Soil, that complex mixture of minerals, organic particles, chemicals, and anthropogenic materials, is ubiquitous and easily transferred, which gives it forensic relevance. Forensic soil analysis has traditionally included the identification and comparison of both organic and inorganic components, the inorganic portion being most commonly examined by light microscopy and other instrumental techniques. Predominantly, the inorganic materials in soil are naturally-occurring minerals, contributed as grains from the surrounding bedrock, which gives a soil its regional character. Recent genomic studies of microbiomes have revealed an overwhelming number of biosynthetic genes of unknown function. Most of these “cryptic” biosynthetic genes are not expressed in laboratory monocultures of individual microbes. Thus, there remains tremendous untapped potential for natural products discovery. Here we employ mixed microbial culture (MMC) as a simple yet powerful approach to awaken cryptic biosynthetic gene clusters. Our preliminary studies demonstrated that arrays of metabolites could be induced in MMCs upon environmental cues, such as surface adhesion. Immobilized-enzyme reactors (IMERs) are flow-through devices containing enzymes that are physically confined or localized with retention of their catalytic activities. IMERs can be used repeatedly and continuously and have been applied for (bio)polymer degradation, proteomics, biomarker discovery, inhibitor screening, and detection. Online integration of IMERs with analytical instrumentation, such as high-performance liquid chromatography (HPLC) systems, reduces the time needed for multi-step workflows, reduces the need for sample handling, and enables automation. Enzymes and other related proteins are promising targets as biomarkers for cancer diagnostics, and it is therefore important to be able to quantify intracellular enzyme activity levels, not just presence of enzyme. Thus, there is a great need for the development of methods that offer spatiotemporal quantification of intracellular enzyme activity levels in respiring 2- and 3-dimensional cultures or their sample-processed variants. The compounds and chemical compositions of synthetic cannabinoids and designer cathinones are designed to mimic the intoxicating effects of D9-tetrahydrocannabinol and amphetamines, respectively. In order to skirt existing drug laws, non-controlled ingredients are used, and the original chemical structures of current drugs are being modified using analogs or derivatives. Applications of SLE (supported liquid extraction) is a relatively new extraction technique that is not widely used in forensic toxicology. SLE is also easier and less time consuming than traditional LLE (liquid-liquid extraction) and SPE (Solid phase extraction). This work will compare the extraction efficiencies between SLE and SPE for methamphetamine from urine with detection and quantitation using GC-MS (gas chromatography- mass spectrometry). The SLE cartridges provide better analyte recoveries, eliminate emulsion formation, and allow for shorter processing time than other extraction techniques. In this dissertation, I conducted various untargeted and targeted metabolomics and lipidomics studies using mass spectrometry based analytical platforms. Firstly, I investigated the androgen depletion effects on fecal metabolites in a mouse model of systemic lupus erythematosus and metabolite biomarkers for colorectal cancer using two-dimensional gas and liquid chromatography-mass spectrometry. Carbon fiber ultramicroelectrodes (CF-UMEs) are commonly used as electrochemical probes and sensors due to their small size, fast response, and high signal-to-noise ratio. Surface modification strategies are often employed on CF-UMEs to improve their selectivity and sensitivity for desired applications. However, many modification methods are cumbersome and require expensive equipment. In this study, a simple approach known as soft nitriding is used to prepare nitrogen-doped CF-UMEs (N-CF-UMEs). Ion selective electrodes (ISEs) are analytical sensors that monitor the interactions between an ionophore within a polymeric membrane and ions in various solutions. The sensitivity and selectivity of ISEs is directly related to the chemical components. One type of ionopohore utilized in ISEs are metalloporphyrins. Metalloporphyrins have unique binding characteristics, which make them useful in sensors. Biomaterials without exogenous cells or therapeutic agents often fail to achieve rapid endogenous bone regeneration with high quality. Here, we reported a class of three-dimensional (3D) nanofiber scaffolds with hierarchical structure and controlled alignment for effective endogenous cranial bone regeneration. 3D scaffolds consisting of radially aligned nanofibers guided and promoted the migration of bone marrow stem cells from the surrounding region to the center in vitro. These scaffolds showed the highest new bone volume, surface coverage, and mineral density among the tested groups in vivo. The United States Pharmacopeia sets the standards for the manufacturing, storage, and analysis of medicinal formulations. One analysis, dissolution testing evaluates the rate at which the medicinal formulation forms a solution to predict in vivo drug release. Dissolution testing on ibuprofen tablets alone and in the presence of ascorbic acid or caffeine was performed to mimic the administration using orange juice or caffeinated soft drinks to assess their impact on the dissolution rate of ibuprofen. The Fukushima Daiichi Nuclear Powerplant was struck by an earthquake and a tsunami resulting in the meltdown of four of the six reactor cores operating at the plant. As a result, nuclear waste was released from the plant, contaminating the soil in the region. Most of the contamination was sequestered within the few inches of the soil but unluckily the contamination, radiocesium formed a non-exchangeable bond. The Japanese government bagged this soil and has stored it in fields surrounding the exclusion zone. Long-term storage facilities have not been determined. We investigate the optical response of graphene nanoribbons (GNRs) using the broadband nonlinear generation and detection capabilities of nanoscale junctions created at the LaAlO3/SrTiO3 interface. GNR nanoclusters measured to be as small as 1–2 GNRs in size are deposited on the LaAlO3 surface with an atomic force microscope tip. Time-resolved nonlinear optical probes of GNR nanoclusters reveal a strong, gate-tunable second and third harmonic response, as well as strong extinction of visible to near-infrared light at distinct wavelengths, similar to previous reports with graphene.

A microfluidic paper-based analytical device (µPAD) is developed in this work to analyze hydroquinone in facial whitening creams using phloroglucinol. The µPAD features a hydrophobic barrier for detection and was fabricated using a wax printer with Whatman chromatographic paper. Detection was achieved by colorimetry based on the formation of an orange hydroquinone–phloroglucinol complex. The colored reaction product formed on the detection zone of the µPAD was scanned, and the images obtained were processed with Image-J software to determine their color intensity (RGB value). The solution resistance (Ru) between the working electrode (WE) and the reference electrode (RE) may lead to significant Ohmic drop (iRu) and deviation of actually applied potential at the WE to the desired ones in electrochemical measurement. Surfaces with microscale roughness can entail dualscale hierarchical structures such as the recently reported nano/ microstructured surfaces produced in the laboratory (Wang et al.
