The effects of adsorbed gold nanoparticles on the surface-enhanced infrared absorption (SEIRA) difference spectra of redox-induced cytochrome (Cyt) c in AuNPs/Cyt c/Au sandwich architecture were monitored by SEIRA spectroscopy. The results indicated that the intensity of SEIRA difference spectrum for the vibration of the heme was significantly increased due to the adsorption of gold nanoparticles on the Cyt c. This was induced by the electron transfer between the heme and the gold nanoparticles, which also promoted electron transfer of adsorbed protein. This study suggested a new technique for optimizing the electrochemical property of adsorbed protein.

The Electrocatalytic Performance Of Pd Catalysts Supported On Pvp Modified Mwcnts For Formic Acid Oxidation, Jing Zhang, Xiao Zhao, Chang-Peng Liu, Wei Xing
Journal of Electrochemistry
In this paper the MWCNTs were modified by Poly (N-vinyl-2-pyrrolidone) (PVP) to support Pd nanoparticles, which were used as the catalyst for formic acid electrooxidation. And the performance of Pd/PVP-MWCNTs catalysts was studied by electrochemical measurements. The observations from Fourier transforms infrared spectrometer (FTIR) and transmission electron microscopy (TEM) illustrated that the Pd nanoparticles with a small size and narrow size distribution were highly dispersed on PVP-MWCNT support. Therefore, the Pd/PVP-MWCNTs catalysts showed excellent catalytic activity for formic acid electrooxidation.

The detailed molecular composition of laboratory generated limonene ozonolysis secondary organic aerosol (SOA) was studied using ultrahigh-resolution Fourier transform ion cyclotron resonance (FT-ICR) mass spectrometry. Approximately 1200 molecular formulas were identified in the SOA over the mass range of 140 to 850 Da. Four characteristic groups of high relative abundance species were observed; they indicate an array of accretion products that retain a large fraction of the limonene skeleton. The identified molecular formulas of each of the groups are related to one another by CH2, O and CH2O homologous series. The CH2 and O homologous series of the low molecular …

Design And Synthesis Of Protein-Protein Interaction Inhibitor Scaffolds, David B. Badger
Design And Synthesis Of Protein-Protein Interaction Inhibitor Scaffolds, David B. Badger
USF Tampa Graduate Theses and Dissertations
Many currently relevant diseases such as cancer arise from altered biological pathways that rely on protein-protein interactions. The proteins involved in these interactions contain certain functional domains that are responsible for the protein's biological activities. These domains consist of secondary structural elements such as α-helices and Β-sheets which are at the heart of the protein's biological activity. Therefore, designing drugs that inhibit protein-protein interactions by binding to these key secondary structural elements should provide an effective treatment for many diseases. Presented in this dissertation are the designs, syntheses, and biological evaluations for both novel α-helix and novel Β-sheet mimics.

We have previously derived seven requirements for the formulation of effective stain etchants and have demonstrated that Fe3+, Ce4+, and VO2 (+) + HF solutions are highly effective at producing nanocrystalline porous silicon. Here, we show that Cl-2, Br-2, I-2, ClO3 (-), BrO3 (-), IO3 (-), I-, and I-3 (-) induce etching of silicon when added to HF. However, using the strict definition that a pore is deeper than it is wide, we observe little evidence for porous layers of significant thickness but facile formation of pits. Iodate solutions are extremely reactive, and by the combined effects of IO3 (-), …

Mineral aerosol deposition is recognized as the dominant source of iron to the open ocean and the solubility of iron in the dust aerosol is highly variable, with measurements ranging from 0.01–80%. Global models have difficulty capturing the observed variations in solubility, and have ignored the solubility dependence on aerosol size. We introduce two idealized physical models to estimate the size dependence of mineral aerosol solubility: a diffusion‐controlled model and a surface‐area‐controlled model. These models produce differing time‐ and space‐varying solubility maps for aerosol Fe and Al given the dust age at deposition, size‐resolved dust entrainment fields, and the aerosol …

Chagas disease is a neglected tropical disease caused by the protozoan parasite Trypanosoma cruzi. Here we report crystal structures of the galactofuranose biosynthetic enzyme UDP-galactopyranose mutase (UGM) from T. cruzi, which are the first structures of this enzyme from a protozoan parasite. UGM is an attractive target for drug design because galactofuranose is absent in humans but is an essential component of key glycoproteins and glycolipids in trypanosomatids. Analysis of the enzyme-UDP noncovalent interactions and sequence alignments suggests that substrate recognition is exquisitely conserved among eukaryotic UGMs and distinct from that of bacterial UGMs. This observation has implications for inhibitor …

Hierarchy Of Supramolecular Synthons In The Of Design Multi-Component Crystals, Padmini Kavuru
USF Tampa Graduate Theses and Dissertations
Most of the biological systems in nature are sustained by molecular self-assemblies which are the finest examples of supramolecular architectures. Non-covalent interactions are key concepts which govern these molecular assemblies. Inspired by these examples crystal engineering emerged as an important tool in supramolecular chemistry which aids in the invention of new molecular structures with desired properties. Understanding of how the molecules interact at the molecular levels enables one to rationally design novel solid forms with modulated physicochemical properties. This feature of crystal engineering has heightened its position in materials chemistry and is currently one of the most well studied fields …

Their catalytic activity depending solely of their factes, it is recomendable to achieve six-fold symmetry along with edge sites, ideal places for the attachment of molecules. The aim of this project is to achieve the synthesis of NP-Ag and their functionalizatiin using supported on silica to later perform an oxidation model reaction of diobenzothiophene (DBT) to diobenzenethiophene-sulfone. The DBT oxidation reaction was performed in a reflux system using DBT and silica solved in …
The Importance Of The Rotor In Hydrazone-Based Molecular Switches, Xin Su, Timo Lessing, Ivan Aprahamian
The Importance Of The Rotor In Hydrazone-Based Molecular Switches, Xin Su, Timo Lessing, Ivan Aprahamian
Dartmouth Scholarship
The pH-activated E/Z isomerization of a series of hydrazone-based systems having different functional groups as part of the rotor (R = COMe, CN, Me, H), was studied. The switching efficiency of these systems was compared to that of a hydrazone-based molecular switch (R = COOEt) whose E/Z isomerization is fully reversible. It was found that the nature of the R group is critical for efficient switching to occur; the R group should be a moderate H-bond acceptor in order to (i) provide enough driving force for the rotor to move upon protonation, and (ii) stabilize the obtained Z configuration, to …

During the aerosolization process at the sea surface, halides are incorporated into aerosol droplets, where they may play an important role in tropospheric ozone chemistry. Although this process may significantly contribute to the formation of reactive gas phase molecular halogens, little is known about the environmental factors that control how halides selectively accumulate at the air-water interface. In this study, the production of sea spray aerosol is simulated using electrospray ionization (ESI) of 100 nM equimolar solutions of NaCl, NaBr, NaI, NaNO(2), NaNO(3), NaClO(4), and NaIO(4). The microdroplets generated are analyzed by mass spectrometry to study the comparative enrichment of …

Marine leeches, specific to sea turtles, have been implicated as potential vector organisms in the spread of fibropapillomatosis (FP), a pandemic neoplastic disease with green turtles (Chelonia mydas) having the highest affliction rate. Polymerase chain reaction identified two independent, seemingly functional histone H3 loci for marine turtle leeches Ozobranchus branchiatus collected from C. mydas in Florida and Hawaii. Primers were developed to amplify each product separately. These novel markers will be useful in identifying ectoparasites in FP research, evaluating other histone variants, and chromatin dynamics regulation studies.

Nylon nucleic acids containing oligouridine nucleotides with pendent polyamide linkers and flanked by unmodified heteronucleotide sequences were prepared by DNA templated synthesis. Templation was more efficient than the single-stranded synthesis; coupling step yields were as high as 99.2%, with up to 7 amide linkages formed in the synthesis of a molecule containing 8 modified nucleotides. Controlled digestion by calf spleen phosphodiesterase enabled the mapping of modified nucleotides in the sequences. a combination of complete degradation of nylon nucleic acids by snake venom phosphodiesterase and dephosphorylation of the resulting nucleotide fragments by bacterial alkaline phosphatase, followed by LCMS analysis, clarified the …

Aggregates of amyloid-β (Aβ) peptides have been implicated in the etiology of Alzheimer disease. Among the different forms of Aβ aggregates, low molecular weight species ranging between ∼2- and 50-mers, also called “soluble oligomers,” have emerged as the species responsible for early synaptic dysfunction and neuronal loss. Emerging evidence suggests that the neurotoxic oligomers need not be formed along the obligatory nucleation-dependant fibril formation pathway. In our earlier work, we reported the isolation of one such “off-pathway” 12–18-mer species of Aβ42 generated from fatty acids called large fatty acid-derived oligomers (LFAOs) (Kumar, A., Bullard, R. L., Patel, P., Paslay, L. …

Multiple myeloma (MM) is an incurable B lymphocyte cancer. To date, a comparative analysis of global protein metabolism for the MM cell line CCL-155 (RPMI-8226) and the non-cancerous B lymphocyte cell line CCL-156 (RPMI-1788) has not been published. Here, we report that both global protein synthesis and degradation occur at a higher rate in MM cells and demonstrate that alkylating agents can reduce global protein degradation in both cell lines, but the effect is greater in CCL-156 cells. Treatment with melphalan plus the proteasome inhibitor MG132 reduced global protein degradation for MM cells to roughly 60% of that seen without …

N-Hydroxylating monooxygenases (NMOs) are essential for pathogenesis in fungi and bacteria. NMOs catalyze the hydroxylation of sine and ornithine in the biosynthesis of hydroxamate-containing siderophores. Inhibition of kynurenine monooxygenase (KMO), which catalyzes the conversion of kynurenine to 3-hydroxykynurenine, alleviates neurodegenerative disorders such as Huntington's and Alzheimer's diseases and brain infections caused by the parasite Trypanosoma brucei. These enzymes are examples of flavin-dependent monooxygenases, which are validated drug targets. Here, we describe the development and optimization of a fluorescence polarization assay to identify potential inhibitors of flavin-dependent monooxygenases. Fluorescently labeled ADP molecules were synthesized and tested. An ADP-TAMRA chromophore bound to …

The testing of drug dissolution rates from solid dosage forms is a very important area of research within the pharmaceutical industry. The ability to produce drugs with a given dissolution rate will lead to improved performance in the treatment of patients and will be of economic benefit to the pharmaceutical industry. However, dissolution testing in laboratories, aimed at reflecting in-vivo conditions, can be both time consuming and costly. Currently, most simulations of drug dissolution take place in standardized USP (United States Pharmaceutical) apparatuses. A number of these apparatuses exist, and it is the aim of this thesis to analyse drug …

Eight fluorescent indicators were evaluated for their ability to detect the corrosion of aluminum metal substrates. The fluorophore Rhodamine-salicylaldehyde (RSA) was selected as a candidate for further study based on its ability to bind and detect aluminum ions at low concentrations, its selectivity for aluminum ions, its long-term stability, its solubility in our solvent-based epoxy formulation, and its compatibility with our testing method.
A recent publication suggested that an alternative method of fluorescence activation was possible—an acid-promoted ring opening that occurred in the absence of metal ions. To prove the capability of RSA to bind aluminum ions, thorough 1H …

Polymer matrices are commonly used as guest-host systems for organic chromophores for use in non-linear optical materials. The chromophores must be aligned or poled in an electric field in order to impart anisotropy and non-linear activity to the material. This poling process raises several issues, the two largest being the eventual relaxation of the chromophores back into random orientations due to brownian motion, and high molecular weight polymer matrices limiting chromophore mobility during poling. The prevention of this relaxation process is an area of significant interest, especially in applications that require long term coating stability and activity. In this study, …

Design And Synthesis Studies Of New S-Adenosyl-L-Methionine Analogues, Shanshan Wu
Theses
S-Adenosyl-L-Methionine (SAM), which is involved in methyl group transfers, is the second most abundant coenzyme in the human body. It is made from adenosinetriphosphate (ATP) and methionine catalyzed by adenosyltransferase. Methyltransferases (MTs) transfer the activated methyl group from the sulfur center to a specific position in a variety of substrates, e.g., DNA, RNA, proteins, and secondary metabolites. Methyltransferases have been linked to various diseases, including cancer.
In recent years, more and more SAM analogues have been reported. These analogues show activities in biological target labeling. The purpose of this thesis is to design and synthesize new SAM analogues to inhibit …
