Aims: To identify isosteres and bioisosteres suitable for substitution on the molecular scaffold of meclofenamic acid and tolfenamic acid. The compounds will be studied to determine drug-likeness and other properties.
Study Design: Isosteres and bioisosteres were selected and emplaced on the scaffold of meclofenamic acid and tolfenamic acid to ascertain drug-likeness outcome. Drug candidates were selected based on favorable drug-likeness.

Aims: To synthesize small molecule alkylating compounds and analyze the kinetics of the alkylation in aqueous solution. Determine molecular properties and the drug likeness of these four compounds as potential antineoplastic agents and apply statistical analysis to identify interrelationships of properties.
Study Design: Four compounds were synthesized, characterized, and studied for alkylation capability. The alkylation kinetics were elucidated, as well as drug likeness properties. The interrelationships of properties were examined by statistical methodology.

Cancer is designated as the leading cause of mortality worldwide and lung cancer is responsible for nearly 30% of all cancer related deaths. Over the last few decades mortality rates have only marginally increased and rates of recurrence remain high. These factors, among others, suggest the need for more innovative treatment modalities in lung cancer therapy. Methamphetamine (METH) abuse is a serious problem in the United States and worldwide. The reward experienced by METH users is due to the increase in extracellular dopamine (DA) concentrations caused by an interaction between METH and the DA transporter (DAT) as well as the Vesicular Monoamine Transporter-2 (VMAT2). The reward felt by users of METH leads to further use of the drug and subsequent abuse. Many pharmaceutical compounds being developed in recent years are poorly soluble in water. This has led to insufficient oral bioavailability of many compounds in vitro. The amorphous formulation is one of the promising techniques to increase the oral bioavailability of these poorly water-soluble compounds. However, an amorphous drug substance is inherently unstable because it is a high energy form. In order to increase the physical stability, the amorphous drug is often formulated with a suitable polymer to form an amorphous solid dispersion. Rationale
Ultra-performance liquid chromatography (UPLC) coupled with coordination ion spray tandem mass spectrometry was used for the analysis of air samples containing triglycidyl isocyanurate. The method is not affected by any chromatographic interference and the filter extract is compatible with the UPLC system, as opposed to approaches using high-performance liquid chromatography coupled with ultraviolet detection and gas chromatography coupled with mass spectrometry.

Two methods of transition metal catalysis were investigated as a means to synthesize biologically interesting heterocyclic molecules. The gold-catalyzed intermolecular cyclization of allenes and 1,3-dipoles was first explored. An allene, ethyl-3-hexyl-3,4-pentadienoate was subjected to a combination of one of five 1,3-dipoles ((Z)-N-benzylidenemethanamine oxide, 1-benzylidene-3-oxopyrazolidin-1-ium-2-ide, (Z)-N-benzylidenebenzenamine oxide, (E)-N-benzylidenebenylamine, and (E)-N-Benzylidene-4-methoxybenzenamine) and one of three gold catalysts (1,3-bis(2,6-di-isopropylphenyl)imidazole-2-ylidenegold(I)chloride, chlorotriphenyl-phosphinegold(I), and gold(III) chloride). No heterocyclic product resulted from these catalytic reactions. The goal of this project was to study novel synthetic procedures for heterocycles. This was achieved through a two-part investigation. In the first part, a variety of conditions for the gold catalyzed cyclization of allenes were studied. This work was focused on the lesser studied intermolecular reactions of allenes. A variety of allenes were synthesized, and then reacted with 1,3-dipoles and a gold catalyst. These reactions were determined to be unsuccessful through the analysis of nuclear magnetic resonance spectroscopy data. Chemically patterned surfaces were fabricated using a combination of molecular self-assembly and particle lithography to generate billions of nanostructures of organosilane self-assembled monolayers (SAMs). Monodisperse mesospheres were used as surface masks to prepare nanostructures on flat surfaces using the simple benchtop chemistry steps of mixing, centrifuging, evaporation, and drying. Periodic arrays of well-defined organosilane nanostructures serve as discrete surface sites for the selective deposition of polymers and magnetic nanoparticles.

The work described herein is centered on reductive methylation, a chemical modification reaction, which allows for the introduction of methyl groups at the primary amines of the lysine side chain and at the N-terminus. The resulting methyl groups are effective probes for the study of protein structure and dynamics using nuclear magnetic resonance spectroscopy (NMR). The work presented in this dissertation will relate the photorecombination dynamics to photoionization dynamics in SF6 utilizing the self-probing mechanism, high harmonic generation (HHG). This work specifically aims to establish parameters for which the comparison is applicable and the macroscopic complications inherent in HHG do not interfere with the extraction of dynamic information. The first part of this work established the macroscopic experimental apparatus necessary for consistent spectroscopic observation. A limiting factor in the digitization of dental devices is the availability of materials suitable for use in both dentistry and the new digital technologies. As a rapidly growing industry, three-dimensional printing (3DP) has the potential to disrupt traditional manufacturing and prototyping methods. A review of both restorative materials and the current 3DP materials has lead to a focus on fiber- reinforced composites in the exploration for a new 3DP material. In addition, another area worth exploring and investing in would be 3D bioprinting as it opens up the possibility of regenerative dentistry.

The reaction of thione 3 or its 2-methylthio derivative 4 with hydrazonoyl halides 5a-l, in the presence of triethylamine, yielded the corresponding triazolo[4,3-$a$]pyrimidin-5(1$H)$-ones 8a-l. The structure of compounds 8a-l was further confirmed by the reaction of 3 with the appropriate active chloromethylenes 11a-c followed by coupling of the products with benzenediazonium chloride to afford the azo-coupling products 6b, f, and j, which were converted in situ to 8b, f, and j. 2-Hydrazinyl-pyrido[3',2':4,5]thieno[3,2-$d$]pyrimidin-4(3$H)$-one (13) was prepared and condensed with different aldehydes 14a-f to give the corresponding hydrazone derivatives 15a-f. SMRT/HDAC Associated Repressor Protein interacts with the long noncoding RNA, produced by SRA, by binding at the RRMs. Three projects were done to prepare the truncated proteins for use in 2D and 3D NMR experiments. The first focuses on RRM 3 and its optimization during the purification process. The second focuses on RRM 2-4, which was found to be missing two alpha helices that may be important for protein stability. Notch1 is a transmembrane protein involved in cell signaling, which is found to be linked to various genetic and neurodegenerative diseases. The extracellular domain of the Notch1 protein consists of 36 tandem epidermal growth factor-like (EGF) repeats, which are directly involved in ligand binding and are heavily glycosylated. EGF 24-29 repeats lie in the Abruptex region of the Notch1 protein, and regulate the ligand binding activity through structural changes. The evolution of powder particle size, crystal structure, and surface chemistry was evaluated for micron scale NiO powders subjected to impact milling with commonly employed milling additives: methanol, Vertrel XF, and amorphous carbon. The effect of the different comminution protocols on sintered body microstructure was evaluated for high temperature sintering in inert atmosphere (N2). X-ray photoelectron spectroscopy showed that NiO powder surface chemistry is surprisingly sensitive to milling additive choice. In particular, the proportion of powder surface defect sites varied with additive, and methanol left an alcohol or alkoxy residue even after drying. This study aimed to synthesize an oligo(azomethine-ether) with benzothiazole moiety in organic medium by means of chemical oxidative polycondensation (OP). Optical properties were examined by photoluminescence (PL) and UV-Vis measurements both in solutions and solid film involved a poly (methyl methacrylate) (PMMA) matrix. Oligomer film in the PMMA matrix emitted a fine green light with a fluorescence quantum yield (QY) of 3.30%. Spectral and thermal observations showed a high rate of C--O--C coupling (cross-linking) for the oligomer. SEC results indicated low molecular weight (\sim3200--4650 g mol^{-1}) and the oligomer was soluble in organic solvents with high polarity. Electrochemical behavior was …

A facile one-pot synthesis of novel oxindole derivatives bearing benzothiazolylmethyl-2-thioxothiazolidin-4-one was accomplished via one-pot reaction of 5-oxoindolinylidene rhodanine-3-acetic acid derivatives, 2-aminothiophenol, and triphenyl phosphite in the presence of tetrabutylammonium bromide (TBAB) and nano silica-bonded 5-n-propyl-octahydro-pyrimido[1,2-a]azepinium chloride (NSB-DBU) as heterogeneous reusable nanocatalyst. The target compounds were obtained in excellent yields (85%--92%) and short reaction times under fairly mild reaction conditions.

CeO$_{2}$ nanoparticles were used as an efficient catalyst for the preparation of cyclic $\beta $-aminoesters by three-component reaction between primary amines, ethyl acetoacetate, and chalcones in ethanol. Atom economy, low catalyst loading, reusable catalyst, and high yields of products are some of the important features of this protocol.

The reaction of 5-amino-3-(4-methoxyphenylamino)-$N$-aryl-1$H$-pyrazole-4-carboxamides \textbf{1a}--\textbf{c} with ethyl acetoacetate \textbf{2} and 2-(4-fluorobenzylidene)malononitrile \textbf{6} yielded pyrazolo[1,5-$a$]pyrimidines \textbf{5a}--\textbf{c} and \textbf{9a}--\textbf{c}, respectively. On the other hand, Schiff bases \textbf{11a}--\textbf{f} were obtained upon treatment of carboxamides \textbf{1a}--\textbf{c} with some selected aldehydes \textbf{10a }and\textbf{ b}. The newly synthesized compounds were characterized and confirmed by analytical and spectroscopic data (IR, MS, $^{1}$H NMR, and $^{13}$C NMR). Pyrazolo[1,5-$a$]pyrimidines \textbf{5a}--\textbf{c} and \textbf{9a}--\textbf{c} and Schiff bases \textbf{11b}--\textbf{f} were investigated for their cytotoxicity against four human cancer cell lines (colon HCT116, lung A549, breast MCF-7, and liver HepG2) according to SRB assay and the structure--activity relationship was discussed.

We have developed a simple synthetic methodology for bis-spirocycles and spiroindole derivatives starting with a commercially available 6-bromo-2-tetralone. Here, we have used Fischer indolization, ring-closing metathesis, and Suzuki--Miyaura cross-coupling as key steps to assemble a variety of spirocyclic frameworks. The methodology developed here is simple and it may be useful to prepare various spirocycles containing indole moiety.

A green and efficient protocol has been developed for the Pd(OAc)$_{2}$-catalyzed ligand-free Suzuki--Miyaura reaction of $N$-methyliminodiacetic acid (MIDA) boronates in water. In the presence of Pd(OAc)$_{2}$ as a catalyst and ($i$-Pr)$_{2}$NH as a base, the cross-coupling reactions of aryl bromides with aryl MIDA boronates proceeded smoothly in water without any surfactant, and various functional groups were tolerated under the optimized conditions.

In the present study, relationships are developed for determining bond orders (also referred to as bond valences or bond numbers) from published bond lengths for carbon-carbon (C-C) and carbon-oxygen (C-O) bonds. The relationships are based on Pauling’s empirical formula s = exp((Ro-R)/b)), where s is the bond order, R is the corresponding bond length, Ro is the unit valence bond length, and b is a fitting parameter. We use a recently derived relationship for the b parameter in terms of the bonding atoms’ published atomic orbital exponents. In the theoretical investigation of the electrical breakdown of a gas, we apply a one-dimensional, steady state, constant velocity, three component fluid model and consider the electrons to be the main element in propagation of the wave. The electron gas temperature, and therefore the electron gas partial pressure, is considered to be large enough to provide the driving force. The wave is considered to have a shock front, followed by a thin dynamical transition region. Indoles and N-alkylindoles undergo Friedel–Crafts addition to aldehydes in the presence of trimethylsilyl trifluoromethanesulfonate and a trialkylamine to produce 3-(1- silyloxyalkyl)indoles. Neutralization of the reaction mixture with pyridine followed by deprotection under basic conditions with tetrabutylammonium fluoride provides the 1:1 adduct as the free alcohol. This method prevents spontaneous conversion of the desired products to the thermodynamically favored bisindolyl(aryl)methanes, a process typically observed when indoles are reacted with aldehydes under acidic conditions.
