Compounds containing the pyrrole ring as a core structural motif continue to show significant biological activity, and both natural product derivatives and novel chemical scaffolds show potential for use as pharmaceuticals in treating a variety of cancers, infections, and inflammation. Given the widespread and important applications of compounds containing this motif, novel, rapid, and selective methods for the synthesis of multifunctional pyrroles is of some importance. The research conducted in the Leopold Bioanalytical and Nanomaterials Analytical Chemistry Lab prior to the summer of 2013 focused on the synthesis of a functional biosensor equipped with a nanoparticle network embedded in a xerogel film for the detection of glucose. Xerogel-based films featuring embedded glucose oxidase enzyme and doped with alkanethiolate-protected gold NPs, known as monolayer protected clusters (MPCs), exhibit significantly enhanced performance compared to analogous systems without NPs including higher sensitivity, faster response time, and extended linear/dynamic ranges.

"Single particle (SP)-ICP-MS methods were developed to characterize and quantify Ti-containing, titanium dioxide, silver, and gold NP concentration, size, size distribution and dissolved metal element concentration in surface water and treated drinking water. The effectiveness of conventional drinking water treatments (including lime softening, alum coagulation, filtration, and disinfection) to remove NPs from surface water was evaluated using six-gang stirrer jar test simulations. Six-gang stirrers were used to simulate drinking water treatments including lime softening, alum coagulation, powdered activated carbon sorption, filtration, and disinfection by free chlorine. Nanocomposites of TiO$_{2}$/chitosan (nTC) were synthesized by hydrothermal process at 200 $^{\circ}$C. Photocatalytic activity of the nanocomposites was studied with degradation of malachite green (MG) in aqueous medium. Structural and physico-chemical properties of the obtained nanocomposites were characterized by XRD, SEM, BET, FTIR, and particle size analyzer. Complete photodegradation of MG was successfully achieved with the aid of the nanocomposites. The optimum photocatalytic degradation conditions, irradiation time, irradiation power, and amount of catalyst were studied. We performed the optimization of conditions in chemical bath deposition using the design of experimental methodology. Factorial design (2$^{3})$ was used for finding the optimal conditions of preparing vanadium(V) oxide xerogel thin films with better electrochromic properties suitable for electrochemical applications. The thin films were prepared by acidification of NH$_{4}$VO$_{3}$ aqueous solution with chemical bath deposition. Numerous efficient synthetic methodologies have been elaborated for the synthesis of $\beta $-blockers since the introduction of propranolol (a beta-blocker) in 1968. In this review, focus is placed on the more concise asymmetric and bioenzymatic synthetic approaches attempted towards the synthesis of beta-blockers (betaxolol, metoprolol, sotalol, and timolol).

Synthetic Access To Some New Benzothiazole-Based 1,3,4-Thiadiazole And 1,3-Thiazole Derivatives, Kamal M. Dawood, Eman A. Ragab, Korany A. Ali

1-(Benzothiazol-2-yl)-3-phenylthiourea\textbf{ 2} was prepared and treated with hydrazonoyl chlorides \textbf{3a}--\textbf{e }to yield the corresponding 5-(benzothiazol-2-ylimino)-1,3,4-thiadiazole derivatives \textbf{6a}--\textbf{e}, respectively. Reaction of the thiourea derivative \textbf{2} with ethyl 2-chloro-3-oxobutanoate \textbf{9} afforded the corresponding 2-(benzothiazol-2-ylimino)thiazole-5-carboxylate derivative \textbf{11}. The newly synthesized heterocyclic derivatives were confirmed from their elemental and spectral analyses.

Synthesis And Evaluation Of Some Novel Thiazoles And 1,3-Thiazines As Potent Agents Against The Rabies Virus, Magda Abdalla, Sobhi Gomha, Mohamad Abd Elaziz, Nany Serag

A series of novel thiazoles and 1,3-thiazine derivatives were synthesized in good yield via reaction of ethyl 3-(1-(2-thiocarbamoylhydrazono)ethyl)-1,5-diphenyl-1$H$-pyrazole-4-carboxylate with hydrazonoyl halides and arylidenemalononitriles, respectively. The structure of the newly synthesized products was elucidated via elemental analysis, spectral data, and alternative routes whenever possible. Moreover, the antiviral screening of the products was evaluated and the results revealed that some of them have strong to moderate potency against the rabies virus compared with the reference drug.

Prostereoisomerism (PIM), representing a subgroup within the achiral class of molecules, is manifested in a chiral environment. Although this is regardless of the orientation of the prostereoisomeric (PIC) molecule, PIM is manifested most emphatically in relation to a chiral surface (`two-dimensional chirality'). Then PIM is tantamount to `de facto chirality' as attachment of the PIC molecule to the surface leads to diastereomeric possibilities (cf. Ogston's hypothesis). Furthermore, the formation of host--guest complexes requires a steric complementarity between the component molecules. The fact that the weak dispersive forces involved therein require an optimum distance implies a `snugness of fit' criterion. This …

SiO$_{2}$ supported core@shell nanoparticles (CSNs) have recently attracted great attention due to their unique, tunable, optical, photocatalytic, and higher adsorption properties. In this study, SiO$_{2}$@CeO$_{2}$~CSNs were synthesized using a chemical precipitation technique and characterized by Fourier transform infrared (FT-IR), X-ray diffraction (XRD), scanning electron microscope (SEM), and transmission electron microscope (TEM) analysis. XRD analysis showed that SiO$_{2\, }$particles were the core while CeO$_{2}$ particles were the shell. The reaction kinetics of CO$_{2}$ absorption into new carbon dioxide binding organic liquids (CO$_{2}$BOLs) was comprehensively studied to evaluate their potential for CO$_{2}$ removal. A stopped-flow apparatus with conductivity detection was used to determine the CO$_{2}$ absorption kinetics of novel CO$_{2}$BOLs composed of DBN (1,5-diazabicyclo[4.3.0]non-5-ene)/1-propanol and TBD (1,5,7-triazabicyclo[4.4.0]dec-5-ene)/1-butanol. A modified termolecular reaction mechanism for the reaction of CO$_{2}$ with CO$_{2}$BOLs was used to calculate the observed pseudo-first-order rate constant k$_{0}$ (s$^{-1})$ and second-order reaction rate constant k$_{2}$ (m$^{3}$/kmol.s). Thirty-one complexes of bis-(benzimidazole, benzothiazole, and benzoxazole) compounds with Zr(IV) metal centers were synthesized, characterized, activated with methylaluminoxane (MAO), and then tested for catalytic ethylene polymerization. The activities of the various catalysts were found to be functions of the heteroatoms in the ligand frameworks and the structure around the active metal center. The highest activity was obtained with 38/MAO (424 kg E/mol cat. h). The produced polyethylenes showed high molecular weights (41/MAO, 1.9 $\times$ 10$^{6}$ g/mol) and broad molecular weight distributions (38/MAO, M$_{w}$ = 9.64 $\times $ 10$^{5}$ g/mol, PD = 23). The speciation and preconcentration of selenium were carried out by solid phase extraction using nanozirconium oxide/boron oxide (ZrO$_{2}$/B$_{2}$O$_{3})$. Electrothermal atomic absorption spectrometry (ETAAS) was used for detection. This preconcentration and/or speciation method depends on the retention of Se(IV) ions selectively as its ammonium pyrrolidine dithiocarbamate (APDC) complex on the sorbent. The effects of pH, flow rate, and volume of sample solution and eluent type were investigated for determining the optimum preconcentration and speciation conditions. Oxidation of bisphenol-A (BPA) was investigated using a sonophoto Fenton-like hybrid process under visible light irradiation in the presence of iron-containing perovskite LaFeO$_{3}$ catalysts. For this purpose, firstly the perovskite catalyst (LaFeO$_{3})$ was prepared by the sol-gel method and calcined at different temperatures (500, 700, and 800 $^{\circ}$C). The prepared catalysts were characterized using XRD, SEM, FTIR, nitrogen adsorption, UV-vis DRS, and ICP/OES measurements. A series of novel positional oximes (2a-2d), $O$-methyl oxime ethers (3a-3d), and $O$-benzyl oxime ethers (4a-4d) were synthesized in high yields starting from their corresponding methyl 3-, 4-, 6-, and 13-keto tetradecanoates. The synthesized compounds were characterized by $^{1}$H NMR, $^{13}$C NMR, FT-IR, mass, and elemental analyses for their structures and $(E)$/$(Z)$ isomerizations. Their DNA binding abilities were investigated in vitro by agarose gel electrophoresis. The antibacterial and antifungal activities were tested also in vitro against eleven bacterial strains and three fungal strains. A novel method for the determination of mercury(II) is suggested. The procedure is based on the formation of an ion associate between the bromide complex of Hg(II) and Astrazon Red 6B dye and vortex-assisted liquid-liquid microextraction of the ion associate formed, with subsequent spectrophotometric detection. Cloud point extraction (CPE), which is the most popular coacervate-based extraction method, was utilized for the separation and preconcentration of trace amounts of cadmium, lead, and nickel. Quinalizarin was used as complexing agent and Triton X-114 was used as surfactant. After extraction, analyte ions were determined by flame atomic absorption spectrometry. The detection limits (3$s)$ of 1.8 $\mu $g L$^{-1}$ for Cd(II), 3.2 $\mu $g L$^{-1}$ for Pb(II), and 2.8 $\mu $g L$^{-1}$ for Ni(II) were obtained using 50 mL of solutions. A new method based on the combination of carrier element-free coprecipitation and flame atomic absorption spectrometric determination has been investigated for simultaneous separation and preconcentration of Pb(II) and Cu(II) ions in some environmental solid and liquid samples. The 2-{3-(4-methylbenzyl)-4-[2-(1H-indol-3-yl)ethyl]-5-oxo-4,5-dihydro-1H-1,2,4-triazol-1-yl} -N'-(phenylmethylene) acetohydrazide (MITA) was utilized as an organic coprecipitating agent without using a carrier element for coprecipitation of the analyte ions. The effects of experimental conditions including aqueous solution pH, MITA quantity, sample volume, standing time, and centrifugation rate and time were investigated in detail and optimized. The anodization of Aluminium was investigated in the low-melting, mixed halide molten salt system, lialbr4-naalcl4-kalcl4 (30-50-20 m/o), and in the Lewis acidic Chloroaluminate ionic liquid, aluminum chloride-1-ethyl-3-methylimidazolium chloride (alcl3-etmeimcl) using rotating disk electrode voltammetry. In both cases, at modest overpotentials, the anodization reaction proceeds under mixed kinetic/mass-transport control. However, at larger positive overpotentials and correspondingly higher anodization rates, the reaction transitions to a mass transport-limited process governed by the dissolution of a passive layer of alcl3(s) and/or albr3(s) on the electrode surface. In alcl3-etmeimcl, the passive current density scales linearly with the concentration of alcl4- in the ionic liquid. The …

The protozoan parasite, Trypanosoma cruzi, the etiologic agent of Chagas disease (ChD), has a cell surface covered by immunogenic glycoconjugates. A synthetic glycoarray containing non-reducing α-galactopyranosyl moieties related to mucin O-glycans of the parasite T. cruzi was evaluated by a chemiluminescent enzyme-linked immunosorbent assay (CL-ELISA) with sera from patients with chronic ChD, revealing the disaccharide Galα(1,3)Galβ as the immunodominant glycotope (Chapter 2). Organic materials with conjugated Ï€-electron networks are increasingly being studied as an alternative to inorganic semiconductors. In order to effectively transport charge as a semiconductor, the organic materials should maintain a planar conformation, exhibit Ï€ conjugation, and be rigid to rotational interconversion. Both furan and thiophene systems have been investigated as potential organic semiconductors but difficulties with solubility, synthesis, and effectiveness hampered these efforts. Dimethylaminopyridine (DMAP) can act to catalyze numerous organic reactions. To increase its recovery and reuse, it has been tethered to dendrimers and to micellar systems, creating nano-scale catalytic pockets for reactions to proceed. Previously developed systems show diminished reactivity over time as the products of the reaction fill up the nano-scale reactor. Thus, the goal of my research is to use RAFT polymerization methods to synthesize a hydrophobic polymer chain bearing 4-methylaminopyridine units and to attach it to a hydrophilic polymer chain to create a polymer system capable of micelle formation in solution. Tomas Lindahl was recognized for studies that uncovered the inherent instability of DNA as well as the mechanism of the base excision repair pathway, Paul Modrich for characterization of the mismatch repair pathway, and Aziz Sancar for mechanistic elucidation of the nucleotide excision repair (NER) pathway. Poly(arylene ether sulfone)s with pendent aryl iodide group was successfully synthesized via Nucleophilic aromatic polycondensation of 1-(3,5-difluorophenyl sulfonyl)-4-iodobenzene using 4,4’-difluorodiphenylsulfone (DFDPS) and Bisphenol A in the ratio 25:75:100. Iodo monomer was prepared by reacting 3,5-difluorodiphenyl sulfone with N-iodo succinimide (NIS). Using GC/MS and NMR spectroscopy, the quantitative analysis was done to this monomer. Modification was done to iodo copolymer using sodium azide and copper sulfate as catalyst to form azide copolymer. Cadherins are calcium dependent glycoproteins whose homophilic interactions mediate cell-cell adhesion in solid tissues. They are comprised of an extracellular region, a transmembrane region and a cytoplasmic region. The extracellular region plays a critical role in cadherin-mediated cell adhesion, and has five tandemly repeated ectodomains (ec1-ec5), with three calcium binding sites situated in each interface between the domains. Dimerization of cadherin occurs through formation of adhesive interactions between extracellular domains of cadherins from neighboring cells. Adhesive interaction occurs at the interfaces of ec1 domains of two molecules originating from different cell surfaces. Dimerization is critically dependent on the binding of …

The use of cell penetrating peptides (CPPs) as biomolecular delivery vehicles holds great promise for therapeutic and other applications, but development has been stymied by poor delivery and lack of endosomal escape. We have developed a CPP-adaptor system capable of efficient intracellular delivery and endosomal escape of user-defined protein cargos. The cell penetrating sequence of HIV transactivator of transcription was fused to calmodulin, which binds with subnanomolar affinity to proteins containing a calmodulin binding site. Our strategy has tremendous advantage over prior CPP technologies because it utilizes high affinity noncovalent, but reversible coupling between CPP and cargo. Three different cargo …
