Aims

Studies dealing with plants’ mineral nutrient status under elevated atmospheric CO2concentration (eCO2) are usually conducted in closed hydroponic systems, in which nutrient solutions are entirely renewed every several days. Here, we investigated the contribution of the fluctuation of concentrations of N ([N]), P ([P]), and K ([K]) in nutrient solutions in this short period on their concentrations in cucumber plants exposed to different [CO2] and N levels.

Methods

Cucumber (Cucumis sativus L.) plants were hydroponically grown under two [CO2] and three N levels. [N], [P], and [K] in nutrient solutions …

Line-Of-Sight Winds And Doppler Effect Smearing In Ace-Fts Solar Occultation Measurements, Chris D. Boone, Johnathen Steffen, Jeff Crouse, Peter F. Bernath Jan 2021

Line-Of-Sight Winds And Doppler Effect Smearing In Ace-Fts Solar Occultation Measurements, Chris D. Boone, Johnathen Steffen, Jeff Crouse, Peter F. Bernath

Chemistry & Biochemistry Faculty Publications

Line-of-sight wind profiles are derived from Doppler shifts in infrared solar occultation measurements from the Atmospheric Chemistry Experiment Fourier transform spectrometers (ACE-FTS), the primary instrument on SCISAT, a satellite-based mission for monitoring the Earth’s atmosphere. Comparisons suggest a possible eastward bias from 20 m/s to 30 m/s in ACE-FTS results above 80 km relative to some datasets but no persistent bias relative to other datasets. For instruments operating in a limb geometry, looking through a wide range of altitudes, smearing of the Doppler effect along the line of sight can impact the measured signal, particularly for saturated absorption lines. Implications …

Potentiometric Biosensing Of Ascorbic Acid, Uric Acid, And Cysteine In Microliter Volumes Using Miniaturized Nanoporous Gold Electrodes, Christopher J. Freeman, Borkat Ullah, Md. Shafiul Islam, Maryanne M. Collinson Jan 2021

Potentiometric Biosensing Of Ascorbic Acid, Uric Acid, And Cysteine In Microliter Volumes Using Miniaturized Nanoporous Gold Electrodes, Christopher J. Freeman, Borkat Ullah, Md. Shafiul Islam, Maryanne M. Collinson

Chemistry & Biochemistry Faculty Publications

Potentiometric redox sensing is a relatively inexpensive and passive approach to evaluate the overall redox state of complex biological and environmental solutions. The ability to make such measurements in ultra-small volumes using high surface area, nanoporous electrodes is of particular importance as such electrodes can improve the rates of electron transfer and reduce the effects of biofouling on the electrochemical signal. This work focuses on the fabrication of miniaturized nanoporous gold (NPG) electrodes with a high surface area and a small footprint for the potentiometric redox sensing of three biologically relevant redox molecules (ascorbic acid, uric acid, and cysteine) in …

Using multidecadal time series of ground-based and satellite Fourier transform infrared measurements of inorganic fluorine (i.e., total fluorine resident in stratospheric fluorine reservoirs), we investigate stratospheric circulation changes over the past 20 years. The representation of these changes in five modern reanalyses is further analyzed through chemical-transport model (CTM) simulations. From the observations but also from all reanalyses, we show that the inorganic fluorine is accumulating less rapidly in the Southern Hemisphere than in the Northern Hemisphere during the 21st century. Comparisons with a study evaluating the age-of-air of these reanalyses using the same CTM allow us to link this …

Respiration in intertidal salt marshes generates dissolved inorganic carbon (DIC) that is exported to the coastal ocean by tidal exchange with the marsh platform. Understanding the link between physical drivers of water exchange and chemical flux is a key to constraining coastal wetland contributions to regional carbon budgets. The spatial and temporal (seasonal, annual) variability of marsh pore water exchange and DIC export was assessed from a microtidal salt marsh (Sage Lot Pond, Massachusetts). Spatial variability was constrained from 224Ra : 228Th disequilibria across two hydrologic units within the marsh sediments. Disequilibrium between the more soluble 224Ra …

Mitochondrial Energetics With Transmembrane Electrostatically Localized Protons: Do We Have A Thermotrophic Feature?, James Weifu Lee Jan 2021

Mitochondrial Energetics With Transmembrane Electrostatically Localized Protons: Do We Have A Thermotrophic Feature?, James Weifu Lee

Chemistry & Biochemistry Faculty Publications

Transmembrane electrostatically localized protons (TELP) theory has been recently recognized as an important addition over the classic Mitchell’s chemiosmosis; thus, the proton motive force (pmf) is largely contributed from TELP near the membrane. As an extension to this theory, a novel phenomenon of mitochondrial thermotrophic function is now characterized by biophysical analyses of pmf in relation to the TELP concentrations at the liquid-membrane interface. This leads to the conclusion that the oxidative phosphorylation also utilizes environmental heat energy associated with the thermal kinetic energy (kBT) of TELP in mitochondria. The local pmf is now calculated to be in …

With the increased occurrence of forest fires around the world, interest in the chemistry of pyrogenic organic matter (pyOM) and its fate in the environment has increased. Upon leaching from soils by rain events, significant amounts of dissolved pyOM (pyDOM) enter the aquatic environment and interact with microbial communities that are essential for cycling organic matter within the different biogeochemical cycles. To evaluate the bio-reactivity of pyDOM, aqueous extracts of laboratory-produced chars were incubated with soil microbes and the molecular changes to the composition of pyDOM were probed using ultrahigh resolution mass spectrometry (Fourier transform – ion cyclotron resonance – …

Atoms remaining as recognizably distinct constituents of bulk condensed phases can have properties modified from those of the isolated species. Dense helium bubbles at high pressures are a common form of radiation damage degrading the mechanical and electrical properties of host materials. Detailed knowledge is critical for predicting their long term performance. Modifications of the ground and first singlet excited states of confined compressed helium are investigated using an entirely non-empirical theory based on the results of ab initio self-consistent field calculations with corrections for the effects of electron correlation. For finite sized portions representing bulk condensed fcc and bcc …

Examination of dinitrogen (N2) fixation in the Eastern Tropical South Pacific oxygen deficient zone has raised questions about the range of diazotrophs in the deep sea and their quantitative importance as a source of new nitrogen globally. However, technical considerations in the deployment of stable isotopes in quantifying N2 fixation rates have complicated interpretation of this research. Here, we report the findings of a comprehensive survey of N2 fixation within, above and below the Eastern Tropical South Pacific oxygen deficient zone. N2 fixation rates were measured using a robust 15N tracer method (bubble removal) …

Phytoplankton iron contents (i.e., quotas) directly link biogeochemical cycles of iron and carbon and drive patterns of nutrient limitation, recycling, and export. Ocean biogeochemical models typically assume that iron quotas are either static or controlled by dissolved iron availability. We measured iron quotas in phytoplankton communities across nutrient gradients in the Pacific Ocean and found that quotas diverged significantly in taxon‐specific ways from laboratory‐derived predictions. Iron quotas varied 40‐fold across nutrient gradients, and nitrogen‐limitation allowed diatoms to accumulate fivefold more iron than co‐occurring flagellates even under low iron availability. Modeling indicates such “luxury” uptake is common in large regions of …

Submarine groundwater discharge (SGD) is now recognized as an important process of the hydrological cycle worldwide and plays a major role as a conveyor of dissolved compounds to the ocean. Naturally occurring radium isotopes (Ra-223, Ra-224, Ra-226 and Ra-228) are widely employed geochemical tracers in marine environments. Whilst Ra isotopes were initially predominantly applied to study open ocean processes and fluxes across the continental margins, their most common application in the marine environment has undoubtedly become the identification and quantification of SGD. This review focuses on the application of Ra isotopes as tracers of SGD and associated inputs of water …

Chemistry & Biochemistry Theses & Dissertations

Transition metal complexes with Schiff base ligands offer a wide application in the field of development of catalysis and material. The straightforward synthesis allowed the structural modification and helped to optimize in various application of such complexes. Titanium-containing complexes have been reported to be important for their catalytic and material applications through the coordination of a tetradentate Schiff base ligand, viz. N, N’-ethylene bis(salicylideneiminate) dianion (salen). Studies reporting the characterization of achiral titanium(IV) salen complexes are scarce due to their intricate nature. Such complexes would be comparatively less expensive and easier to prepare synthetically and thus could represent an …

Advances In The Understanding Of Sourcing And Fate Of Pyrogenic Organic Matter In The Environment, Aleksandar Ivaylov Goranov

Chemistry & Biochemistry Theses & Dissertations

With higher occurrences of forest fires worldwide, there has been an increase in scientific interest surrounding the chemistry of pyrogenic organic matter (pyOM). The main structural components of pyOM, the condensed aromatic compounds (ConAC), exhibit intriguing physico-chemical properties and have been one of the main focuses of biogeochemical research. The overwhelmingly large number of scientific articles regarding pyOM and ConAC are guided by the assumption that ConAC in the environment are exclusively of pyrogenic origin, even though some recent studies have suggested that some of these ConAC could also be derived from non-pyrogenic radical-driven processes. To evaluate this controversial proposition, …

Implementation of dynamic spectrum access (DSA) in cognitive radio (CR) systems requires the unlicensed secondary users (SU) to implement spectrum sensing to monitor the activity of the licensed primary users (PU). Energy detection (ED) is one of the most widely used methods for spectrum sensing in CR systems, and in this paper we present a novel ED algorithm with an adaptive sensing threshold. The three-event ED (3EED) algorithm for spectrum sensing is considered for which an accurate approximation of the optimal decision threshold that minimizes the decision error probability (DEP) is found using Newton’s method with forced convergence in one …

Protonic Capacitor: Elucidating The Biological Significance Of Mitochondrial Cristae Formation, James Weifu Lee Jun 2020

Protonic Capacitor: Elucidating The Biological Significance Of Mitochondrial Cristae Formation, James Weifu Lee

Chemistry & Biochemistry Faculty Publications

For decades, it was not entirely clear why mitochondria develop cristae? The work employing the transmembrane-electrostatic proton localization theory reported here has now provided a clear answer to this fundamental question. Surprisingly, the transmembrane-electrostatically localized proton concentration at a curved mitochondrial crista tip can be significantly higher than that at the relatively flat membrane plane regions where the proton-pumping respiratory supercomplexes are situated. The biological significance for mitochondrial cristae has now, for the first time, been elucidated at a protonic bioenergetics level: 1) The formation of cristae creates more mitochondrial inner membrane surface area and thus more protonic capacitance for …

The principal bioeffect of the nanosecond pulsed electric field (nsPEF) is a lasting cell membrane permeabilization, which is often attributed to the formation of nanometer-sized pores. Such pores may be too small for detection by the uptake of fluorescent dyes. We tested if Ca2+, Cd2+, Zn2+, and Ba2+ ions can be used as nanoporation markers. Time-lapse imaging was performed in CHO, BPAE, and HEK cells loaded with Fluo-4, Calbryte, or Fluo-8 dyes. Ca2+ and Ba2+ did not change fluorescence in intact cells, whereas their entry after nsPEF increased fluorescence within <1 ms. The threshold for one 300-ns pulse was at 1.5–2 kV/cm, much lower than >7 …

Lignin: A Likely Precursor Of A Significant Fraction Of Humic Substances Via Oxidative Transformations, Seyyedhadi Khatami

Chemistry & Biochemistry Theses & Dissertations

Lignin is a major component of decaying terrestrial vegetation in soils and has been arguably reported to contribute substantially to the formation of soil carbon humus, and natural dissolved organic matter (DOM). To better understand the process by which this humification occurs, lignin and lignin-derived monomers were subjected to both biotic and abiotic oxidation processes. Two well-known oxidative transformation strategies were employed. The first involved the fungal degradation of brown-rot degraded wood subjected to a white-rot fungus (Phanerochaete chrysosporium) whose enzymes are particularly effective in lignin degradation via enzymatic oxidation. This enzymatic attack was monitored by ultrahigh resolution …

A Halogen Bonding Perspective On Iodothyronine Deiodinase Activity, Eric S. Marsan, Craig A. Bayse

Chemistry & Biochemistry Faculty Publications

Iodothyronine deiodinases (Dios) are involved in the regioselective removal of iodine from thyroid hormones (THs). Deiodination is essential to maintain TH homeostasis, and disruption can have detrimental effects. Halogen bonding (XB) to the selenium of the selenocysteine (Sec) residue in the Dio active site has been proposed to contribute to the mechanism for iodine removal. Polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) are known disruptors of various pathways of the endocrine system. Experimental evidence shows PBDEs and their hydroxylated metabolites (OH-BDEs) can inhibit Dio, while data regarding PCB inhibition are limited. These xenobiotics could inhibit Dio activity by competitively …

Modeling Iron And Light Controls On The Summer Phaeocystis Antarctica Bloom In The Amundsen Sea Polynya, Hilde Oliver, Pierre St-Laurent, Robert M. Sherrell, Patricia L. Yaeger

CCPO Publications

Of all the Antarctic coastal polynyas, the Amundsen Sea Polynya is the most productive per unit area. Observations from the 2010-2011 Amundsen Sea Polynya International Research Expedition (ASPIRE) revealed that both light and iron can limit the growth of phytoplankton (Phaeocystis antarctica), but how these controls manifest over the bloom season is poorly understood, especially with respect to their climate sensitivity. Using a 1-D biogeochemical model, we examine the influence of light and iron limitation on the phytoplankton bloom and vertical carbon flux at 12 stations representing different bloom stages within the polynya. Model parameters are determined by …
