The glass transition phenomenon and free volume behavior below and above the glass transition temperature of various polymeric systems have been investigated. Several novel polymeric systems were considered for this study. Two generations of hyperbranched polyols, H40 and H20, were selected due to large number of hydroxyl groups on the periphery and within the bulk. The effect of hydrogen bonds and molecular weight was related with the glass transition and free volume behavior for the whole range of experimental temperature. Strategies to influence the steric and electronic properties of three classes of ligands were investigated. First, a compound class known as amino-dithiaphospholanes was studied. It is structurally related to phosphoramidites, which have been successfully applied in catalysis. The synthesis of amino-dithiaphospholanes was envisaged as an approach to electronically tuned phosphoramidites. General access to a variety of structurally modified amino-dithiaphospholanes from commercially available starting materials was developed. The electron donating properties of the new ligands were characterized by NMR. They were found to be more electron-donating than the corresponding phosphoramidites. This work is divided into 2 parts: a) total synthesis of (±)-cyclophostin, (±)-cyclipostin P and their monocyclic analogs b) development of a metathesis based methodology for transformation of vinyl phosphonates. Cyclophostin (1) and cyclipostins (2) are bioactive natural bicyclic organophosphates. Cyclophostin is a powerful inhibitor of acetyl cholinesterase (AChE), whereas members of the cyclipostins are potent inhibitors of hormone sensitive lipase (HSL). It consists of two parts: (a) modification and synthesis of oxetane (OXE), and oxolane (OXO) substituted chitosan (CHI) macromonomer, and (b) design, and synthesis of self-repairing polyurethane (PUR) networks consisting of modified chitosan. Alzheimer's Disease (AD) is a progressive, neurodegenerative disorder which is ranked as a leading cause of death among Americans. AD is characterized by the presence of intracellular neurofibrillary tangles and extracellular plaques made of amyloid ß (Aß). Together these pathologies lead to severe memory impairment in patients, but research has implicated the presence of the Aß deposits as likely causes for AD progression. Sixteen novel, Ru-based olefin metathesis catalysts bearing pH responsive ligands were synthesized. The pH-responsive groups employed with these catalysts included dimethylamino (NMe2) modified NHC ligands as well as N-donor dimethylaminopyridine (DMAP) and 3-(o-pyridyl)propylidene ligands. These pH-responsive ligands provided the means by which the solubility and/or activity profiles of the catalysts produced could be controlled via acid addition. Gas transport and free volume properties of photopolymerized thiol-ene networks for various applications are reported. For basic, commercially-available thiol-ene formulations, oxygen permeation was strictly dependent on the difference between Ttest (room temperature for this work) and network Tg. Networks with Tg near Ttest demonstrated the lowest values of permeability, diffusivity, and solubility. A robust, high-barrier network was selected for further modification. New networks with embedded functionality were formed using a two-step approach. Thio-Michael addition of a tetrathiol to monofunctional acrylates formed new functional thiol monomers. The primary goals of this study were modification of existing Nafion® membranes and characterization of newly developed hydrocarbon-based membranes for high temperature fuel cell applications. Various Nafion®/silicate nanocomposites were formulated via in situ sol-gel reactions for tetraethylorthosilicate. Different silicate composition profiles generated across membrane cross-sections were investigated by EDAX/ESEM. Composite water uptake, proton conductivity and fuel cell performance were comparable to that of unmodified Nafion®. Tafel analysis showed better electrode kinetics for composites having more silicate in the middle and less or no silicate at electrolyte-electrode interfaces. The core of this dissertation lies in the search of analytical tools which can be used to detect and quantify the high explosives confiscated from suspects in transportation hubs and from soil and water bodies where these explosives pose a greater threat to public health and safety. High explosives, namely, hexamethylene triperoxide diamine (HMTD), triacetone triperoxide (TATP), trinitrotoluene (TNT), and pentaerythritol tetranitrate (PETN), were detected and quantified by electrochemical methods such as electrogenerated chemiluminescence (ECL) and cyclic voltammetry (CV).

Trap grease offers a promising and less expensive alternative to food grade vegetable oils for biodiesel production, but its commercial marketability could be affected by its intense and offensive odor. Identification, quantitation and eventual elimination of the odorant compounds are vital challenges that need to be addressed before commercialization can be achieved. A rapid method using HS-SPME and GC/MS was developed in this study to identify and quantify the major volatile odorant compounds present in trap grease. Type 2 diabetes is mediated by insulin resistance, the inability of insulin to elicit a normal biological response in insulin responsive tissues. Several cellular models have been utilized to determine the mechanism of induction of insulin resistance but questions remain unanswered. One model, implicates the products of the Hexosamine Biosynthetic Pathway (HBP) in the induction of insulin resistance under hyperglycemia. The major end product of HBP, UDP-GlcNAc, is the substrate for O-GlcNAc transferase, an enzyme that catalyzes the O-GlcNAcylation of numerous proteins. Despite the increasing interest in developing sensors for nerve gas agents, efficient detection remains challenging. Among the various sensors developed so far, fluorescence sensors play an important role due to their simplicity. We developed six new rhodamine-based compounds that can be used as fluorescent turn-on sensors. Compound 1 and 3 gave high fluorescent enhancement with diethyl chlorophosphate (DCP) compared to the other compounds. Very high selectivity and sensitivity were observed as these compounds did not show significant fluorescent enhancement with dimethyl methylphosphonate (DMMP), HC1 and transition metal ions. Histopathological hallmarks that characterize AD are senile plaques formed by extracellular deposition of Amyloid beta (Aβ) peptide and intracellular aggregates of hyperphosphorylated tau protein. The plaques, which are found in the brain parenchyma, comprise both 40 and 42 residue Aβ. Aggregation of Aβ is an established pathogenic mechanism in AD, but little is known about the initiation of this process in vivo. Various piano-stool ruthenium complexes bearing phosphoramidite ligands have been synthesized and characterized spectroscopically and in some cases structurally. Reaction of phosphoramidite ligands with an appropriate metal precursor gives new piano-stool ruthenium complexes [RuCl(L)(arene)(phosphoramidite)], where L = Cl, PPh3, or others. The novel complexes are tested for their ability to activate propargylic alcohols catalytically as well as stoichiometrically. Specifically, catalytic substitution of propargylic alcohols via allenylidene intermediates is envisioned. Solid-state structure, crystalline morphology, crystallization kinetics, thermal, free volume, and gas transport properties of semicrystalline syndiotactic polystyrene (sPS) and ethylene vinyl alcohol copolymers (EVOH) have been investigated. Solid-state structure of sPS after crystallization from the melt and glassy state was examined by differential scanning calorimetry (DSC), density and wide angle x-ray diffraction analysis (WAXD). The measurements confirmed low density of syndiotactic polystyrene crystalline forms, which in the case of α and δe was smaller and in the case of β and γ crystalline forms was slightly larger than the density of the glassy amorphous sPS. The purpose of this research is to develop a multiscale understanding of crosslinked amorphous matrices, connecting molecular level events to macroscopic properties. To accomplish this goal, our methodology was to identify network architectures that influence molecular level energy dissipation through mechanisms such as bond rotations, torsions, and ring flips and then relate those molecular motions to macroscopic properties. Sulfonated poly[styrene-b-ethylene-co-butylene-b-styrene] (sSEBS) block copolymers/inorganic nanocomposite materials were synthesized via in situ formation of inorganic fillers and characterized particularly for their dielectric properties and proton conductivities.
In preparation of sSEBS/SrTiO3 nanocomposites, titanium (IV) isopropoxide [Ti(OPri)4] complex was diffused into sSEBS film, followed by subsequent hydrolysis of [Ti(OPr i)4], diffusion of strontium cations in sSEBS domains, and in situ formation of crystalline SrTiO3. sSEBS with sulfonation degree of 38.1% and 65.0% were employed, and relevant sSEBS/SrTiO3 composites contain SrTiO3 of 10-15 wt%. The research presented in this dissertation involves the synthesis and polymerization of heterocyclic monomers which pave the way to biodegradable polyester nanocomposites, functional polyesters or poly(ester amide)s, functional polyamides and supramolecular polymers. The key monomers are ε-caprolactone, γ-acetamido-ε- caprolactone, γ-ethylene ketal-ε-caprolactam and α-amino-ε-caprolactam.
Poly(ε-caprolactone) organo-modified montmorillonite nanocomposites were prepared by in-situ polymerization using dibutyltin dimethoxide as initiator/catalyst. The montmorillonite was first modified with 1-decyl-2-methyl-3-(11-hydroxyundecyl)- imidazolium cation. Semiconductor and metal clusters in the nanometer size regime display interesting optical, electronic and chemical properties that are size dependant. Highly monodisperse gold nanoparticles of different sizes have been synthesized and isolated by newly developed methodologies, using tiopronin as the capping ligand. Titanium dioxide nanoparticles (Ti02) have been used as the photocatalytic center and gold nanoparticles have been attached on Ti02 to make a nanocomposite. The development of synthetic procedures that produce nanoscale materials with controlled morphology is a significant area of investigation in nanotechnology. We have developed new synthetic procedures for fabrication of isotropic and/or anisotropic nanomaterials composed of palladium, ruthenium, nickel and alloys of palladium-ruthenium. The nanomaterials were fully characterized and were found to display unique size dependent properties. The electrochemical properties of isotropic palladium, ruthenium and palladium-ruthenium nanoparticles were investigated and the results showed that the particles were capable of charge storage and charge transfer on demand.
