Mysteries surrounding the most important mineral for the vertebrate biology, hydroxyapatite, are many. Perhaps the Greek root of its name, απαταo, meaning ‘to deceive’ and given to its mineral form by the early gem collectors who confused it with more precious stones, is still applicable today, though in a different connotation, descriptive of a number of physicochemical peculiarities exhibited by it. Comparable to water as the epitome of peculiarities in the realm of liquids, hydroxyapatite can serve as a paradigm for peculiarities in the world of solids. This research studied the preparation of highly dispersed platinum nanoparticles on polyimide films or Pt/PI by surface modification method. Firstly, polyimide film was hydrolyzed in basic solution, followed by being immersed in platinum-ion solution to allow ion exchange process. Platinum ions on the film were subsequently reduced using proper reducing agent to give Pt/PI. After that, morphological and other information of the Pt/PI was investigated by X-ray diffraction analysis, scanning electron microscopy-energy dispersive X-ray spectroscopy, transmission electron microscopy, and inductively coupled plasma-optical emission spectroscopy. Proper collection of charges at the electrodes is an important process in organic solar cells. Interfacial layers between the electrodes and the photoactive layer have been design to improve charge collection and ohmic contacts. The most studied interfacial layer between the anode and the photoactive layer is the negatively charged polymer poly(3,4-ethylendioxythiophene):poly(styrenesulfonate) (PEDOT:PSS), that possesses good hole transporting ability. Characterization of crude oil and some of its refined products has always been a challenge in the refining industry. This is mainly due to the high molecular complexity of crude oil. In this research, Heavy and Light Resins extracted from Penn Grade crude oil were analyzed with a wide variety of techniques, ranging from X-Ray Diffraction, Electron Microscopy, Fourier Transformed Infrared, C-13 Nuclear Magnetic Resonance, and chromatographic methods. An asphaltene extraction protocol was performed on both products. Only the Heavy Resin sample yielded a low amount of asphaltene. Increased levels of nitrosative stress intracellularly within the endoplasmic reticulum is a key factor involvled in the pathogenesis of both Parkinson's (PD) and Alzheimer's (AD) diseases. Previous in-vitro studies in our lab showed that increased levels of nitrosative stress lead to aggregation of misfolded proteins and formation of Lewy Bodies, the main biomarker of PD and AD diseases. Although this was mainly through nitrosylation of Protein Disulfide Isomerase (PDI), the chief endoplasmic reticulum (ER) resident oxidoreductase chaperone responsible for maturation of disulfide-bond-containing proteins, we demonstrate in this project that increased levels of nitrosative stress has an additional direct effect on …

Previous studies investigating the effects of cerium oxide nanoparticles (nanoceria, nCeO2) on plants have primarily focused on the physiological and biochemical changes at early growth stages. Comprehensive information on the effects of nCeO2 through the entire life cycle of plants and the nutritional quality of the edible tissues is limited. No studies have been reported on the interactions between nCeO2 and common beans (Phaseolus vulgaris). Common beans are leguminous crops, which are societally important due to their nutritional benefits. Optimization Of Molecular Beacon-Based Multicomponent Probes For Analysis Of Nucleic Acids, Maria Stancescu

Detection of single nucleotide substitutions (SNS) in DNA and RNA has a growing importance in biology and medicine. One traditional approach for recognition of SNS takes advantage of hybridization probes that bind target nucleic acids followed by measuring ?Tm, the difference in melting temperatures of matched and mismatched hybrids. The approach enables SNS differentiation at elevated temperatures (usually 40-65oC) often only in a narrow range of < 10oC and requires high-resolution melting devices. Here we demonstrate that a specially designed DNA probe (X sensor) can broaden ?Tm from ~10oC to ~16oC and distinguish SNS in the interval of ~5-40oC. Therefore, there is no need for heating or measuring Tm for accurate SNS differentiation. Our data indicate that this wide differentiation range is in part due to the non-equilibrium hybridization conditions. Further we explored the idea that it is possible to improve the performance of an X sensor operable in close to equilibrium conditions by shifting its operability to non-equilibrium conditions. One way to achieve this is to introduce as many as possible structured ligands in analyte's dissociated state. Here we show that by introducing the maximum possible conformational constraints in X probe it is possible to shift its operation to non-equilibrium conditions and to improve its selectivity at temperatures < 15oC. Thus, this work points towards a new strategy for the design of highly selective hybridization sensors which operate in non-equilibrium conditions at close to room temperature. The X sensors could be utilized in qPCR, microarrays, as well as RNA analysis in living cells and for ambient temperature point-of-care diagnostics. In the last part of this work, X sensors were used in real time detection of PCR products. The sensors were optimized to operate in PCR buffer with optimal Mg2+ concentration. They were able to detect the target amplicon together with nonspecific products. The results presented here suggest that X sensors might be adopted for real time PCR format.

In forensic casework it is often necessary to obtain genetic profiles from crime scene samples that contain increasingly smaller amounts of genetic material, called Low Template DNA (LTDNA). Two examples of LTDNA sources are touch DNA evidence and dust bunnies. Touch DNA refers to DNA that is left behind through casual contact of a donor with an object or another person. Touch DNA can be used to prove a suspect was present at a crime scene. The title compound [Co(C22H26N4O4)]PF6, commonly known as [Co(bpcd)]PF6, where bpcd2- is derived from the historical ligand name N,N'-bis­(2-pyridyl­meth­yl)-trans-1,2-di­amino­cyclo­hexane-N,N'-di­acetate, crystallized by slow evaporation of a saturated aceto­nitrile solution in air. The cation of the hexa­fluorido­phosphate salt has the CoIII atom in a distorted octa­hedral coordination geometry provided by an N4O2 donor atom set. The acetate groups, which are oriented trans with respect to each other, exhibit monodentate coordination whereas the pyridyl N atoms are coordinating in a cis configuration. This project was initiated in conjunction with Dr. Justin Youngblood from the University of North Texas. The goal was to synthesize a molecule to be used in solar cell testing. The target molecule involved combining donor and acceptor groups to make a combination that would be able to transfer electricity in a unilateral direction to increase efficiency in solar cells. My project involved the donor portion, a perylene monoimide donor with a long alkyl swallowtail to impart solubility. Single stranded DNA that forms secondary structures has been a popular area of research in recent years. Much research has also been done in the field of heavy metals binding to DNA. Recently, it has been discovered that mercury in the presence of thymines will form T-HgII-T bonds. It is our hypothesis that single stranded DNA rich in thymine residues will form into a structure similar to i-motifs, formed from C-rich single strands, in the presence of mercury. Recent studies have illustrated the presence of several secondary, non-B DNA structures that are associated with the promoter regions of several oncogenes, such as the intensely studied MYC gene. Two such secondary structures, G-quadruplexes and i-Motifs, have become topics of interest due to their prevalence within oncogenic promoters and their potential accessibility for future drug targeting due to their globular natures.
