Breast cancer cells overexpress EpCAM that can be used as a marker to select breast cancer CTCs from heterogeneous clinical samples, even when present in low abundance. There is currently no available molecular diagnostic test for stroke; the common modality for diagnosis consists of computed tomography or magnetic resonance imaging. Unfortunately, the use of these diagnostic regimens can delay proper therapeutic treatment, which requires administration within the first 3 h of a stroke event. We are developing a molecular assay that can report, in near real time and at the point-of-care, the presence or absence of biomarkers specifically targeted for the diagnosis of ischemic or hemorrhagic stroke. Frontal polymerization (FP) is when the monomer is polymerized by a zone of reaction propagating through the monomer. The reaction zone is fueled by the heat given off by the polymerization of the monomer. FP is a promisig method for curing adhesives because it allows for cure-on-demand and fast cure times. The monomers selected for the adhesive is the main factor in determining the adhesive’s properties. Trimethylolpropane triacrylate (TMPTA), pentaerythritol tetraacrylate (PETA), and other multifunctional monomers make the polymer formed more brittle. Liposomes have had a long and fruitful application of drug delivery devices in the treatment of select tumors and cancer types. Third-generation liposomes possess endogenous triggering methods that hold the ability for site-specific unloading of liposomal contents, thus providing necessary temporal and spatial control over contents release that can be tuned to match the drug efficacy for decreased side effects during treatment. Of these new endogenous triggers, redox-active liposomes that can be triggered to unload contents via two-electron reduction by enzymes or chemical means have demonstrated promise for the encapsulation and delivery of cargo. This dissertation explores the relationship between ionic conductivity, viscosity, and thermal properties of several MePEG and MePPG based copolymer electrolytes. In particular two main copolymer modifications have been investigated namely copolymerization with "bulky groups" and cross polymerization of MePEG and MePPG monomers. The modifications were made to vary the fractional free volume of the MePEG polymers. All of the copolymers obeyed the Doolittle equation. For both the bulky copolymers and MePEG/MePPG copolymers, increases in the FFV corresponded to increases in the viscosity and decreases in FFV correspond to decreases in viscosity. The monooxovanadium(V) complex of composition [VO(OC_6H_3(NO_2)_2-2,4)_3] (I) was synthesized by the reaction of VOCl_3 with Me_3SiOC_6H_3(NO_2)_2-2,4 in toluene and characterised by elemental analyses, molar conductance, and IR, ^1H- and ^{13}C-NMR, and mass spectral studies. The reactions of (I) with alkali metal 2,4-dinitrophenoxides [MOC_6H_3(NO_2)_2-2,4] (M = Li, Na, K) yield complex salts of composition M[VO(OC_6H_3(NO_2)_2-2,4]_4 authenticated by physicochemical and IR spectral studies. Thermal behaviour of complexes was studied by TG and DTA techniques. In order to investigate the enantiomeric recognition abilities toward 2 chiral alkylammonium perchlorates (AmI, AmII) by ^1H-NMR titration method in CDCl_3, 4 chiral lariat ethers 8-11 with a (p-methoxyphenoxy) methyl flexible side arm were used. The most effective enantiomeric recognition was obtained by LCEs 9 and 11 toward AmII, by K_R/K_S 6.58 and K_S/K_R 6.63, respectively. The effect of macroring size, subunit of macroring, and side arm appeared to have strong influence on the binding ability of these alkylammonium ions.

A new and simple differential pulse polarographic method was developed for the trace determination of As(III) and As(V) species in the same sample. Among several electrolytes, 0.1 M HCl was found to be the most suitable. As(III) had 3 peaks in this medium, and the peak at --0.82 V was chosen because of its peak shape and response to standard additions. In this medium, the limit of detection for As(III) was found to be 1.5 \times 10^{-7} M. Since As(V) is not electroactive, it had to be reduced first into As(III). The results of computational calculations on the aromaticity of the monosubstituted diazabenzenes (pyridazine, pyrimidine, and pyrazine) are reported herein. The aromaticity of the parent heterocycle was enhanced by substitution of strong electron-withdrawing groups. The effects of the position of the substituent on the aromaticity and the stability of the system were also investigated by studying all possible derivatives of the systems.

Drug delivery by a controlled-release mechanism offers several advantages over traditional delivery systems such as improved therapeutic activity, reduced number of drug administrations, as well as minimizing the risk of over-dose or under-dose. In this study, attempt has been made to develop a novel controlled release device from glycerol crosslinked poly(acrylic acid) hydrogels. Glycerol crosslinked poly(acrylic acid) was fabricated by polymerizing acrylic acid and crosslinking the polymer with glycerol in the same reaction pot in the presence of benzoyl peroxide and Novozym 435. A versatile AFM local oxidation lithography was developed for fabricating clean protein patterns ranging from nanometer to sub-millimeter scale on octadecyltrichlorosilane (OTS) layer of Si (100) wafer. This protein patterning method can generate bio-active protein pattern with a clean background without the need of the anti-fouling the surface or repetitive rinsing.

As a model system, lysozyme protein patterns were investigated through their binding reactions with antibodies and aptamers by AFM. Polyclonal anti-lysozyme antibodies and anti-lysozyme aptamer are found to preferentially bind to the lysozyme molecules on the edge of a protein pattern before their binding to the interior ones. A novel amphiphilic, unsymmetrically substituted N-((2S)-2-aminohexanoic acid)-N'-[(S)-1-phenylethyl]-3,4,9,10- perylenetetracarboxydiimide with 2 different chiral centers was synthesized and characterized. Chiral amphiphilicity, which can be tunable between hydrophilic and hydrophobic parts, is responsible for controlled self-assembly through intermolecular hydrogen bonding (N-H \cdots O). The product showed high thermal stability but partial solubility in common organic solvents. We observed 2 isosbestic points at 533 and 611 nm, confirming the presence of overlapped monomer and excimer emissions in the temperature range of 10-80 °C. In-vitro inhibition activity screening and in-silico 3D molecular modeling approaches are important tools for design and development of specific histone deacetylase (HDAC) inhibitors. The objective of this study was to investigate the consistency between these 2 approaches. The HDAC 8 inhibition activities of 8 randomly selected different carboxylic acid derivatives were screened and in-vitro experimental results were compared with in-silico molecular modeling calculations. This study demonstrated that there is no sole gold standard technique for inhibitor design, and it was concluded that a combination of molecular modeling and activity screening assays will ensure more comprehensive and dependable results.

In the present study, some new 2-(pyridin-2-yl)thieno[2,3-d] pyrimidin-4(3H)-ones derivatives (IIa-o) were synthesized. The target compounds (IIa-o) were synthesized through the acid catalyzed condensation of 2-cyano, 3-cyano, and 4-cyano-pyridines with various 2-amino-3-carbethoxythiophenes (Ia-e). All thiophene derivatives were synthesized by Gewald reaction. The structures of the newly synthesized compounds were confirmed by UV-Visible, FT-IR, ^1H-NMR, and mass spectral studies. All synthesized compounds were evaluated for their antimicrobial activity against various gram-positive and gram-negative bacterial and fungal strains. Amongst the synthesized compounds IIa, IIb, IId, IIe, and IIm were found to be active.

Spectrophotometric determination of Ni(II) was carried out using picolinaldehyde-4-phenyl-3-thiosemicarbazone (PAPT) as a complexing/chromogenic reagent. The possibility of using a micellar system was explored to enhance the degree of detection and allow a more sensitive spectrophotometric method for the determination of nickel ions. The anionic surfactant Triton X-100 greatly enhanced the molar absorptivity and limit of detection relative to that observed with bulk water or an organic solvent. The linear calibration graphs were obtained for 0.5-2.0 mg L^{-1} and 0.001-1.0 mg L^{-1} of Ni(II) in aqueous and surfactant media, respectively. An evaluation was made of different digestion methods for the determination of arsenic and antimony in table salt samples prior to hydride generation atomic absorption spectrometric analysis. Microwave acid digestion, classical wet digestion, dry ashing, and fusion were applied to the decomposition of salt samples and optimum conditions were investigated. Samples were decomposed by changing heating time, digestion techniques, and the amount and composition of acid, and then the concentrations of arsenic and antimony in an unrefined salt sample were measured. This research concentrates on the preparation, characterization, and photoluminescence properties of new Co(II), Ni(II), Cu(II), and Zn(II) complexes of N-aminopyrimidine-2-thione. The ligand and metal complexes were characterized by elemental analysis and spectroscopic studies such as IR, UV/Vis, API-ES, NMR, fluorescence, and magnetic susceptibility measurements. The ligand and the resulting metal complexes gave intense emissions (\lambda_{max} = 408 nm) upon irradiation by ultraviolet light. The photoluminescence quantum yields and long excited-state lifetimes of the ligand and its complexes were calculated. Polychlorinated biphenyls (PCBs) are persistent organic pollutants. Due to their properties, PCBs accumulate in the food-chain and post a threat to the health of human beings and wildlife. Hydroxylated PCBs (OH-PCBs) are oxidative metabolites of PCBs and are more hydrophilic than their parent PCBs. This work probes radiative polaritons in thin oxide layers as a mean to capture and absorb broadband infrared radiation and transform it into heat. A heat recovery mechanism, based on the Seebeck effect, is used as the tool of the investigation. Heat production challenges the current understanding which views the excitation of radiative polaritons as only accompanied by the emission of electromagnetic radiation. In preliminary screens, several functions, most notably, genes involved in the control of reactive oxygen species (ROS), were identified as playing a role in the process of growth rate compensation of branch density. The maintenance of branch density under growth at various temperatures was examined in a selection of mutants in genes known to be important in the control of ROS. In all ROS control mutants tested, growth was shown to branch tighter when grown at higher temperatures (which result in faster growth rates) and looser when grown at lower temperatures (which results in slower growth rates). In 1979, the Dow Chemical Company published an excerpt from a speech by H. Peter Metzger that announced an emerging conflict in American ideals and public policy. He stated that a new kind of individual inhabited Washington, people from the counterculture who were “coercive utopians” because they sought to achieve their agenda through covert actions and hoped to end the American free market economy. This paper deals with the synthesis and spectroscopic investigation of homometallic dinuclear ruthenium(II) complex containing chlorophenanthroline and bipyridine ligands. This bimetallic ruthenium polypyridine complex may be useful for biological electron transfer studies. Heteroleptic ruthenium monomer complex Ru(bpy)2(Cl-phen) (where bpy = 2,2’-bipyridine and Cl-phen = 5-chloro- 1,10-phenanthroline) was prepared in a two step procedure previously developed in our laboratory. This monomer complex was used to prepare the ruthenium dimer complex, (bpy)2Ru(phen-phen)Ru(bpy)2, by utilizing the Ni-catalyzed coupling reaction. Thiol-alkyne 'click' chemistry is a modular, efficient mechanism to synthesize complex A2B 3-arm star polymers. This general motif is similar to a phospholipid where the A blocks correspond to lypophilic chains and the B block represents the polar head group. In this communication we employ thiol-yne chemistry to produce polypeptide-based A2B lipid mimetics. The utility of the thiol-yne reaction is demonstrated by using a divergent and a convergent approach in the synthesis. We have explored in silico the potential energy surfaces of the C5Linn-6 (n = 5, 6, and 7) clusters using the Gradient Embedded Genetic Algorithm (GEGA) and other computational strategies. The most stable forms of C5Li5-- and C5Li6 are two carbon chains linked by two lithium atoms in a persistent seven membered ring capped by two Li atoms. Our aim was to develop the synthetic potential and reaction chemistry of Ir3+ and Ru2+ electrophiles by preparing well-characterized complexes whose properties are controllable by modification of the ancillary ligand environment Specifically, we prepared a series of ruthenium complexes to serve as selective hydrogenation and hydrogenolysis catalysts of furan derivatives. We also expanded the synthesis of electrophilic Ir3+ di-thiolate complexes. These types of compounds could eventually serve as catalysts precursors for the addition of weak nucleophiles to alkynes and nitriles.

The tetradentate ligand 6,6'-diacetyl-2,2'-bipyridine dioxime (L) was obtained in high yield by the condensation reaction of 6,6'-diacetyl-2,2'-bipyridine with hydroxyl amine in alkaline solution. Density functional calculations and 3D modeling of the structure at the B3LYP/6-31G(d) level of theory for L revealed that the energy difference between the global trans conformer (trans-L), which possesses the C_{2h} point group, and the distorted cis conformer (cis-L) was 5.214 kcal/mol. Synthesis and characterization of the metal complexes with Cu(I), Cu(II), Pt(II), and Pd(II) metal ions were reported.
