A confirmatory method has been developed to allow for the analysis of eleven nitroimidazoles and also chloramphenicol in milk and honey samples. These compounds are classified as A6 compounds in annex IV of council regulation 2377/90 and therefore prohibited for the use in animal husbandry. Milk samples are extracted by acetonitrile with addition of NaCl, while honey samples are first dissolved in water before a similar extraction. Honey extracts undergo a hexane wash to remove impurities. Both milk and honey extracts are evaporated to dryness and reconstituted in initial mobile phase. Various anticancer and antibacterial agents have been synthesized via azide chemistry by taking advantage of carbohydrate. Starting from the synthesis of 14 glycosyl azides, a library of carbohydrate-oxazolidinone conjugates and a library of carbohydrate-cyclopamine conjugates with biological interests were synthesized based on a highly efficient "click reaction" assisted by sonication. Some of the conjugates have improved solubility and enhanced anticancer activity.

A library of neomycin B derivatives with various modifications at the 5" position has been synthesized. Two leads exhibit prominent activity against both methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE). Chronic childhood lead exposure, yielding blood lead levels consistently below 10 μg/dL, remains a major public health concern. Low neurotoxic effect thresholds have not yet been established. Progress requires accurate, efficient, and cost-effective methods for testing large numbers of children. The LeadCare® System (LCS) may provide one ready option. The comparability of this system to the “gold standard” method of inductively coupled plasma mass spectrometry (ICP-MS) for the purpose of detecting blood lead levels below 10 μg/dL has not yet been examined. Paired blood samples from 177 children ages 5.2–12.8 years were tested with LCS and ICP-MS. Triplicate repeat tests …

Biodegradable polymer such as Polyethylene glycol (PEG) is frequently used as encapsulation material. This study has tried to produce microencapsulation product of Imipenem antibiotics in Polyethylene Glycol.Commercially available product called Tienam® was used as a major source of Imipenem. A slightly branched urethane acrylate macromer (UAM) was synthesized by reacting palm oil-based polyol with excess amounts of polymeric methylene diphenyl diisocyanate (MDI) to form urethane prepolymer, then reacted with 2-hydroxyethyl methacrylate (HEMA) and FT-IR spectroscopy confirmed the urethane and grafted acrylate groups. 0.2% by weight of camphoroquinone (CQ) and 0.8% by weight of ethyl-4-N,N-dimethylamino-benzoate (4EDMAB) by weight were added to render the experimental resins light-curable. Their viscosity, percentage of degree of conversion (%DC) and cross-linking density (CLD), percentage of volumetric polymerization shrinkage (%VPS), water sorption and solubility, flexural strength, modulus of elasticity and toughness were determined and compared to …

Aglaia is a small genus of trees that belong to the family Meliaceae. They are widely distributed in India, Malay Peninsula and throughout Indomalesia. The chemistry of Aglaia has been extensively investigated and most of the researches done on these plants focused on their effect as insecticides, antifeedant, growth inhibitors, cytotoxicity and others. Tryptamine (1H-indole-3-ethyleneamine) Schiff bases were prepared by the condensation reaction of ethanolic solution of tryptamine with: salicylaldehyde (TS), 5 chlorosalicylaldehyde (TCS), 5-nitrosalicylaldehyde (TNS), 3,5-di-tert-butyl-2-hydroxy benzaldehyde (TTET) and the ketones: 2-hydroxyacetophenone (THAP), 5-methyl-2- hydroxyacetophenone (TMeHAP), 5-methoxy-2 hydroxyacetophenone (TOMeHAP), 5- chloro-2-hydroxyacetophenone (TClHAP) and 5-bromo-2 hydroxyacetophenone (TBrHAP). The ligands were coordinated to Zn(II), Cu(II) and Ni(II) by the reaction of metal acetates with the corresponding ligand in a 1 : 2 metal to ligand ratio. The extraction of alkaloids from the Malaysian Cryptocarya densiflora has been carried out in this study. The alkaloids were extracted from the bark and leaves of this species using acid base extraction and the crude alkaloids obtained were subjected to extensive chromatographic techniques such as thin layer and column chromatography. Four thiosemicarbazones ligands (1-4) have been prepared with good yield by refluxing 2,4-dihydroxybenzaldehyde with thiosemicarbazide or N(4)-substituted thiosemicarbazide in ethanol or methanol. Aryl hydrocarbon receptor (AHR) is a sensor protein, activated by aromatic chemical species for transcriptionally regulating xenobiotic metabolizing enzymes. AHR is also known to be involved in a variety of pathogenesis such as cancer, diabetes mellitus, cirrhosis, asthma, etc. The AHR signaling induced by xenobiotics has been intensively studied whereas its physiological role in the absence of xenobiotics is poorly understood. The integrity of an organism's genome depends on the fidelity of DNA replication and the efficiency of DNA repair. The DNA mismatch repair (MMR) system, which is highly conserved from prokaryotes to eukaryotes, plays an important role in maintaining genome stability by correcting base-base mismatches and insertion/deletion (ID) mispairs generated during DNA replication and other DNA transactions. Mismatch recognition is a critical step in MMR. Two mismatch recognition proteins, MutSα (MSH2-MSH6 heterodimer) and MutSβ (MSH2-MSH3 heterodimer), have been identified in eukaryotic cells. The focus of this dissertation is centered on the mass spectral analysis of lipids and changes occurring in keeping with the concept of homeoviscous adaptation [1]. Homeoviscous adaptation is the process of modification of membrane lipids in response to environmental stimuli [1]. Dissertation investigations applied this concept to prokaryotic and eukaryotic organisms, and expanded the perception of environmental factors from exogenous organic solvents to intracellular environment.

During atherogenesis and vascular inflammation quiescent platelets are activated to increase the surface expression and ligand affinity of the integrin αIIbβ3 via inside-out signaling. Diverse signals such as thrombin, ADP and epinephrine transduce signals through their respective GPCRs to activate protein kinases that ultimately lead to the phosphorylation of the cytoplasmic tail of the integrin αIIbβ3 and augment its function. The signaling pathways that transmit signals from the GPCR to the cytosolic domain of the integrin are not well defined. Many people are diagnosed with thyroid related disorders, and many more are unaware of their existing thyroid problems. T1AM, a naturally occurring metabolite of the thyroid hormone (TH), has been shown to activate the Trace Amine Associated Receptor 1 (TAAR1) and exhibits effects that oppose those of the TH. It is, therefore, likely that there is regulatory relationship between T1AM and the TH. These data suggest that iron(II) reactivity for a set of homologous spiroadamantyl 1,2,4-trioxolane, 1,2,4-trioxane, and 1,2,4-trioxepane peroxide heterocycles is a necessary, but insufficient, property of animalarial peroxides. Heme alkylation efficiency appears to give a more accurate prediction of antimalarial activity than FeSO4-mediated reaction rates, suggesting that antimalarial activity is not merely dependent on peroxide bond cleavage, but also on the ability of reactive intermediates to alkylate heme or other proximal targets.

A major limitation of successful radiation therapy in cancer treatment is the increase in normal tissue damage as higher doses are used to achieve greater tumor destruction. Radiation dose optimization in cancer therapy requires achieving maximum tumor destruction with minimal damage to normal tissue Antioxidants have been shown to protect normal tissues against radiation damage, as radiation-induced tissue damage results predominantly from reactive oxygen species that directly damage cellular components. However, for effective use as normal tissue radioprotectants in radiotherapy, these antioxidants must not protect the tumors. Cisplatin is a common therapeutic agent used in cancer treatment. Unfortunately, resistance to cisplatin in addition to severe side effects limits its use in cancer treatment. Two novel cisplatin analogues, 4DB and 4TB were shown to have varying cytotoxicity in lung, breast and prostate cancer cells. The hypothesis for this study states that the differences in 4DB and 4TB cytotoxicity among different tissue types is due to the type and efficiency of DNA repair mechanisms involved in response to these drugs.

Singlet Oxygen Delivery Through The Porous Cap Of A Hollow-Core Fiber Optic Device, Matibur Zamadar, David Aebisher, Alexander Greer

The development of the first photosensitizer/fiber optic device is reported. An oxygen-flowing, fiber-capped configuration is used for the application of heterogeneous, spatially confined singlet oxygen delivery in aqueous media. This is a unique device, unlike other heterogeneous photosensitizers, in which local concentrations of singlet oxygen can be delivered via introduction and withdrawal of the fiber tip.

The placenta forms in eutherian mammals, and is responsible for the nutrition of the developing fetus. However, maternal exposure to environmental pollutants both before and during pregnancy may result in the passage of toxins through the placental barrier and into fetal tissues. The placenta is the only organ derived from both maternal and fetal tissues, and establishes a link between the fetus and the environmental exposures of the mother. The analysis of placentae for the presence of environmental pollutants offers the possibility of exposure measurements in both the mother and the developing fetus. The synthesis and characterization of some substituted benzyl N-nitrosocarbamates with an N-2-(methylthio)ethyl or a bis(2-aminoethyl)sulfide functionality is reported, as a part of a long-term goal to design and prepare novel photolabile structures that could be used as substances for controlled release of alkylating and/or crosslinking agents. The synthesis was accomplished by reaction of benzyl chloroformates with the corresponding amines, resulting in the preparation of carbamates. The latter were subsequently nitrosated, utilizing two different N-nitrosation methods, to yield the target structures.

A specific and sensitive liquid chromatography (LC)-tandem mass spectrometric method for quantitative determination of methylprednisolone (MP) in rat plasma and liver was developed and validated using triamcinolone acetonide as an internal standard. Liquid-liquid extraction using tert-butyl methyl ether was used to extract the drug and the internal standard from plasma and liver. The separation of MP was performed on a C(18) column with a mobile phase of acetonitrile:0.5% formic acid aqueous solution (85:15, v/v) over 4 min.
