Chitosan is examined as a non-viral vector capable of efficient gene delivery due to its favorable properties such as biodegradability, biocompatibility, and is non-toxic to mammalian cells. Cell modifications and vector permeability are enhanced in non-viral vectors that incorporate electrostatic interactions. This work synthesizes cationic (N+, P+) chitosan derivatives, to enhance electrostatic interactions towards nucleotides, using amide coupling agents EDC and HOBt. Herein, I present my work towards two goals: shear force-responsive materials and sugarconjugated fluorescent probes for bacterial identification applications. The first work is on the development of stimuli-responsive polymers that allow for precise control over the release of encapsulated material. Stimuli-responsive polymers are becoming increasingly important in drug delivery and other applications. However there have been few reports on shear force-responsive micellar systems. I show the synthesis, and characterization by 1H and 13C NMR of four macro-chain transfer agents (macro-CTAs), one of which has not been previously reported. Autocatalytic intramolecular isopeptide bonds have been found in nature in certain gram-positive bacterial pilus structures. Recently, splitting of these domains that are capable of autocatalytic intramolecular isopeptide bond formation have been applied to create stable, selective, bio-orthogonal Catcher/Tag systems. The CnaB2 domain found in the FbaB pilus structure of Streptococcus pyogenes, has yielded the Catcher/Tag, Protein/Peptide systems termed SpyCatchter and SpyTag. Recent study has focused on tag optimization, stability and bio-orthogonality evaluation, along with applications in bioconjugation. Every year many international students come to study in the United States. In addition to all the challenges associated with coming to a new country, non-native English speakers (NNES) face the additional challenge of having to learn their content in English. Therefore, in this project, I recruited NNES participants from general chemistry courses since preliminary inquiries indicated that they have the largest enrollments. Because there were relatively few students at MSU who are non-native English speakers studying chemistry, I used a case study design to be able to extract generalizations from an in-depth study of a few individuals. Electronic structure theory has progressed significantly within the last few decades, venturing far from the early days of the Hartree-Fock self-consistent field method. Modern electronic structure theory focuses on compound methods, which operate under the idea that we can take a lower level of theory computation (typically, a result from Hartree-Fock, Configuration Interaction, Coupled Cluster or Moller-Plesset perturbation theory) and add in higher level of the theory corrections such as extrapolations to the infinite basis set limit, as well as, relativistic effects.

This paper documents modifications to an existing vacuum system to allow laser-induced fluorescence spectroscopy measurements within simulated atmospheres under a variety of conditions. This added capability will expand the laboratory’s ability to experimentally validate a computational model that calculates the effects of radiation within the atmosphere. The computational model could reveal radiation-induced chemical products that can be used to develop an alternative detection method that can be implemented from a safe distance. Quercetin, a common dietary flavone, is a competitive inhibitor of glucose uptake and is also thought to be transported into cells by GLUT1. In this study, we confirm that quercetin is a competitive inhibitor of GLUT1 and also demonstrate that newly synthesized compounds, WZB-117 and BAY-876 are robust inhibitors of GLUT1 in L929 cells. To measure quercetin interaction with L929 cells, we develop a new fluorescent assay using flow cytometry. The binding of quercetin and its inhibitory effects on 2-deoxyglucose (2DG) uptake showed nearly identical dose dependent effects, with both having maximum effects between 50 and 100 μM and similar …

Natural product total synthesis provides an alternative method for obtaining medicinally relevant compounds in a more efficient process with higher yields than what nature can provide. Natural products pose significant synthetic challenges due to the unique heterocyclic skeletons with fused and spirocyclic ring systems. While early efforts have been made to explore the chemistry of neutral multitopic nonchelating N-heterocyclic carbenes (NHCs), limited progress has been made in the development of their anionic counterparts. This thesis describes the synthesis and characterization of a novel trianionic NHC. The corresponding thione was generated through an NMR experiment of the free carbenes with an excess of sulfur. The utility of this novel triscarbene as a ligand in organometallic chemistry has been proven by the construction of homotrimetallic systems. Gene therapy has emerged as a promising precision nano-medicine strategy in the treatment of numerous diseases including cancer. At the forefront of its utility are the applications of short-interfering RNA (siRNA), that silence oncogenic mRNA expression leading to cancer cell death through the RNA interference (RNAi) pathway. Despite the therapeutic potential, siRNAs are limited by poor pharmacological properties, which has hindered their translation into the clinic. The electrogenerated chemiluminescence (ECL) of octaethylporphyrin (OEP), tetraphenylporphyrin (TPP) and a series of metal-ligand porphyrin complexes (Zinc, Copper, Palladium, Platinum, Ruthenium, Vanadium, Cobalt and Nickel) in CH2Cl2 is reported. ECL was generated upon sweep to positive potentials using tri-n-propylamine (TPrA) as an oxidative-reductive coreactant. ECL efficiencies were between 0.010 and 0.770 using Ru(bpy)3(PF6)2 (bpy = 2,2’-bpyridine) as a relative standard (ecl = 1). The ECL intensity peaks at a potential corresponding to oxidation of the complexes and TPrA, suggesting that the same excited states are formed in both photoluminescence and ECL. General and accurate computational methodologies are currently lacking for large chemical systems. This is primarily due to the computational expense required to perform calculations on systems with one hundred or more atoms. Calculated spectroscopic properties could aid in the process of elucidating structural features of large biologically relevant molecules if accurate and inexpensive methods are developed. Towards this end the first steps were taken to design a general methodology for predicting NMR chemical shifts of large nucleic acid systems. The Pacific Northwest National Laboratory (PNNL) prototype bubble chamber is intended to address issues encountered with the current PICO dark matter search detectors and improve the functionality of future experimental designs. The PNNL bubble chamber accomplishes this with a simplified interface between the hydraulic pressure controls and the target vessel and altering the standard chamber design such that it can be easily exchanged and replaced with vessels of various sizes and materials for testing purposes. Due to enhanced properties such as large surface area-to-volume ratio, metal nanoparticles are often employed as catalysts for various applications. However, most studies involving nanoparticle catalysts have been conducted on collections of particles rather than single nanoparticles. Ultramicroelectrodes (UMEs) (limiting dimensions <~25 μm) and nanoelectrodes (<~100 nm) exhibit enhanced electrochemical properties compared to macroscopic electrodes. Their small sizes and enhanced properties make them well-suited for various interesting and important applications such as measuring redox-active species in nonaqueous solvents, studying intermediates of fast electrochemical reactions, and investigating electrochemical and electrocatalytic properties of single nanoparticles. While UMEs are commercially available, nanoelectrode fabrication is still largely confined to research labs. Various methods for constructing nanoelectrodes have been reported and continue to be developed, but most require considerable expertise, and comparisons between different fabrication processes are lacking. In this work, a comparison of laser-assisted pulling and chemical vapor deposition (CVD) methods of electrode fabrication is made with the aim of optimizing production of carbon nanoelectrodes for single nanoparticle electrochemical measurements. By examining effects of pulling parameters, post-pulling treatments, and CVD processing, electrodes as small as ~50 nm were successfully produced.

Hydrolysis of cellulosic biomass is an important and ongoing subject of research due to generating precursors for biofuel synthesis. This work involves hydrolysis of microcrystalline cellulose and Arundo donax to glucose. Methods for acid hydrolysis include the use of sulfuric acid, phosphomolybdic acid, blue molybdenum using low concentrations at 40-100 °C. For comparison purposes, enzymatic hydrolysis was also carried out using cellulase. Products were quantified by measuring total organic carbon and reducible sugars using dinitrosalicylic acid. Use of phosphomolybdic acid, blue molybdenum and sulfuric acid even at 6.0 M did not increase the amount of glucose compared to using water …

Biomarkers are measurable indicators of health status or disease state that can be used for diagnosis and may help guide patient treatment strategies. Enzyme-linked immunosorbent assays (ELISA) and other many clinical techniques currently used for measuring biomarker proteins lack sensitivity, demand high analysis cost, are often not well-suited for measuring multiple biomarkers in a single sample, and require long analysis times. Photocatalytic carbon dioxide reduction transforms CO2 to useful chemicals and fuels, reducing CO2 emissions and making fossil fuels more renewable. Due to a lack of earthabundant sensitizers, we want to design new earth-abundant sensitizers to go with the many known carbon dioxide reduction catalysts. Zn(II) dipyrrin complexes strongly absorb visible light, but their excited state properties have not been widely studied. To investigate their photophysical properties, two Zn dipyrrin complexes, with and without heavy atoms, were synthesized and characterized by NMR and mass spectrometry. Natural product synthesis has many applications in the field of chemical biology, due to the protein binding affinity to the complex three-dimensional structure. However, synthetic and structure-activity relationship studies concerning certain drug targets are scarce. For example, members of the ipomoeassin family have exhibited anti-cancer properties, but cost-effective optimization of these compounds has not yet been extensively studied. Ipomoeassin F, a natural glycoresin isolated from the leaves of the Ipomoea squamosa plant in the Suriname rainforest, has shown high cytotoxicity, with IC50 values measured at the low nanomolar range. Glutathione, a peptide found in microbes, plants and animals including human plays a key role in maintaining healthy cells. The peptide exists in both reduced and oxidized forms. Synthesis of GSH occurs in the cytosol of cells, and the extent of glutathione synthesis relies on various factors, such as amino acid availability, protein activity etc. Once synthesized, glutathione exists in both forms: oxidized (GSSG) and reduced (GSH). This work presents electrospun nanofibers from synthetic spider silk protein, and their application as both a mechanical vibration and humidity sensor. Spider silk solution was synthesized from minor ampullate silk protein (MaSp) and then electrospun into nanofibers with a mean diameter of less than 100 nm. Then, mechanical vibrations were detected through piezoelectric characteristics analysis using a piezo force microscope and a dynamic mechanical analyzer with a voltage probe. The piezoelectric coefficient (d33) was determined to be 3.62 pC/N. The resin glycoside, ipomoeassin F has been shown to be extremely potent against multiple cancer lines (IC50 = 4.2-36 nM). However, the mechanism of action of this potent and complex natural product is still not fully understood. The α,β-unsaturated esters of the glucosyl moiety have been shown to be vital for the overall cytotoxicity of ipomoeassin F. Nevertheless, the importance of the tigloyl ester of the glucosyl moiety is still largely unknown. The research in this dissertation documents connections between the primary amino acid sequence of proteins, the dynamics of proteins, and their catalytic function. This research project studied two proteins called protein-tyrosine phosphatases (PTPs): the human enzyme PTP1B, and the bacterial enzyme YopH. PTP1B is a human enzyme that down regulates the insulin receptor on the outer cellular membrane, and causes the insulin receptor to be less responsive to insulin. A deeper knowledge of how PTP1B is different from other human PTPs might be useful in designing drugs to increase insulin sensitivity in diabetics. Yersinia Pestis is the bacteria that caused …

Rate coefficients for pure rotational quenching in H2(ν 1 = 0, j 1) + H2(ν 2 = 0, j 2) collisions from initial levels of j 1 = 2–31 (j 2 = 0 or 1) to all lower rotational levels are presented. We carried out extensive quantum mechanical close-coupling calculations based on a recently published H2–H2 potential energy surface (PES) developed by Patkowski et al. that has been demonstrated to be more reliable than previous work. Singlet fission (SF), a multiple exciton generation process that generates two triplet excitons after the absorption of one photon, can potentially enable more efficient solar cell designs by harvesting energy normally lost as heat. While low-bandgap conjugated polymers are highly promising candidates for efficient SF-based solar cells, few polymer materials capable of SF have been reported because the SF process in polymer chains is poorly understood. Using transient spectroscopy, we demonstrate a new, highly efficient (triplet yield of 160-200%) isoindigo-based donor–acceptor polymer and show that the triplet pairs are directly emissive and exhibit a time-dependent energy evolution.
