In industrial polymer and synthetic rubber production facilities, workers are exposed to 1,3-butadiene. This compound is converted in vivo to 1,2,3,4-diepoxybutane (DEB) and has been linked to increased incidences of cancer in these individuals. Carcinogenesis has been attributed to formation of DEB induced DNA interstrand cross-links. Previous studies have demonstrated that DEB cross-links deoxyguanosine residues within 5'-GNC sequences in synthetic DNA, in restriction fragments, and in defined sequence nucleosomes. The current study utilized the polymerase chain reaction (PCR) to examine DEB damage frequencies within nuclear genes, found within "open" regions of chromatin, as compared to regions of unexpressed sequence that …

This thesis reports the first complete synthesis of monriporynes A and B. isomeric diacetylenes that bear α, β unsaturated ketone functionalities. Recently isolated from the stony coral Montipora sp. these compounds are reported to possess in vitro cytotoxic activity against several human solid tumor cells. We have synthesized both isomers in three simple steps. In the first step, l-iodononyne was prepared by reacting 1-nonyne with N-iodosuccinimide and a catalytic amount of silver nitrate. This iodoalkyne was then coupled with propargyl alcohol using copper iodide as the catalyst to give 2,4dodecadiynyl alcohol. Finally, the diacetylenic alcohol was subjected to the Swern …

In 1952, Dwyer and coworkers began testing a series of metal complexes for potential inhibition of cancer cell proliferation in animals.[l] The complexes tested were unsuitable for such studies due to their high toxicity. Therefore, no further work was done on the project. However, in 1965, Rosenberg and coworkers revisited the possibility of potential metal-based drugs. Serendipitously, they discovered that cis-diamminedichloroplatinum(lI) (cisplatin) inhibits cell division in E. coli.[2] Further studies of this and other platinum compounds revealed inhibition of tumor cell lines sarcoma 180 and leukemia LI2l0 in mice.[l] Cisplatin was approved by the Food and Drug Administration in 1970 …

The Trout Valley Formation of Emsian-Eifelian age, outcropped in Baxter State Park, Maine, consists offluvial and coastal deposits preserving early land plants. Massive, crudely bedded conglomerate represents deposits of proximal braided channels on an alluvial fan complex. Lithic sandstone bodies in channel-form geometries represent deposits of river channels draining the Acadian highlands whereas associated siltstones represent overbank deposits, intertidal flats, and tidal channels. Localized lenticular quartz arenites represent nearshore shelf bar deposits that were storm influenced. The majority of plant assemblages preserved mainly in siltstone lithologies are allochthonous and parautochthonous, with only one autochthonous assemblage identified in the sequence. Plant …

Alkene (6) was produced in three steps from 3-hydroxybenzaldehyde (8), and 3-bromobenzyl bromide (10). A variety of strategies were employed to synthesize the desired boronic acid (13), but as of this time none have been successful. The synthesis of this boronic acid is necessary to study the feasibility of applying the Chan-Evans copper mediated formation of diaryl ethers to a bis(bibenzyl) synthesis. The synthesis of both cis and trans-RuCl?(DMSO)? has been reported in literature, and is currently the only way of acquiring these compounds. Because these metallodrugs have shown similar antitumor properties to cisplatin, they are currently under investigation for use in clinical trials as potential chemotherapeutics. It is believed that these drugs act in a way similar to cisplatin by causing intrastrand crosslinking of adjacent guanine residues in DNA. In an attempt to better understand any Ru/DNA interaction for these compounds, both cis and trans-RuCl?(DMSO)? were synthesized with reasonable yields (~40%). HMG1 is a nonhistone chromosomal protein that binds preferentially to some types of globally modified DNA. HMG1 has been implicated in the cellular response to the anti-cancer agent cisplatin, functioning to block excision repair of specific distorted platinated DNA lesions and resulting in enhanced cytotoxicity. We have investigated whether other DNA-binding agents also produce lesions that are recognized by HMG1. Specifically, we used polyacrylamide gel shift assays to monitor the potential HMG1 binding of diepoxybutane interstrand cross-links, which have been suggested to induce DNA bending. 6-Thioguanine (65G) has been used for the past thirty years as an anti-cancer agent in maintenance chemotherapy for childhood acute lymphoblastic leukemia as well as acute myeloid leukemia. Despite its long-standing clinical use, little is understood of the molecular basis of the cytotoxicity associated with ?sG. Research has shown that ?sG is incorporated into DNA during replication after which it may exert its cytotoxicity by inducing rapid mutation or by affecting DNA-protein interactions. 65G has also been shown to impair HIV replication by inhibiting HlV-l reverse transcriptase and to inhibit human telomerase activity. Efforts have been made at generating the parent α-acetoxycarbene from a non-nitrogenous precursor. Baeyer-Villiger oxidation of the ketone, 1-(1a,9b-dihydro-lHcyclopropa[1] phenanthren-1-yl)-ethanone, available in two steps from phenanthrene, led to the ester, 1-(1 a,9b-dihydro-lH-cyclopropa[/]phenanthren-l-yl)-acetate, which is a photochemical precursor of the parent α-acetoxycarbene intermediate. Photolysis of the ester in cyclohexene afforded an isomeric mixture of endo and exo acetates resulting from the carbene adding to the α-bond. Photolysis of the ester in trans-3-hexene yielded a single trans-cis acetate, a product of the same cycloaddition process. The formation of methyl glyoxal, via the l,2-acyl shift of the carbene, is observed through its derivatives. …

Two macrocyclic ligands with a pendant phenyl group -N-[2-(l-phenyl)ethyl]-1-aza-4,8di thiacyclodecane (L1) and N-[2-0-phenyl)propyl]-1-aza-4,8-dithiacyclodecane, (L2) has been synthesized and characterized. The copper(I)-triphenylphosphine complex [LICu(PPhJ)]OTf , (2) was synthesized from L1 1/2(C6H6)[CuOTf]2, and PPh3. The complexes [LICu]OTf 1 and [L2Cu]OTf 3 were also synthesized and characterized using DC and IH NMR, UV-vis and lR spectroscopy, and ESI-MS. In CD2Cl2, 1 was found to have a fluxional behavior with the barrier ΔG to the process estimated at 9-11 kcal/mol. Variable temp. NMR spectroscopy of 1 gave compelling evidence that the phenylappended group is bound in an intramolecullar η2-fashion with the copper center. At …

A flow injection system using the chemiluminescent reagent 2-methyl-6-(4methoxyphenyl)- 3,7-dihydroimidazo[1,2-a]pyrazin-3-one (MCLA) was used to quantify aqueous concentrations of superoxide. Stock solutions of superoxide were produced by photolysis of a solution of 2-propanol and benzophenone. The concentration of superoxide was quantified in the stock solutions by direct measurement of the absorbance at 240 nm. Quantified additions of superoxide to the flow injection analysis system reacted with MCLA at pH 5.1 demonstrated a detection limit of 0.074 nM superoxide. Selective superoxide detection was confirmed by pH-dependent decay of superoxide, which was in excellent agreement with published rates.

2-(1a,9b-Dihydro-lH-cyclopropa[l]phenanthren-1-yl)-propan-2-ol was synthesized by two different but related synthetic routes and characterized by melting point, IR GC/MS, CHN, and NMR spectroscopy. Photolysis of the alcohol in benzene d-6 led to the formation of 2-butanone, 1-methylcyclopropanol, and 2,2-dimethyloxirane in a 26:10:1 ratio via 2-hydroxy-2methylpropylidene, a novel ?-hydroxycarbene. These products are a consequence of intramolecular insertions of the carbene into C-C, C-H, and O-H bonds. Molecular orbital calculations performed at the Becke-3LYP/6-31G(d) level indicate that the singlet form of this carbene is more thermodynamically stable than the triplet. Furthermore, the transition states to the three products from the singlet species were modeled …

As alternative structures to the double helix, G-tetrads are thought to occur in DNA with many repeating deoxyguanosines (dG). These tetrads have one dG on each corner of a plane and can stack into tetraplexes that can be aptamers and may be found in telomeres. G-tetrads are destabilized by incorporation of 6-thiodeoxyguanosine (?sdG). Addition of metal ions that bind sulfur may stabilize these ?sdG-tetrads. We have carried out a survey of metal ion titrations into ?sG. Stable ?sG:MD? complexes were found for Hg2? (2:1), Cd2? (4:1), Ag2? (0.7:1) and Pt2? (thought to be 2:1). The focus of this research was the synthesis of two natural products, 1-methyl-s-(1,2,2-trimethy1cyclopentyl)-benzene (formally cuparene), and 3-(1,2dimethylcyclopenty1)-5-methylbenzene-l,2-diol (formally herbertene diol). Cuparene possesses anti-fungal and anti-arthritic properties. This molecule has been the target of a number of syntheses. Herbertene diol, a natural product itself, represents one retrosynthetic step back from mastigophorane; a compound isolated form the liverwort Mastigophora diaclolades. Mastigophorane has been shown to have neural network forming capabilities and has warranted complete synthetic efforts. The structural beauty and interesting bonding properties of polycyclic cage compounds have been a source of fascination to many chemists. In this work, progress toward the synthesis of large oxabowl systems, such as hexaoxa[6]peristylane and its derivative dipbenylhexaoxa[6)peristylane, is described. Thus, the adduct obtained from the cycloaddition of maleic anhydride and cyc!ooctatetraene is converted into key dicarbonyl intermediates. Subsequently, the ozonolysis of these intermediates is carried out in order to generate hexaoxa(6)peristylane as well as diphenylhexaoxa[6]peristylane. Molecular modeling studies, performed at the semi-empirical level, are also described.

Atrazine and 2,4-D are common herbicides used for crop, lawn, and rangeland management. Photochemical degradation has been proposed as one safe and efficient remediation strategy for both 2,4-D and Atrazine. In the presence of iron(llI) and hydrogen peroxide these herbicides decay by both thermal and light induced oxidation. Past studies have focused primarily on sun light as an energy source. This work provides a mechanistic description of herbicide degradation incorporating intermediate degradation products produced in the dark and under well-defined light conditions.

Metal substitution has been proposed as an aid in the characterization of the structure of Low-Molecular-Weight Chromium-binding substance (LMWCr), which appears to be the biologically active chromium species. Preparations of bovine liver were diced and suspended in an isolation solution containing either a cobalt or a chromium salt. Peptide samples from these preparations were isolated by precipitation and centrifugation and purified by liquid chromatography. Several methods were used to analyze these samples, which may contain LMWCr or the cobalt-substituted form (LMWCo), in order to determine their purity, molecular weight (MW), and metal-to-peptide ratio. The Fe(II)-catalyzed oxidation of luminol has been a well-accepted chemiluminescence method for the trace and ultra-trace analysis of Fe(II) and Fe(III) in natural waters. Investigation into this chemiluminescence system has shown that COlg) is essential to Fe-catalyzed luminol chemiluminescence. In absence of CO2(g) there is no chemiluminescent signal. These reactions worked well in the past because CO2 is present in most laboratory H20 supplies. Intentional saturation of CO2 results in a 5-fold increase for this conventional system. Aziridines are three-membered rings containing one nitrogen atom. They are useful as synthetic intermediates in many organic reactions. Much work has been devoted to synthesizing their analogs, cyclopropanes and epoxides, but little work has been done on the aziridination reaction. This study endeavored to probe the efficiencies of novel copper aziridination catalysts with N3 and S3 macrocyclic ligands compared to an NS2 analog for the reaction with the nitrene precursor (N-(paratosylsulfonyl) imino)phenyliodinane, PhINTs, and the olefin styrene. The complexation of metal ions in natural waters plays a critical role in defining the metal's biogeochemistry, Numerous investigators have used various experimental techniques to measure complexation constants in the laboratory, but many constants are still poorly undefined or have yet to be investigated due to the difficulty of some of the experiments. A computational approach to obtaining these constants for complexes of Fe(lII), Fe(D), Cr(lli), Cu(lI), Zn(II), and Cd(II) was explored with varying degrees of success. Nizatidine, N-[2-[[[2-[(dimethylamino)methyl]-4-thiazoly]]methy1]thio]ethyl]-N'- methyl- 2-nitro-I,l, ethenediamine, has been investigated for its potential to act as a host to small molecules through hydrogen bonding and pi-pi interactions. In previous work, pyridoxine (vitamin B6) was hypothesized to have a high guest-host binding constant with nizatidine. This was supported through experimental methods involving one and two dimensional NMR spectroscopy, UV-visible spectroscopy, and high performance liquid chromatography. Computer aided molecular design and three dimensional conformationally flexible database searching led to the discovery of other possible guests, including albuterol, which has exhibited even higher binding constants from HPLC.

DNA interstrand cross-linking agents have been recognized for many years to be powerful antitumor chemotherapeutics. While the interactions between free DNA and many anticancer drugs have been explored extensively, there is less infonnation available about these interactions with nucleosomal DNA. Nitrogen mustards have been reported to exhibit relatively similar cross-linking sites and efficiencies of binding in free and nucleosomal DNA, but mitomycin C exhibits markedly reduced cross-linking near the center of nucleosomal DNA. This research has focused on the total synthesis of the iron--chelating natural product 1-(2,3-hydroxyphenyl)-4-methoxy-I-butanone. An extract of the iron-cleficient fungus Gleophyllwn lraebeum, this molecule is thought to bind to Fe3+ in a hexacoordinate system to aid in transport through cell walls in order to help regulate iron levels in such iron-deficient fungi. Since the proposed structure of the molecule is based only on the spectral data of a very small sample, our hope is to synthesize and confirm its structure as the extract of the brown rot fungus. In order to understand further the interactions that exist between specific organometallic complexes and DNA, reactions of double-stranded calf-thymus DNA with bis(cyclopentadienyl)titanium dichloride [Cp₂TiC1₂] (TDC), bis(t-butylcyclopentadienyl)titanium dichloride [(C₄H₉Cp)₂TiC1₂] (r-butyl TDC), bis(i-propylcyclopentadienly)titanium dichloride [(C₃H₆Cp)2TiCl₂ a-propyl mc), bis(pentamethylcyclopentadienyl) titanium dichloride [(Cp"₂TiC1₂ (pentarnethyl TDC), triphenyltin chloride (Ph₃SnCl), diphenylrin dichloride (Ph₂SnC1₂) , and triphenyltin acetate (Ph₃SnOAc) were performed. Nizatidine, N-[2-[[[2-[(dimethylamino)metbyl]-4-thiazolyl]methyl]thio]ethyl]-N'methyl- 2-nitro-l,l-ethenediamine (Axid"), has been investigated for its potential to act as a host in guest-host complexes. Nizatidine binds with the guests through hydrogen bonding and pi-pi interactions. Guest-host studies were conducted using fluorescence quenching, calorimetry, NMR and invisible experiments. The lowest energy conformation of nizatidine is found to be U-shaped through theoretical calculations. Fluorescence experiments indicated that tryptophan and pyridoxine had large association constants. Further studies using calorimetry and NMR techniques concluded that no complexation had occurred. Complexation experiments have been done to find guests that complex with nizatidine. Caffeine, luminol, nicotinamide, phenylalanine, pyridoxine, pyrogallol, and tryptophan were …

In this investigation, a method was developed for the determination of purity of the drug PNU-10483 using the technique of capillary zone electrophoresis (CZE). CZE was seen as a potential alternative to the formerly used procedure of ion-pair reversed phase highperformance liquid chromatography (HPLC). Nearly five years earlier, a similar study was performed, in which an attempt was made to develop a method that used CZE to determine the purity of bulk drug and the potency of drug tablets. Any transition metal nitrosyl complex may release nitric oxide and thus has potential as a vasodilator. Nitroprusside, Na2[Fe(CN)5(NO)], is currently used, but dosages are limited because of the potential release of cyanide. Research in our group has been directed towards the synthesis of iron and ruthenium complexes containing the nitrosyl ligand and polypyridyl coligands such as 2,2-dipyridyl (bpy) and 2,2': 6',2"-terpyridine (tpy). These coligands are much less toxic than cyanide. Both [(bpy)(tpy)Fe(NO)]3+ and [(bpy)(tpy)Ru(NO)]3+ have been synthesized and characterized using infrared and ultraviolet spectroscopy, conductivity, and magnetic susceptibility.
