Environmentally persistent free radicals (EPFRs) from combustion-generated PM have been demonstrated to form via reactions of molecular precursors with redox-active transition metals at 150-500C thermal reactions in a few seconds reaction times. While ambient temperatures are much lower, soils/sediments contain similar transition metals, and reaction times of contaminants in soils/sediments are years, rather than seconds. This questions whether EPFRs could be formed at ambient temperatures in soils/sediments from Superfund sites that are contaminated with hazardous materials. Superfund soils contaminated with PCP from Georgia and Montana, and sediments contaminated with PAHs from Washington. Using EPR spectroscopy, EPFR concentrations and structural assignments …

The goal of this dissertation is to investigate electroosmotic flow (EOF) and electric field dynamics during capillary electrophoresis (CE) experiments using methods based on periodic photobleaching of fluorophores added to the separation buffer at nanomolar concentrations. The methods provide time resolved EOF and local electric field information during an experiment, which can be applied to fundamental studies to provide better understanding of CE techniques. The potential of the EOF monitoring method to improve CE migration reproducibility was investigated in Chapter 2. The mechanism and kinetics of the interaction of humic acids (HAs) with a 1-palmitoyl-2-oleoyl-Sn-glycero-3-phosphatidylcholine (POPC) large unilamellar vesicle (LUV) model biomembrane system were studied by fluorescence spectroscopy. All three HAs studied induced a perturbation to the biomembrane bilayer structure at pH 4.8. Concentration dependence studies revealed that biomembrane perturbation increased with increasing HA concentrations for Suwannee River HA (SWHA) from 0 to 20 mg C/L. For both Leonardite HA (LAHA) and Florida Peat HA (FPHA) aggregation influenced biomembrane perturbation at concentrations above 5 and 7.5 mg C/L, respectively. This dissertation describes the first total synthesis of alloviroidin in trace amounts, along with that of three analogs containing L-proline (Pro), trans-3-hydroxyproline (3-Hyp) or cis-4-hydroxyproline (4-hyp) residue substituting for 2,3-trans-3,4-trans-dihydroxyproline in the natural product. We report herein an efficient strategy that provides a dipeptide containing a (2S,4S)-4,5-dihydroxyleucine (dihyLeu) residue, including a diastereoselective dihydroxylation. Nα-Carbobenzyloxy-(2S)-4,5-dehydroleucine was coupled with valine ethyl ester to give a dipeptide that was subjected to a Sharpless asymmetric dihydroxylation to introduce the diol. Infrared lasers are highly efficient at sample removal and post-ablation ionization of materials removed by IR laser can be used to improve ionization efficiency. Fundamental studies of the physical processes in laser ablation mass spectrometry can help elucidate the mechanism. Porphyrins and their derivatives are often used as photosensitizers in photodynamic therapy (PDT), which is a noninvasive antitumor treatment. The photochemical process for PDT involves exciting a photosensitizing agent with visible light, which induces cytotoxicity in the presence of oxygen as a result of forming reactive oxygen species (ROS). The ROS are the responsible components for invoking cell death and destruction of tumors. Although this mechanism is an effective cancer therapeutic, it still has many shortcomings. Over the last few decades, liposomes have generated a lot of interest as drug delivery vehicles to address the need for providing both increased therapeutic efficacy and decreased systemic exposure, simultaneously. Despite having the advantage of easy and fast thin film manufacturing, solution techniques have intrinsic drawbacks, such as disordered and poor molecular organization in the film (often resulting in poor device performance), as well as insufficient stabilities of the films and resulting devices. The first part of the work is dedicated to the selective detection of Homocysteine (Hcy). In order to better understand this colorimetric detection method, a thermodynamic model for the pH dependence of the effective reduction potential of Hcy derived á-carbon radicals has been proposed. And using viologen indicators of varying reduction potentials, we demonstrate that colorimetric changes occur at experimental pH values consistent with the model. The one-carbon bridge stereochemistry of bicyclo[3.3.1]nonane framework is critical to make the third ring of the tricyclic precursor of vinigrol. Recently, there have been reports on biological and solid-state nanopores that identify single biopolymers or their constituent monomers by analyzing changes in ionic current blockades when they block the flux of buffer ions while travelling through the nanopores. There is currently no available molecular diagnostic test for stroke; the common modality for diagnosis consists of computed tomography or magnetic resonance imaging. Unfortunately, the use of these diagnostic regimens can delay proper therapeutic treatment, which requires administration within the first 3 h of a stroke event. Frontal polymerization (FP) is when the monomer is polymerized by a zone of reaction propagating through the monomer. The reaction zone is fueled by the heat given off by the polymerization of the monomer. FP is a promisig method for curing adhesives because it allows for cure-on-demand and fast cure times. The monomers selected for the adhesive is the main factor in determining the adhesive’s properties. Trimethylolpropane triacrylate (TMPTA), pentaerythritol tetraacrylate (PETA), and other multifunctional monomers make the polymer formed more brittle. Liposomes have had a long and fruitful application of drug delivery devices in the treatment of select tumors and cancer types. Third-generation liposomes possess endogenous triggering methods that hold the ability for site-specific unloading of liposomal contents, thus providing necessary temporal and spatial control over contents release that can be tuned to match the drug efficacy for decreased side effects during treatment. A hybrid imaging mode for characterization of magnetic nanomaterials has been developed, using atomic force microscopy (AFM) combined with electromagnetic sample actuation. Instead of using a coated AFM probe as a magnetic sensor; our strategy is to use a nonmagnetic probe with contact mode AFM to characterize the vibration of magnetic and superparamagnetic nanomaterials responding to the flux of an AC electromagnetic field. Naturally self-assembled mesospheres provide a practical route for controlling the arrangement of materials on surfaces at the nanoscale. Periodic arrays of well-defined nanostructures can be produced with different nanomaterials and interpattern spacings. Results presented in this dissertation demonstrate particle lithography methods developed for fabricating arrays of organosilane nanostructures. Surfaces were designed for the selective deposition of polymers and nanoparticles to produce multicomponent nanopatterns. The formation of environmentally persistent free radicals in combustion system was investigated from chemisorptions of chlorine- and hydroxy-substituted benzenes on transition metal oxide surface under post-combustion conditions. This manuscript reports the formation of EPFRs on silica particles containing 5% Fe(III)2O3, Ni(II)O, and Zn(II)O. The EPFRs are formed by the chemisorption of substituted aromatic molecular adsorbates on the metal cation center followed by electron transfer from the adsorbate to the metal ion at temperatures from 150 to 400 oC. Scanning probe microscopy (SPM) provides unique capabilities for surface visualization and measurements that reach atomic and molecular dimensions. My research focus is directed toward applying and developing new measurements for analytical and surface chemistry with SPM. Two distinct goals based on studies with atomic force microscopy (AFM) will be described within this dissertation. The primary goal was to develop and apply a new AFM imaging mode for ultrasensitive measurements of the superparamagnetic properties of proteins. Structural disorder and dimensionality play central roles in the characterization of structure and properties of crystalline materials. Although structural disorder is commonly considered an undesirable quality, structural disorder may be desirable when searching for new materials with exotic properties. Disorder can be used as a tunable parameter when considering atomic sizes, coordination preferences, and electronegativity differences can be varied by substituting elements into a given structure. Dimensionality can be viewed as another adjustable parameter when searching for new materials presenting unique challenges when characterizing new materials. My research focus was to investigate alligator blood using mass spectrometry-based proteomics methods to understand their innate immune systems. The first goal was to sequence peptides and proteins from the blood serum and leukocytes using tandem mass spectrometry and de novo sequencing. The second goal was to determine the function of these biological molecules and their relationship to the immune system. One- and two-dimensional gel electrophoresis was used to separate proteins from alligator leukocytes, which were enzymatically digested. The peptides were measured using reversed phase nano-high performance liquid chromatography coupled with tandem mass spectrometry (nano-HPLC-MS/MS) followed with de novo sequencing. …
