Chronic inflammation and dysfunctional inflammatory responses are believed to be contributing factors to the severity of many diseases. Since the emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), it has been recognized that levels of inflammatory markers and the severity of coronavirus disease-2019 (COVID-19) cases are closely related. The work within this dissertation begins with establishing an alternative platform for the diagnosis of COVID-19 and explores ways to enhance the diagnostic utility of this platform. Over the past decades, nucleic acids have emerged as crucial frameworks with significance in biological systems and functional material engineering. Cytosine-rich deoxyribonucleotide sequences can form quadruplex structures a.k.a. i-motifs. Recently, i-motifs have attracted scientific interest in biological and material design areas.

In this work, we systematically investigate an effect of structured and unstructured modifications such as hairpins, stems, loops and overhangs on the kinetic and thermodynamic properties of DNA i-motifs. We demonstrate that the modifications can influence i-motif’s stability and folding/unfolding rates: thus, double stranded stems and hairpins are the best at stabilizing i-motifs and increasing kinetic rates of folding. …

Perovskites have emerged as a promising candidate for low-cost production of solar cells. However, the most critical barrier for commercialization of perovskite solar cells (PSCs) is the inadequate stability of the organic metal halide perovskites (OMHPs). The degradation of OMHPs is induced by light, heat, air, and electrical bias. The known degradation pathways involve the oxidation of I- and Sn2+, dissolution of perovskites by moisture, irreversible reactions with water, ion migration, and ion segregation. The work outlined in this dissertation describes acid-base catalyzed projects facilitating the construction of key synthetic analogs with vast therapeutic applications. Three major projects have been covered demonstrating various synthetic routes developed by our research group to furnish key derivatives of heterocyclic compounds, including coumarins, carbohydrate moieties, and beta-lactams. By their catalytic nature, acids, and bases are known to enhance the reactivities of compounds. We investigate switching of photoluminescence (PL) from PbS quantum dots (QDs) crosslinked with two different types of photochromic diarylethene molecules, 4,4'-(1-cyclopentene-1,2-diyl)bis[5-methyl-2-thiophenecarboxylic acid] (1H) and 4,4'-(1-perfluorocyclopentene-1,2-diyl)bis[5-methyl-2-thiophenecarboxylic acid] (2F). Our results show that the QDs crosslinked with the hydrogenated molecule (1H) exhibit a greater amount of switching in photoluminescence intensity compared to QDs crosslinked with the fluorinated molecule (2F). With a combination of differential pulse voltammetry and density functional theory, we attribute the different amount of PL switching to the different energy levels between 1H and 2F molecules which result in different potential barrier …

YO bands are conspicuous in the spectra of S stars. The laboratory spectrum of the A2Π–X2Σ+ electronic transition has been recorded with a Fourier transform spectrometer using a composite-wall hollow cathode lamp source. The A2Π–X2Σ+ transition with v ' ≤ 4 and v'' ≤ 4 has been rotationally analyzed using the modern spectral fitting program PGOPHER. Diarylethenes (DAEs) are an important class of photoswitchable compounds that typically undergo reversible photochemical conversions between the open and closed cyclized forms upon treatment with UV light or visible light. In this study, we introduced thioacid functional groups to several photochromic dithienylethene (DTE) derivatives and established a method that can be used to prepare these photoswitchable thioacids. Four thioacid-functionalized diarylethene derivatives were synthesized through the activation of carboxylic acids with N-hydroxysuccinimide, followed by reactions with sodium hydrosulfide with yields over 90%. In this study, reproducible label-free detection and quantification of the antiretroviral drug emtricitabine (FTC) down to 78 ng/mL in human plasma by surface enhanced Raman spectroscopy (SERS) is presented. A novel plasma sample pretreatment method using silver nitrate and silver colloidal nanoparticles (Ag CNPs) was used to prepare the plasma samples for analysis. The pretreated plasma samples were evaporated to dryness on an aluminum surface and a computer-controlled Raman scanning system was used to collect spatially resolved SERS spectra of the entire surface. "Pathological processes often involve complex biochemical changes which can be assessed using advanced mass spectrometry. In this present dissertation, two fields were studied: traumatic brain injury (TBI), and water contamination by L. pneumophilia. TBI is a pressing public health concern for which current clinical tools remain inadequate. We present newly developed mass spectrometric methods to access metabolites associated with blast induced TBI. We applied these methods to the biofluids of soldiers conducting explosives training. Significant changes in several metabolites were observed between pre- and post-blast specimens, including changes that increased with repeated exposure. These changes point to the possibility of …

ZrO is a well-studied metal monoxide because of its astrophysical importance in characterizing S stars. The bands of the d³Φ–a³Δ system (γ system) with v′≤4 and v" ≤ 5 are rotationally analyzed using the PGOPHER program to provide spectroscopic constants. The high-resolution ZrO emission spectrum was recorded with a Fourier transform spectrometer using a high-temperature carbon furnace source. New ab initio calculations of the transition dipole moment were carried out in order to determine the vibronic band strengths. Tape evidence is often used in criminal cases involving violent crimes, kidnappings, improvised explosive devices (IEDs), and drug trafficking. This evidence can reveal potential links between suspects, items, or scenes. The forensic examination of electrical tape can provide investigative leads or offer support to alternative hypotheses evaluated in the courtroom. "Shape-memory poly(isocyanurate-urethane) (PIR-PUR) aerogels are nanoporous solids that can return to their original shape after being compressed, through a heating actuation step. This thesis compares the effectiveness of various metal ions as catalysts in the formation of PIR-PUR aerogels, and explores the correlation between catalytic activity, nanomorphology, and mechanical properties of the resulting aerogels. The gelation rate was found to increase from Fe to Cu and then decline from Cu to Ga in the periodic table. CuCl2 was found to be the fastest catalyst, and FeCl3 the slowest. "The development of highly sensitive and efficient analytical methods utilizing advanced instrumentation is necessary to help improve disease diagnosis and therapeutics. A major neuro-consequence of traumatic brain injury (TBI) is oxidative stress from the generation of reactive oxygen species and the depletion of antioxidant defenses. Alteration in concentrations of certain small molecules also occurs with the disease progression and can help understand TBI pathophysiology. Two analytical methods employing liquid chromatography with tandem mass spectrometry (LC-MS/MS) were developed and validated to monitor the potential small-molecule TBI biomarkers at sub-ppb levels. Light-driven molecular switches that can be turned on and off on demand have found many biological applications. For DNA, RNA, and synthetic derivatives, the switch can be used to interfere with a hybridization event with a complementary strand. To explore this potential, we have synthesized three light-sensitive PNA which have an azobenzene group (PNA-5azo5 and PNA-4azo4), and two azobenzene groups (PNA-2azo5azo2) as part of the PNA backbone. Ytterbium(III)-doped glasses are widely known for their application in near-infrared (NIR) lasers and nowadays attract attention for solar spectral conversion in photovoltaic cells. In this study, the effects of ytterbium(III) concentration on the physical, structural, optical, and thermal characteristics of Yb-doped phosphate glasses were investigated. The glasses were synthesized using the melt-quenching technique with nominal compositions of 50P2O5:(50 – xBaO):xYb2O3 (x = 0, 0.5, 1.0, 2.0, 3.0 and 4 mol%).
