Theses and Dissertations
Receptors are important drug targets for the pharmaceutical industry, since they are involved in many illnesses. Nuclear receptors are a subset of receptors that control a variety of physiological events, by expression of genes from DNA. One of the nuclear receptors, the androgen receptor, is a steroid binding receptor that controls a variety of regulatory and development functions in the body. It is also the cause of some types of cancer, so the inhibition of this receptor is crucial. One of the processes studied to inhibit the function of the receptor is to see if an allosteric ligand, TRIAC, can …

Development Of Electrochemical Sensors Suitable For In Vivo Detection For Neurotransmitters, Mengjia Hu
Graduate Theses and Dissertations
The electrochemical method of redox cycling was exploited to achieve new discoveries in neurotransmitter detection and to advance its suitability toward in vivo use. Redox cycling has advantages in signal amplification, selectivity of species based on their electrochemical reaction mechanisms, and limited or no background subtraction. Distinction of dopamine from norepinephrine in a mixture with an electrochemical method at unmodified electrodes was demonstrated for the first time in vitro. This ability resulted from a series of fundamental studies of redox cycling behavior of the catecholamines (dopamine, norepinephrine and epinephrine) using different electrode configurations. Taking advantage of the ECC’ mechanism associated …

Analysis Of Scientific Argumentation In Two Physical Chemistry Classrooms Using The Pogil Approach, Alena C. Moon
Open Access Dissertations
The benefits of facilitating argumentation in science education have been well reported (Jimenez-Aleixandre & Erduran, 2007). Engaging in argumentation has shown to model authentic scientific inquiry as well as promote development of content knowledge. However, less emphasis has been placed on facilitating argumentation in upper level undergraduate courses, though it is important for evaluating undergraduate curricula to characterize upper level students’ scientific reasoning. This work considers two implementations of the POGIL physical chemistry curriculum and evaluates the classroom argumentation. The researchers aimed to consider the content of the arguments and dialectical features characteristic of socially constructed arguments (Nielson, 2013). To …

Purification And Preparation Of Intrinsically Disordered Proteins For Nmr Spectroscopy, Kyle Chamberlain
Open Access Theses
NMR spectroscopy has recently become a promising field for protein characterization and dynamic studies. As the technology and pulse sequences improve for tracking proteins, a greater demand for developing effective purification protocols to produce NMR grade protein samples have arisen. This thesis explores two proteins: histone H4 tail and Mucin 1; Two very different proteins that require different methods of expression and purification to achieve a high enough yield for NMR analysis. H4 is a water soluble protein that weighs ~2.7 kDa, and has no extinction coefficient. Since the protein is too small for many methods of expression and purification, …

Targeted Metabolomics On The Shikimate And Aromatic Amino Acid Biosynthetic Pathways, Robert E. Wheeler
Open Access Theses
The shikimate and aromatic amino acid biosynthesis pathways are some of the most studied biosynthetic pathways in nature due to their fundamental importance. Interest in this field stems from synthesis of the essential amino acids, tryptophan, tyrosine and phenylalanine. The pathways also are important as generating the precursors of thousands of secondary metabolites and lignin. Inhibiting the pathway has led to discovery of herbicides, pesticides as well as tuberculosis drugs. However there is currently a lack of a comprehensive targeted method for detecting and quantifying the majority of the intermediates in these pathways. Often multiple extraction and detection methods are …

Known primarily for its single electron chemistry, controlling the electronics at uranium to facilitate multi-electron processes remains a fundamental challenge in the actinide sciences. Our group has implemented the use of redox-active ligands to assist in the stabilization of low-valent uranium analogues, capable of performing multi-electron chemistry at a single uranium center. Of particular interest to our group are high-valent imido complexes, which are ubiquitous in organometallic and coordination chemistry due to their unique bonding properties and their ability to mediate bond-forming reactions. Uranium imido complexes, in particular, are the subject of much research to determine the degree of f- …

Mass spectrometry (MS) stands in an outstanding position in analysis of biological specimens owing to its abundant structural information, high accuracy, incomparable sensitivity, high speed, and the large variety of its applications. The ion source, an instrumental part for converting the analyte into ions, has played an important role in analyzing biological specimens by MS. However, the performance of conventional spray-based ionization methods always suffers from chemical interferences derived from complex biological matrices. A series of sample extraction, purification, and separation steps is required before the ionization, so as to ensure excellent performance of MS analysis. In order to simplify …
Synthesis Of Dmap Tethered Polymer Chains For Reversible Micelle Formation, Richard Arthur Fredricks
Synthesis Of Dmap Tethered Polymer Chains For Reversible Micelle Formation, Richard Arthur Fredricks
Master's Theses and Doctoral Dissertations
Dimethylaminopyridine (DMAP) can act to catalyze numerous organic reactions. To increase its recovery and reuse, it has been tethered to dendrimers and to micellar systems, creating nano-scale catalytic pockets for reactions to proceed. Previously developed systems show diminished reactivity over time as the products of the reaction fill up the nano-scale reactor. Thus, the goal of my research is to use RAFT polymerization methods to synthesize a hydrophobic polymer chain bearing 4-methylaminopyridine units and to attach it to a hydrophilic polymer chain to create a polymer system capable of micelle formation in solution. These functionalized micelles are designed to be …

Graduate Theses and Dissertations
An improved synthetic method was developed for symmetric ruthenium(II) polypyridine complexes with the form L2Ru(diphen)RuL2. The scope of the reaction was investigated in regards to the ligands, bridging ligands, and starting materials. Several ligands were successful in the synthesis, including 2,2’-bipyridine (bpy), 4,4’-dimethyl-2,2’-dipyridyl (dmb), 1,10-phenanthroline (phen), 4,7-diphenyl-1,10-phenanthroline (dpphen), and 3,4,7,8-tetramethyl-1,10-phenanthroline (tmphen). Ligands that did not react to form symmetric dimeric complexes were 2,2’-bipyrazine, bpz, and 2,2’-bipyrimidine, bpm.
Dpp, 2,3-bis(2-pyridyl)-pyrazine, effectively replaced diphen as the bridging ligand to produce (phen)2Ru(dpp)Ru(phen)24+. However, replacing the [Ru(CO)2Cl2]n with Ru(DMSO)4Cl2 did not prove successful. The newly developed synthesis was also applied to the synthesis of …

Asymmetric Synthesis And Transition Metal-Catalyzed Cross-Coupling Arylations Of Selected Organolithiums, Barry Kyle Sharp
Graduate Theses and Dissertations
My former boss, Dr. Gawley, always loved to say, “The world is chiral” (à la Pasteur). From DNA and proteins to hands and feet, it is obviously true. Also, a wide variety of chemical products exist as single enantiomers. Advances in chemical technology have greatly accelerated asymmetric synthesis in the past quarter century, and namely, organolithiums, have been shown to provide a versatile route to chiral natural products and biologically active molecules. Versatility arises from the array of methods that produce a chiral organolithium. Dynamic thermodynamic resolution (DTR) is considered one of the most practical methods, but among the others …

Designing Fret Assays To Study Electrostatic Interactions Pertaining To The Binding Of Intrinsically Disordered Proteins, Ashley Ann Howard
Graduate Theses and Dissertations
Fibroblast growth factor receptor plays a major role in several biological processes. Without FGFR, a human cannot live. FGFR is involved in cell differentiation and wound healing. Of course, if FGFR signaling becomes unregulated, it causes severe distress in the body. Several cancers are contributed to high signaling levels, as well as developmental conditions like rickets and Kallmann’s syndrome. FGFR is thought to undergo an auto-inhibition (or self-regulatory) process in order to try to facilitate regulation. The exact method of this inhibition is currently unknown, but is proposed to involve the unstructured acid box region of FGFR. We developed a …

Graduate Theses and Dissertations
Quantum dots are a valuable tool in many research applications and commercial development. Their applications are far reaching and as such have become a topic of great interest. One of their greatest assets is the ability to utilize them as water soluble fluorescent labels. Rendering the nanocrystals water soluble can be accomplished in several ways, but one very popular method is by capping them with water-soluble multi-functional organic ligands. However, these ligands, when attached to the quantum dot surface, are labile and can be exchanged which can be problematic when using the quantum dots in research applications. This projects investigates …

Graduate Theses and Dissertations
Near-infrared (NIR) responsive nanoparticles (NPs) like gold nanorods (GNRs) are important in biomedical fields because of their transparency for biological tissues. Although GNRs are sought after as contrast agents for theranostics in cancer studies, capping ligands like cetyltrimethylammonium bromide (CTAB) for the GNR synthesis are toxic for biological tissues. The need for an alternative to toxic GNRs is of interest to alleviate the problem.
This work aimed to optimize the synthesis of NIR responsive nanosphere composites (NSCs) by inertial force (g-force) using colloidal gold NPs as model, elucidate the mechanism for the NSC formation, and study their detailed physicochemical characteristics. …

Effect Of Crimping, Multilayering, And Twisting On Polymer Fibers For Acl Replacement, Mariana Ocampo
Theses and Dissertations - UTB/UTPA
The Anterior Cruciate Ligament (ACL) is the most common knee injury in the United States; roughly 150,000 ACL surgeries are performed annually. The ACL lacks self-healing properties due to its location inside the knee’s intra – articular environment (IA). Treatments for ACL ruptures involve the use of autogenous grafts or allografts. More suitable treatments can be found in new bio engineered tissue scaffolds, composed of natural and synthetic polymers. The polymers provide stronger mechanical properties and the ability to promote cell adhesion. It is believed that mimicking the crimping pattern of human ligaments will enhance polymer properties. Four polymers were …

Agricultural residues constitute the bulk of potential feedstock available for cellulosic fuel production. On a global scale, rice straw is the largest source of agricultural residues and is therefore an ideal crop model for biomass deconstruction studies. Lignocellulosic biofuel production involves the processes of biomass conditioning, enzymatic saccharification, microbial fermentation and ethanol distillation, and one of the major factors affecting its techno-economic feasibility is the biomass recalcitrance to enzymatic saccharification. Preconditioning …
The Effects Of Molecular Chaperones On Tau Fibril Assembly, Ahmed Omran
The Effects Of Molecular Chaperones On Tau Fibril Assembly, Ahmed Omran
Electronic Theses and Dissertations
The accumulation of microtubule-associated protein tau into fibrillar aggregates is the hallmark of Alzheimer’s disease and other neurodegenerative disorders, collectively referred to as tauopathies. Fibrils can propagate from one cell to the next and spread throughout the brain. However, a study shows that only small aggregates can be taken up by cultured neuronal cells. The mechanisms that lead to the breakage of fibrils into smaller fragments remain unknown. In yeast, the AAA+ chaperone HSP104 processes the reactivation of protein aggregates and is responsible for fragmentation of fibrils. This study focused on investigating the effects of molecular chaperones on tau fibrils …
