A confirmatory method has been developed to allow for the analysis of fourteen prohibited medicinal additives in pig and poultry compound feed. These compounds are prohibited for use as feed additives although some are still authorised for use in medicated feed. Feed samples are extracted by acetonitrile with addition of sodium sulphate. The extracts undergo a hexane wash to aid with sample purification. The extracts are then evaporated to dryness and reconstituted in initial mobile phase. The samples undergo an ultracentrifugation step prior to injection onto the LC-MS/MS system and are analysed in a run time of 26 minutes. The …

The vitamin folic acid (folate, FA) has been extensively explored as a targeting ligand to deliver a variety of diagnostic/prognostic/therapeutic agents into various tumors through the assistance of its receptor – the folate receptor (FR). FR is over-expressed in many types of human cancer and can mediate internalization of FA-conjugates through an endocytic pathway. The discovery of small interfering RNA (siRNA), which is cable of inducing potent gene silencing in a sequence-specific manner, provides an excellent molecular tool to suppress aberrant gene expression in malignancies, and therefore siRNA has the potential to revolutionize cancer therapeutics. Solid-state structure, crystalline morphology, crystallization kinetics, thermal, free volume, and gas transport properties of semicrystalline syndiotactic polystyrene (sPS) and ethylene vinyl alcohol copolymers (EVOH) have been investigated. Solid-state structure of sPS after crystallization from the melt and glassy state was examined by differential scanning calorimetry (DSC), density and wide angle x-ray diffraction analysis (WAXD). The measurements confirmed low density of syndiotactic polystyrene crystalline forms, which in the case of α and δe was smaller and in the case of β and γ crystalline forms was slightly larger than the density of the glassy amorphous sPS. Thermodynamic and adsorption properties of protein monolayer electrochemistry (PME) are examined for Pseudomonas aeruginosa azurin (AZ) immobilized at an electrode modified with a networked film of monolayer-protected clusters (MPCs) to assess if nanoparticle films of this nature offer a more homogeneous adsorption interface compared to traditional self-assembled monolayer (SAM) modified electrodes. Specifically, electrochemistry is used to assess properties of surface coverage, formal potential, peak broadening, and electron transfer (ET) kinetics as a function of film thickness. Triacetone triperoxide (TATP) is an organic peroxide that has received widespread attention in recent years. TATP is a ketone peroxide with a high active oxygen content. It is relatively shock sensitive, with explosive decomposition easily initiated, and is therefore considered a primary explosive. However, TATP is also a powerful explosive possessing about 83 % of the power of TNT. TATP can be prepared quickly and easily from inexpensive household chemicals in the absence of any specialized facilities, making it the explosive of choice for modern day terrorists. Cerium oxide (ceria, CeO2-x where x is 0 to 0.5) has been one of the most widely used heterogeneous catalysts particularly in three way catalytic converters. Most of the catalytic traits can be attributed to two properties of ceria: first, the high mobility and storage capacity of oxygen within the lattice; second, the ease with which cerium changes between Ce3+ and Ce4+ states. These properties, combined with the abundance of cerium on earth, make ceria a low-cost highly effective alternative to noble metal catalysts. Part I. The use of chemical inhibitors to manipulate the level of amino acids in cells has proven to be invaluable in the mechanistic study of gene expression in bacteria and fungi. Here we present a new approach to the synthesis of δ‐N‐ (phosphonoacetyl)‐L‐ornithine (PALO), a potent ornithine transcarbamylase inhibitor, using a new amino acid protecting group, 9‐borabicyclononane (9‐BBN). Starting from commercially available reagents and utilizing mild reaction conditions, we were able to form PALO in fewer synthetic steps and in greater yields than previous attempts.

Studies were conducted to investigate the use of spectral reflectance by foliage of common sunflower as a potential indicator of arsenic contamination of soil. Germination method was developed for sunflower seeds, and cohorts of sunflower seedlings in hydroponic tanks were established. The cohorts were exposed to 0 ppm, 5 ppm, 7.5 ppm, and 10 ppm treatments of As (V) and reflectance measurements of foliage were collected using a spectroradiometer during two experiments.Results demonstrated the feasibility of using spectral reflectance by foliage of common sunflower as a potential indicator of arsenic contamination. The purpose of this research is to develop a multiscale understanding of crosslinked amorphous matrices, connecting molecular level events to macroscopic properties. To accomplish this goal, our methodology was to identify network architectures that influence molecular level energy dissipation through mechanisms such as bond rotations, torsions, and ring flips and then relate those molecular motions to macroscopic properties. Studies were accomplished on two aerospace grade matrices: the epoxy, diglycidyl ether of bisphenol A (DGEBA) cured with two amines, 3,3’- diaminodiphenyl sulfone (33DDS) and 4,4’-diaminodiphenyl sulfone (44DDS). Sulfonated poly[styrene-b-ethylene-co-butylene-b-styrene] (sSEBS) block copolymers/inorganic nanocomposite materials were synthesized via in situ formation of inorganic fillers and characterized particularly for their dielectric properties and proton conductivities.

In preparation of sSEBS/SrTiO3 nanocomposites, titanium (IV) isopropoxide [Ti(OPri)4] complex was diffused into sSEBS film, followed by subsequent hydrolysis of [Ti(OPr i)4], diffusion of strontium cations in sSEBS domains, and in situ formation of crystalline SrTiO3. sSEBS with sulfonation degree of 38.1% and 65.0% were employed, and relevant sSEBS/SrTiO3 composites contain SrTiO3 of 10-15 wt%. The ability to rapidly detect and identify hazardous analytes in the field has become increasingly important. One of the most important analytical detection methods in the field is gas chromatography-mass spectrometry (GC-MS). In this work, a hand-portable GC-MS system is described that contains a miniature toroidal ion trap mass analyzer and a low thermal mass GC. The system is self-contained within the dimensions of 47 x 36 x 18 cm and weighs less than 13 kg. A convenient synthesis of sidechain-modified phytosterols is achieved via a temporary masking of the stigmasterol 5,6-alkene as an epoxide. Following performance of the desired modification, the alkene is regenerated through a mild deoxygenation. The approach is applied to the syntheses of β-sitosterol and campesterol acetate, and suggests a facile route to the (Z)-isomers of Δ22–23 phytosterols.

This dissertation explores the use of UV-Visible spectroscopy and Time Resolved Laser Induced Fluorescence spectroscopy as near real time process monitors of uranium and plutonium concentrations in aqueous reprocessing trains. The molar absorptivities and linear ranges of these metals were investigated under total nitrate and acid concentrations similar to those found in current reprocessing systems. Concurrent to this, a new multiple wavelength monitor was derived that is capable of determining the total nitrate concentration spectroscopically. Highly reactive radical species such as hydroxyl radical cause oxidative stress, resulting in chronic or degenerative diseases. In the biological Fenton reaction, iron(II) generates hydroxyl radical, OH, but only within a specific electrochemical potential window (-324 mV to 640 mV). Selenium and sulfur compounds have been extensively studied for their antioxidant properties, and they may exert their effects by binding iron(II) and shifting its electrochemical potential out of this window. Neomycin is an aminoglycoside antibiotic known for its high affinity for RNA duplex structures. Recent developments from our labs have indicated that aminoglycoside binding is not limited to RNA, but to nucleic acids that, like RNA, adopt conformations similar to A-form. Our group sought to further expand the utility of aminoglycoside binding to B-DNA structures by conjugating neomycin, an aminoglycoside antibiotic, with the B-DNA minor groove binding ligand Hoechst 33258. Envisioning a dual groove-binding mode, we have extended the potential recognition process to include a third, intercalative moiety. There have been rapid advances in the development and applications of semiconductor quantum dots (QDs) represented by CdSe/ZnS. However, a serious limitation of these QDs is the necessary use of toxic heavy metals. Fluorescent 'carbon dots' (surface passivated carbon nanoparticles) are developed as alternative to classical semiconductor QDs.
The carbon dots could be made to be highly fluorescence, with emission quantum yields close to 60%. Their optical properties resemble bandgap transitions found in nanoscale semiconductors, suggesting that carbon particles at the nanoscale acquire essentially semiconductor-like characteristics.

The preparation and electrochemical properties of donor - acceptor systems based on trimetallic nitride endohedral metallofullerenes (TNT-EMFs) for enhanced solar energy conversion is described. It was found that TNT-EMFs have an enhanced ability to stabilize charge separated states formed upon light irradiation when compared to empty cage fullerene analogues. The influence of the linker connecting the donor and acceptor moieties was studied in two different systems. Optimal conditions for preparing cyclopropanated Bingel-Hirsch derivaties of the most common TNT-EMFs were found; this functionalization approach is a very useful alternative to the 1,3-dipolar cycloaddition reactions that yield pyrrolidine derivatives, because the latter …

Microfluidic systems are of great interest in analytical chemistry for the development of lab-on-a-chip devices. This dissertation describes fundamental investigations of magnetohydrodynamics with added redox species (redox-MHD) as a new microfluidic approach for use in analytical applications. Redox-MHD is attractive because it offers a unique combination of desirable features that are otherwise only available separately with individual micropumps. Redox-MHD is compatible with aqueous and nonaqueous solutions, does not require moving parts, easily reverses flow direction, and requires only low voltages. The flame retardancy and thermal stability of ammonium polyphosphate/tripentaerythritol (APP/TPE) intumescent flame retarded polystyrene composites (PS/IFR) combined with organically-modified layered inorganic materials (montmorillonite clay and zirconium phosphate), nanofiber (multiwall carbon nanotubs), nanoparticle (Fe2O3) and nickel catalyst were evaluated by cone calorimetry, microscale combustion calorimetry (MCC) and thermogravimetric analysis (TGA). Cone calorimetry revealed that a small substitution of IFR by most of these fillers (≤2%) imparted substantial improvement in flammability performance. The necessity of preparing single enantiomer drugs in an optically pure form has triggered much research, especially in the pharmaceutical industry. On-board hydrogen storage presents a challenging barrier to the use of hydrogen as an energy source because the performance of current storage materials falls short of platform requirements. Because boron is one of the lightest chemical elements that can form strong covalent bonds with hydrogen, it provides an excellent opportunity to design new lightweight materials on the basis of novel boron hydride building blocks. Realizing this potential requires an understanding of the electronic structure, chemical bonding, and stability of neutral and anionic BxHy clusters with variable stoichiometry. In 1977, Shirakawa and coworkers discovered the first conductive polymers. Their conductive polymers have been extensively studied. Though some of these first conductive polymers such as polyactelyne seemed to be a promising organic conductor, the commercial applications of these polymers were severely limited due the polymer’s highly air-sensitive nature. Thus, the current focus has shifted towards the development of heterocyclic aromatic polymers which are air-stable and have a low band gap. Nuclear Magnetic Resonance Affinity Screening Methods For Functional Annotation Of Proteins And Drug Discovery, Matthew D. Shortridge

With nearly 1,350 complete genome sequences available our understanding of biology at the molecular level has never been more complete. A consequence of these sequencing projects was the discovery of large functionally unannotated segments of each genome. The genes (and proteins they encode) found in these unannotated regions are considered “hypothetical proteins”. Current estimates suggest between 12%-50% of the known gene sequences are functionally unannotated. Incomplete functional annotation of the various genomes significantly limits our understanding of biology. Pragmatically, identifying the functions of these proteins could lead to new therapeutics; making functional annotation of paramount importance.

he effect of calcined LDH-MgAl-CO3 on rebar corrosion behavior in chloride ion contaminated neutral concrete pore solutions was investigated in this paper.Corrosion potentials of rebars in pore solutions after treatment with CLDH were higher than before treatment.According to polarization tests,passivated region of rebars in pore solutions after treatment with CLDH was found apparently instead of activation.EIS tests showed that CLDH made the impedance of specimens increase.The measurements of pH and chloride ion concentration after treatment indicated that / of pore solutions decreased by 4~5 magnitude orders.
