The photochemically-induced destruction of ground-level Arctic ozone in the Arctic occurs at the onset of spring, in concert with polar sunrise. Solar radiation is believed to stimulate a series of reactions that cause the production and release of molecular halogens from frozen, salty surfaces, though this mechanism is not yet well understood. The subsequent photolysis of molecular halogens produces reactive halogen atoms that remove ozone from the atmosphere in these so-called “Ozone Depletion Events” (ODEs). The chemical composition of particulate matter (PM), a known contributor to air pollution, is highly variable, and elemental analysis reveals information about local and regional sources, as well as how air masses and climate influence PM compositions. Seasonal changes in climate, such as temperature, amount of daylight, or meteorological patterns influence source emissions (increased residential heating activities, decreased natural soil emissions) and the relative importance of certain chemical pathways in the atmosphere. Histones are essential chromosomal proteins with large numbers of variants and post-translational modifications (PTMs). PTM levels of histones are known to correlate with different stages of cancer. Due to the lack of resolution on intact histone separations, conventional methods for histone analysis require time-consuming digestion, which often leads to the loss of PTM information. Slip flow chromatography with orderly packed nonporous silica particles has been shown to greatly increase the efficiency of protein separation by reducing eddy diffusion and resistance to mass transfer. Fibronectin is an important biomolecule due to its role in cell differentiation, growth, kinesis, and adhesion. Such biological responses are mediated through membrane recognition and signaling; where fibronectin is found. Studying the outer molecular surface of fibronectin allows deeper insight into the microbiological reactions that occur during these processes. In situ mass spectrometry analysis in aqueous solution accurately represents fibronectin’s chemical components, made possible by a vacuum compatible microfluidic reactor, SALVI (System for Analysis at the Liquid Vacuum Interface). The replacement of traditional bead-based chromatography by membranes has gained recent interest in the bioseparations industry. However, membranes have drawbacks such as lower binding capacity that make them unrealistic for some applications. The goal of this work is to compare the adsorption kinetics of anion exchange bead-based resin and membranes both on a theoretical modeling level and experimental level. Bovine serum albumin (BSA) was adsorbed to Q Sepharose FastFlow resin and Sartobind Q membrane to obtain adsorption equilibrium characteristics and kinetic data. Single-scattering albedo (SSA) and absorption Ångström exponent (AAE) are two critical parameters in determining the impact of absorbing aerosol on the Earth's radiative balance. Aerosol emitted by biomass burning represent a significant fraction of absorbing aerosol globally, but it remains difficult to accurately predict SSA and AAE for biomass burning aerosol. Black carbon (BC), brown carbon (BrC), and non-absorbing coatings all make substantial contributions to the absorption coefficient of biomass burning aerosol. SSA and AAE cannot be directly predicted based on fuel type because they depend strongly on burn conditions. It has been suggested that SSA can be effectively parameterized …

Organic matter (OM) in the environment acts as a nutrient, but may also act as a transport vector for harmful chemical compounds and bacteria. Acetate is a labile form of OM produced during fermentation in anaerobic lagoons used to store animal fecal-waste from concentrated animal feeding operations (CAFOs). Dry and liquid fertilizers from CAFOs pose a threat to groundwater by introducing excessive amounts of nutrients (e.g. OM, nitrate and ammonia), metals, and antibiotic compounds. Vitamin pathways exogenous to the human body represent an attractive target for drug development since the enzymatic machinery is solely and ubiquitously to many infectious agents. Haloacetic acids, which are generated from disinfection processes, are one of the major groups in disinfection by-products. Because they induce potential harmful effects for human health, the US government has already established regulations to control the levels of haloacetic acids in drinking water. Cannabinoids are a class of chemical compounds that act on cannabinoid receptors in cells. These receptors have different kinds of ligands, including endocannabinoids (produced in the body by humans and animals), phytocannabinoids (found mainly in Cannabis sativa), and synthetic cannabinoids. At least 85 different phytocannabinoids have been isolated; different cannabinoids are believed to have different effects and possible benefits in medical treatments, and it is therefore fundamental to classify cultivars prior to their medical use. Antibiotics play a vital role in fighting infectious disease caused by bacterial species. Antimicrobial resistance has been shown for every current antibiotic and poses a major health risk. To combat these growing threats to human health; new molecules must be investigated to treat bacterial infection. Chemical inhibition of the vitamin B1 biosynthetic pathway in the bacterial cell was hypothesized to inhibit the survival of bacteria. Au–Fe3O4 nanocomposites are of great interest for technological reasons due to their combined optical and magnetic properties. Nanosized Fe3O4 particles are superparamagnetic and can also be coated with a layer of Au, which supports localized plasmon resonances for useful optical properties. Ion–molecule reactions in a flowing afterglow are used to examine the electronic structure of 3- and 4-pyridinylnitrene-n-oxide radical anions. Reactions with nitric oxide are generally similar to those reported previously for other aromatic nitrene radical anions. In particular, phenoxide formation by nitrogen–oxygen exchange is observed with both isomers. Oxygen atom abstraction by NO is also observed with both isomers. Very significant differences in the reactivity are observed in the reactions of the two isomers with carbon disulfide. The reactivity of the 3-noxide isomer with CS2 is similar to that observed previously for nitrene radical anions, and reactions of the n-oxide …

There is an increasing demand for protein detection and quantitation in biological fluids for disease detection, protein therapeutics monitoring, and drug development response control. Current methods use highly sensitive and specific triple quadrupole mass spectrometry (QqQ-MS) to quantify protein digest peptides to predict original intact protein concentrations from the sample. The bottom-up protein quantitation format requires protein digestion which is often incomplete and can introduce errors into protein quantitation methods. Therefore, absolute protein quantitation is impossible without including expensive isotopically labeled protein standards. This dissertation focuses on two chromatographic techniques, gas chromatography (GC) and high performance liquid chromatography (HPLC). The goal of the GC work is to describe advances in separation methodologies focusing on the separation and quantitation of commercially related compounds (i.e. fatty acids, fatty amines, water, and ethanol). Four ionic liquid (IL) columns were evaluated for rapid analysis and improved resolution of long-chain methyl and ethyl esters of omega-3, omega-6, and additional positional isomeric and stereoisomeric blends of fatty acids found in fish oil, flaxseed oil, and potentially more complicated compositions. Peracetic acid is a simple molecule that works as a powerful oxidant in many applications including food safety and water disinfection. The synthesis of the compound is fairly well understood due to its straightforward synthesis by mixing acetic acid and hydrogen peroxide. However, the decomposition of the compound has not been well studied. Peptoids are isomers of peptides where the side chains are on the nitrogens instead of the carbons, which have been developed for a variety of biomedical, nano, and materials applications. Conventional peptoid synthesis uses molar concentrations of reagents and hours of reaction time per residue, thus practically limiting the lengths of peptoids. Recently, we have managed to improve yields and make peptoids of unprecedented lengths while reducing the reaction times and concentrations of reagents. Ready-to-use (RTU) grout is becoming more important to the finish and remodeling construction industry. Market research shows it is a fast-growing product that not only is creating its own space, but is beginning to supplant existing technology.
The original intent of this research was to investigate formulation parameters and how they affect grout performance. It was learned that particle size and oil absorption (OA) value are important filler properties that affect performance as much as adequate packing density and optimal pigment volume concentration (PVC) without going beyond critical PVC (CPVC).

A number of chemosensors have been designed and synthesized to target cations (Zn2+ions), neutral molecules (cathinones), charged molecules (aminoindanes), and anions. The Zn2+ ion sensor featured bistriazole designed binding unit and ferrocene signaling units. Characterization of Zn2+ ion binding was carried out with electrochemical techniques (CV and DPV), 1H-NMR, mass spectrometry, and molecular modelling. It exhibited a 1:1 binding stoichiometry with Zn2+ and had an affinity for ZnCl2 (Log K1:1 = 4.1 ± 0.02) over other Zn2+ salts.

The synthesis of substituted phenylpropene dimers using a one-pot, tandem olefin metathesis and isomerization sequence has been studied. This sequence relies on the facilitated, in-situ conversion of a ruthenium carbene species (Ru=C) to a ruthenium hydride species (Ru-H) upon addition of an inorganic hydride source. Three separate reactions occur within one reaction flask: 1) olefin metathesis of the starting phenylpropene to yield phenylpropene dimer via Ru=C catalyst, 2) conversion of Ru=C to Ru-H via addition of an inorganic hydride source, 3) isomerization of phenylpropene dimer via insertion and β-hydride elimination to yield conjugated product. Heparin is a polydispersed sulfated molecule that is part of the family called glycosaminoglycans found in the extracellular matrix and cell surfaces. This molecule is extremely important for the activation of proteins and protein-receptor interactions that are responsible for downstream cell signaling pathways. Heparin has been isolated from porcine intestine and used as an anticoagulant for the prevention of embolisms, heart thrombosis, and clotting during heart surgeries. This so-called miracle drug was in use until 2008, when isolated batches were found to be contaminated with other glycosaminoglycans similar to heparin. From 2008, there has been a dire need for a …

Aryl iodides have become widely recognized as versatile synthetic intermediates, owing to aromatic iodine’s excellent ability to participate in oxidative addition reactions. Iodoarenes readily undergo a variety of synthetic transformations including metal catalyzed cross-coupling reactions, diaryliodonium chemistry, formation of organometallics through reduction or metal-halogen exchange, as well as classical SN2 type chemistry. Because a wide array of transformations are available for aryl iodides, organic molecules containing this moiety often serve as vital precursors to highly desirable pharmaceuticals.

This dissertation describes studies in the asymmetric synthesis of the C21 – C34 fragment of the natural product, antascomicin B. Antascomicin B is structurally related to FK506, binds strongly to FKBP12, yet does not shown immunosuppressive activity. Small ligand FKBP12 binding complexes were shown to have potent neuroprotective and neuroregenerative properties in mouse models of Parkinson’s disease. The highlighted chemical reactions include an asymmetric transfer hydrogenation (ATH), Ireland-Claisen rearrangement (ICR), directed hydrogenation and allylic diazene rearrangement.

This work explores the derivatization of C60 and Sc3N@C80 with S-, P-, and C- centered radicals, and is the first exploration into the functionalization of Sc3N@C80 with sulfur and phosphorous. The number of addends bound to C60, as a function of reaction time and molar ratios, as well as, structure-property relationships including thermal stability, solubility, and singlet oxygen generation were explored. The low temperature (298 – 700 K) oxidation characterizations of a series of fuel additives (ETBE and isooctane) were studied at advanced light source in Lawrence Berkley National lab, using a multiplexed chemical kinetics photoionization mass spectrometer with tunable synchrotron radiation. Using Cl atoms as initiators in presence of oxygen, photoionization data were collected. Data analysis was performed via characterization of the reaction species photoionization spectra and kinetic traces, from which the products and also the intermediates were identified. Current lithium ion batteries are comprised of organic liquid electrolytes - a mixture of lithium salts and binary solvents such as ethylene carbonate (EC) and dimethyl carbonate (DMC). The main drawbacks of this liquid mixture related to safety are flammability of the organic solvents and chemical instability with the electrode materials.
