In developing an immunometric assay of intact parathyrin (parathyroid hormone, PTH), we found that affinity chromatography is a useful tool in purifying and optimizing the labeling conditions for acridinium ester-labeled antibodies. ... In summary, affinity chromatography was found to be useful in the purification of acridinium ester-labeled antibodies, particularly for removing denatured antibodies from the prepared label and for monitoring the amount of active labeled antibodies produced.

A simple, reliable method is described for the routine measurement of non-steroidal anti-inflammatory drugs (NSAIDs) as inhibitors of prostaglandin synthase. Appropriate concentrations of inhibitor are incubated with ram seminal vesical prostaglandin synthase, sodium arachidonate and reduced glutathione. Prostaglandin E2 (PGE2) production is then quantitated by and enzyme linked immunosorbant assay (ELISA), in which anti-PGE2 antibody is utilized in a binding competition between test sample and adsorbed conjugate of PGE2-BSA. Telomere Structure In Euplotes Crassus: Characterization Of Dna-Protein Interactions And Isolation Of A Telomere-Binding Protein, Carolyn M. Price

The nucleoprotein structure of telomeres from Euplotes crassus was studied by using nuclease and chemical footprinting. The macronuclear telomeres were found to exist as DNA-protein complexes that are resistant to micrococcal nuclease digestion. Each complex encompassed 85 to 130 base pairs of macronuclear DNA and appeared to consist of two structural domains that are characterized by dissimilar DNA-protein interactions. Dimethyl sulfate footprinting demonstrated that very sequence-specific and salt-stable interactions occur in the most terminal region of each complex. DNase I footprinting indicated that DNA in the region 30 to 120 base-pairs from the 5' end lies on a protein surface; …

Lithium has been known to be an effective medication for people with bipolar disorder. The mechanisms of action of lithium in the brain is not very well understood. NMR spectroscopy and imaging are effective both in determining lithium levels in tissue and brain. We have monitored lithium levels in red blood cells. We have been able to separate intra- and extracellular compartments of lithium using shift reagents, thereby obtaining T^1 's of both the compartments. Lithium uptake as a function of hematocrit was monitored weekly over a 3 week period. The time constant of 50 mM lithium uptake at 25°C …

Drugs such as adrenoceptor antagonists and certain centrally acting agents are known to lower blood pressure and thus serve as clinically useful antihypertensive agents. Norepinephrine 4, a natural ∝-adrenoceptor agonist, is the major agent associated with increased blood pressure in the periphery. Based on a careful structure-activity comparison, a series of substituted oxazolidines, 20, 21, 22 and 23, have been designed as possible antihypertensive candidates. Specifically, 22 and 23 have been synthesized to serve as potential norepinephrine antagonists.
Preliminary evaluation of these compounds indicated that they were not antihypertensive in nature. The more active compound, 23, …

Commercially pure titanium rods were machined into hollow cylinders and gas-phase permeation and diffusion measurements obtained with a quadrupole residual gas analyzer at temperatures between 400-800°C and pressures between 0.533 and 266.6 Nrn-2. Pressure dependence measurements established that phase boundary reactions as well as lattice diffusion control the transport process. Lag-time data yield excellent agreement with the diffusivity data of Wasilewski and Kehl and Papazoglou and Hepworth for the case where phase boundary reactions are taken into account. The best value of the diffusivity is D = 2.1 x 10-2 exp (-50,40O/RT). Analysis of lag-time data reveals …

This paper describes a screening process used to determine the overall effect of anti-cancer drugs on a tumor more rapidly than other established methods. This technique measures the rate of oxygen consumption by cells when a solution of acrylamide and bisacrylamide is polymerized.

In recent years, interest in an enzyme which catalyzes the oxidation of a variety of aldehydes and purines has been growing steadily. In fact, this enzyme commonly known as xanthine oxidase, has been implicated by some (1) in the regulation of nucleic acid synthesis and cellular growth - an important consideration in the study of caner. Unfortunately, detailed investigations directly pertaining to biological aspects of this enzyme have as yet been somewhat limited. Nevertheless, the amount of information concerning the nature of the enzyme and its catalytic effects upon different classes of substrates, through far from complete, is indeed significant.

This thesis, submitted to Xavier University in partial fulfillment of requirements for a Maters degree in chemistry in 1952, reports the synthesis and characterization of itaconic acid derivatives of sulfanilamide and amino phenyl sulfone compounds. The research describes reaction procedures and the isolation and purification of a series of novel chemical compounds, with experimental data including melting points, percent yields, elemental analysis, and molecular weights reported for numerous derivatives. The thesis presents conclusions regarding the chemical properties and reactivity of the synthesized compounds.
