We show that a quantum-mechanics/molecular-mechanics strategy based on ab initio (i.e., first principle) multiconfigurational perturbation theory can reproduce the spectral properties of a tryptophan residue embedded in the contrasting hydrophobic and hydrophilic environments of parvalbumin and monellin, respectively. We show that the observed absorption and emission energies can be reproduced with a less than 3 kcal mol(-1) error. The analysis of the computed emission energies based on a protein disassembly scheme and protein electrostatic potential mapping allows for a detailed understanding of the factors modulating the tryptophan emission. Reactor R&D: Synthesis And Optimization Of Metallic Nitride Fullerenes And The Introduction Of Two New Classes Of Endohedral Metallofullerenes, Metallic Nitride Azafullerenes And Oxo-Metallic Fullerenes, Curtis Earl Coumbe

Metallic nitride fullerenes (MNFs) were discovered in 1999. This class of endohedral fullerenes show promise in a new diverse range of useful applications. Since then, focus has shifted to the selective synthesis of these molecules with yields that would accommodate adequate sample distribution. Using the electric arc method, the traditional yield of these molecules has been very low (i.e. < 5 mg), and only a small percentage of the fullerene products (i.e. < 5%). This dissertation introduces the novel CAPTEAR (Chemically Adjusting Plasma Temperature, Energy, And Reactivity) method that allows the targeted synthesis of MNFs in high purity and yield. This method utilizes a nontraditional oxidizing method for fullerene synthesis that has not only provided optimization of MNFs, but also resulted in the discovery of two new classes of fullerenes: metallic nitride azafullerenes (MNAFs) and oxo-metallic fullerenes (OMFs). Evidence suggests that the nitrogen of the MNAF cage provides stability for the trimetallic nitride clusters, while the OMFs are the first fullerenes to encapsulate oxygen and incorporate a seven atom cluster inside a Cgo cage.
Other efforts to increase yields resulted from scaling up production of fullerenes by using larger quantities of starting materials. These larger quantities required energy (electrical current) beyond the capacity of the traditional electric arc generator. Therefore, a new electric arc generator …

Thermal frontal polymerization is a process that involves a propagating front travelling through a monomer/initiator solution and converting monomer into polymer. The effects of different reactive and non-reactive additives on front temperature, front velocity, and pot life were studied in a thermal frontal polymerization system of multifunctional acrylates. One issue with thermal frontal polymerization of acrylate monomers is the amount of smoke and fumes produced due to high front temperatures. The effect of thermally-expandable microspheres was studied utilizing a variety of monomers. This study was designed to achieve better understanding of (1) how carboxysome genes are regulated and expressed to yield with precise relative ratios and (2) the in vivo roles of two sets of conserved bacterial microcompartment genes, namely the three csoSl and two csoS4 genes of H. neapolitanus, in the biogenesis and function of the carboxysome. For the first goal, a detailed transcriptional profile of carboxysomal genes in H. neapolitanus was established using absolute quantification real-time RT-PCR and transcript ends analysis. Concept mapping was used with Donna High School students to determine its effect on the Texas Assessment of Knowledge and Skills (TAKS) test. Concept mapping was used with models such as the Sheltered Instruction Observation Protocol model (SIOP), the 5- E model, and the Learning Groups model. This measure was taken because the models used have not helped students improve their TAKS scores significantly. The findings of this research reveals that using the process of concept mapping together with the SIOP model helps students clearly see related concepts and motivates them to think more deeply. The synthesis of disubstituted and homologated unnatural amino acids and the development of a mass spectrometry based assay for enantioselectivity are presented here. Disubstituted and homologated unnatural amino acids have proven to be effective for the treatment of diseases and for use as imaging agents. Peptides that contain unnatural amino acids have also proven to be effective treatments for various disease states including cancer. The potential to evaluate such unnatural amino acids for pharmaceutical properties is limited by the available syntheses. Molecular modeling is a powerful tool to better understand the intermolecular interactions of carbon nanostructures. It provides structures and energies not easily obtainable from experiments and predicts properties that can be tested experimentally. Intermolecular interactions play an important role in the aggregation of various carbon nanomaterials. Three molecular modeling studies of carbon nanomaterial dispersions are presented in this dissertation, with an emphasis on illustrating how effective these theoretical techniques are in providing insight on the selection of dispersion additives. While characterizing various splice forms of p120 catenin, we observed what appeared to be a novel posttranslational modification of p120, resulting in a higher molecular weight form that was dependent on the splicing pattern. Further investigation revealed the higher molecular weight form to be a fusion protein between sequences encoded by the retroviral vector and p120. We found that the publicly available sequence of the vector we used does not agree with the experimental sequence. We caution other investigators to be aware of this potential artifact.

This thesis concerns characterization of synthetic manganese oxides belonging to mineral phases known as Birnessite and Cryptomelane. Presented here are the results of an experiment designed to examine the influence of sodium, potassium, chloride, and sulfate ions on the reduction of potassium permanganate under acidic conditions at room temperature to produce Birnessite and Cryptomelane. The experiments used KMnO4 as the source of Mn and the resulting Birnessite and Cryptomelane precipitates were washed with 18 Mn/cm NANOpure water at the end of syntheses. Silver nanoparticles exhibit size, shape and the surrounding dielectric medium dependent optical properties (scattering and absorption) in the visible spectral region. These optical properties can be explained in terms of the collective oscillation of the 'free conduction band electrons' or 'surface plasmons' induced by an external electromagnetic field. Dipole and Quadrupole Plasmon resonances have been observed for silver particles less than 100nm in diameter prepared using various synthetic techniques. Mesoporous carbon materials of carbon xerogel (CX) and silica-templated carbon (MC) were synthesized and explored as catalyst supports alternative to the most-commonly-used carbon black (CB) support, for polymer-electrolyte-membrane fuel cell (PEMFC) application. Pt catalyst was loaded on these carbons and electrodes were fabricated from them. These Pt-loaded carbon supports were characterized with XRD, TEM, ex-situ and in-situ cyclic voltammetry, etc. The fabricated electrodes were evaluated in single-cell testing in comparison with commercial CB-supported Pt catalyst fabricated electrodes. Developing chemical bonding models in clusters is one of the most challenging tasks of modern theoretical chemistry. There are two reasons for this. The first one is that clusters are relatively new objects in chemistry and have been extensively studied since the middle of the 20th century. The second reason is that clusters require high-level quantum-chemical calculations; while for many classical molecules their geometry and properties can be reasonably predicted by simpler methods.

Background Spinal Muscular Atrophy (SMA) is the leading genetic cause of infantile death. It is caused by the loss of functional Survival Motor Neuron 1 (SMN1). There is a nearly identical copy gene, SMN2, but it is unable to rescue from disease due to an alternative splicing event that excises a necessary exon (exon 7) from the majority of SMN2-derived transcripts. While SMNΔ7 protein has severely reduced functionality, the exon 7 sequences may not be specifically required for all activities. The Pt nanoflower/ZnO(PtNF/ZnO) nanorod composites arrays were electrochemically prepared by controlling the hydrophobicity and the hydrophilicity of ZnO nanorods arrays.The Pt nanoflowers grew on the tip of ZnO nanorods,which maintain the vertical array of Zno nanorods with a high degree of size uniformity and high surface area.Compared with Pt nanoparticle covered ZnO nanorod arrays,which were prepared by controlling the hydrophilicities of ZnO nanorods and Pt nanoparticles,the PtNF/ZnO displayed highest electrocatalytic activities for the oxidation of methanol.

The cyclic voltammgram of Mb/AAO/Au Colloid/GCE shows a pair of well-defined and nearly reversible peaks at about-0.20 V(vs.Ag/AgCl) in 0.1 mol/L NaAc-HAc buffer(pH = 5.4),being the characteristic peaks of the Mb heme Fe(Ⅲ) / Fe(Ⅱ)redox couples.The electron transfer between Mb and the electrode was greatly enhanced under present testing conditions.The effects of scan rate,pH and buffers concentration on the electrochemical behaviors of Mb were studied in detail.Position of the Soret absorbance band suggested that Mb in chitosan-colloidal gold hept its secondary structure similar to the native states.Oxygen was catalytically reduced by Mb in chitosan-colloidal gold films.

Self-organized anodic anatase TiO2 nanowire arrays doped with nitrogen have been successfully fabri-cated and their photoelectrochemical(PEC) properties have been characterized and found to be substantially im-proved compared to undoped nanowires or commercial P25 nanoparticles.Photocurrent measured with monochro-matic incident light showed that the incident photon-to-current efficiency(IPCE,%) values of nanowire arrayelectrodes with or without N-doping were obviously higher than that of commercial P25 nanoparticle electrodes,and nitrogen-doped TiO2 nanowire arrays(NTNA) had noticeable absorption in the visible region.The NTNAelectrodes showed the highest photocurrent density and power conversion efficiency under 100 mW/cm2 visiblelight illumination.A maximumolphotoconversion efficiency of 0.52% was achieved for the …

In this work,the electrocatalytic activities of MmNi3.2Al0.2Mn0.6Co1.0, NaOH treated MmNi3.2Al0.2Mn0.6Co1.0 and Au-MmNi3.2Al0.2Mn0.6Co1.0 obtained after NaOH treatment(Mm: misch metal) for NaBH4 oxidation were reported. The effects of dihexadecyl phosphate(DHP) and multi-walled carbon nanotubes(MWNTs) on the oxidation of guanine mediated by[Ru(bpy)3]2+ on the ITO electrode have been investigated by cyclic voltammetry and differential pulse voltammetry.The results showed that [Ru(bpy)3]2+ can mediate the oxidation.In the range from 0.01 to 0.15 mmol·L-1,peak currents for guanine oxidation mediated by [Ru(bpy)3]2+ increased with the rise of DHP concentrations.On the contrary,cationic surfactant HTAC has the ability to weaken the mediated oxidation of guanine.In addition,the oxidative mechanism of guanine mediated by [Ru(bpy)3]2+ with the participation of DHP …

The Ni-Pd alloy catalysts with Pd-enriched surface were synthesized through a modified polyol process.The synthesized materials were investigated by XRD and electrochemical methods,and tested as electrocatalysts for CH3OH-tolerant oxygen reduction.The results indicated that the synthesized Ni-Pd nanoparticles possess multiple phases containing Pd-Ni alloy and pure Ni.Comparing with Pd/C,the prepared Ni-Pd/C nanoparticles exhibited better resolved voltammetric peaks between the hydrogen absorption and adsorption,and positively shifted current peaks for the reduction of Pd oxide.The prepared Ni-Pd alloys with Pd-enriched surface have significantly enhanced ORR activity and improved CH3OH tolerance as comparing with Pd/C catalysts.

A simple procedure involved the complexing of PdCl2 with sodium citrate followed by ethylene glycol reduction has been employed for the preparation of carbon-supported Pd nanoparticles(Pd/C).The XRD and TEM characterizations indicate that Pd nanoparticles with a small particle size were well-dispersed on carbon support.The mean particle sizes of the Pd/C catalysts were found to be increased from 2.7 to 5.8 nm with heat-treatments at different temperature.The mass activity of the Pd/C catalyst for formic acid electrooxidation increased with the decrease in Pd particle size.However,the specific activity increased with Pd particle size.Furthermore,the Pd/C catalyst with a particle size of ca.3.6 …

A Pd-Ni alloy film was electrodeposited on Au disc electrode at a constant potential from a weakly basic bath containing PdCl2,NiSO4 and ethylenediamine.The as-deposited film was then subjected to potential cycling in a perchloric acid solution for surface dealloying treatment.The dealloyed Pd-Ni film electrode was examined for the electrocatalytic oxidation of formic acid,and compared with the as-deposited one as well as a Pd film electrode.The FE-SEM and electrochemical measurements on the Pd-Ni films revealed that the dealloying treatment caused a surface roughening and an increase in specific surface reaction area.The ICP-AES and XPS analyses demonstrated that the …
