We examine radical mediated pathways in electrophilic addition to the simplest alkene, ethylene, where the structure of the radical intermediate has been extensively debated. Starting from the π-complex with a dihalogen, C2H4⋯I2, isolated in an inert matrix, we initiate reaction by photolytically cleaving the I2 bond. We succeed in trapping and spectroscopically interrogating the symmetrically bridged radical complex, which calculations confirm is the global minimum on the C2H5I Potential Energy Surface (PES). This research encompasses the detailed investigation of the design and synthesis of metal organic framework (MOF) materials involving the M(I,II)/ polyazaheterocycle/anion system in order to expand our understanding of the principles which render the chemistry more controllable and predictable. In addition, this research investigates the development of properties in these classes of compounds. Furthermore, it has been demonstrated that hydrothermal reaction conditions of stoichiometry, pH, and temperature can influence the identity of the products.

The antitumor effects of platinum(IV) complexes, considered prodrugs for cisplatin, are believed to be due to biological reduction of Pt(IV) to Pt(II), with the reduction products binding to DNA and other cellular targets. In this work we used pBR322 DNA to capture the products of reduction of oxoplatin, c,t,c-[PtCl2(OH)2(NH3)2], 2, and a carboxylate-modified analog, c,t,c-[PtCl2(OH)(O2CCH2CH2CO2H)(NH3)2], 3, by ascorbic acid (AsA) or glutathione (GSH). Conserving the environment is an issue that is gaining popularity day by day. Phosphorus transfer from agricultural soils is an important environmental issue that is being closely observed as the transport of phosphorous to water bodies is adversely affecting water quality due to accelerated eutrophication. Heterocyclic organic and organometallic compounds (i.e. polypyrrole), and their derivatives, have been of great interest for conductive polymers due to their novel properties and environmental stability as compared to their non-aromatic analogs (i.e. polyacetylene). Our current interest focus upon the potential role of metal ligand bound pyridazines as the next generation of electronic devices that utilize the metal ligands bound to organics as the semiconducting material. Glycosylation involves the post-translational addition of carbohydrates to protein molecules and is an intricate and indispensable biochemical process. Study of this complicated network of interactions is hindered by the lack of a coding template analogous to the genetic code, and by the vast structural complexity inherent to carbohydrate polymers. We use lectins (non-enzymatic carbohydrate-binding proteins of non-immunological origin) as microarray probes to identify carbohydrate features expressed on cellular surfaces. Specifically, we utilized lectin microarray technology to investigate the differences in carbohydrates expressed by the cell lines of the Nation Cancer Institute’s NCI-60 panel. The pentiptycene receptors form edge-to-face complexes with a variety of aromatic guests including nitroaromatics. X-ray diffractometry revealed that compounds 1, 2, and 3 form host-guest assemblies with a thienyl fragment (from a neightboring molecule of 1), benzene and nitrobenzene, respectively. X-ray studies of the three crystal structures reported here strongly suggest the edge-to-face to be a predominant binding mode between the aromatic guests and electron-rich faces of the pentiptycene aromatic cavity.

A supramolecular assay based on two fluorescent cucurbit[n]uril probes enables the recognition and quantification of nitrosamines, including cancer-associated nitrosamines, compounds that are difficult to recognize. The cross-reactive sensor leverages weak interactions and competition among the probe, metal, and guest, yielding high information density in the signal output (variance) and enabling the recognition of structurally similar guests.

A three and a half day conference focusing on nucleic acid structures called G-quadruplexes (G4s) and other guanine-based assemblies was held in Sorrento. Italy (June 28-July 1, 2011) and featured 35 invited talks and over 89 posters. The G-quadruplex field continues to expand at an explosive rate with the emergence of new connections to biology, chemistry, physics, and nanotechnology. Following the trend established by the previous two international G4 meetings, the conference touched upon all these areas and facilitated productive exchanges of ideas between researchers from all over the world.

A detailed discussion on the development of an ambient ionization mass spectrometry method called paper spray (PS-MS) and leaf spray (LS-MS) mass spectrometry will be presented. This project aims to understand how surface polishing affects different materials in the context of art conservation. These materials include both those used in the creation and preservation of art objects, and those used to house and display them in a museum. Conservators at SBE Conservation and Cooper Hewitt have been working on two series of art objects, Still Life #46 and Still Life #54 by Tom Wesselman, for the past 6 years. Tryptophan is a fluorescent amino acid that is commonly used as a probe in protein structure-dynamics studies. It is also widely used as a convenient standard for measurements of fluorescence quantum yields of organic and bioorganic molecules. Literature values of the fluorescence quantum yield range from 0.13 to 0.15 upon excitation of tryptophan at 270 nm in aqueous solution. Even though the fluorescence quantum yield of tryptophan is known with a high degree of accuracy upon excitation at 270 nm (i.e., ca. ± 0.02), this value is being used indiscriminately in the literature for the determination of the fluorescence quantum …

Reactions with 2-methylenetetrahydropyrans are underutilized in the literature, especially with respect to general methods demonstrating their nucleophilic nature. This work provides three general methods toward the synthesis of 2-β-oxygenated pyranyl substrates, which are key structural components of many biologically active polyketides and natural products.
The use of exocyclic enol ethers as nucleophiles was first demonstrated as a general process in three-component coupling reactions with secondary nucleophiles. The 2-β-hydroxy tetrahydropyran products were obtained in good to excellent yields through promotion with stoichiometric levels of titanium(IV) chloride at reduced temperatures. This dissertation involves work both on the syntheses of small molecules with potential therapeutic applications and synthetic studies on natural products with unusual core skeletons.
The first part of this work involves the synthesis, design, and biological evaluation of aminosteroids as inhibitors for the SH2-containing inositol phosphatase 1 (SHIP1). SHIP1, a 145 kDa protein, is involved in regulating the formation of new blood cells. High- throughput screening identified several SHIP inhibitors including NSC23922, a mixture of 3a- and 3β-aminocholestane. Protein therapeutics like insulin and glucagon-like peptide-1 analogues are currently used as injectable medications for the treatment of diabetes mellitus. An orally administered protein therapeutic is predicted to increase patient adherence to medication and bring a patient closer to metabolic norms through direct effects on hepatic glucose production. The major problem facing oral delivery of protein therapeutics is gastrointestinal tract hydrolysis/proteolysis and the inability to passage the enterocyte. Clostridium difficileinfections (CDI) have emerged as a leading cause of hospital-associated complications. CDI is the major cause of antibiotic-related cases of diarrhea and nearly all cases of pseudomembranous colitis. The infective form of C. difficileis the spore, a dormant and hardy structure that forms under stress. Germination of C. difficile spores into toxin producing bacteria in the GI tract of susceptible patients is the first step in CDI establishment. Patient susceptibility occurs with a disruption of the natural gut microbiota by broad-spectrum antibiotics. Antibiotic treatments usually resolve CDI but refractory cases are on the rise. Waterborne coatings are formulated with a number of different ingredients; water, latex polymers, pigments, surfactants, dispersants, defoamers, biocides, coalescing aids, and rheology modifiers or thickeners. Rheology modifiers are necessary in order to improve the physical properties of the coating before, during, and after application to a substrate. There are two kinds of rheology modifiers used in waterborne coatings; associative thickeners and non-associative thickeners. The photophysical properties of the canonical DNA/RNA nucleobases can be modified by selective functionalization. The carbonyl functional group at the C6 position of guanine or 2-amino-6-oxopurine, for example, can be removed to form 2-aminopurine, thus replacing the ultrafast internal conversion pathway to the ground state observed in guanine with slow internal conversion and intersystem crossing pathways in 2-aminopurine.1 Similarly, substitution of the carbonyl oxygen in guanine with a heavier atom results in a redshift of the absorption spectrum and a remarkable modulation of the electronic relaxation pathways of the DNA nucleobase. Intramolecular BO-chelated azadipyrromethenes are promising organic semiconductors. Here, we evaluated the electron and hole mobility of a series of BO-chelated azadipyrromethenes using the Space Charge Limited Current(SCLC) method. In order to determine the mobility of the material, Mott Gurney's law was applied using the film thickness and slope of the J1/2 versus voltage plot in the SCLC graph. The best electron mobility observed for BO-chelated materials is 4.64—10-6 cm2V-1s-1 and the best hole mobility is 7.38—10-4 cm2V-1s-1. These results suggest that BO-chelated materials are promising p-type semiconductors for electronic applications.

A common reaction to yield ethers relies on an alkoxide nucleophile and a carbon electrophile (Williamson ether formation). Ethers can also be formed by reactions with carbon nucleophiles and oxygen electrophiles by way of the peroxide functional group. We screened a number of carbon nucleophiles suitable for attack on dialkyl peroxides to form new C-O bonds. Also a novel approach using a peroxyketal electrophile is shown to increase the efficiency of this intermolecular reaction. This method of C-O bond formation can also be implemented intramolecularly, not seen before in the literature, to yield cyclic ethers. This approach requires the generation …

Many rare earth compounds have photoluminescent characteristics for various important applications. In this study, europium doped La2Zr2O7 nanoparticles were first synthesized morphology-controllably by using a molten-salt synthesis method. These nanoparticles were used as the core for coating with YBO3 shell. We then explored the effect of different experimental processing parameters, such as annealing temperatures, reaction times and different ratios of salt mixtures on the structure and morphology of these nanoparticles. The overall core size and shell thickness can be controlled by varying the reactant concentration both the core and shell material. When developing a model polymeric system to facilitate in the detection of molecular and microscopic events that preface macroscopic corrosive failure; a better understanding of how polymers can indicate corrosion was accomplished. Initially, the thought that molecular chain scission as a necessity for corrosion to occur had to be tested. Through the utilization of high molecular weight thermoplastic (HMWTP) model polymers, it was found that corrosion protection did not correlate to the quantity of weak bonds within an epoxy-amine polymer matrix. Therefore more sensitive methods of detecting corrosion had to be developed since changes within the polymer matrix did not …

Synthesis of polymer brushes to decorate a surface with desired functionality typically involves surface-initiated polymerization (SIP) of functional, but non-reactive monomers. This approach suffers major drawbacks associated with synthesizing sufficiently thick polymer brushes containing surface-attached polymer chains of high molecular weight at high grafting density (i.e. cost, synthetic effort and functional group intolerance during polymerization). The research herein seeks to circumvent these limitations by the decoration of surfaces with polymer chains bearing specific pendent functional groups amenable to post-polymerization modification (PPM). While many methods exist to separate and analyze radionuclides from a variety of environmental matrices, the performance of all of these methods is often limited by other interfering constituents that are consistently found in most of these samples. The presence of such constituents can significantly reduce the recovery of the radioisotopes of interests and lead to incomplete separations.

Pyridinium salts, both molecular and polymeric, are an interesting class of multifunctional materials that exhibit liquid-crystalline and light-emitting properties. Moreover, their properties can be easily tuned by introducing new types of anions or by modifying their chemical structures. This dissertation describes synthesis and characterization of poly(pyridinium salt)s containing macrocounterions and fluorene moieties in their backbones, synthesis and characterization of nanocomposites of poly(pyridinium salt)s with single-walled carbon nanotubes via non-covalent interactions, and synthesis and characterizations of pyridinium salts having different aliphatic linkages and their application in organic acid sensing.

Concerns over the proliferation and instances of nuclear material in the environment have increased interest in the expansion of nuclear forensics analysis and attribution programs. A new related field, molecular forensic science (MFS) has helped meet this expansion by applying common scientific analyses to nuclear forensics scenarios. In this work, MFS was applied to three scenarios related to nuclear forensics analysis. In the first, uranium dioxide was synthesized and aged at four sets of static environmental conditions and studied for changes in chemical speciation. The implications of H+H2 cooling in astrophysics is important to several applications. One of the most significant and pure applications is its role in cooling in the early universe. Other applications would include molecular dynamics in nebulae and their collapse into stars and astrophysical shocks. Shortly after the big bang, the universe was a hot primordial gas of photons, electrons, and nuclei among other ingredients. By far the most dominant nuclei in the early universe was hydrogen. In fact, in the early universe the matter density was 90 percent hydrogen and only 10 percent helium with small amounts of lithium …
