The work described here provides measured data to improve the understanding of the interaction between a pesticide and leaf surface after application. Two methods were developed, one using a newly introduced instrument, for the extraction of pesticides from leaves. This is required to measure the concentration of pesticide in the leaves. Using one of the developed extraction methods, measurements were made to determine how a pesticide equilibrates between a leaf and the air above it. These measurements were incorporated into a pesticide fate model that predicts how a pesticide moves through the environment after application to an agricultural field. The …

LMWGs self-assemble through noncovalent interactions such as hydrogen bonding, halogen bonding, van der Waals interactions and hydrophobic forces in solvents. The development of new therapeutic options against Clostridioides difficile (C. difficile) infection is a critical public health concern, as the causative bacterium is highly resistant to multiple classes of antibiotics. C. difficile, an anaerobic spore-forming Gram-positive pathogenic bacterium, is a major cause of hospital-acquired infections. C. difficile persists in the environment and spreads the infection to new hosts in the form of dormant spores and can persist within hosts as surface-attached biofilms. These studies investigate bacterial vegetative cell survival, biofilm formation, and sporulation in response to stress. Spin polarized photocathodes are necessary to examine parity violations and other fundamental phenomena in the field of high energy physics. To create these devices, expensive and complicated growth processes are necessary. While integral to accelerator physics, spin polarized electrons could have other exciting applications in materials science and other fields of physics. The current study probes the structure, dynamics, and rheological behavior of associating polymers including ionomers in melts and solutions as well as conjugated polymers confined into nanoparticles, using molecular dynamics (MD) simulations and neutron scattering techniques. The study focuses on two families of associative polymers, ion containing macromolecules and conjugated polymers.
Polymers that consist of ionizable groups along their backbone found uses in a broad range of applications. Examples include light weight energy storage and generation systems, and biomedical applications, where the polymers act as ion exchange membranes, and actuators. In the search for a sustainable method to meet increasing energy needs, solar energy emerges as an underutilized, plentiful resource. Solar intermittency and requirements for transportation necessitate storing solar energy in the form of chemical bonds via artificial photosynthesis. Photoelectrochemical (PEC) water splitting generates hydrogen fuel from solar energy and water. A semiconducting material that successfully meets the complex requirements for building an industrially scalable PEC device has yet to emerge. Trypanosoma cruzi, the causal agent of Chagas disease (CD), currently affects 6-8 million people worldwide, predominantly in Latin America, where the parasite is endemic. The treatment of CD is based on two drugs, benznidazole (BZN) and nifurtimox (NFX). These drugs cause adverse effects and are more effective in the acute phase of the disease. Conventional serology assays can be used for diagnosis, but they are not suitable for determining chemotherapy effectiveness because patients take approximately 10-20 years to exhibit negative seroconversion. Protein aggregation that results in the formation of amyloid fibrils has been linked to many neurodegenerative disorders, including Alzheimer’s disease and Parkinson’s disease. The sulfur atoms in methionine (Met) and cysteine (Cys) residues of proteins can be readily oxidized, significantly affecting their properties. Oxidation of sulfur-containing amino acids has recently been shown to affect protein fibrillation. This work presents novel findings on Cys and Met redox reactions that are related to the formation of amyloid fibrils and on the polymorphism of a model fibrillogenic protein, hen egg white lysozyme (HEWL). Ion mobility spectrometry - mass spectrometry (IMS-MS) has potential for the investigation of structure and dynamics in large biopolymers, which will come to full fruition only with a firmer understanding of how to interpret the experimental data. Numerous studies have employed elements of nucleic acid (NA) secondary structure, such as duplexes and hairpins, to explore the relationships between structure, experimental conditions, and actual observations. When combined with molecular dynamics simulations (MDS), IMS-MS can be effectively employed to perform structural elucidation of biomolecules that are not readily amenable to established techniques employed for structural analysis.

Plants of the Veratrum genus have been used throughout history for their emetic properties, rheumatism, and for the treatment of high blood pressure. However, inadvertent consumption of these plants, which resemble wild ramps, induces life-threatening side effects attributable to an abundance of steroidal alkaloids. Several of the steroidal alkaloids from Veratrum spp. have been investigated for their ability to antagonize the Hedgehog (Hh) signaling pathway, a key pathway for embryonic development and cell proliferation. Uncontrolled activation of this pathway is linked to the development of various cancers; most notably, basal cell carcinoma and acute myeloid leukemia. Additional investigation of Veratrum …

Allostery (1) is the process through which proteins self-regulate in response to various stimuli. Allosteric interactions occur between nonadjacent spatially distant residues (1), and they are exhibited through the correlated motions (2) and momenta of participating residues. The location of allosteric sites in proteins can be determined experimentally but computational methods to predict the location of allosteric sites are being developed as well (2-4, 10). Our laboratory has previously shown that nicotine oxidoreductase (NicA2), a member of the flavin-containing amine oxidase family, uses a cytochrome c protein (CycN) as its electron acceptor rather than characteristically accepted molecular oxygen. Within the Pseudomonas putida S16 genome, the nicA2 gene forms an operon with cycN and pseudooxynicotine amine oxidase (pnao), another flavin containing amine oxidase enzyme. The first two steps in the nicotine catabolism pathway are catabolized by NicA2 and Pnao. Through redox chemistry, NicA2 converts nicotine to pseudooxynicotine and Pnao converts pseudooxynicotine to 3-succinoylsemialdehyde-pyridine. Nanomaterials offer abundant applications not accessible in regular bulk structures. A nanotubular structure (nanoscroll or nanotube) is a 1D architecture with distinct properties varying from that of the parent material. The new structure for example provides more surface area and presents more active sites on the structure exterior, interior, or edges. In this research, different nanotubular materials were targeted including nanopeapods (NPPs), nanoscrolls (NScs), and their various derivatives.

Halogen bonding, the attractive interaction of an electrophilic region on a halogen atom with a nucleophilic region on another atom or molecule, provides a highly directional tool in forming solid-state motifs. This interaction, along with the related chalcogen bonding interactions, forms powerful synthons, which, when combined with other typical intermolecular attractions such as hydrogen bonding, allow for the design of supramolecular structures and inputs to crystal engineering. This dissertation research serves two primary purposes: (1) to catalog halogen and chalcogen bonding interactions with various donor molecules and (2) to utilize these interactions to probe interesting organic transformations.

Irinotecan is a cancer medication approved for medical use in the United States in 1996. It is currently administered via intravenous injection. However, intravenous dosing has disadvantages such as requirement of administration by a trained professional in a medical facility, possible severe adverse effects, pain at the injection site, and possible hemolysis if injected too rapidly. Previously oral administration was not feasible because the p-glycoprotein (p-GP) on the cell membrane of stomach cells acts as a defense mechanism against harmful substances by pumping the irinotecan back into the intestinal lumen for excretion. Factor XIII (FXIII) is an emerging target for treating blood clotting and cardiovascular related diseases. FXIII can be activated non-proteolytically by the presence of elevated Ca2+ levels (2-100 mM) or proteolytically by thrombin-cleavage of the Activation Peptide along with low mM Ca2+. The studies herein utilized fluorescence to examine how monovalent and divalent ions influence the transglutaminase activity and conformation of FXIII. Monodansylcadaverine assays revealed that increasing ionic radius (Cs+ > K+ > Na+ > Li+) and increasing ionic strength (XCl- levels and SO42- > Cl-) elevated FXIII-A transglutaminase activity. Healthcare-associated infections (HCAIs) affect 1.7 million hospitalized patients each year, resulting in over 98,000 deaths and anywhere from $28.4 to $45 billion in treatment costs. Furthermore, it has been established that more than 80% of these infections are caused by biofilms While HCAIs can be bacterial, viral, fungal, or parasitic, there are between 12 and 17 species cause 80 to 87% of all HCAIs. Among the most commonly isolated microorganisms are Pseudomonas aeruginosa (P. aeruginosa) and Staphylococcus aureus (S. aureus). Our group has shown that reduction of titanium complexes supported by the sterically encumbering N-donor ligands 1,3â??bis(Dipp)imidazolinâ??2â??iminato (ImDippNâ??) and ketimine guanidinate (ketGuan = [(tBuC=N)C(NDipp)2]-) gives access to electron rich platforms featuring a metal coordinated, reduced, dearomatized arene substituent, which act as an electronic reservoir capable of facilitating multi-electron transfer chemistry driven by the rearomatization driving force. For instance, the arene-masked Ti(II) synthon (DippketGuan)(η6-ImDippN)Ti (Dipp = 2,6-iPr2C6H3) (TiDipp) is a versatile and potent one- and two-electron reductant capable of distinctive late-metal type reactivity such as transfer hydrogenation and catalytic olefin hydrogenation. Recent emphasis on carbon dioxide utilization has necessitated the exploration of different catalyst compositions other than copper-based systems that can significantly improve the activity and selectivity towards specific CO2 reduction products at low applied potential. In this study, a binary CoTe has been reported as an efficient electrocatalyst for CO2 reduction in aqueous medium under ambient conditions at neutral pH. CoTe showed high Faradaic efficiency and selectivity of 86.83 and 75%, respectively, for acetic acid at very low potential of − 0.25 V vs RHE. YidC is a membrane protein that facilitates the insertion of newly synthesized proteins into lipid membranes. Through YidC, proteins are inserted into the lipid bilayer via the SecYEG-dependent complex. Additionally, YidC functions as a chaperone in protein folding processes. Several studies have provided evidence of its independent insertion mechanism. However, the mechanistic details of the YidC SecY-independent protein insertion mechanism remain elusive at the molecular level. This project suggests improvements to the treatment methods utilized for the sample preparation and evaluation of minerals in soil samples encountered in forensic science casework and research. The evaluation of pre-treated and post-treated soil samples were performed using Fourier-Transform Infrared Spectroscopy with Attenuated Total Reflection (FTIR-ATR) and Scanning Electron Microscopy-Energy Dispersive Spectroscopy (SEM-EDS). Because soil samples contain a significant amount of organic matter and water which interfere with examination of these minerals while using these instruments, pretreatment methods are necessary to isolate the mineral fraction for examination and analysis.

In the present study, Zea mays seedlings grown under nano Cu(OH)2 (nCu), bulk Cu(OH)2 (bCu), and ionic CuSO4 (iCu) compound exposure were harvested after six days. The nutritional profile was determined to be significantly disrupted in the roots by 1000 ppm bCu treatment, resulting in a 58.7% reduction in potassium compared to the control. In the shoots, a significant decrease of manganese was observed for 10 and 1000 ppm iCu treatments with 55.7% and 64.2% reductions, respectively. The overall protein content and catalase (CAT) enzymatic activity, however, remained unaffected in either roots or shoots, while an absence of polyphenol oxidase …

Hydrogen is one of the most promising alternatives and clean fuel sources, but it has faced several hurdles due to storage and transportation issues. Several methods have been proposed, developed, and are being improved. Key among these is the use of polymers as safe hydrogen storage media.
This work investigated the polymerization of methyl vinyl ketone using different concentrations 2,2’-Azobis(2-methylpropionitrile) as the initiator in a free radical polymerization mechanism.
