, benzothiazole, and benzoxazole) compounds with Zr(IV) metal centers were synthesized, characterized, activated with methylaluminoxane (MAO), and then tested for catalytic ethylene polymerization. The activities of the various catalysts were found to be functions of the heteroatoms in the ligand frameworks and the structure around the active metal center. The highest activity was obtained with 38/MAO (424 kg E/mol cat. h). The produced polyethylenes showed high molecular weights (41/MAO, 1.9 $\times$ 10$^{6}$ g/mol) and broad molecular weight distributions (38/MAO, M$_{w}$ = 9.64 $\times $ 10$^{5}$ g/mol, PD = 23). DNA sensing always has an open meadow of curiosity for biotechnologists and other researchers. Recently, in this field, we have introduced an emerging class of molecules containing azo and guanidine functionalities. In this study, we have synthesized three new compounds (UA1, UA6 and UA7) for potential application in DNA sensing in alcoholic medium. The synthesized materials were characterized by elemental analysis, FTIR, UV-visible, 1H NMR and 13C NMR spectroscopies. Their DNA sensing potential were investigated by UV-visible spectroscopy. “Supramolecular chemistry” powered by non-covalent interactive forces forms the crux in the area of host-guest chemistry. Supramolecular assemblies often have different chemical and physical properties than that of its individual molecular entities and are used to develop novel functional materials. Our expertise involves making functional materials from macrocycles, which contain two urea groups and two rigid C shaped spacer groups. These individual macrocyclic components can self-assemble through hydrogen bonding and other non-covalent interactions to form porous supramolecular assemblies that can be used as confined reaction environments and as ligands to synthesize novel metal organic materials.

The reaction of coordination compounds of palladium, rhodium, ruthenium and nickel with thiolated ortho-carborane derivatives- 1,2-dimercapto-o-carborane (1,2-(HS)2-1,2- C2B10H10 ) and 9,12- dimercapto-o-carborane (9,12-(HS)2-1,2- C2B10H10 ) and with carbornanyl dicarboxylate has been studied. To further investigate the chemistry of o-carboranyl dithiolate, both of the reported literature dithiols have been employed to study the reactivity with some of the above mentioned transition metal coordination compounds. Exploratory crystal growth is a versatile technique and one that can advance materials discovery. Through exploratory crystal growth, it is possible to synthesize many new materials that can have applications in many fields, including solid-state lighting. There exist many adaptations for exploratory crystal growth, including the use of alkali halide flux growth and hydroflux growth, both reported herein. By changing variants in these techniques like reactant ratios, temperature, reaction profile, and, most importantly here, the flux compositions, it is possible to change the products formed or fine tune reactions for product purity.

This dissertation focuses on the synthesis of dauer pheromone analogs for biological evaluation, the development of visible light promoted O-glycosylation methods and the development of an Ir(ppy)3 catalyzed C-H hydroxylation method. Chapter 1 describes the synthesis of dauer pheromone analogs. C.elegans is a small nematode that enters a dauer stage when encountering unfavorable environmental conditions. The dauer stage is initiated by the nematodes chemosensation of the dauer pheromone caused by the down regulation biochemical pathways TGF-â and IGF-1. Creation of the thioether linkage is a fundamental step for the chemical preparation of the bacterial quorum sensing molecule S-D-ribosyl-L-homocysteine (SRH). Although previous preparations of SRH and its analogues have been reported in the literature, they employ assorted methods with varied results. Therefore, a reassessment of the methodology used for synthetic preparation of the thioether bond in SRH-like molecules is here considered. This work examines four methods of thioether generation following two mechanisms, bimolecular nucleophilic substitution (SN2) and radical-initiated thiol-ene coupling, in an attempt to generate SRH in a more efficient and reproducible manner. As state universities feel economic pinches of reduced support from state legislatures, and as sources of federal funding for supported research projects face additional challenges, other sources of support need to be found. Two sources under consideration are royalties and licensing fees for successful patents. A good example of a successful developer of patents was Jonas Kamlet, Ph.D. (1914‐1960), who was an early consultant in chemical and other matters and was the successful co-owner, with his wife Edna, of the Kamlet Laboratories. This study reports the design, synthesis, and calcium channel modulatory activity evaluation of a series of 14 novel fused 1,4-dihydropyridine derivatives. The molecular design of the compounds was based on modifications of nifedipine, which is a calcium channel blocker. The compounds were achieved by one-pot microwave-assisted reaction of 4,4-dimethyl-1,3-cyclohexanedione, 5-chlorosalicylaldehyde/3,5-dichlorosalicylaldehyde, an appropriate alkyl acetoacetate, and ammonium acetate in ethanol according to a modified Hantzsch reaction. The structures of the compounds were confirmed by spectral methods and elemental analysis. A series of new aminophosphines [Ph$_{2}$PHN-C$_{6}$H$_{4}$-R, where R = $o$-SH (4a), $m$-SH (4b) or $p$-SH (4c)] were readily synthesized from cheap starting materials by the phosphorylation reaction of $o$, $m$, and $p$-aminothiophenols with Ph$_{2}$PCl in the presence of triethyl amine. The new compounds were characterized by NMR and IR spectroscopy and microanalysis. In addition, aminophosphine ligands-palladium systems were investigated as precatalysts in C-C coupling reactions. Compounds 4b and 4c were proved to be excellent catalysts for Suzuki and Heck cross-coupling reactions.

Eu2+-containing complexes have considerable applications in synthetic chemistry, medical diagnosis, and materials science. However, Eu2+ is easily oxidized in solution when exposed to air. Allen and coworkers have demonstrated that Eu2+ can be stabilized by functionalized cryptands. Based on this idea, I focused my research on synthesizing cryptands. The progress towards synthesizing several modified cryptands is described in the thesis. The Eu2+-containing complexes of these cryptates have potential applications as magnetic resonance imaging contrast agents, luminescent materials, and magnetic materials.

The goal of this project was to study novel synthetic procedures for heterocycles. This was achieved through a two-part investigation. In the first part, a variety of conditions for the gold catalyzed cyclization of allenes were studied. This work was focused on the lesser studied intermolecular reactions of allenes. A variety of allenes were synthesized, and then reacted with 1,3-dipoles and a gold catalyst. These reactions were determined to be unsuccessful through the analysis of nuclear magnetic resonance spectroscopy data. Methyl 4-((9H-(carbazole-9-carbanothioylthio) benzoate (MCzCTB) was chemically synthesized and characterized by FTIR, ^1H NMR, and ^{13}C NMR. A novel synthesized monomer was electropolymerized on a glassy carbon electrode (GCE) in various initial molar concentrations of [MCzCTB]_0= 1, 3, 5, and 10 mM in 0.1 M NaClO_4/CH_3CN and 1 M H_2SO_4. P(MCzCTB)/GCE was characterized by cyclic voltammetry, Fourier transform infrared-attenuated transmittance reflectance, scanning electron microscopy-energy dispersive X-ray, and electrochemical impedance spectroscopy. The capacitive behavior of the modified electrode was obtained by Nyquist, Bode-magnitude, and Bode-phase plots. Cardiovascular disease has quickly become a major health concern in the United States, with numerous citizens dying from cardiovascular disease each year. Older medications, while effective against cardiovascular disease, are problematic to prescribe. A recently isolated natural product, clavatadine A, selectively inhibits human blood coagulation factor XIa. As a result, the synthesis and biological testing of clavatadine A and synthetic clavatadine A analogues that selectively inhibit factor XIa would represent a new direction in cardiovascular disease research. Synergistic Properties-Driven Synthesis Of Aluminides, Antiomonides, And Bioceramics, Pilanda Watkins-Curry

The synthesis, characterization, and properties of selected intermetallics and ceramic materials are presented in this dissertation. In our quest to grow single crystal intermetallics from the metal flux technique, new strategies were employed to avoid thermodynamically stable phases and to ensure sample homogeneity of the crystalline product for structure elucidation and accurate measurements of their magnetic and electrical properties. The relationship between the complex crystal structures and magnetic properties are reported for LnCo2Al8 (Ln = La-Nd, Sm, Yb), CeCo2-xMnxAl8 (0 < x < 1), and Pr2Fe4-xCoxSb5 (1 ≤ x < 3). The discovery of magnetic frustration in LnCo2Al8 (Ln = Ce, Nd) and complex magnetic ordering coupled with large positive magnetoresistance of up to 150% and 60% at 200 K with relatively low field of H = 1 T in Pr2Fe4-xCoxSb5 (x ≈ 2 and 2.5) is also reported. The demand of new inorganic solid materials as scaffolds for the engineering of bone tissue has led to the synthesis of calcium magnesium silicates such as, diopside (CaMgSi2O6), akermanite (Ca2MgSi2O7), monticellite (CaMgSiO4), and merwinite (Ca3Mg(SiO4)2)) for studying dimensionality and mechanical properties. This dissertation highlights the synergism of solid state chemistry at the interface of condensed matter physics and biological engineering. Nanomaterials derived from a group of uniform materials based on organic salts (GUMBOS) have been introduced into the scientific literature through many analytical, biological, and technological applications. These nanomaterials, referred to as nanoGUMBOS, have been shown to display a number of interesting properties including fluorescence, magnetism, tumor targeting, and optoelectronic properties. Herein, we present major studies on nanoGUMBOS including synthesis and size-control, chiral molecular imprinting in polymers, as well as investigation of optical properties and quantum yield of fluorescent semiconductor-based nanoGUMBOS. In this report the synthesis and characterization of L-valine and L-isoleucine poly(ester ureas) (PEUs) for future use in the synthesis of biodegradable PEUs sponges will be discussed. The method of monomer and polymer synthesis will be explained along with the characterization of the monomer and polymer products. This characterization will be done by the use of Nuclear Magnetic Resonance (NMR) and Size Exclusion Chromatography (SEC). Iron oxide nanoparticles are highly researched for their use in biomedical applications such as drug delivery, diagnosis, and therapy. The inherent biodegradable and biocompatible nanoparticle properties make them highly advantageous in nanomedicine. The magnetic properties of iron oxide nanoparticles make them promising candidates for magnetic fluid hyperthermia applications. The design and synthesis of well-defined magnetic particles with nanoscale dimensions is opening new avenues in understanding their fundamental chemical and physical properties. Our work describes a novel facile procedure for the synthesis of homogenous isotropic and anisotropic magnetic nickel nanoparticles. These nanoparticles were produced using a wet-chemical process and characterized by transmission electron microscopy (TEM), high-resolution transmission electron microscopy (HRTEM), selected-area electron diffraction (SAED), powder x-ray diffraction (XRD), energy dispersive spectroscopy (EDS) and thermogravimetric analysis (TGA). Chalcopyrite CuInS2 (CIS) hierarchical structures composed of nanoflakes with a thickness of about 5 nm were synthesized by a facial solvothermal method. The thermodynamically metastable wurtzite phase CIS would be obtained by using InCl3 instead of In(NO3)3 as In precursor. The effects of the In precursor and the volume of concentrated HCl aqueous solution on the phases and morphologies of CIS nanocrystals have been systematically investigated. Experimental results indicated that the obtained phases of CIS nanocrystals were predominantly determined by precursor-induced intermediate products. Highly oxygenated cyclohexanes comprise the structures of a number of pharmacologically interesting molecules, including potently bioactive natural products and carbapyranosides. The latter, which are unnatural analogues of carbohydrates in which the ring oxygen of the parent sugar is replaced with a methylene group, have attracted interest as hydrolytically stable mimetics of their parent O-glycosides. This research reports the development of two general methodologies for the synthesis of highly oxygenated cyclohexanes: (1) Oxocarbenium Ion Cyclization (OCC) and (2) Ring Closing Metathasis (RCM).
Chapter one gives a review of the literature on the synthesis for highly oxygenated cyclohexanes.

Covalent Organic Frameworks (COFs) were first reported in 2005 and are analogous to Metal Organic Frameworks (MOFs), with proposed applications in separation, storage and sequestration. Since COFs are comprised of lighter elements they are considered more advantageous for weight-sensitive applications. However, COFs are susceptible to degradation by weak nucleophiles such as water (i.e. hydrolysis). The Lavigne group has focused on developing structure-property relationships for COFs. It was previously demonstrated that the porosity of the COFs could be tailored by incorporating small alkyl chains on one monomer. Inclusion of these alkylated monomers into COF materials effectively increased the COF hydrophobicity resulting …

Fructose-1,6-bisphosphate activates ADP-glucose pyrophosphorylase and the synthesis of glycogen in Escherichia coli. Here, we show that although pyruvate is a weak activator by itself, it synergically enhances the fructose-1,6-bisphosphate activation. They increase the enzyme affinity for each other, and the combination increases Vmax, substrate apparent affinity, and decreases AMP inhibition. Our results indicate that there are two distinct interacting allosteric sites for activation. Hence, pyruvate modulates E. coli glycogen metabolism by orchestrating a functional network of allosteric regulators. We postulate that this novel dual activator mechanism increases the evolvability of ADP-glucose pyrophosphorylase and its related metabolic control.

The coordination of pyridyl-tetrazole derivatives containing ester substituents, at either the N-1 or N-2 position of the tetrazole ring, with copper(II) chloride results in the formation of either 1:1 or 1:2 copper to ligand complexes, depending on the ligand. However, when the ester functionality is changed to a carboxylate group, the resulting complexation reactions yield metal-organic frameworks. The resulting structures vary dramatically in pore size, depending on both reaction solvents and position of carboxylate group on the tetrazole ring. Despite the presence of sodium cations in the reaction mixtures, no sodium incorporation was ever observed in any of the complexes. …
