Horseradish Peroxidase-Attapulgite Clay Nanocomposites: Fabrication And Application To Sensing The Extracellular H2O2 Released From Cells, Ping Wu, Chen-Xin Cai

The nanostructured attapulgite clay was employed as a support matrix for immobilizing horseradish peroxidase (HRP). FTIR and electrochemical methods demonstrated that HRP had been effectively assembled on attapulgite surface with the formation of HRP-attapulgite nanocomposites. The HRP-attapulgite hybrid was deposited on the glassy carbon (GC) electrode forming the HRP-attapulgite/GC electrode. Cyclic voltammetric results showed a pair of well-defined redox peaks, which were ascribed to direct electron transfer (DET) of HRP, with the formal potential E0′ = -335 mV (vs. SCE) in the phosphate buffer solution (PBS, pH 7.0). Due to the unique optical, electronic, magnetic and catalytic properties, functional nanomaterials provide new opportunities for the rapid development in a variety of disciplines and show considerable promise in different fields. Specifically, constructions of the novel electrochemical interface using various nanomaterials and exploring their advanced electrochemical properties have received great attention to develop electrochemical devices with potential applications. In this paper, the uniform ZnO nanorod arrays were prepared by galvanostatic electrodeposition on Ti Substrates. The morphology and structure of samples were characterized by scanning electron microscopy (SEM) and X-ray diffraction (XRD). The effects of the concentration of Zn(NO3)2 and the deposition current on the morphology of the ZnO nanorod arrays were investigated. Photocatalytic properties of ZnO nanorod arrays were investigated by degradation of methyl orange (MO) under UV-light irradiation. The analysis revealed that the concentration of Zn(NO3)2 and the deposition current strongly influenced the morphology of the ZnO nanorod arrays. Compared with …

The deoxidation speed of a solid oxide cathode in molten CaCl2 can be estimated by the PRS steady diffusion model of O2-, which correlates the deoxidation speed with the precursor porosity, P, the metal-to-oxide molar volume ratio, R, and the cathode volume shrinkage S. The PRS model indicates that the porosity of the oxide cathode has important influence on the deoxidation speed, and provides a very simple equation for the prediction of the optimal cathode porosity. For the electrolysis of Ta2O5, the porosity of the cathode is better to be within 50%. …

A series of model molecules with 4 kinds of conformational transformations have been investigated as optical molecular switches by using density functional theory combined with nonequilibrium Green’s function method. The theoretical calculations show that molecules after conformational transformations have photoswitching characteristics. We find that the photochromic molecules with the same conformational transformation usually have a similar current on/off state when they are applied as photoshwitches. Among these transformations, the molecular switch with E(“trans”)/Z(“cis”)-isomerisation of the NN double bond has the highest current on-off ratio. Although the molecular structures are similar to each other, the redox property of Co(CN)63- at solid/liquid interface is not so good as Fe(CN)63-. In this paper, scanning electrochemical microscopy (SECM) was adopted to construct an electrochemical microsystem with a volume of fL~pL, in which a micropipette with micrometer-sized orifice was employed as the scanning tip. Due to the evaporation of water, the NaCl electrolyte is concentrated to form a microcrystal. The Na2MnPO4F is one of the promising cathode materials for the sodium ion batteries. In the paper, we employed the ex situ X-ray diffraction and solid state NMR techniques to study the charge and discharge processes of this material, including the crystal structure and sodium sites changes. The ex situ x-ray diffraction patterns showed that two new diffraction peaks could be observed at 31o and 36o indicating an intermediate phase formed with the extraction of Na+. DFT calculation is used to study various possible steps in the oxygen reduction reaction (ORR), including the adsorption and dissociation process of O2 and the serial protonation process of dissociated products to form H2O on the Pt(111) surface. By using slabs of different sizes, and different numbers of pre-adsorbed oxygen atoms, the coverage effects on the pathways are investigated. The calculated results using different hydrated proton models are also compared. Electrochemistry and electroanalytical chemistry at a liquid/liquid interface have attracted much attention in the past few decades since they have been closely related to many applications including exploration in mechanisms of extraction processes and chemical sensing, as well as investigations in phase transfer catalysis, drug release and mimicking the function of biological membranes. In this review, recent progresses in this field in the last decade, especially the investigations in microstructure of liquid/liquid interface, charge (ion and electron) transfer reactions and functionalization of liquid/liquid interfaces, are reviewed.

Inductively coupled plasma-mass spectrometers (ICP-MS) have become the workhorses of many analytical labs over the past few decades. Despite the instruments' high sensitivities and low detection limits there is still a demand for improvements in several aspects of their performance. Perturbations in metabolite concentrations have been used to reflect the activity of corresponding enzymes or proteins. Nuclear magnetic resonance (NMR) spectroscopy is a well-known approach for the structure determination of biological macromolecules. Although type IV collagen is heavily glycosylated, the influence of this posttranslational modification on integrin binding has not been investigated. In the present study, galactosylated and non-galactosylated triple-helical peptides have been constructed containing the α1(IV)382-393 and α1(IV)531-543 sequences, which are binding sites for the α2β1 and α3β1 integrins, respectively. All peptides had triple-helical stabilities of 37 °C or greater. The galactosylation of Hyl393 in α1(IV)382-393 and Hyl540 and Hyl543 in α1(IV)531-543 had a dose dependent influence on melanoma cell adhesion which was much more pronounced in the case of α3β1 integrin binding. Background: To determine how diets high in saturated fat could increase polyp formation in the mouse model of intestinal neoplasia, ApcMin/+, we conducted large-scale metabolome analysis and association study of colon and small intestine polyp formation from plasma and liver samples of ApcMin/+ vs. wild-type littermates, kept on low vs. high-fat diet. Label-free mass spectrometry was used to quantify untargeted plasma and acyl-CoA liver compounds, respectively. Differences in contrasts of interest were analyzed statistically by unsupervised and supervised modeling approaches, namely Principal Component Analysis and Linear Model of analysis of variance. Optical devices were fabricated by sandwiching a “monolithic” poly(N-isopropylacrylamide-co-N-(3-aminopropyl) methacrylamide hydrochloride) (pNIPAm-co-APMAH) microgel layer between two semitransparent Au layers. These devices, referred to as etalons, exhibit characteristic multipeak reflectance spectra, and the position of the peaks in the spectra primarily depends on the distance between the Au surfaces mediated by the microgel layer thickness. Here, we show that the positively charged microgel layer can collapse in the presence of negatively charged single stranded DNA (ssDNA) due to ssDNA induced microgel crosslinking. The experimental Raman spectra of three phosphorus-substituted phosphine boranes with bulky hydrocarbon substituents are presented and compared to the results of electronic structure computations by using the M06-2X method and the 6-311G(2df,-2pd) basis set. Total-energy distributions (TEDs) are calculated to describe the degree of mixing of the dative-bond stretching vibration with other simple internal coordinates. This level of theory is found to accurately reproduce the B-P stretching frequency in all three crystalline solids. The Raman spectra of five smaller B-P-containing molecules, including BH3PH3, are also simulated at this level of theory and compared to previous experimental …

Additional experimental evidence is presented for in vitro generation of hydroxyl radicals because of redox cycling of environmentally persistent free radicals (EPFRs) produced after adsorption of 2-monochlorophenol at 230 °C (2-MCP-230) on copper oxide supported by silica, 5% Cu(II)O/silica (3.9% Cu). A chemical spin trapping agent, 5,5-dimethyl-1-pyrroline-N-oxide (DMPO), in conjunction with electron paramagnetic resonance (EPR) spectroscopy was employed. Experiments in spiked O17 water have shown that ∼15% of hydroxyl radicals formed as a result of redox cycling. This amount of hydroxyl radicals arises from an exogenous Fenton reaction and may stay either partially trapped on the surface …

The prevalent trend in chemistry instruction relies on what has been described as the classroom game. In this model, students take a passive role and the instructor does all the explaining (thinking), and learning is trivialized to knowing the correct answers (memorizing) and being able to produce them when prompted (regurgitating). The generation of explanations is central to scientific and technological development. Gangliosides are a family of glycosphingolipids characterized by mono- or polysialic acid-containing oligosaccharides linked through 1,3- and 1,4-β glycosidic bonds with subtle differences in structure that are abundantly present in the central nervous systems of many living organisms. Their cellular surface expression and physiological malfunction are believed to be pathologically implicated in considerable neurological disorders, including Alzheimer and Parkinson diseases. Isobutylene is a molecule of great interest for modeling an explosive detection scheme. Previous analyses in the Reeve lab include low resolution (0.125 cm -1) FTIR scans of the 7 and 28 bands of isobutylene. These scans were used as a baseline for calibrating high resolution (0.01 - 0.001 cm -1) QCL scans of the 7 band. Amplitude modulation and frequency modulation techniques were utilized, though the latter was primarily used in this work. In order to determine the distance between the antigen-binding site and the fragment crystallizable (Fc) region of an antibody using single-molecule Förster Resonance Energy Transfer, dyes must be attached to these locations. To accomplish this, the variable regions of the light and heavy chains of an antibody can be modified and a dye-reactive amino acid introduced into the Fc region. Initial attempts to alter the variable light chain (VL) gene included trying to ligate a 350bp variable gene into an 11,000bp plasmid. Tubulin and heat shock protein 27 (Hsp27) are well-characterized molecular targets for anti-cancer drug development. We previously identified lead compounds that inhibited both Hsp27 and tubulin. These compounds exhibited extensive anti-cancer activities against the proliferation of various human cancer cell lines. In the current study, a systematic ligand based structural optimization led to new analogs that significantly inhibited the growth of a panel of breast cancer cell lines. Furthermore, the most potent compounds were examined with tubulin polymerization assay and Hsp27 chaperone activity assay. This thesis reports the crystallization of two new quasiracemic crystals, the synthesis of a new oxadiazinanone, and the identification of crystals of two new compounds. In previous research the quasiracemates, (5,R,6,S)-4-cyclohexyl-5-methyl-6-phenyl-2H-1,3,4-oxadiazinan-2-one and (5S,6R)-4-isopropyl-5-methyl-6-phenyl-2H-1,3,4-oxadiazinan-2-one, were mixed in solution and crystallized. Instead of fractionally crystallizing, as would be expected, the compounds formed quasiracemic co-crystals. This suggested that other mixtures of oxadiazinanones would be possible candidates for studying compounds which fail to fractionally crystallize.

Diisocyanates (dNCO) such as methylene diphenyl diisocyanate (MDI) are used primarily as cross-linking agents in the production of polyurethane products such as paints, elastomers, coatings and adhesives, and are the most frequently reported cause of chemically induced immunologic sensitization and occupational asthma (OA). Immune mediated hypersensitivity reactions to dNCOs include allergic rhinitis, asthma, hypersensitivity pneumonitis and allergic contact dermatitis.

Eukaryotic initiation factor (eIF) 4F binding to mRNA is the first committed step in cap-dependent protein synthesis. Barley Yellow Dwarf Virus (BYDV) employs a cap-independent mechanism of translation initiation which is mediated by a structural element BTE (BYDV translation element) located in the 3’ UTR of its mRNA. eIF4F bound the BTE and a translational inactive mutant with high affinity; thus questioning the role of eIF4F in translation of BYDV. To examine the effects of eIF4F in BYDV translation initiation, BTE mutants with widely different in vitro translation efficiencies ranging from 5-164% compared to WT were studied. We developed a series of catalysts, composed of metal nanoparticles immobilized on basic supports for the hydrogenation of heteroaromatics of relevance to cleaner fossil fuels and biodiesel, and for the dehydrogenation of heteroaromatics of relevance to hydrogen storage in organic liquids. Our catalyst design involves nanostructured catalysts composed of metal particles immobilized on basic supports capable of ionic mechanism that may avoid catalyst poisoning and enhance catalytic activity.

Orthogonally protected azalanthionines were successfully synthesised by the ring-opening of N-activated aziridine-2-carboxylates with protected diaminopropanoic acids (DAPs). The required DAPs were also prepared by ring-opening of N-activated aziridine-2-carboxylates with para-methoxybenzylamine, but it was found that the choice of aziridine protecting groups dictated both the success of the reaction as well as the regioselectivity of the isolated products. Attempts to extend the methodology to the preparation of the more sterically demanding b-methyl azalanthionines have, so far, been unsuccessful.
