Simple water soluble lanthanum and europium complexes are effective at detecting neutral sugars as well as glyco- and phospholipids. In solutions at physiologically relevant pH the fluorescent lanthanum complex binds neutral sugars with apparent binding constants comparable to those of arylboronic acids. Interference from commonly occurring anions is minimal. The europium complex detects sialic acid-containing gangliosides at pH 7.0 over an asialoganglioside. This selectivity is attributed, in large part, to the cooperative complexation of the oligosaccharide and sialic acid residues to the metal center, based on analogous prior studies. My graduate work has focused on the development of new fluorophore materials towards addressing several interdisciplinary research problems. Selective detection of sialic acid via a boron-amide interaction. Concentration changes of sialic acid can be monitored ratiometrically in the visible region using a boronic acid based chemosensor. The technique shows great promise for the detection of sialic acid, a critical cell surface residue involved in cancer, graft rejection and virulence in certain bacteria. Chapter 1 presents a concise introduction to porphyrins, especially on the synthesis of meso-tetraphenylporphyrins, which were synthesized during the course of my research and were used for further purposes in the following chapters. Chapter 2 describes the synthesis of a series of cobaltacarborane-porphyrin conjugates for the boron neutron capture therapy (BNCT) of tumors. Using an efficient and high yield ring-opening reaction, N-substituted porphyrins were also synthesized. The spectroscopic and structural properties of these compounds are presented. Guar gum is a naturally occurring polysaccharide; guar gum and its derivatives, such as carboxymethyl guar gum (CMG) are widely used in the oil and gas industry. A hydraulic fracturing fluid for rock formations to enhance oil recovery (EOR) is comprised of viscosifier, gelled CMG, and sand particles (proppants). The gel is mixed with proppant (sand) and pumped at high rate and high pressure into an oil/gas well to the hydrocarbon-producing zone. Platinum is the metal present in various classes of inorganic medicinal agents directed at important diseases such as cancer or viral infections. Several new Pt(II) complexes of polyamine ligands bearing a dansyl moiety linked by a sulfonamide group were prepared; in some cases the sulfonamide nitrogen formed a bond to Pt. The sulfonamido group donates strongly to Pt and places the dansyl group close to a vacant coordination position, allowing it to stack with bound guanine base (G) when G binds. Isoporphyrins are tautomers of porphyrins with an interrupted macrocyclic conjugation owing to the presence of a sp3 hybridized meso-carbon. Due to their absorption at long wavelengths (~800nm), isoporphyrins are potential candidates as photosensitizers in photodynamic therapy (PDT), a ternary modality of cancer treatment. Biologically damaging semiquinone-type radicals have been reported in cigarette smoke, their likely origin being hydroquinone (HQ) and catechol (CT) molecular precursors contained by the tobacco. Since other biomass contains HQ and CT-type species, it is likely that combustion of other biomass will also form semiquinone-type radicals. All hydrocarbon fuels will form phenol and substituted phenol that can form substituted phenoxyl radicals. Because each of these radicals has the potential to be environmentally persistent and biologically active, their formation and stabilization from various molecular precursors was studied with the focus on surface-bound radicals formed in association with combustion-generated fly-ash particles. ABSTRACT Cardiovascular disease, primarily atherosclerosis involves a number of distinct processes that are associated with plaque development. The use of differential gel electrophoresis to determine differences between proteins produced in native and bypass coronary arteries from the same heart has been studied. The analytical techniques presented in this study to characterize plaque samples include two dimensional gel electrophoresis, sodium dodecylsulfate polyacrylamide gel electrophoresis, mass spectrometry, differential gel electrophoresis, and high performance liquid chromatography. Multiple regression modeling techniques allow for rapid and accurate prediction of migration times and resolution values for micellar electrokinetic chromatography (MEKC) as well as the development of quick screening methods using steady-state fluorescence spectroscopy. All studies reported in this dissertation include optimization of calibration models and predictions of dependent variables by the use of validation samples. The root-mean-square percent relative error (RMS%RE) is used as a figure of merit for characterizing the performance of the calibration models. This research seeks to understand the structure and shear responses of poly(ethylene oxide)-Laponite nanocomposite hydrogels and to relate the macroscopic physical properties to the nanoscopic structural changes. Understanding that relationship is necessary, in order to tailor material properties to specific applications. Information on the structure at rest on multiple length scales was obtained by combining microscopic and neutron scattering techniques. The association mechanism between hydrophobic organic compounds (HOC) and dissolved humic materials (DHM) has continued to be one of the interesting areas in environmental applications. The goal of the research reported in this dissertation is to investigate the association behavior of HOC with DHM. The first part of this research involves steady-state fluorescence measurements of these multi-component systems to study the effect of DHM content on the association mechanism of HOC, in particular using pyrene as a model. Cα,α-disubstituted amino acids (ααAAs) are widely utilized to conformationally constrain peptides. Several pentapeptides containing dipropylglycine (Dpg) at alternating positions and their α-amino acid counterpart L-norvaline (Nva) analogues were synthesized to fully investigate the impact of Dpg on peptide backbone structure in aqueous solution. CD, VCD and NMR spectral analysis suggest that Dpg containing peptides adopt more ordered structures relative to their Nva containing analogues. In this dissertation, novel dendronized cellulose derivatives with biodegradable backbones were created by using sodium carboxymethyl cellulose (CMC) as the starting material. CMC was first transformed into its corresponding tetrabutyl ammonium salt, which reacted with 2-chloro-N-methylpyridinium iodide (CMPI) to produce an activated ester of [2-(N-methyl pyridium) carboxymethyl cellulose] iodide (MPCMCI). The reaction of MPCMCPI and di-tert-butyl 4-[2-(ter-butoxycarbonyl)ethyl]-4-aminoheptanedicarboxylate (BA) produced a new derivative with BA dendron elaborated on cellulose backbone (CMCBA). This dissertation describes the crystal growth, chemical structures, and efforts made towards the understanding of the physical properties of novel ternary rare-earth intermetallics, which may display signature behavior such as superconductivity, heavy fermion behavior, intermediate valence, and/or large magnetoresistance. A striking example: Alzheimer’s disease is caused by the self-assembly of the â-amyloid fragment of amyloid precursor protein. Such natural amphiphiles normally are very expensive and complicated to isolate. Synthetic mimics that are much cheaper and easier to prepare can facilitate the development of efficient characterization methods, even as they suggest new materials science applications. In Chapter 2, a simple and reproducible technique for constructing perfectly aligned gaps in fused-silica capillaries has been developed for postcolumn reagent addition with capillary electrophoresis (CE) to take advantage of laser-induced fluorescence (LIF) detection. ABSTRACT This dissertation focuses on the structural stability, magnetic and transport properties of ternary lanthanide compounds grown using indium and gallium flux. Single crystals of TbRhIn5 were synthesized using indium flux. TbRhIn5 is isostructural to the well known LnnMIn3n+2 (n = 1, 2, ∞; Ln = La, Ce; M = Rh, Ir) and adopts the HoCoGa5 structure type and crystallizes in the space group P4/mmm, Z = 1. Lattice parameters are a = 4.6000(6) Å and c = 7.4370(11) Å, V = 157.29(6) Å3. A sharp antiferromagnetic transition is observed at TN = 48 K for TbRhIn5. Single crystals of …

MAO is the co-catalyst in the metallocene catalytic systems, which are widely used in single site olefin polymerization due to their high stereoselectivity. To date, the precise structure of MAO, the particular compound or compounds catalytically active in MAO, have eluded researchers. Understanding of the fundamentals behind desorption and ionization can lead to improvements in the technique and to new applications. Experiments aimed at advancing this goal are wavelength studies and two-laser infrared/ultraviolet matrix-assisted laser desorption/ionization experiments. Alzheimer's disease is a progressive neurological ailment affecting 4.5 million aging Americans. The disease is characterized by the presence in the brain of self-assembled fibrils consisting of beta amyloid protein (Aβ). Soluble Aβ protein is present in normal human cerebrospinal fluid, but it is unclear what makes the protein aggregate into insoluble plaques. The goal of the research presented in this dissertation is to develop novel water soluble carboranylporphyrins that can deliver therapeutic dosage of boron specifically to tumors for application in boron neutron capture therapy. In order to accomplish this goal novel water soluble tetra(nido-carboranyl)porphyrin and the octa(nido-carboranyl)porphyrin with high boron contents are synthesized by the condensation of the carborane substituted benzaldehyde with the pyrrole under acidic conditions. These porphyrins are made water soluble by the deboronation of the closo-carboranes to afford the corresponding amphiphilic nido-carborane cages. A major task in proteomics is to identify proteins from a biological sample using two-dimensional (2-D) separation prior to mass spectrometry of peptides generated via proteolytic digestion of the proteins. For 2-D separations, microfluidic devices are superior to bench top and capillary-based systems since they potentially provide higher separation efficiencies due to the minimal dead volumes produced during peak transfer between the two separation dimensions. In addition, fast separations can be envisioned because the column lengths are typically shorter in microfluidic platforms without scarifying peak capacity. This dissertation describes the integration of micro-scale separations to matrix assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI TOF MS) for protein analysis. MALDI MS provides unsurpassed accurate mass measurements of intact bio-molecules, for example peptides and proteins, which in turn generate high molecular specificity enabling the identity, function and structure of these molecules to be characterized.
