Objective: To assess the chemical composition of cerumen by flash pyrolysis-gas chromatography/mass spectrometry.
Study Design: Collected earwax specimens were fractionated into residue and supernatant by means of deoxycholate. This natural bile acid produces significantly better disintegration of earwax in vitro than do presently available ceruminolytic preparations, and also has demonstrated excellent clinical results in vivo to date.
Patients: The sample for analysis was obtained from a patient with clinical earwax impaction.
Results: The supernatant is composed of simple aromatic hydrocarbons, C5-Cl 7 straight-chain hydrocarbons, a complex mixture of compounds tentatively identified as diterpenoids, …

A non-aqueous titration method was developed to assay Loratadine bulk active and a High Performance Liquid Chromatography (HPLC) method was developed to determine Loratadine's manufacturing impurity. The HPLC method was also suitable for simultaneously assay and related substance determination of Loratadine pharmaceutical preparations. In the tritation method, glacial acetic acid was chosen as an amphiprotic solvent. Loratadine ionised in glacial acetic acid; the conjugated base of acetic acid (Ac·), was then titrated with standardised perchloric acid. The chemical content of the 'leaves and bark of Des mos dumosus has been carried out in this study. The leaves yielded nine isoquinoline type of alkaloids and two flavones. The isoquinoline alkaloids isolated were pronuciferine 37, stepharine 39, nomuciferine 40, (-) - 3 - hydroxynomuciferine 41, norlirioferine 42, asimilobine 43, liriodenine 44, lysicamine 45 and O - methylmoschatoline 46. The flavones were 5 - hydroxy - 6,7 - dimethoxyflavone 47 and 5 - hydroxy - 7,8 - dimethoxyflavone 48. The same flavones, 47 and 48 and the alkaloids 45 and 46 were also present in the bark of this …

Enalapril and its metabolite Enalaprilat were determined by gas chromatography - mass spectrometry (GC - MS) and high - performance liquid chromatography (HPLC). For HPLC, the detection limit of enalapril and enalaprilat was 10n g/ml in methanol, whereas for the GC - MS, the detection limit was 6.25ng/ml in plasma. These methods were compared and GC - MS was selected for routine analysis. Enalapril and Enalaprilat in plasma were extracted and cleaned up by using Sep - Pak C 18. Captopril is a highly specific competitive inhibitor of the angiotensin-convening enzyme (ACE). This enzyme converts angiotensin I, a relatively inactive decapeptide, to angiotensin II , a potent endogenous vasoconstrictor substance. Captopril is widely used for the treatment of hypertension and congestive heart failure. This drug, which contains a sulphydryl group binds readily to albumin and to other plasma proteins. Captopril is unstable in blood and plasma ex vivo because of the reactivity of sulphydryl group which results in oxidation and rapid formation of disulphides. Therefore a fixative or a stabiliser must be added to each blood sample immediately following collection. …

Levuglandin (LG) E2, a cytotoxic seco prostanoic acid co-generated with prostaglandins by nonenzymatic rearrangements of the cyclooxygenase-derived endoperoxide, prostaglandin H2, avidly binds to proteins. That LGE2-protein adducts can also be generated nonenzymatically is demonstrated by their production during free radical-induced oxidation of low density lipoprotein (LDL). Like oxidized LDL, LGE2-LDL, but not native LDL, undergoes receptor-mediated uptake and impaired processing by macrophage cells. Since radical-induced lipid oxidation produces isomers of prostaglandins, isoprostanes (isoPs), via endoperoxide intermediates, we postulated previously that a similar family of LG isomers, isoLGs, is cogenerated with isoPs. Acid digested brain containing 4 mg added 27Al was ashed at 1000°C to prepare an Al2O3 target for accelerator mass spectrometry (AMS) analysis of 26Al. A glass-like material usually resulted which was thought to be aluminum (Al) oxyphosphate. The separation of Al and phosphate was investigated. Al, but not phosphate, was bound by a cation exchange resin (AG 50-X8). Hydrofluoric acid eluted the Al from the resin. Removal of phosphate from acid digested brain by this method produced an amorphous material after ashing that was easier to recover from the porcelain crucible and had a …

When the immune system is compromised, opportunistic fungi can flourish and become fatal. Current antifungal treatments are limited and often toxic(2,3). In addition, strains of fungi resistant to available antifungals are emerging(4,5). Flash pyrolysis-gas chromatography/mass spectrometry was used to assess the chemical composition of the head louse's nit sheath. The pyrolyzate of the female insect's secretions, which form a cement-like cylinder holding the egg onto the hair, is dominated by amino acid derivatives and fatty acids. No chitin-specific compounds were detected in the sheath. These results, contrary to previous reports, show that the polymeric complex of the sheath is composed of proteinaceous moieties, possibly cross-linked to aliphatic components. This study constitutes the first chemical characterization of the pyrolysis products of insect (louse) glue and unequivocally confirms that louse sheaths are not chitinous, …

Gypsum is produced as a by-product during the citric acid production process at ADM Ringaskiddy. The particle size and filterability of the gypsum has a critical bearing on the overall citric acid production rate. Prior to August 1996, research in the area of gypsum crystal optimisation at ADM had concentrated on the application of vortex mixing to perform the gypsum formation reaction. While the gypsum crystals produced with vortex mixing were found to be desirably clear of microcrystalline gypsum, the crystals were considerably smaller and more difficult to filter and wash. Objective: To determine aneuploidy frequencies in pellet and swim-up semen fractions from 10 infertile men with severe oligoasthenoteratozoospermia (OAT) who were donating sperm for intracytoplasmic sperm injection and to determine whether the swim-up isolation method would successfully separate aneuploid from haploid sperm.
Design: Prospective study.
Setting: Infertility clinic and molecular genetics laboratory.
Patient(s): Ten patients with severe OAT.
Intervention(s): Cytogenetic analyses by fluorescence in situ hybridization to determine aneuploidy frequencies for chromosomes 1, 13, 18, 21, X, and Y in sperm from swim-up and pellet fractions.

Affinity chromatography is a type of liquid chromatography that makes use of biological-like interactions for the separation and specific analysis of sample components. This review describes the basic principles of affinity chromatography and examines its use in the testing of clinical samples, with an emphasis on HPLCbased methods. Some traditional applications of this approach include the use of boronate, lectin, protein A or protein G, and immunoaffinity supports for the direct quantification of solutes. Active transport via Na+, K+-ATPase plays an important role in establishing and maintaining proper ionic gradient across all mammalian cell membranes. A strict regulation of Na+, and K+ ion levels is especially important in excitable cells such as neurons and myocytes. Inhibition of the enzyme is associated with several pathological conditions and has serious detrimental effects on cerebral cortical cells. The N-terminal amino acid residues on polypeptides and proteins were determined in wastewaters prior to chlorination. The terminal amino groups'ere first derivatized with the well known derivatizing agent dansyl chloride, and then the resulting dansyl amino acid hydrolyzed from the peptide chain by a propionic acid/ hydrochloric acid mixture. The resulting dansyl amino acids were then separated and detected using reversed phase high performance liquid chromatography with fluorescence detection. The majority of N-terminal residues detected were the more polar amino acids. Concentrations ranged from 1 x 10-10 to 3 x 10-7 moles/liter. It is suggested that the shorter …

A novel PCR technique used to amplify Euplotes crassus macronuclear chromosomes was developed to provide scientific proof that an Euplotes crassus 1.0 kb gene is not genetically related to an Oxytricha nova 1.8 kb β-telomere protein gene. Theories and experimental procedures associated with the development of this PCR technique were the product of research that will have a profound impact on future studies of telomere protein genes and telomeric DNA in a multitude of eukaryotic cells. The hypothesis that a β-telomere protein does not exist in Euplotes crassus was supported by the result of this study.

Currently, amylase determinations are nonspecific for the organ source and are based entirely on the enzymatic properties of amylase to produce a measurable product or byproduct. The determination of pancreatic amylase is important in the diagnosis of acute pancreatitis. Most commercially available tests for amylase employ the measurement of the change in NADH absorbance at 280 nm or of the p-nitrophenol released from a maltotetrose substrate. These are nonspecific measurements of pancreatic amylase and often necessitate other tests to be run such as a serum lipase.

Sodium dodecyl sulfide (SDS) non-acrylamide gel-filled capillary columns are a new technology being used for analysis and separation of biotechnology-derived proteins. This research was to compare this new technology to the current methods of SDS polyacrylamide gel electrophoresis (SDS-PAGE) and high-performance size-exclusion chromatography (HPSEC). The molecular mass of four different recombinant proteins were determined by two commercialized SDS non-acrylamide gel-filled capillary columns, SDS-PAGE, and HPSEC. The data obtained showed that the SDS non-acrylamide gel-filled capillary columns were compatible with the SDS-PAGE technique for molecular mass determination. Grape seed extract is being marketed in France for cardiovascular use and skin care. The active constituents of both these drugs are certain polyphenols called procyanidins. A similar seed extract has now been obtained from Virginia Chardonnay grape seeds. Ten of the major components in this extract have been isolated in their pure form by the semi-preparative HPLC separation on a YMC ODS-AQ (Octadecylsilane-AQ) column. The ODS-Aq column has been shown to have a better resolving capacity compared to the Spherisorb ODS (Octadecylsilane) column previously described in the literature. The macrolide rapamycin (RAP) is a potent inhibitor of interleukin-2 (IL- 2)-induced T-cell proliferation. Current models suggest that RAP, when complexed to its intracellular receptor, FK506-binding protein, interferes with an IL-2 receptor-coupled signaling pathway required for cell-cycle progression from G1- to S-phase. Here we show that RAP treatment inhibits the growth of an IL-2-dependent cytotoxic T-cell line, CTLL-2, in late G1- phase, just prior to entry of the cells into S-phase. In contrast, RAP- treated CTLL-2 cells retained the ability to respond to IL-2 with enhanced cytolytic activity, indicating that RAP was not a general suppressant of cellular responsiveness to …

Four experiments were performed to determine the ability of marine higher fungi to grow in and degrade crude oil. Eight species, previously determined to be hydrocarbonoclastic, were grown in artificial sea water medium (ASM) and southern Louisiana crude oil. The dry mycelial weights were measured and consumption of crude oil was measured gravimetrically. The two species with the best growth, Corollospora maritima and Zalerion maritimum, were grown in ASM containing crude oil spiked with three known hydrocarbons. To this was added glucose (0.15%) and incubated 49 days to test for possible cometabolism of hydrocarbons. Hydrocarbons remaining in the growth …
