Tissue engineering is a scientific methodology that provides the means for fabrication of vital tissues in a laboratory with the ultimate goal of translating the tissue to a patient in a clinic. It is a desirable technique in clinical medicine because it eliminates the need for patient tissue grafts and transplants, which have the potential for donor site morbidity and rejection. We seek to discover new drug candidates for the excitatory amino acid transporter 3 (EAAT3) for the treatment of Obsessive Compulsive Disorder (OCD). We hypothesize that decreasing the glutamatergic transmission via EAAT3 intervention will create an amelioration of the symptoms of OCD. For this we have prepared in-silico binding calculations to identify a better compound that can stimulate EAAT3 and decrease glutamatergic transmission. This research will help us further understand and elucidate the role of glutamate in the neural mechanisms of this illness, which to date remain inconclusive. N-acyl-7-nitroindolines are photoreactive compounds that are known to acylate a number of nucleophiles in inert aprotic solvents under neutral conditions when illuminated at 350 nm. We have assembled a MUC1-derived peptide and Tn or STn-containing MUC1 glycopeptides with a C-terminal photoreactive 7-nitroindoline linker attached to Sieber amide resin by solid phase peptide synThesis using the Fmoc strategy. This type of construction allows for the cleavage of fully protected photoreactive peptides/glycopeptides from the resin with dilute trifluoroacetic acid, or for the direct photorelease of protected peptide/glycopeptide derivatives. p-Indolequinones are important heterocycles since they display interesting bioactivities such as anticancer activities as well as the ability to trigger drug release in drug targeting research. Additionally, they are also useful in precursors for the synThesis of bioactive compounds. Over decades, many methodologies have been investigated for the synThesis of p-indolequinones. Among those, cyclization of quinones plays an important role in convenient synThesis of these compounds. Endocrine disrupting compounds (EDCs) are a class of compounds that are of concern for their potentially adverse effects on fish, wildlife, and humans. They can cause developmental and reproductive anomalies at concentrations in the low ppb to ppt levels. Studies have shown the primary route of exposure of EDCs within the environment is through the discharge of effluent wastewater. Soil contamination that results from elemental sulfur (S0) deposition is a new occurrence. Elemental sulfur is stockpiled as a byproduct from the oil and gas industry and is deposited by biodegradable trap and skeet targets at shooting ranges. Chemolithotrophs and mixotrophs can oxidize S0 to H2SO4. Consequently, in areas where excess S0 is deposited or stockpiled, soils can acidify to pH values observed with acid mine drainage.

Characterization Of Pm2.5 From Residential Woodstove Use For Source Apportionment Application, Virginia M. Porden

Results are presented for the comparative analysis of the PM2.5 (Particulate Matter <2.5 µm) emissions from an EPA certified and a Traditional style wood stove using western larch. A total of 92 Quartz QMA 47mm filters were collected using a BGI PM2.5 SSC (Sharp Cut Cyclone) sampler from each stove type and analyzed on a gas chromatograph-mass spectrometer against blank and deuterated internal standards. The results were analyzed using a Welch’s t-test (α > 0.05) to statistically differentiate between stove designs for temperature, mass, levoglucosan, resin acids (abietic and dehydroabietic), and polycyclic aromatic hydrocarbons [PAH] (acenaphthene, anthracene, benz(a)anthracene, pyrene, and retene). There was no statistical difference in levoglucosan PM2.5 mass fraction between the stove types, yielding a mean levoglucosan fraction of 9.25% and 95% confidence interval of 8.43% …2.5>

The lack of a rapid and quantitative autophagy assay has substantially hindered the development and implementation of autophagy-targeting therapies for a variety of human diseases. To address this critical issue, we developed a novel autophagy assay using the newly developed Cyto-ID fluorescence dye. We first verified that the Cyto-ID dye specifically labels autophagic compartments with minimal staining of lysosomes and endosomes. We then developed a new Cyto-ID fluorescence spectrophotometric assay that makes it possible to estimate autophagy flux based on measurements of the Cyto-ID-stained autophagic compartments. This paper presents wavelength configurable on-chip solid-state ring lasers fabricated by a single-mask standard lithography. The single- and coupled-ring resonator hosts were fabricated on a fused-silica wafer and filled with 3,3′-Diethyloxacarbocyanine iodide (CY3), Rhodamine 6G (R6G) and 3,3′-Diethylthiadicarbocyanine iodide (CY5)-doped polymer as the reconfigurable gain media. The recorded lasing threshold was ~220 nJ/mm2 per pulse for the single-ring resonator laser with R6G, marking the lowest threshold shown by solid-state dye-doped polymer lasers fabricated with a standard lithography process on a chip. The imidazolium salt (IS) and silver carbene complex (SCC) studied in this report are 3-(pyridin-2-ylmethyl)-1-(quinolin-2-ylmethyl)imidazolium chloride (1) and 3-(pyridin-2-ylmethyl)-1-(quinolin-2-ylmethyl)imidazole – silver acetate complex (2). It was anticipated that the anti-proliferative activity of 1 and 2 would be similar to other characterized ISs while having increased solubility. The SCC (2) was synthesized in order to simulate the ligand-metal coordination similar to that of cisplatin.

Numerous studies by our lab and others demonstrate that epidermal growth factor receptor (EGFR) plays critical roles in primary breast cancer (BC) initiation, growth and dissemination. However, clinical trials targeting EGFR function in BC have lead to disappointing results. In the current study we sought to identify the mechanisms responsible for this disparity by investigating the function of EGFR across the continuum of the metastatic cascade. We previously established that overexpression of EGFR is sufficient for formation of in situ primary tumors by otherwise nontransformed murine mammary gland cells. Various ions of poly-L-lysine and polystyrene sulfonic acid were successfully observed first within a matrix-assisted laser desorption/ionization mass spectrometer and subsequently in an electrospray ionization mass spectrometer. The successful ionization of these polymers allowed for studies to be performed on the noncovalent interactions between the two. Noncovalent interactions were effectively studied on both instruments, but further quantified using tandem mass spectrometry found within the MALDI system and ion-mobility mass spectrometry found within the electrospray ionization system. As a result of employing the use of ion-mobility, various different stoichiometries and charges of polyelectrolyte complexes could be distinguished.

Several aldehydes (butanal, pentanal, hexanal, 4-hydroxybenzaldehyde) were reacted with 1,3-bis(naphthalen-2-ylmethyl)-imidazolium bromide (1) to produce novel C2 substituted imidazolium salts for the potential use against non-small cell lung cancer in humans. Compounds 2-(1-hydroxypentyl)-1,3-bis(naphthalen-2-ylmethyl)-imidazolium bromide (3) and 2-(1-hydroxyhexyl)-1,3-bis(naphthalen-2-ylmethyl)-imidazolium bromide (5) were successfully synthesized with structures supported by NMR and mass spectrometry. Characterization by 1H NMR showed evidence of 1 in both compounds. The tumor cell growth inhibition of 3 against non-small cell lung cancer lines NCI-A549, NCI-H460, HCC827, and NCI-H1975 was tested and found to be comparable to cisplatin as measured by MTT assay. …

Several chlorine/oxygen exchange reactions of [PCl2N]3 were investigated with the goal of forming [PON]3, a precursor to PON materials. The reaction of MeSO3H and Et3N with hexachlorocyclotriphosphazene ([PCl2N]3) has been claimed to produce a [PON]3 single molecule. This reaction could not be reproduced. Only partially oxygenated phosphazenes have been obtained. Similarly, in the reaction of [PCl2N]3 and dimethylformamide (DMF) both partial and full oxygenation occurs. Karlotoxins (KmTxs), produced by a toxic marine/estuarine phytoplankton, the dinoflagellate Karlodinium veneficum, are known to be ichthyotoxic, thus being associated with numerous fish deaths events worldwide. They have also been reported to show a variety of biological activities such as hemolytic, cytotoxic, and anti-fungal activities. Recently, the Hamann group successfully assigned the absolute configurations of KmTx2, the first complete structure elucidation among the congeners. In a structural sense, karlotoxins are a family of linear polyketides with three distinctive regions; a bis-tetrahydropyran core fragment, a long, highly oxygenated carbon chain, and a lipophilic chlorodiene unit. A series of N,N’-bis(arylmethyl)benzimidazolium salts with hydrophilic and lipophilic substituents have been synthesized, characterized, and tested against select non-small cell cancer cell lines. Substituent variations on the imidazole ring have shown that lipophilicity and hydrophilicity can influence the imidazolium salts’ anti-proliferative activity and aqueous solubility.

While spectrophotometers are commonly found in many universities and laboratories, they are not available to all high schools or laboratories in developing countries. The objective of this project was to design a high-quality, 3D-printable, iPhone-compatible spectrophotometer that uses a cell phone camera and interface to collect data. Following recent work on heterometallic titanocene–gold complexes as potential chemotherapeutics for renal cancer, we report here on the synthesis, characterization and stability studies of new titanocene complexes containing a methyl group and a carboxylate ligand (mba ¼ S–C6H4–COO) bound to gold(I)-phosphane fragments through a thiolate group [(h-C5H5)2TiMe(m-mba)Au(PR3)]. The compounds are more stable in physiological media than those previously reported and are highly cytotoxic against human cancer renal cell lines. A novel method was developed to measure mercury (Hg) directly in environmental solids using inductively coupled plasma mass spectrometry (ICP-MS) with online sample ashing. The method combines a direct mercury analyzer (DMA) that is based on sample combustion, Hg pre-concentration by amalgamation with gold, and atomic absorption spectrometry (AAS), with a high resolution ICP-MS. Unlike AAS, the ICP-MS is capable of individual isotope measurements allowing for accurate quantitation by isotope dilution mass spectrometry (IDMS) and for isotope tracer experiments. Electrospun microfibers (MFs) composed of pH and temperature responsive polymers can be used for controllable and variable delivery of ibuprofen. First, electrospinning technique was employed to prepare poly(ε-caprolactone) (PCL) and poly(N-isopropylacrylamide-co-methacrylic acid) (pNIPAM-co-MAA) MFs containing ibuprofen. It was found that drug release rates from PCL MFs cannot be significantly varied by either temperature (22–40°C) or pH values (1.7–7.4). In contrast, the ibuprofen (IP) diffusion rates from pNIPAM-co-MAA MFs were very sensitive to changes in both temperature and pH. Titanium dioxide (TiO2) nanoparticles are one of the most highly manufactured and employed nanomaterials in the world with applications in copious industrial and consumer products. The liver is a major accumulation site for many nanoparticles, including TiO2, directly through intentional exposure or indirectly through unintentional ingestion via water, food or animals and increased environmental contamination. The aggregation of the 37-amino acid polypeptide islet amyloid polypeptide (IAPP, amylin), as either insoluble amyloid or as small oligomers, appears to play a direct role in the death of pancreatic β-islet cells in type 2 diabetes. It is believed that inhibiting the aggregation of IAPP may slow down, if not prevent entirely, the progression of this disease. Extracts of 13 different common fruits were analyzed for their ability to prevent the aggregation of amyloidogenic IAPP. Thioflavin T binding, immuno-detection and circular dichroism assays were performed to test the in vitro inhibitory potential of each extract. Atomic force microscopy was …
