A peptide-based electron-transfer system has been designed in which the specific positions of redox-active metal complexes appended to either an a-helix, or an a-helical coiled-coil, can be reversed to test the effect of the helix dipole in controlling photoinduced electron-transfer rates. Two 30-residue apopeptides were prepared having the following sequences: (1) Ac-K-(IEALEGK)(ICALEGK)(IEALEHK)-(IEALEGK)-G-amide, and (II) Ac-K-(IEALEGK)(IHALEGK)-(IEALECK)(IEALEGK)-G-amide. Each apopeptide was reacted first with [Ru(bpy)(2)(phen-ClAc)](2+), where bpy = 2,2'-bipyridine and phen-ClAc = 5-chloroacetamido-1,10-phenanthroline, to attach the ruthenium polypyridyl center to the cysteine side-chain of the polypeptide. The isolated products were then reacted with [Ru(NH3)(5)(H2O)](2+) to yield the binuclear electron-transfer metallopeptides ET-I and ET-II. …

Two species of shark found in the coastal waters Karachi (Pakistan) were studied, Eusphyra blochii (winghead snark) and Carcharhinus bleekeri (sandbar shark) for their liver oil fatty acid composition. Since liver has high lipid content and traditionally the liver oil of these species been used to relieve muscular pain and arthritis in Pakistan this study was conducted. The isolation, identification and characterization of these fatty acids were carried out by gas liquid chromatography (GLC) and a combination of the thin layer chromatography (TLC)-GLC technique. The viscosity measurements of micelles composed of mixtures of anionic/nonionic surfactants were carried out as a function of the mole fraction of surfactants. The systems studied were sodium dodecyl sulfate (SDS)-polyoxyethylene lauryl ether (C_{12}POE_{n} n = 4, 10 and 23) in water and 0.1 M sodium chloride solution. In the single systems, the relative viscosity of nonionic surfactant is greater than that of SDS, and increases with the increasing level of ethylene oxide. In the mixed surfactant systems, the relative viscosities vary at the mixed molar fraction of SDS, between 0.2 and 0.3, and then the relative viscosities of mixed …

Adsorbent powders were prepared from the uncatalysed hydrolysis-condensation reactions of different complexes that were synthesized by the reactions of Zr(OPr^{n})_{4} with different chelating ligands (such as allylacetic acid [AAH], vinylacetic acid [VAH] and citraconic acid [CAH]). The complex products were characterized by FT-IR, ^{1}H-NMR and ^{13}C-NMR spectra. The maximum combination ratios of AAH/Zr(OPr^{n})_{4}, VAH/Zr(OPr^{n})_{4} and CAH/Zr(OPr^{n})_{4} were 1.7, 0.9 and 1.3, respectively. The hydrolysis-condensation products of these complexes were calcinated at 900 °C and half of them were coated with 3-glycidyloxypropyltrimethoxysilane (GLYMO). A number of regulatory mechanisms are involved in increasing citric acid production under certain conditions. The accumulation of citric acid takes place under the high concentration of carbon sources. The effect of sucrose as a carbon source on the accumulation of citric acid and tyrosine phosphorylated proteins during high glycolytic conditions was investigated using Aspergillus niger ATCC 11414. When mycelia are treated by different sucrose concentrations, a protein of \sim 27 kDa becomes tyrosine phosphorylated. This tyrosine phosphorylation takes place in response to signal transduction from the extracellular milieu. A possible role of this \sim 27 kDa protein in signal …

The In Vitro Effects Of Dexamethasone On Sheep Lens Glucose-6-Phosphate Dehydrogenase, Şükrü Beydemi̇r, Destan Ni̇l Kulaçoğlu, Mehmet Çi̇ftçi̇, Ömer İrfan Küfrevi̇oğlu

Glucose-6-phosphate dehydrogenase (G6PD) enzyme was purified from sheep lenses, and the in vitro effects of dexamethasone on the enzyme's activity were investigated. Sheep lens glucose-6-phosphate dehydrogenase was purified 10,000-fold by using ammonium sulphate fractionation and 2',5'-ADP-Sepharose 4B affinity gel chromatography for a yield of 83.8\% and a specific activity of 7.6 EU/mg protein. Enzyme activity was determined by Beutler's method. Dexamethasone strongly inhibited the enzyme under in vitro conditions. The I_{50} value of the dexamethasone was 3.05 mM.

Silver Removal From Aqueous Solution By Adsorption On Concrete Particles, Shakila Begum

The ability of concrete to remove silver from its aqueous solutions was studied. Silver removal is favored by low concentration and high pH at room temperature. The intrinsic acidity constants, average pK_{a1}^{int} and average pK_{a2}^{int} were determined. The calculated pH_{PZC} (5.4) is in agreement with the pH_{PZC} values determined from zeta potential measurements, indicating that the concrete particulate behaves amphoterically. The total acidity capacity for the negative and positive surface was 0.5 \muC/cm^2. A surface complex formation model (SCFM) was used to describe the adsorption of silver onto concrete particles. Hypothalamic proopiomelanocortin (POMC) neurons are critical for controlling homeostatic functions in the mammal. We used a transgenic mouse model in which the POMC neurons were labeled with enhanced green fluorescent protein to perform visualized, whole-cell patch recordings from prepubertal female hypothalamic slices. The mouse POMC-enhanced green fluorescent protein neurons expressed the same endogenous conductances (a transient outward K current and a hyperpolarization-activated, cation current) that have been described for guinea pig POMC neurons. In addition, the selective -opioid receptor agonist DAMGO induced an outward current (maximum of 12.8 1.2 pA), which reversed at K equilibrium potential (EK), in the majority …

C75, a recently derived compound that potently suppresses feeding and induces weight loss, has been proposed to act mainly by inhibiting fatty acid synthase (FAS) in central neurons that control feeding. For example, normal, fasting- associated, hypothalamic increases in neuropeptide Y (NPY)/Agouti-related protein (AGRP) expression and decreases in proopiomelanocortin (POMC)/cocaine and amphetamine regulated transcript (CART) expression were reported to be blocked by C75. All three title compounds, prepared from bis(3,5-dimethyl-2- nitrophenyl)ditellurium, exhibit high degrees of intramolecular TeÐO coordination. Their TeÐO distances increase in the order C8H8BrNOTe < C8H8BrNO2Te < C8H8Br3NO2Te, with distances of 2.165 (3), 2.306 (1) and 2.423 (6) AÊ , respectively, indicating that C8H8BrNOTe may be more aptly described as 1-bromo-4,6-dimethyl-2,1,3-benzoxatellurazole.

A Solvent-Free Claisen Condensation Reaction For The Organic Laboratory, John J. Esteb, Matthew B. Stockton

An experiment involving the Claisen condensation reaction for a first-year organic chemistry laboratory is presented. Claisen condensations are routinely covered in organic textbooks but owing to the long reaction times required to reach equilibrium in solution they are seldom explored in the undergraduate teaching laboratory. In this experiment, potassium tert-butoxide and ethyl phenylacetate are heated to 100 °C for 30 minutes under solvent-free conditions to produce 2,4-diphenyl acetoacetate in 80% yield. The solvent-free nature of this procedure greatly reduces the quantity of waste generated by students relative to typical carbonyl condensation experiments.

Diepoxybutane, diepoxyoctane, and mechlorethamine are cytotoxic DNA crosslinking agents that vary in their carcinogenic versus chemotherapeutic potentials. Interstrand cross-linking occurs between the N7 positions of deoxyguanosine residues on opposite strands of the DNA duplex. Each synthetic DNA oligomer used in this study contained four 5'-N\GN2CN3 sites (within a 32 base sequence) and we have systematically varied the bases in the N1 and N2 positions to determine the resulting cross-linking efficiencies of each cytotoxic agent. Each duplex was 5' -end labeled and incubated with cross-linking agent. Interstrand cross-links were purified through denaturing polyacrylamide gel electrophoresis and then subjected to piperidine cleavage. …

Purpose: Since preclinical studies have shown more than additive cytotoxicity and DNA damage with the combination of gemcitabine and 5-fluoro-2′-deoxyuridine (FUDR), we studied this combination in a phase I trial. Methods: Gemcitabine alone was given in cycle 1 as a 24-h, 2-h or 1-h i.v. infusion weekly for 3 of 4 weeks; if tolerated, a 24-h i.v. infusion of FUDR was added with oral leucovorin. The cycle was aborted for grade 3 thrombocytopenia, grade 4 neutropenia, and grade 2 or worse nonhematologic toxicity. The novel three legged piano stool chelating carbene complex, Cp'(CO)Mn{C(OEt)CH2PPh2}, 1, exhibits interesting structural, spectroscopic and electrochemical properties. It also readily undergoes reaction with CO to produce the three legged piano stool complex Cp'(CO)2{PPh2C(OEt)=CH2}, 2. This is in contrast to the analogous non-chelating complex Cp(CO)(PPh3)Mn{C(OMe)CH2CH3}, 3, which does not react with CO. This paper discusses the results of Fenske-Hall approximate molecular orbital calculations on model complexes for 1 and 3. The differences in spectroscopic and electrochemical properties are explained using molecular orbital analysis. Possible reasons for the enhanced reactivity of 1are also presented.

Phycotoxins are potentially toxic substances, which are produced by marine phytoplankton and which may accumulate in shellfish or finfish. Students learn more if they are actively involved in the learning process, particularly in a cooperative manner. Several UMR faculty have operated course-based learning centers (LCs) as part of the campus-wide Learning Enhancement Across Disciplines (LEAD) Program of student learning assistance and enhancement. LCs are designed to assist large numbers of students in a cost- and time-efficient manner that promotes student engagement without requiring undue amounts of faculty time. Thin polymer films are used in applications such as electronics, biomedical devices and coatings. As the trend to make the devices smaller continues, a film's influence on a material's overall properties becomes more prominent. Previous studies have shown that thin polymer films have characteristics slightly different from those of bulk polymers. Film behavior has been characterized by examining thermal expansion, diffusion, conformation, density and glass transitions. The dynamics of glassy polystyrene has been investigated using a wide variety of theoretical and experimental methods. A great amount of work has been done on the configuration of surface adsorbed poly(methyl methacrylate) (PMMA), especially on surfaces to which they have a strong attraction. Some studies have been make on the relationships between configuration and tacticity, molecular weight, adsorbed amount, and density of surface hydroxyl groups. The effect of tacticity on the conformation of alumina-adsorbed PMMA in solution and dry spin-coated films were studied by Grohen et al. Their results showed that surface-adsorbed PMMA chains adopted a relatively flat configuration, but iso-PMMA (isotactic) adsorbed on aluminum, with a much larger fraction of the bound carbonyl groups, had …

The recent popularity of methods for solid-phase peptide synthesis that use the 9-fluorenylmethyloxycarbonyl (Fmoc) group for Nα-amino protection has created a need for compatible anchoring linkages and handles. In an effort to develop mild new methods for use in solid-phase peptide synthesis (SPPS), a new ferrocene containing linker or “handle”, the 1’1-ferrocenyl carboxaldehyde handle was designed, synthesized, characterized and tested. This linker is analogous to those commercially available and developed by Barany. The ferrocenyl amine linker(FAL) releases C-terminal peptide amides upon acidolysis. We report on argon excimer emission from high-pressure microdischarges formed inside metal capillaries with or without gas flow. Excimer emission intensity from a single tube increases linearly with gas pressure between 400 and 1000 Torr. Higher discharge current also results in initial intensity gains until gas heating causes saturation or intensity drop. Argon flow through the discharge intensifies emission perhaps by gas cooling. Emission intensity was found to be additive in prealigned dual microdischarges, suggesting that an array of microdischarges could produce a high-intensity excimer source.

This thesis reports the first complete synthesis of monriporynes A and B. isomeric diacetylenes that bear α, β unsaturated ketone functionalities. Recently isolated from the stony coral Montipora sp. these compounds are reported to possess in vitro cytotoxic activity against several human solid tumor cells. We have synthesized both isomers in three simple steps. In the first step, l-iodononyne was prepared by reacting 1-nonyne with N-iodosuccinimide and a catalytic amount of silver nitrate. In industrial polymer and synthetic rubber production facilities, workers are exposed to 1,3-butadiene. This compound is converted in vivo to 1,2,3,4-diepoxybutane (DEB) and has been linked to increased incidences of cancer in these individuals. Carcinogenesis has been attributed to formation of DEB induced DNA interstrand cross-links. Previous studies have demonstrated that DEB cross-links deoxyguanosine residues within 5'-GNC sequences in synthetic DNA, in restriction fragments, and in defined sequence nucleosomes. The Trout Valley Formation of Emsian-Eifelian age, outcropped in Baxter State Park, Maine, consists offluvial and coastal deposits preserving early land plants. Massive, crudely bedded conglomerate represents deposits of proximal braided channels on an alluvial fan complex. Lithic sandstone bodies in channel-form geometries represent deposits of river channels draining the Acadian highlands whereas associated siltstones represent overbank deposits, intertidal flats, and tidal channels. Localized lenticular quartz arenites represent nearshore shelf bar deposits that were storm influenced. The majority of plant assemblages preserved mainly in siltstone lithologies are allochthonous and parautochthonous, with only one autochthonous assemblage identified in the sequence. Plant …

In 1952, Dwyer and coworkers began testing a series of metal complexes for potential inhibition of cancer cell proliferation in animals.[l] The complexes tested were unsuitable for such studies due to their high toxicity. Therefore, no further work was done on the project. However, in 1965, Rosenberg and coworkers revisited the possibility of potential metal-based drugs. Serendipitously, they discovered that cis-diamminedichloroplatinum(lI) (cisplatin) inhibits cell division in E. coli.[2] Further studies of this and other platinum compounds revealed inhibition of tumor cell lines sarcoma 180 and leukemia LI2l0 in mice.[l] Cisplatin was approved by the Food and Drug Administration in 1970 …

Degradation of chlorobenzene, dichlorobenzenes and chloronitrobenzenes (ClB, x-Cl2B and x-ClNO2B) was tested by trying to make them react with metallic iron, Fe (0), in water solvent.
The experiments consisted of optimizing the parameters of the reactions (like minimum mass of iron needed) and analytical detection conditions. Analytical techniques used were gas and liquid chromatography (GC and HPLC), mass spectrometry (MS) and UV-Vis spectroscopy.
No substances other than the initial ClB and x-Cl2B were detected in their tests with Fe (0) after shaking for periods of up to three days, although 10%weight average decreases in concentration were observed.

TiO2-coated conducting glass electrodes can be modified by attachment of redox-active molecules to the TiO2 surface. This thesis describes the synthesis and characterization of a polypyridyl Ru(II) complex that can be covalently bound to a TiO2 surface through the carboxylate groups of 2,2'-bipyridine-4,4' -dicarboxylic acid, H2dcbpy. In addition, the bound complex contains a redox-active alloxazine derivative, pptd, which is capable of 2e-/2H+ transfers. Di(p-tol)phosphido-bridged complexes, (OC)4M[μ-P(p-tol)2]2M(CO)4 (M = Mo, W), have been synthesized for the first time. These have been obtained from the reaction of (OC)5MP(p-tol)2H with BuLi, followed by air oxidation. The choice of solvent is very important as the intermediate anion, [(OC)5MP(p-tol)2] is unstable in THF. An ideal solvent for these preparations is CH3OCH2CH2OCH3 (monoglyme) from which yields of 68% and 76% were realized for the tungsten and molybdenum complexes, respectively. …

3-Phenylbenzofuranone, when deprotonated with sodium hydride, forms an extended enolate system which, when treated with excess alkyl chloroformates, affords only oxygen-acylated products, as opposed to the usually desired carbon-functionalized derivatives. It was discovered that such molecules, when treated with a catalytic amount of 4- (N, N-dimethylamino)pyridine (DMAP), rearrange quantitatively to the carbon acylated isomers.
These migrations, which are accompanied by a deep blue color, are proposed to involve a reactive intermediate. This intermediate, which is postulated as an ion pair charge transfer (IPCT) complex, has been closely studied in order to gain a better understanding of the …
